Vielfältig lässt sich die Bedeutung von Vorstrafen zeigen; jenseits ihrer gesellschaftlichen Wirkungen beschäftigen sie Straf-, Arbeits- und Zivilgerichte. Dieser Beitrag untersucht, welche Implikationen die Registrierung einer Verurteilung hat. Dabei soll aus strafrechtlicher wie auch aus grundrechtlicher Perspektive belegt werden, dass der Staat als Autor der Verurteilung alle ihre Folgen bedenken und bestimmten Konsequenzen gegebenenfalls entgegenwirken muss. Zur Einstimmung sollen fünf Fallskizzen Schlaglichter auf den Problemkreis werfen:
Es wurden die Lokalisation des (P)RR/ATP6ap2 in PC12 Tumorzellen neuronalen Ursprungs, kardialen und renalen Zellen und Geweben sowie die funktionellen Folgen nach Downregulation oder Überexpression (paradoxe Downregulation) des Rezeptors untersucht. Er weist sowohl eine ER-spezifische, Membran-assoziierte als auch eine Lokalisation in der löslichen Fraktion auf.
Als akzessorische Untereinheit der vakuolären H+ ATPase beeinflusst der Rezeptor ihre Integrität und Aktivität. Die Vielzahl differenter Daten zwischen den mit einem V ATPase-Inhibitor behandelten und den (P)RR/ATP6ap2-downregulierten PC12 Zellen verdeutlicht, dass die Funktion des (P)RR/ATP6ap2 insbesondere im Metabolismus über eine bloße Regulation der V ATPase-Aktivität hinausgeht und möglicherweise durch die Integration des (P)RR/ATP6ap2 im Wnt-Pathway bestimmt wird.
Environmental activism, defined as a range of difficult pro-environmental behaviors, is analyzed within the conceptual framework of Significance Quest Theory (SQT). In Study 1, 40 interviews were carried out on two groups of people in the European Union: Committed Actors for Nature (CANs, n = 25) versus Committed Actors for Society (CASs, n = 15). Results demonstrated that Significance Quest (SQ) motivates each group to be strongly committed to their chosen action and the main difference between them being in their ideology (pro-social vs. pro-environmental). In Study 2 (N = 131), the relationship between SQ and intention to enact difficult pro-environmental behaviors was assessed. Results suggested that the higher the SQ, the higher the tendency to enact difficult pro-environmental behaviors, but not average or easy ones. Moreover, the higher the pro-environmental ideology, the stronger the indirect effect of SQ on difficult behavior through willingness to sacrifice.
Introduction: Hearing and vision loss are highly prevalent in elderly adults, and thus frequently occur in conjunction with cognitive impairments. Studies have shown that hearing impairment is associated with a higher risk of dementia. However, evidence concerning the association between vision loss and dementia, as well as the co-occurrence of vision and hearing loss and dementia, has been inconclusive.
Objectives: To assess the association between: (i) either hearing or vision loss and the risk of dementia, as well as between; and (ii) the combination of both sensory impairments and the risk of dementia.
Methods: This case-control study was based on a 5-year data set that included patients aged 65 years and older who had initially been diagnosed with dementia diseases by one of 1,203 general practitioners in Germany between January 2013 and December 2017. In total, 61,354 identified dementia cases were matched to non-dementia controls, resulting in a sample size of 122,708 individuals. Hearing loss and vision loss were identified using the ICD-10 diagnoses documented in the general practitioners’ files prior to the initial dementia diagnosis. Multivariate logistic regression models were fitted to evaluate the associations between visual and/or hearing impairment and the risk of dementia and controlled for sociodemographic and clinical variables.
Results: Hearing impairment was documented in 11.2% of patients with a dementia diagnosis and 9.5% of patients without such a diagnosis. Some form of vision impairment was documented in 28.4% of patients diagnosed with dementia and 28.8% of controls. Visual impairment was not significantly associated with dementia (OR = 0.97, CI = 95% 0.97–1.02, p = 0.219). However, patients with hearing impairment were at a significantly higher risk of developing dementia (OR = 1.26, CI = 95% 1.15–1.38, p < 0.001), a finding that very likely led to the observed significant association of the combination of both visual and hearing impairments and the risk of dementia (OR = 1.14, CI = 95% 1.04–1.24, p = 0.005).
Discussion: This analysis adds important evidence that contributes to the limited body of knowledge about the association between hearing and/or vision loss and dementia. It further demonstrates that, of the two, only hearing impairment affects patients’ cognition and thus contributes to dementia risk.
Das gram-negative Stäbchenbakterium Burkholderia pseudomallei, Erreger der Melioidose, ist ein fakultativ intrazelluläres Pathogen. Ein detailliertes Wissen über das Proteom dieses Pathogens liefert eine wichtige Grundlage für das Verständnis der Pathomechanismen. Im Rahmen dieser Arbeit wurden detailierte „reference maps“ des intra- und extrazellulären Proteoms von B. pseudomallei Stamm E8 erstellt. Es wurden hierbei insgesamt 353 intrazelluläre und 154 extrazelluläre Proteine identifiziert und anschließend einer funktionellen Klassifizierung unterzogen. Die „reference maps“ stellen ein wichtiges Werkzeug für weitere vergleichende Proteomanalysen dar. In weiterführenden Experimenten wurde das Proteom dreier Wildtypstämme verglichen. Hierbei zeigten sich insgesamt nur geringe Unterschiede zwischen den drei analysierten Stämmen E8, E212 und K96423. Die Proteomanalysen bestätigten die Ergebnisse der Genotypisierung – beide Ansätze ergaben eine engere Verwandtschaft der Stämme E212 und K96423 als mit Stamm E8.
Im Infektionsverlauf von B. pseudomallei ist das sonst aerob lebende Bakterium in der Lage sich auch unter anaroben Bedingungen auszubreiten. Über die anaerobe Physiologie von B. pseudomallei ist bisher wenig bekannt. Es konnte ein Screeningsystem entwickelt werden, mit dem aus insgesamt 2344 Transposonmutanten 16 Mutanten mit reduziertem Wachtum unter anaeroben Bedingungen identifiziert wurden. Die vier Mutanten mit dem am stärksten verringerten Wachstum wurden für weitere Analysen im Rahmen dieser Arbeit ausgewählt. Drei der Mutanten hatten Molybdän bezogene Gendefekte und bei einer Mutante betraf der Gendefekt eine putative Sensor-Kinase. Die untersuchten Mutanten konnten erfolgreich komplementiert werden und somit polare Effekte ausgeschlossen werden. Die Komplentanten wiesen wieder dem Wildtyp entsprechendes anaerobes Wachstum auf und wiesen auch bei allen weiteren Experimenten, wie z.B. Infektionsversuchen, die Eigenschaften des Wildtyps auf. Dass unter den 2344 gescreenten Transposon Mutanten drei der vier auffälligsten Mutanten Molybdän bezogene Gendefekte aufwiesen, unterstreicht die Wichtigkeit von Molybdän und im Zusammenhang damit der anaeroben Nitratreduktase. Die Attenuierung der drei Mutanten im C57BL/6-Maus-Infektionsmodell deutet auf die Wichtigkeit des anaeroben Stoffwechsels bei chronischen Infektionsverläufen von B. pseudomallei hin. Bei der weiteren näher untersuchten Mutante liegt ein Defekt einer putativen Sensorkinase vor. Diese gehört zu einem Zwei-KomponentenSignaltransduktionssystem, welches Homologien zum RegB-RegA-System aufweist. Es konnte gezeigt werden, dass die hier untersuchte Sensorkinase einen entscheidenen Einfluss auf die Virulenz von B. pseudomallei hat. Der exakte Mechanismus der Attenuierung durch die Mutation von BPSL0201 bleibt vorerst jedoch noch ungeklärt.
Zusammenfassend lässt sich sagen, dass mit dem hier entwickelten Screeningsystem erfolgreich Transposonmutanten identifiziert werden konnten, welche ein reduziertes Wachstum unter anaeroben Bedingungen aufweisen. Durch die anschließende Charakterisierung konnten tiefere Einblicke in den anaeroben Lebensstil von B. pseudomallei gewonnen werden. Die Ergebnisse deuten auf einen Zusammenhang zwischen der Fähigkeit des anaeroben Wachstums von
B. pseudomallei und der vollen Virulenz hin. Die gewonnenen Erkenntnisse stimulieren weitere Untersuchungen im Hinblick auf den Einfluss des anaeroben Wachstums von B. pseudomallei bei der Pathogenese der Melioidose.

Research on self-stigma in substance use disorders (SUD) is still scarce, especially regarding the role of childhood trauma and subsequent posttraumatic disorders. Objectives: The present study investigated the progressive model of self-stigma in women with SUD and posttraumatic stress disorder (PTSD), and the predictive value of PTSD severity and childhood trauma experiences on self-stigma. Method: In a cross-sectional study with 343 women with SUD and PTSD, we used the Self-Stigma in Alcohol Dependency Scale, the Childhood Trauma Questionnaire (CTQ), the PTSD Symptom Scale Interview (PSS-I), and to control for SUD severity and depression, the Addiction Severity Index Lite and the Beck Depression Inventory-II. Hierarchical regression analyses were conducted for each stage of self-stigma (aware-agree-apply-harm). Results: The interrelated successive stages of self-stigma were largely confirmed. In the regression models, no significant effects of the PSS-I- and the CTQ-scores were observed at any stage of self-stigma. Agreeing with negative stereotypes was solely predicted by younger age, applying these stereotypes to oneself was higher in women with younger age, higher depression and SUD severity, and suffering from the application (harm) was only predicted by depression. Conclusions: The progressive model of self-stigma could be confirmed in women with SUD and PTSD, but PTSD severity and childhood trauma did not directly affect this process. Self-stigma appears to be related to depression in a stronger way than PTSD is related to women with SUD and PTSD.

The bacterium Staphylococcus aureus is a notorious pathogen that causes dangerous and difficult-to-treat infections. This applies especially to methicillin-resistant S. aureus, better known as MRSA. MRSA infections were originally associated with healthcare settings as a consequence of clinical antibiotic therapy. However, in recent years MRSA infections have become more common among healthy individuals in the community. The community-associated (CA-)MRSA lineages are generally more aggressive than hospital-associated (HA-) lineages. Therefore, it is alarming that such CA-MRSA lineages are now emerging in hospitals. This raises the fundamental question of how CA-MRSA adapts to this new niche. Further, since the originally distinguishing features of CA- and HA-MRSA are losing discriminative value, it is important from a healthcare perspective to identify novel distinctive markers for early recognition and elimination of hospital-adapted CA-MRSA. In the present PhD research, these challenges were tackled with a ‘multi-omics’ approach focused on the USA300 lineage of MRSA, originally identified as CA, but now also causing hospital outbreaks. The results show that hospital-adapted USA300 isolates produce an altered spectrum of virulence factors, changed their metabolism, and exploit human immune cells as a protective environment against antibiotics. Importantly, hospital-adapted CA-MRSA strains can be recognized through distinctive patterns of gene expression and secreted virulence factors. Altogether, these observations show that the epidemic behaviour of MRSA is a multi-factorial trait, and they provide new insights into the missing links between epidemiology and pathophysiology of S. aureus. Moreover, they highlight the benefits of multi-omics technologies for protecting patients and frail individuals against the aggressive CA-MRSA.
The aim of the present study was to construct a biological age score reflecting one’s physiologic capability and aging condition with respect to tooth loss over 10 y. From the follow-up to the population-based Study of Health in Pomerania (i.e., SHIP-2), 2,049 participants were studied for their baseline biomarker measures 10 y before (i.e., in SHIP-0). Metabolic and periodontal data were regressed onto chronological age to construct a score designated as “biological age.” For either sex separately, the impact of this individualized score was used to predict tooth loss in the follow-up cohort in comparison with each participant’s chronological age. Outcome data after 10 y with respect to tooth loss, periodontitis, obesity, and inflammation were shown to be better for biologically younger subjects than as expected by their chronological age, whereas for the older subjects, data were worse. Especially for tooth loss, a striking increase was observed in subjects whose biological age at baseline appeared to be higher than their chronological age. Biological age produced significantly better tooth loss predictions than chronological age (P < 0.001). Areas under receiver operating characteristic curves for tooth loss of ≥3 teeth in men during follow-up were 0.811 and 0.745 for biological and chronological age, respectively. For women, these figures were 0.788 and 0.724. For total tooth loss, areas under the curve were 0.890 and 0.749 in men and 0.872 and 0.752 in women. Biological age combines various measures into a single score and allows identifying individuals at increased risk of tooth loss.
Phenolics and its derivatives are aromatic compounds with a wide range of industrial applications. Gallic acid, protocatechuic acid, catechol or pyrogallol are only a few examples of industrially relevant aromatics. The production of bulk fine chemicals primarily for chemical and pharmaceutical industry has put a strong emphasis on optimizing manufacturing conditions. Commercial production of many chemicals is still based on organic chemical synthesis using petroleum derivatives as starting material. Since these processes are considered environmentally unfriendly and posing an irresponsible strain on limited fossil resources, much attention is paid to the development of new microbial factories for the bioproduction of industrially relevant chemicals using renewable sources or organic pollutants as starting material. Arxula adeninivoras is a non-conventional yeast possessing attractive properties for industrial application such as thermo- and osmotolerance. Another major advantage of this organism is its broad substrate spectrum with tannin at the forefront. The present project is dedicated to the study of the tannic acid degradation pathway in A. adeninivorans. Two genes encoding enzymes annotated as gallic acid decarboxylase (AGDC1) and catechol-1,2-dioxygenase (ACDO1) have been selected and investigated. Both enzymes were characterized and their function in tannin catabolism analyzed.
Über 40% der derzeit verwendeten Arzneimittel beinhalten Amine als Wirkstoff. Vor allem die Chiralität dieser Moleküle stellt eine immer größere Bedeutung dar. Chirale Moleküle unterscheiden sich in der räumlichen Anordnung der Atome um das chirale Zentrum. Nicht selten besitzen Naturstoffe ein solches chirales Zentrum und sind asymmetrisch aufgebaut. In diesem Zusammenhang ist es nicht verwunderlich, dass die in der Medizin eingesetzten Wirkstoffe einen unterschiedlichen Wirkungsgrad je nach chiraler Konfiguration aufweisen.
Ziel dieser Arbeit war es neue Methoden zur stereoselektiven Synthese chiraler Amine zu untersuchen. Im Gegensatz zu herkömmlichen chemischen Synthesen, die beispielsweise auf Übergangsmetalle als Katalysatoren setzen, stellen Enzyme als Katalysatoren eine interessente Alternative dar. Stereo-, Regio- und Chemoselektivität ist Enzymen oft von Natur aus gegeben. Im Mittelpunkt der enzymatischen asymmetrischen Synthese optisch aktiver Amine standen bisher Amintransaminasen (ATA), die eine Aminogruppe von einem Amin (Aminodonor) auf ein Keton (Aminoakzeptor) transferieren. Diese Enzyme sind jedoch auf die Synthese primärer Amine beschränkt, sekundäre und tertiäre Amine sind nicht zugänglich. Eine Alternative hierzu stellen Iminreduktasen (IREDs) dar. Dabei handelt es sich um NADPH-abhängige Enzyme, die eine Reduktion von Iminsubstraten zu optisch aktiven Aminen katalysieren. Vor allem die IRED-katalysierte reduktive Aminierung steigerte das Interesse dieser Enzymklasse. In einer reduktiven Aminierung wird nicht das Imin selbst als Substrat eingesetzt, sondern eine prochirales Keton. Dieses formt mit einem Aminsubstrat (Nukleophil) ein Imin und wird anschließend reduziert. Durch diesen Reaktionsweg sind IREDs nicht nur auf zyklische Substrate beschränkt, auch instabile azyklische Imine werden zugänglich.
Die reduktive Aminierung mittels Iminreduktase wurde erstmalig im Jahr 2014 beschrieben und war zu Beginn dieser Arbeit nur als "Proof of Concept" gezeigt worden. Im Rahmen dieser Promotionsarbeit konnte gezeigt werden, dass diese Enzyme die Möglichkeit bieten, optisch aktive Amine mit hohen Umsätzen und Enantiomeren- bzw. Diastereomerenüberschüssen zu synthetisieren.
Abstract
Metabolomics studies now approach large sample sizes and the health characterization of the study population often include complete blood count (CBC) results. Upon careful interpretation the CBC aids diagnosis and provides insight into the health status of the patient within a clinical setting. Uncovering metabolic signatures associated with parameters of the CBC in apparently healthy individuals may facilitate interpretation of metabolomics studies in general and related to diseases. For this purpose 879 subjects from the population‐based Study of Health in Pomerania (SHIP)‐TREND were included. Using metabolomics data resulting from mass‐spectrometry based measurements in plasma samples associations of specific CBC parameters with metabolites were determined by linear regression models. In total, 118 metabolites significantly associated with at least one of the CBC parameters. Strongest associations were observed with metabolites of heme degradation and energy production/consumption. Inverse association seen with mean corpuscular volume and mean corpuscular haemoglobin comprised metabolites potentially related to kidney function. The presently identified metabolic signatures are likely derived from the general function and formation/elimination of blood cells. The wealth of associated metabolites strongly argues to consider CBC in the interpretation of metabolomics studies, in particular if mutual effects on those parameters by the disease of interest are known.
Als hochmaligner Tumor stellt das Neuroblastom ausgehend vom sympathischen Nervensystem den häufigsten soliden, extrakraniellen Tumor des Kindesalters dar. Aufgrund seiner heterogenen Charakteristik und teils höchst aggressiven Wachstums bedarf es besonders im Hinblick auf die hohe Rezidivrate von Hochrisikopatienten neuer innovativer Therapien, die einen langanhaltenden Anti-Tumoreffekt induzieren. Durch zunehmende Erkenntnisse auf dem Gebiet der Tumorimmunologie sind dabei besonders immuntherapeutische Behandlungsstrategien immer weiter in den Vordergrund gerückt. Diese führen zur spezifischen Aktivierung des körpereigenen Immunsystems verbunden mit der Erkennung und Eliminierung der Tumorzellen. In der Praxis ist eine wirksame Anwendung der aktiven Immuntherapie jedoch mit einigen Herausforderungen verbunden. Dazu gehören die Identifikation von geeigneten Tumorantigenen, die optimale Aufnahme, Prozessierung und Präsentation der Antigene durch Immunzellen sowie die Überwindung der Immuntoleranz zur Induktion zytotoxischer und Gedächtnis-T-Zellen. Auch die natürlichen Regulationsmechanismen sollten nicht außer Acht gelassen werden, da diese einerseits die anti-tumoralen Effekte einer resultierenden Immunantwort eindämmen können, jedoch andererseits gesundes Gewebe vor einer Autoimmunreaktion schützen. Ziel dieser Arbeit war es daher die bereits bekannte präklinische Wirksamkeit einer Tyrosinhydroxylase (TH)-gerichteten Immuntherapie gegen das Neuroblastom durch eine zusätzliche Immunstimulation mit dem Zytokin IL-15 und einer Immuncheckpoint-Blockade zu verbessern. Dafür wurde ein bicistronisches DNA-Vakzin generiert, welches sowohl für die cDNA der TH als auch die von IL-15 kodiert. Um dabei die Herstellung und MHC-I-vermittelte Präsentation antigener TH-Peptide, effektiv zu gestalten und die Immunigenität des Vakzins zu erhöhen, erfolgte die upstream-Kopplung einer modifizierten Ubiquitin-Sequenz an die der xenogenen TH. Die Integration der IL-15-Sequenz in das DNA-Vakzin ermöglicht eine auf die Mikroumgebung der Antigenpräsentation beschränkte lokale Applikation und kann so die Aktivierung zytotoxischer T-Zellen unterstützen ohne systemische Zytokin-bedingte Nebenwirkungen zu verursachen. Nach der Generierung des Vakzins erfolgte die In-vitro-Charakterisierung durch Transfektion von CHO-Zellen mit dem DNA-Vakzin, wobei die Plasmid-vermittelte Synthese der ubiquitinierten TH und Freisetzung von bioaktivem IL-15 bestätigt wurden. Die Evaluierung der anti-tumoralen Eigenschaften in vivo erfolgte in einem syngenen Mausmodell, wobei attenuierte S. typhimurium SL7207 als orales Applikationsvehikel für das DNA-Vakzin dienten, die durch die Stimulierung des mukosalen Immunsystems dem DNA-Vakzin eine gesteigerte Immunogenität verleihen. Im Rahmen dieser Arbeit konnte der S. typhimurium-vermittelte Genransfer sowie die effektive Expression der im Vakzin enthaltenen Gene in den APC der Peyer Plaques nachgewiesen werden. Weiterhin konnten die durch zytotoxische T-Zellen vermittelte anti-tumoralen Effekte einer TH-basierten Immuntherapie, repräsentiert durch ein reduziertes Primärtumorwachstum sowie verminderter Metastasierung, durch die Ko-Expression von IL-15 in einem syngenen Mausmodell gesteigert werden. Interessanterweise führte die Immuncheckpoint-Blockade durch eine anti-PD-1-AK-Therapie sowohl mit als auch ohne Kombination mit dem DNA-Vakzin zu einem signifikanten Anstieg der anti-tumoralen Effekte. Dieses Ergebnis unterstreicht die nachteilige Rolle immunregulatorischer Mechanismen bei der Ausbildung einer zellulären Immunantwort durch eine Immuntherapie, was ergänzend durch den Nachweis immunsuppressiver Zytokine in der Tumormikroumgebung bestätigt wurde. Bei der Anwendung von Behandlungsstrategien zur aktiven Immunisierung ist daher die Blockierung der natürlichen Regulationsmechanismen sowie der tumor-induzierten Immunsuppression von enormer Bedeutung.
Zusammenfassend war das in dieser Arbeit generierte xenogene DNA-Vakzin in der Lage durch die Ko-Expression von IL-15 die NB-spezifischen Effekte der TH-gerichteten Immuntherapie zu steigern. Des Weiteren konnten mittels einer anti-PD-1-gerichteten Immuncheckpoint-Blockade immunregulatorische Prozesse unterbunden und dadurch die CTL-vermittelten anti-tumoralen Effekte der Target-gerichteten DNA-Vakzinierung verstärkt werden. Gerade bei schwach immunogenen Tumoren wie dem Neuroblastom sind Kombinationstherapien notwendig, welche eine TAA-gerichtet Immunantwort zur Induktion eines Immunologischen Gedächtnisses in Gang bringen und gleichzeitg die Regulationsmechanismen unterbinden, um so eine effektive Tumorbekämpfung zu ermöglichen. Diese vielversprechende Vakzinierungsstrategie könnte somit neuer immunologischer Ansatz für eine adjuvante Erhaltungstherapie für eine minimale Resterkrankung bei refraktären Neuroblastompatienten darstellen.
Transition wird bezeichnet als zielgerichteter, geplanter Wechsel der Kinder und Jugendlichen mit chronischen Erkrankungen von dem kindzentrierten zum erwachsenenzentrierten Gesundheitssystem (Blum et al., 1993). Transition soll ein strukturierter, gut implementierter, geplanter und absichtsvoller Prozess sein, welcher die Jugendlichen mit chronischen Erkrankungen, ihre Eltern und die involvierten Gesundheitsexpert*innen befähigt, den Wechsel von der Pädiatrie zur Erwach¬senen¬medizin erfolgreich abzuschließen (Blum et al., 1993; Huang et al., 2014; Kennedy et al., 2007).
Es ist notwendig den Prozess der Transition an die individuellen Bedürfnisse der Jugendlichen anzupassen und somit deren Unterstützung flexibler zu gestalten zu können, um dies zu erreichen, werden entsprechende Instrumente zur Erfassung von Konstrukten in der Transition benötigt. Von besonderem Interesse ist die Transitionsbereitschaft von Jugendlichen mit chronischen Erkrankungen.
Um die individuelle Bereitschaft zur Transition eines Jugendlichen mit chronischer Erkrankung feststellen zu können, fehlen zudem änderungssensitive Instrumente, die zugleich auch allgemein anwendbar sind, sodass eine Vergleichbarkeit zwischen verschiedenen Erkrankungen bzw. Versorgungsbereichen ermöglicht wird. Von diesem methodischen Defizit der Transitionsforschung ausgehend werden in der vorliegenden Arbeit folgende Konstrukte sowie deren Operationalisierung und Erfassung im Kontext der Transition psychometrisch genauer analysiert: gesundheitsbezogene und krankheitsspezifische Lebensqualität, Versorgungszufriedenheit, Transitionskompetenz, Patientenaktivierung und Patienten-empowerment.
Zusammenfassend zeigt sich, dass die Konstrukte krankheitsspezifische Lebens-qualität, Versorgungszufriedenheit, Transitionskompetenz, Patientenaktivierung und Patienten¬empowerment zur Erfassung von transitionsbezogenen Veränderungen bei Jugendlichen mit chronischen Erkrankungen geeignet sind. Lediglich die Erfassung die gesundheitsbezogene Lebensqualität sollte für zukünftige Studien geprüft werden.
Das Pankreasadenokarzinom (PDAC) ist aufgrund seiner hohen Letalität die vierthäufigste Todesursache unter den Krebserkrankungen in der westlichen Welt. PDAC ist durch eine erhebliche desmoplastische Stromareaktion mit geringer Vaskularisierung gekennzeichnet, so dass die Karzinomzellen ständigem Nährstoffmangel und Hypoxie ausgesetzt sind. In gesunden Zellen wird die Autophagie, welche konstitutiv auf einem basalen Niveau aktiv ist, als Reaktion auf umweltbedingte Stressfaktoren als Recyclingmechanismus aktiviert, um die Zellen mit Nährstoffen zu versorgen. In dieser Arbeit konnte gezeigt werden, dass die Basalautophagie in den Karzinomzellen so hoch war, dass eine zusätzliche Aktivierung durch Mangelzustände nur schwach oder überhaupt nicht möglich war. Als Ursache hierfür ist die fehlende Sensitivität von mTOR zu nennen, wodurch eine verminderte Reaktion auf Nährstoffmangel oder Hypoxie bedingt ist. Ebenso sind Mutationen im KRAS-Gen, welche den RAS-Signalweg konstitutiv aktivieren, dafür verantwortlich, die Autophagie auch unter normalen Bedingungen zu steigern, so dass eine zusätzliche Induktion kaum möglich ist. Diese erhöhte Basalautophagie ist für die gesteigerte Proliferationsrate der Karzinomzellen sowie für die Versorgung der Zellen mit Aminosäuren notwendig. Weiterhin konnte gezeigt werden, dass sich Karzinomzellen durch einen uneinheitlichen oxidativen/ glykolytischen Stoffwechsel auszeichnen, wobei die oxidative Phosphorylierung eine größere Bedeutung innehat. In der vorliegenden Arbeit konnte gezeigt werden, dass die Proliferation der Karzinomzellen unter OXPHOS-Inhibitoren unterdrückt wurde. Dieses Ergebnis weist darauf hin, dass die Aufrechterhaltung der oxidativen Phosphorylierung der Schlüsselmechanismus ist, über den Autophagie und Nährstoffversorgung zum Zellwachstum beitragen. Dies stellt einen Widerspruch zur Warburg-Hypothese dar. Die Ergebnisse dieser Arbeit zeigen auf, dass der Mechanismus der Autophagie ein therapeutisches Target für eine unterstützende Therapie im Pankreaskarzinom darstellt, welches in nachfolgenden Arbeiten weiterführend evaluiert werden muss.
A 2-day workshop of ORCA and the IADR Cariology Research Group was organized to discuss and reach consensus on definitions of the most commonly used terms in cariology. The aims were to identify and to select the most commonly used terms of dental caries and dental caries management and to define them based on current concepts. Terms related to definition, diagnosis, risk assessment, and monitoring of dental caries were included. The Delphi process was used to establish terms to be considered using the nominal group method favored by consensus. Of 222 terms originally suggested by six cariologists from different countries, a total of 59 terms were reviewed after removing duplicates and unnecessary words. Sixteen experts in cariology took part in the process of reaching consensus about the definitions of the selected caries terms. Decisions were made following thorough “round table” discussions of each term and confirmed by secret electronic voting. Full agreement (100%) was reached on 17 terms, while the definitions of 6 terms were below the agreed 80% threshold of consensus. The suggested terminology is recommended for use in research, in public health, as well as in clinical practice.

The first part of my work comprises empirical findings and theoretical foundations on stress in its historical development and socio-emotional and behavioural factors.
The first study of my dissertation focuses on the relationship between perceived stress in adolescence, the context variables of perceived helpfulness and competition and socio-emotional and behavioural strengths and difficulties (i.e., emotional problems, symptoms of hyperactivity, problems with peers, prosocial behaviour and conduct problems) from early to middle adolescence. I postulated a moderation or mediation of the effect of perceived stress on socio-emotional and behavioural strengths and difficulties through the two context variables. My hypotheses were tested using a latent moderating structural equation model (moderation analysis) and a multi-group structural equation model taking into account the gender and age of the students (mediation analysis). The theoretical basis of my first study is the transactional stress model by Lazarus and Folkman (1984).
The second study of my dissertation is dedicated to the question whether perceived stress is a moderator in the relationship between depressive symptoms in early adolescence and socio-emotional and behavioural strengths and difficulties in middle adolescence. Based on the cognitive vulnerability-transactional stress theory of Hankin and Abramson (2001), which assumes reciprocal and dynamic relationships between the individual and the environment, I investigated this question and tested the relationships using a latent moderating structural equation model.
The third study of my dissertation is based on Bronfenbrenner’s socio-ecological model (1975). Using a cross-lagged panel design, I investigate the within time and longitudinal relationships between variables of emotional (in-)stability (i.e., depressive symptoms, perceived stress and loneliness) and socio-environmental factors (i.e., sense of belonging, student-student and teacher-student-relationship) from early to middle adolescence.
At the end of my work there is a summary of all results, a discussion and an outlook for future research.
Das Ziel des Projektes war die Untersuchung der erworbenen Chemotherapie-Resistenz des Pankreaskarzinoms gegenüber den Zytostatika 5FU und Gemcitabin in Hinblick auf eine potentielle Regulation durch microRNAs.
Im ersten Schritt wurden Zelllinien mit einer intrinsischen Sensitivität identifiziert. Sieben etablierte Zelllinien des Pankreaskarzinoms (BXPC-3; COLO 357; PATU8988-S; PATU8988-T; CAPAN-1; MIA-PaCa-2; PANC-1) wurden mit Hilfe einer Annexin-V/DAPI-Färbung auf eine 5FU- oder Gemcitabin-induzierte Apoptose untersucht. Vier Zelllinien mit einer intrinsischen Sensitivität (BXPC-3; COLO 357; PATU8988-S; PATU8988-T) konnten identifiziert werden. Es folgte die Etablierung einer sekundären Chemotherapie-Resistenz in diesen sensitiven Zelllinien und die Bestätigung durch einen Proliferationsassay. Um die Hypothese einer veränderten microRNA-Expression in den sekundär resistenten Zelllinien zu untermauern, wurde die Expression der microRNAs miR-21, miR-200b und miR-214 untersucht. Eine veränderte Expression wurde gefunden und eine microRNA-Transkriptom-Analyse folgte. Diese deckte eine aberrante Expression von neun microRNAs (miR-7-5p; miR-23a-3p; miR-24-3p; miR-100-5p; miR-125b- 5p; miR-301a- 3p; miR-3158-5p; miR-3621; miR-3940- 5p) auf. Die Validierung dieser microRNAs via RTPCR zeigte eine Überexpression von miR-100 und miR-125b in allen resistenten Tumorzellen. Dies ist die erste Studie, welche valide Beweise für die Beteiligung beider microRNAs an der erworbenen Resistenz gegen 5FU und Gemcitabin zeigt. Für die Evaluation ihrer biologischen Rolle wurde die miR-100 oder miR-125b durch eine transiente Transfektion in den resistenten Zellen inhibiert. Eine Einschränkung der Viabilität und ein Anstieg der Caspasen 3 und 7 konnte nach Applikation der Zytostatika gesehen werden. Durch einen Proliferationsassay konnte eine langfristige Inhibition des Tumorzellwachstums, des Metabolismus und der Viabilität nach dem Knock-down der microRNAs bewiesen werden. Ein Zytotoxizitäts-Assay zeigte, dass die microRNA-Inhibition zu einer Wiederherstellung von Sensitivität und Dosis-Wirkungs-Beziehung gegenüber 5FU oder Gemcitabin führt. Es ist somit gezeigt, dass die miR-100 und miR-125b eine entscheidende Rolle in der Ausbildung einer sekundären Antimetabolit-Resistenz spielen. Ob diese microRNAs als Zielmolekül einer zukünftigen Antitumortherapie oder als Screeningparameter genutzt werden können, müssen weitere Studien zeigen.
The site at the southern shore of Krakower See shows the Quaternary geology of the surrounding
area. The local Quaternary sequence comprises a thickness of 50–100m of Quaternary deposits while
the surface morphology is dominated by the ice marginal position of the Pomeranian moraine, which
passes through the area. The bathymetry of the lake basin of Krakower See indicates a predominant
genesis by glaciofluvial erosion in combination with glacial exaration. Past research in this area has focussed
on the reconstruction of Pleniglacial to Holocene environmental changes, including lake-level
fluctuations, aeolian dynamics, and pedological processes and their modification by anthropogenic
land use.
Archaeological discoveries in the Tollense Valley represent remains of a Bronze Age battle of ca.1300–1250 BCE, documenting a violent group conflict hitherto unimagined for this period of time in Europe, changing the perception of the Bronze Age. Geoscientific, geoarchaeological and palaeobotanical investigations have reconstructed a tree- and shrubless mire characterised by sedges, reed and semiaquatic conditions with a shallow but wide river Tollense for the Bronze Age. The exact river
course cannot be reconstructed, but the distribution of fluvial deposits traces only a narrow corridor, in which the Tollense meandered close to the current riverbed. The initial formation of the valley mire dates to the transition from the Weichselian Late Glacial to the early Holocene.
Neurobiological theories suggest that inter-individual differences in vagally mediated heart rate variability (vmHRV) have the potential to serve as a biomarker for inter-individual differences in emotion regulation that are due to inter-individual differences regarding the engagement of prefrontal and (para-)limbic brain regions during emotion processing. To test these theories, we investigated whether inter-individual differences in vmHRV would be associated with inter-individual differences in emotion regulation. We determined resting state vmHRV in a sample of 176 individuals that had also completed a short self-report measure of reappraisal and suppression use. Resting state vmHRV was derived from short-term (300 s) and ultra-short-term (120 s, 60 s) recordings of participants’ heart rate to determine the robustness of possible findings. Irrespective of recording length, we found that an increase in resting state vmHRV was associated with an increase in self-reported reappraisal but not suppression use. However, this association was only evident among male but not female participants, indicating a sex-specific association between inter-individual differences in resting state vmHRV and inter-individual differences in self-reported emotion regulation. These findings, which are consistent with previous ones, support theoretical claims that inter-individual differences in vmHRV serve as a biomarker for inter-individual differences in emotion regulation. Combing (ultra-)short-term measures of resting state vmHRV with short self-report measures of emotion regulation may, thus, be useful for researchers who have to investigate the neurobiological mechanisms of emotion regulation in a time- and resource-efficient manner.

Das Immunsystem des Neugeborenen unterscheidet sich grundlegend von
dem des Erwachsenen. Aufgrund der unvollständig entwickelten adaptiven
Immunantwort sind Neonaten einer erhöhten Infektionsgefahr ausgesetzt,
während sie vorrangig auf das innate Immunsystem bei der Abwehr von
Erregern zurückgreifen müssen. Einen wesentlichen Bestandteil dieser
angeborenen Immunabwehr stellen die neutrophilen Granulozyten dar, die
unter anderem in Form der Phagozytose die Bekämpfung von Pathogenen
im Gewebe realisieren.
Eine weitere, 2004 beschriebene Funktion der Neutrophilen ist die Bildung von „Neutrophil Extracellular Traps“ (NETs). Bei dieser speziellen Form des Zelltods, die auch als NETose bezeichnet wird, nutzen die neutrophilen Granulozyten ihre DNA und intrazelluläre Proteine, um antimikrobiell wirksame, netzförmige Strukturen in den Extrazellularraum auszuwerfen. Diese sind dort in der Lage, Pathogene zu binden und zu zerstören. Ziel dieser Arbeit war, die NETose bei Neugeborenen im Vergleich zu Erwachsenen zu untersuchen und wichtige perinatale Einflussfaktoren auf die NETose zu charakterisieren.
Die Neutrophilen wurden aus Nabelschnurblut isoliert und anschließend
mit entsprechenden Stimuli zur NETose angeregt. Die ausgeworfene DNA
wurde angefärbt, mittels Fluoreszenzmikroskopie analysiert und die
Ergebnisse hinsichtlich Anzahl und Größe der NETs ausgewertet.
In der vorliegenden Studie konnte gezeigt werden, dass die NETose bei
Neugeborenen im Vergleich zu Erwachsenen signifikant vermindert ist.
Darüber hinaus wurde ein Zusammenhang mit dem Gestationsalter
festgestellt, so wiesen unreife gegenüber reifen Neugeborenen ebenfalls eine eingeschränkte NET-Fähigkeit auf. Ferner wurde der Einfluss weiterer Parameter beleuchtet, wie des Geburtsgewichts und des -modus, des Geschlechts oder einer perinatalen Infektion.
Die verminderte Fähigkeit zur NETose ist Teil der unvollständig
entwickelten Immunantwort bei Neugeborenen und könnte so zu deren
Infektionsanfälligkeit beitragen.
Das in dieser Pilotstudie eingeschlossene Patientenkollektiv umfasste 50 Patienten des Institutes für Diagnostische Radiologie und Neuroradiologie der Universitätsmedizin Greifswald, bei denen eine Indikation zur CT-gestützten interventionellen Therapie mittels PRT oder Facetten-/ISG-Infiltration im Rahmen der Schmerztherapie bei Wirbelsäulenerkrankungen gestellt wurde.
Im Rahmen dieser Pilotstudie wurden nach Studieneinwilligung alle Patienten in einem ärztlichen Aufklärungsgespräch über die bevorstehende medizinische Intervention informiert. Bei 25 Patienten erfolgte diese Aufklärung auf dem bisher üblichen Weg unmittelbar von Angesicht zu Angesicht und bei den anderen 25 Patienten wurde dieses Gespräch mittels eines Videotelefonates innerhalb der Klinik durchgeführt. Aufklärungsinhalte sowie die Möglichkeit dem Arzt Fragen zu stellen oder Ängste zu äußern, waren in beiden Kommunikationswegen identisch. Die Akzeptanz gegenüber dem entsprechenden Aufklärungsmedium sowie die verstandenen Inhalte wurden 24 Stunden nach der durchgeführten Intervention telefonisch in Fragebögen erfasst.
In den Variablen der Akzeptanz „Gesprächsklima“, „Inhaltsübermittlung“, „Fragenbeantwortung“ und „Beziehung“ kann kein signifikanter Unterschied zwischen beiden Aufklärungsmedien nachgewiesen werden.
Die Anzahl an erinnerten Nebenwirkungen ist in der Gruppe „Videotelefonie“ verglichen mit der Gruppe „von Angesicht zu Angesicht“ schwach signifikant höher und die Häufigkeitsverteilung für „Ja“-und „Nein“-Antworten der Variable „Strahlenbelastung“ unterscheidet sich schwach signifikant zugunsten der Aufklärung per Videotelefonie. Es konnten keine signifikanten Unterschiede bezüglich der Häufigkeiten von „Ja“- oder „Nein“-Antworten in den Variablen „Behandlungsziel“ und „Fahrtüchtigkeit“ abhängig von der Aufklärungsart nachgewiesen werden.
In Anbetracht dieser Ergebnisse lässt sich feststellen, dass die Aufklärung per Videotelefonie jener von Angesicht zu Angesicht in den hier untersuchten Punkten nicht unterliegt und somit als gleichwertiges Medium für Aufklärungsgespräche genutzt werden könnte.
Periodontitis is one of the most prevalent oral diseases worldwide and is caused by multifactorial interactions between host and oral bacteria. Altered cellular metabolism of host and microbes releases a number of intermediary end products known as metabolites. There is an increasing interest in identifying metabolites from oral fluids such as saliva to widen the understanding of the complex pathogenesis of periodontitis. It is believed that some metabolites might serve as indicators toward early detection and screening of periodontitis and perhaps even for monitoring its prognosis in the future. Because contemporary periodontal screening methods are deficient, there is an urgent need for novel approaches in periodontal screening procedures. To this end, we associated oral parameters (clinical attachment level, periodontal probing depth, supragingival plaque, supragingival calculus, number of missing teeth, and removable denture) with a large set of salivary metabolites (n = 284) obtained by mass spectrometry among a subsample (n = 909) of nondiabetic participants from the Study of Health in Pomerania (SHIP-Trend-0). Linear regression analyses were performed in age-stratified groups and adjusted for potential confounders. A multifaceted image of associated metabolites (n = 107) was revealed with considerable differences according to age groups. In the young (20 to 39 y) and middle-aged (40 to 59 y) groups, metabolites were predominantly associated with periodontal variables, whereas among the older subjects (≥60 y), tooth loss was strongly associated with metabolite levels. Metabolites associated with periodontal variables were clearly linked to tissue destruction, host defense mechanisms, and bacterial metabolism. Across all age groups, the bacterial metabolite phenylacetate was significantly associated with periodontal variables. Our results revealed alterations of the salivary metabolome in association with age and oral health status. Among our comprehensive panel of metabolites, periodontitis was significantly associated with the bacterial metabolite phenylacetate, a promising substance for further biomarker research.
Helicobacter (H.) pylori is the most important cause for peptic ulcer disease and a risk factor for gastric carcinoma. How colonization with H. pylori affects the intestinal microbiota composition in humans is unknown. We investigated the association of H. pylori infection with intestinal microbiota composition in the population-based cohort Study-of-Health-in-pomerania (SHip)-tRenD. Anti-H. pylori serology and H. pylori stool antigen tests were used to determine the H. pylori infection status. the fecal microbiota composition of 212 H. pylori positive subjects and 212 matched negative control individuals was assessed using 16S rRNA gene sequencing. H. pylori infection was found to be significantly associated with fecal microbiota alterations and a general increase in fecal microbial diversity. in infected individuals, the H. pylori stool antigen load determined a larger portion of the microbial variation than age or sex. the highest H. pylori stool antigen loads were associated with a putatively harmful microbiota composition. this study demonstrates profound alterations in human fecal microbiota of H. pylori infected individuals. While the increased microbiota diversity associated with H. pylori infection as well as changes in abundance of specific genera could be considered to be beneficial, others may be associated with adverse health effects, reflecting the complex relationship between H. pylori and its human host.
Analyse der metabolischen Anpassung von Streptococcus pneumoniae an antimikrobielle Umwelteinflüsse

Das Gram-positive Bakterium Streptococcus pneumoniae ist ein humanspezifisches Pathogen des oberen Respirationstraktes. Der opportunistische Krankheitserreger kann jedoch mehrere Organe befallen und tiefer in den Körper vordringen, was zu lokalen Entzündungen wie Sinusitis und Otitis media oder zu lebensbedrohlichen Infektionen wie Pneumonie, Meningitis oder Sepsis führen kann. Für das Bakterium S. pneumoniae wurden bisher kaum Metabolom-Daten erhoben. Daher war das Ziel dieser Dissertation eine umfassende Charakterisierung des Metaboloms von S. pneumoniae. In dieser Dissertation wurden als analytische Methoden die Gaschromatografie (GC) und Flüssigkeitschromatografie (LC) jeweils gekoppelt mit Massenspektrometrie (MS) sowie die Kernspinresonanzspektroskopie (NMR) verwendet, um die Metaboliten zu analysieren. Es sind mehrere Analysetechniken erforderlich, um den Großteil des Metaboloms mit seinen chemisch verschiedenen Metaboliten zu erfassen. Artikel I fasst die Literatur zu Untersuchungen des Metabolismus von S. pneumoniae in den letzten Jahren zusammen. Um eine Momentaufnahme des biologischen Systems zum jeweiligen Zeitpunkt zu erhalten, ist neben dem reproduzierbaren Wachstum während der Kultivierung auch die exakte Probenahme zu beachten. Aus diesem Grund wurde in dieser Dissertation ein Probenahmeprotokoll für das Endometabolom von S. pneumoniae etabliert (Artikel II). Mithilfe des optimierten Protokolls wurde eine umfassende Metabolomanalyse in einem chemisch definierten Medium durchgeführt (Artikel II). Um S. pneumoniae in einer Umgebung ähnlich der im Wirt zu untersuchen, wurde in einem modifizierten Zellkulturmedium kultiviert. Intermediate zentraler Stoffwechselwege von S. pneumoniae wurden analysiert. Das intrazelluläre Stoffwechselprofil wies auf einen hohen glykolytischen Flux hin und bot Einblicke in den Peptidoglykan-Stoffwechsel. Darüber hinaus widerspiegelten die Ergebnisse die biochemische Abhängigkeit von S. pneumoniae von aus dem Wirt stammenden Nährstoffen. Ein umfassendes Verständnis der Stoffwechselwege von Pathogenen ist wichtig, um Erkenntnisse über die Anpassungsstrategien während einer Infektion zu gewinnen und so neue Angriffspunkte für Wirkstoffe zu identifizieren.
Die zunehmende Verbreitung von resistenten S. pneumoniae-Stämmen zwingt zur Suche nach neuen antibiotisch wirksamen Substanzen. Im Zuge dessen wurde in Artikel III die metabolische Reaktion von S. pneumoniae während des Wachstums unter dem Einfluss antibakterieller Substanzen mit dem Ziel der Identifizierung metabolischer Anpassungsprozesse untersucht. Dabei wurden Antibiotika mit unterschiedlichen Wirkmechanismen verwendet, wie die Beeinflussung der Zellwandbiosynthese (Cefotaxim, Teixobactin-Arg10), der Proteinbiosynthese (Azithromycin) sowie Nukleotidsynthese (Moxifloxacin). Es konnten keine Wirkmechanismus-spezifischen Marker-Metaboliten identifiziert werden. Jedes Antibiotikum verursachte weitreichende Veränderungen im gesamten Metabolom von S. pneumoniae. Die Nukleotid- und Zellwandsynthese waren am stärksten betroffen. Besonders vielversprechend sind Antibiotika mit zwei Wirkorten wie Teixobactin-Arg10 und Kombinationen aus zwei Antibiotika. In dieser Dissertation wurde das erste Mal das synthetisch hergestellte Teixobactin-Arg10 mittels einer der modernen OMICS-Techniken untersucht. Die vorliegende umfassende Metabolom-Studie bietet wertvolle Erkenntnisse für Forscher, die an der Identifizierung neuer antibakterieller Substanzen arbeiten.
Insgesamt tragen die Ergebnisse der Dissertation zu einem besseren Verständnis der bakteriellen Physiologie bei.
Der akute Myokardinfarkt auf Grundlage einer koronaren Herzerkrankung ist eine der häufigsten Todesursachen weltweit. Besonders ältere Patienten leiden oft an mehreren kardiovaskulären Grunderkrankungen - so sind Vorhofflimmern und KHK häufige Komorbiditäten. Die pharmakologische Therapie von Vorhofflimmern bei bestehender KHK ist ein therapeutisches Problem. Dronedaron hat sich in zahlreichen klinischen Studien als wirksames Antiarrythmikum herausgestellt und zeigte zudem positive Effekte auf die Inzidenz und Sterblichkeit von akuten Myokardinfarkten (ATHENA-Studie). In verschiedenen Forschungsarbeiten konnte nachgewiesen werden, dass Dronedaron in der Frühphase nach einem akuten Myokardinfarkt eine Reduktion der Infarktgröße bewirken kann. Im Gegensatz dazu scheinen Patienten mit schwerer struktureller Herzerkrankung bezüglich ischämischer Ereignisse nicht von einer Dronedarontherapie zu profitieren (PALLAS-Studie).
In dieser Arbeit sollte unter Nutzung des Schweins als Modellorganismus geklärt werden, ob im Langzeitverlauf vier Wochen nach einem akuten Myokardinfarkt protektive Effekte Dronedarons auf die Infarktgröße und die kardiale Funktion nachgewiesen werden können. Desweiteren wurden mithilfe ventrikulärer Gen- und Proteinexpressionsanalysen Dronedaroneffekte in gesundem Myokard, Infarktgrenze und Infarktnarbe auf molekularbiologischem Level untersucht.
Für den experimentellen Myokardinfarkt wurde mithilfe eines Ballonkatheters der rechte Ramus interventricularis anterior distal des ersten Septalastes für 90 Minuten verschlossen. Nach 4-wöchiger Infarktheilung wurden die Herzen explantiert und Gewebsproben für die Expressionsanalysen entnommen. Die Größe der Infarktnarbe wurde an den formalinfixierten Herzen mithilfe der MRT-Bildgebung bestimmt. Genexpressionsprofile wurden mittels RNA-Microarray und anschließender Validierung mittels RT-qPCR, Proteinexpressionsprofile mittels 2D-DIGE-Analyse und anschließender massenspektrometrischer Proteinidentifizierung erstellt.
Die Infarktgröße wurde durch die Dronedarontherapie nicht signifikant beeinflusst (Kontrolle vs. Dronedaron: 10,1 % vs. 9,2 %, n.s.). Das Schlagvolumen der dronedaronbehandelten Tiere zeigte im Gegensatz zu den Kontrolltieren vier Wochen nach Infarzierung keine signifikante Reduktion. Enddiastolisches Volumen, Ejektionsfraktion sowie Serummarker zeigten keine Unterschiede zwischen den Versuchsgruppen.
In den Expressionsanalysen konnten insbesondere in der Infarktgrenze Effekte der Dronedarontherapie beobachtet werden. Es kam zu einer Expressionssteigerung von TGFβ3, der TGFβ-Rezeptoren 1 und 2 sowie der TGFβ-bindenden Proteine LTBP1 und 2. Die vermehrte Aktivierung des TGFβ-Signalings zeigte sich auch in einer Expressionssteigerung matrizellulärer Proteine, darunter Thrombospondin 1 und 2, Periostin und Fibronektin, sowie diverser extrazellulärer Matrixproteine, v.a. Kollagen-Subtypen.
Die Aktivierung des TGFβ-Signalings sowie die Expressionssteigerung TGFβ-induzierter matrizellulärer Proteine in der Infarktgrenze begünstigen durch antiinflammatorische Effekte und Stimulation der Kollagensynthese die Infarktheilung sowie die Stabilisierung des Narbengewebes. Die Proteine Thrombospondin 1 und 2 verhindern zudem eine Expansion des Infarktes in gesunde Myokardareale. Desweiteren kann insbesondere Periostin die Kardiomyozytenproliferation im infarzierten Myokard stimulieren. So können das TGFβ-Signaling sowie matrizelluläre Proteine zu einer Verbesserung der kardialen Funktionalität nach akutem Myokardinfarkt beitragen. Es ist vorstellbar, dass Calcium-abhängige Mechanismen in Kardiomyofibroblasten die dronedaronabhängige gesteigerte Aktivierung des TGFβ-Signalings und als Folge der matrizellulären Proteine vermitteln.
Die Aufrechterhaltung des Schlagvolumens des linken Ventrikels durch die Dronedarontherapie vier Wochen nach einem Myokardinfarkt spricht für einen Erhalt der systolischen Kontraktionskraft des Herzens. Auch wenn keine signifikante Reduktion der Infarktgröße festgestellt werden konnte, deuten die gefundenen Ergebnisse auf eine protektive Wirkung von Dronedaron auf kardiale Funktionalität und die Infarktheilung bei ansonsten gesunden Patienten hin. Bei Patienten mit bereits strukturell vorgeschädigtem Herzen beispielsweise im Sinne einer schweren Herzinsuffizienz könnte die gefundene vermehrte Aktivierung des TGFβ-Signalings zu einer Verschlechterung der Prognose führen, was die Ergebnisse der PALLAS-Studie erklären könnte.
Species persistence in the face of rapidly progressing environmental change requires adaptive responses that allow organisms to either cope with the novel conditions in their habitat or to follow their environmental niche in space. A poleward range shift due to global warming induced habitat loss in the south has been predicted for the lesser horseshoe bat, Rhinolophus hipposideros. Theoretical as well as numerous empirical studies link range expansion success to increased dispersal and reproduction rates due to spatial sorting and r-selection resulting from low population densities at the expansion front. R. hipposideros females however are highly philopatric and the species’ life history reflects a K- rather than an r-strategy, encompassing a long life span and limited individual annual reproductive output. I therefore investigated if adaptations in these traits determining range expansion success (dispersal and reproduction) can be observed in this bat species of high conservation concern. Genetic diversity presents a critical factor for adaptive responses to global change, both for range expansion and for coping with novel environmental conditions. I hence explored the genetic diversity levels of European R. hipposideros leading edge populations and their drivers for an assessment of these populations’ evolutionary potential and the development of conservation recommendations.
Comparing range expansion traits between an expanding R. hipposideros metapopulation in Germany and a non-expanding one in France revealed that range expansion was associated with an increase in juvenile survival and fecundity, and no decrease in adult survival. These results demonstrate than an increase in reproduction and growth rates is generally possible in R. hipposideros, indicating a potential adaptation (sensu lato) to range expansion. A positive correlation between adult and juvenile survival in the expanding metapopulation suggests higher resource acquisition in the expanding metapopulation, giving rise to the question if the observed demographic changes have a genetic basis or if they are rather induced by differences in environmental conditions between the two metapopulations. Long-term range expansion success requires adaptive evolutionary changes. The relative contribution of the former and that of undirected changes resulting e.g. from differences in resource availability therefore will have to be investigated in more detail in the future to allow predictions about range expansion dynamics in R. hipposideros.
The number of individuals within a radius of approximately 60 to 90 km around a population (as a measure of connectivity) was identified as the main positive driver of the studied populations’ genetic diversity. Overall genetic diversity levels in German R. hipposideros populations were found to be reduced compared to populations in France as a legacy of demographic bottlenecks resulting from severe population declines in the mid-20th century. This finding is alarming as future range expansion can be expected to entail a further decrease in genetic diversity. The resulting loss of genetic diversity can be expected to be particularly strong in R. hipposideros due to the detected dependence of genetic diversity on connectivity, because range expansion often results in small and patchy populations.
Protecting and ideally re-installing genetic diversity in R. hipposideros leading edge populations therefore presents a conservation goal of utmost importance. To achieve this endeavour, conservation efforts should target the protection of extensive networks of well-connected populations. Geographical concentration of individuals should be avoided and populations in key locations that connect clusters must be protected particularly well to prevent populations from becoming isolated. Continuous, regular monitoring of population trends is also important for a quick registration of disturbances or threats, and the subsequent rapid development of countermeasures to preclude further demographic declines.
The reduced levels of genetic diversity in the German metapopulation precluded a reliable quantification of dispersal rates due to the reduced power of discrimination between individuals. While ongoing re-colonization and the establishment of new maternity colonies provide evidence for increased dispersal in the expanding metapopulation, evaluating the expected range expansion velocity of R. hipposideros in relation to the estimated velocity of global warming induced habitat loss will require the confirmation of the existing preliminary dispersal data by employing more genetic markers.
Die Zahl der Menschen mit Demenz (MmD) nimmt auch in den Krankenhäusern zu. Die Stationen sind üblicherweise auf eine Akutbehandlung ausgelegt, dies macht das Eingehen auf MmD schwierig. Sich dadurch ergebende mögliche Konsequenzen erstrecken sich von einer ungeplanten Wiedereinweisung über eine Heimeinweisung bis hin zum Tod des Patienten.
Verschiedene Studien konzentrieren sich auf die kurzfristigen Folgen einer Hospitalisierung
von MmD. Nur einige Studien erfassten Langzeitfolgen. Diese Informationen sind jedoch
bedeutend, um die Versorgung von MmD zu verbessern.
In der vorliegenden systematischen Übersichtsarbeit und Metaanalyse wird ein Überblick zu
den Langzeitfolgen (ungeplante Wiedereinweisung, Institutionalisierung und Mortalität) einer Hospitalisierung von MmD gegeben. Es werden Prädiktoren erfasst, die mit negativen Folgen
eines Krankenhausaufenthaltes assoziiert sind. Um alle relevanten Publikationen zum Thema zu erfassen, wurde eine systematische Literaturrecherche in den Datenbanken PubMed, CENTRAL und ScienceDirect durchgeführt. Die Qualität der einzelnen relevanten Studien
wurde in einem Formular zu Bewertung der Studien dokumentiert. Die Ergebnisse wurden in
einer Tabelle zusammengefasst, die Metaanalyse wurde mittels Review Manager 5.3 der Cochrane Collaboration berechnet.
Die systematische Literaturrecherche führte zu 1108 Artikeln, hiervon erfüllten 20 Artikel die Einschlusskriterien. 10 Studien wurden mit einer Kontrollgruppe durchgeführt und demnach in die Metaanalysen eingeschlossen. Die Inzidenz und das Relative Risiko für die Mortalität von
MmD (RR: 1,74 [95 %-KI 1,50-2,05]) und die Institutionalisierung (RR: 2,16 [95 %-KI 1,31-
3,56]) waren signifikant erhöht im Vergleich zu Menschen ohne Demenz. Die Ergebnisse bezüglich der ungeplanten Wiedereinweisung waren nicht aussagekräftig. Wichtige Faktoren, welche mit diesen Folgen assoziiert waren, waren der Schweregrad der Demenz, die Anzahl der eingenommen Medikamente und der Umfang der benötigten Hilfe bei der Verrichtung der Aktivitäten des täglichen Lebens.
Die Ergebnisse dieser Arbeit sprechen dafür, dass eine umfassendere Betreuung von MmD sowohl im Akutkrankenhaus als auch in der Häuslichkeit nötig ist. Außerdem sollten alle Prozesse an den Schnittstellen dieser Bereiche, insbesondere das Krankenhausentlassungsmanagement, weiter intensiviert und verbessert werden.
In this doctoral thesis, algorithms are presented that are designed for the investigation in the mesopause region between the upper Mesosphere and Lower Thermosphere (MLT). The photochemical models are proposed and applied to represent the oxygen airglow and the oxygen photochemistry in the MLT. Atomic oxygen, O, in the ground state, O(3P), is of special interest because it is a reactive trace gas actively contributing to the Earth’s airglow. The retrievals of O(3P) concentrations, [O(3P)], are based on the nightglow time series of the green line emission measured remotely as in the first part of this thesis and the individual profiles of multiple nightglow emissions of O and molecular oxygen (O2) measured in situ as in the second part of this thesis. To process the complete spectral time series measured by using the satellite-borne instrument SCIAMACHY (SCanning Imaging Absorption spectroMeter for Atmospheric CHartographY), an intricate set of algorithms is developed and applied with the regularized total least squares minimization approach to estimate a set of the optimal regularization parameters and to retrieve a corresponding set of vertical Volume Emission Rate (VER) profiles. Furthermore, these algorithms take emissions of another origin and the Earth's shape into account. Considering not identified states of O2, the established photochemical models are adjusted resulting in two model modifications. Both model modifications are employed to retrieve the [O(3P)] time series on the basis of the VER time series in the MLT. The model input parameters vary in the atmosphere that motivated to propose these two model modifications and to employ available sources of the input parameters. One semi-empirical model, one general circulation model and the satellite-borne instrument SABER (Sounding of the Atmosphere using Broadband Emission Radiometry) are employed as sources of the reference [O(3P)] and input parameters time series. The SABER instrument employed as a source of the input parameters is preferred according to the comparison of the retrieved and reference [O(3P)] time series. Studying the impact of the 11-year solar cycle on O(3P) in the MLT, an algorithm is developed and applied with the Levenberg-Marquardt algorithm to estimate the optimal fit parameters step-wise. The results of the O(3P) sensitivity analysis obtained with respect to the solar activity forcing at the 11 year and 27 day time scales and the lunar gravitational forcing agree with the reference model simulations. The hypothesis regarding vertical shifts between different of Meinel bands at least partly caused by the hydroxyl radical (OH*) quenching with O(3P) is confirmed experimentally. Based on the conclusion drawn in the first part of this thesis that the data sets’ self-consistency is high as for the averaged SABER and SCIAMACHY measurements, a comprehensive set of available data with a higher level of the data sets’ self-consistency is employed in the second part of this thesis. Multiple airglow emissions measured in situ during four campaigns are employed to propose the Multiple Airglow Chemistry (MAC) model. Processed emissions are the Herzberg I, Chamberlain, Atmospheric and Infrared Atmospheric band emissions of O2 and the green line emission of O. Considering all widely known and additionally complemented reactions, the MAC model is proposed to represent the oxygen airglow and the oxygen photochemistry in the MLT. The presented MAC model is based on the hypothesis of Slanger et al. (2004) stating that higher excited states of O2 are coupled with each other through vibronic de-excitation caused by collisions among molecules of this group of O2 states in the MLT. This hypothesis is modified excluding the singlet Herzberg state of O2 from the group of O2 states considered by Slanger et al. (2004). The MAC calculations are carried out sequentially starting with higher excited O2 states to provide the retrieved output concentrations of these O2 states as the input concentrations to the next calculation steps. The final step is only based on concentrations of all species, whereas each of the earlier steps is based on a corresponding VER profile besides of the input concentrations. The oxygen photochemistry in the MLT is represented by all species considered at the final step that makes it possible to adopt the MAC reactions in a general circulation model. Four modifications of the MAC model, i.e. including or excluding the triplet Herzberg states of O2 and including or excluding ozone and odd hydrogen (hydrogen, OH* and hydroperoxy radical), lead to negligible differences in the retrieved [O(3P)] profiles. Based on the MAC calculations verified and validated on the basis of the four rocket campaigns, one of the effective modifications of the MAC model (excluding the triplet Herzberg states of O2, ozone and odd hydrogen) is further reduced to the most effective modification. This implies that for the [O(3P)] retrieval only the O2 Atmospheric band emission, temperature and concentrations of molecular nitrogen (N2) and O2 are sufficient to apply. Calculations carried out by using the most effective modification of the MAC model are verified and validated on the basis of self-consistent in situ measurements obtained simultaneously. The MAC model enables identifying precursors of (1) the three lowest O2 valence states and (2) the second excited O state responsible for (1) the Atmospheric and Infrared Atmospheric band emissions of O2 and (2) the green line emission of O, respectively. Particularly, the singlet Herzberg state of O2 is identified as the major precursor of the second excited O state resulting in the green line emission. In focus of potential further research is an extension of the MAC model with vibrationally excited states of O2 and ionized species.
The term diabetes mellitus comprises a group of metabolic diseases all distinguished by their main characteristic hyperglycaemia. With a steadily increasing prevalence diabetes displays an enormous burden for patients and health systems and is therefore of special interest for research. The development of the two main types of diabetes, type 1 and type 2, is closely linked to the formation of reactive species, especially hydrogen peroxide, inside different compartments of pancreatic beta cells. However, these cells are especially vulnerable towards oxidative stress mediated by hydrogen peroxide due to a low expression of antioxidative enzymes.
The main aims of the present thesis were to analyse the intracellular generation and to enable the site-specific detection of hydrogen peroxide to evaluate its role in the delicate equilibrium between redox signalling and oxidative stress under certain pathophysiological conditions, and moreover to monitor its movement through compartments and subcellular membranes of insulin-producing cells. Additionally, a new methodology for an artificial site-specific generation of hydrogen peroxide inside living cells was developed.
Salivary glands provide secretory functions, including secretion of xenobiotics and among
them drugs. However, there is no published information about protein abundance of drug transporters
measured using reliable protein quantification methods. Therefore, mRNA expression and absolute
protein content of clinically relevant ABC (n = 6) and SLC (n = 15) family member transporters in the
human parotid gland, using the qRT-PCR and liquid chromatography-tandem mass spectrometry
(LC−MS/MS) method, were studied. The abundance of nearly all measured proteins ranged between
0.04 and 0.45 pmol/mg (OCT3 > MRP1 > PEPT2 > MRP4 > MATE1 > BCRP). mRNAs of ABCB1,
ABCC2, ABCC3, SLC10A1, SLC10A2, SLC22A1, SLC22A5, SLC22A6, SLC22A7, SLC22A8, SLCO1A2,
SLCO1B1, SLCO1B3 and SLCO2B1 were not detected. The present study provides, for the first time,
information about the protein abundance of membrane transporters in the human parotid gland,
which could further be used to define salivary bidirectional transport (absorption and secretion)
mechanisms of endogenous compounds and xenobiotics.
Matrix-product-state based methods, in particular the density-matrix renormalization group, are used to numerically investigate several one-dimensional systems, focusing on models with symmetry-protected topological phases that generalize the spin-1 Haldane chain. In the first part, ground state properties such as topological order parameters and the criticality at quantum phase transitions are studied.
The second part deals with dynamic properties of spin chains. Using time-dependent matrix-product-state calculations, the dynamic structure factor, and the transport properties of contacted spin chains are analyzed.
Das Porphyrin-Derivat 5,10,15,20-tetra(m-Hydroxyphenyl)chlorin (mTHPC, Temoporfin), in Europa zugelassen unter dem Handelsnamen Foscan®, stellt einen der vielversprechendsten Photosensibilisatoren (PS) in der Photodynamischen Therapie (PDT) dar. Während einige Publikationen über die Aktivität von Temoporfin in einzelnen Krebszelllinien in der Fachliteratur erschienen sind, fehlen systematische Studien über die Temoporfin-basierte PDT, die mithilfe eines Sets aus mehreren Krebszelllinien durchgeführt wurden. In der vorliegenden Dissertation wurden fünf humane, adhärente Krebszelllinien aus dem Kopf-Hals-Bereich und weiteren PDT-relevanten Geweben eingesetzt, um die Auslösung von oxidativem Stress und die zugrundeliegenden Zelltodmechanismen der Temoporfin-basierten PDT in vitro zu untersuchen. Dafür wurde zunächst die Viabilität der Zellen nach Temoporfin-Behandlung (0,001–5,0 µmol/L) und Bestrahlung mit einer LED-basierter Apparatur (0,2–5,5 J/cm2) im MTT-Test ermittelt und Konzentrations-Wirkungskurven zur Bestimmung von IC50- und IC90-Werten aufgenommen. Zur Untersuchung und zum Vergleich der Zelltodmechanismen in den Zelllinien wurden diese anschließend mit äquitoxischen Konzentrationen des Temoporfins behandelt und mit vorrangig mit einer Lichtdosis von 1,8 J/cm2 (vereinzelt auch 3,5 oder 7,0 J/cm2) bestrahlt. Die Analytik der Zellen erfolgte direkt, 6, 24, oder 48 h nach der Bestrahlung. Die Ergebnisse der vorliegenden Dissertation bestätigen die Auslösung von oxidativem Stress nach der Temoporfin-PDT durch die Detektion eines Totalverlustes des mitochondrialen Membranpotentials (Δψm) sowie einer erhöhten Bildung reaktiver Sauerstoffspezies (ROS). Eine Lipidperoxidation (LPO) sowie die Beteiligung einer Nekrose am Zelltod spielen hingegen in den meisten Zelllinien eine untergeordnete Rolle. Vor allem die ausbleibende Nekrose nach einer hochdosierten PDT mit Temoporfin steht dabei in Kontrast zu den Resultaten vieler publizierter Studien. Als vorherrschender Zelltodmechanismus wurde nach Temoporfin-vermittelter PDT stattdessen über eine Phosphatidylserin-Externalisierung, eine Caspase 3-Aktivierung und eine PARP-Spaltung eine Auslösung von Apoptose identifiziert. Zudem kann parallel zur Apoptose eine Autophagie ausgelöst werden bzw. kann diese zunächst sogar dominieren, bevor sie zu einer Autophagie-assoziierten Apoptose führt. Als Anzeichen der späten Phase einer Apoptose ist zudem früher oder später eine DNA-Fragmentierung nachweisbar, und der Zelltod wird in einigen Fällen von einem Zellzyklusarrest in der G2/M-Phase begleitet.
Neben der Auswertung der Zelltodmechanismen wurde ein weiterer Schwerpunkt der vorliegenden Dissertation auf den Vergleich der Ergebnisse aus verschiedenen, identisch behandelten Zelllinien gelegt. Obwohl die Zellen unter äquitoxischen Bedingungen behandelt wurden und die PDT über eine vergleichsweise unspezifische Bildung von ROS wirkt, wurden sehr heterogene Resultate in den unterschiedlichen Zelllinien erhalten. Die vorliegende Untersuchung zeigt deshalb außerdem, dass generelle Schlussfolgerungen nach einer PDT eine Analytik in mehreren unterschiedlichen Zelllinien benötigt, um verlässlich zu sein. Dieser Umstand ist in der Vergangenheit in In-vitro-Studien zur PDT jedoch viel zu häufig ignoriert worden.
Die Pt(II)-Komplexe Carboplatin (CBDCA), Cisplatin (CDDP) und Oxaliplatin (1-OHP) werden als Zytostatika in einer Vielzahl chemotherapeutischer Verfahren eingesetzt. In der vorliegenden Dissertation wurden die Pt(II)-Komplexe CBDCA, CDDP, oder 1-OHP in fünf humanen, adhärenten Krebszelllinien mit einer Temoporfin-basierten PDT kombiniert. Die Zellviabilität wurde im MTT-Test bestimmt und synergistische Effekte der Kombination mithilfe der Berechnung von Combination Indices (CI) ermittelt. Synergismus wurde dabei nach einigen Kombinationen, z.B. mit 1-OHP in drei der fünf getesteten Zelllinien, beobachtet, aber auch antagonistische Effekte wurden für einige Kombinationen in bestimmten Zelllinien detektiert. Im Fall einer Synergie wurden erhöhte ROS-Level nachgewiesen, jedoch wurde die Auslösung von Apoptose im Vergleich zu einer Behandlung mit den Pt(II)-Komplexen bzw. der PDT allein dadurch nicht notwendigerweise verstärkt. Eine Analyse der Zellzyklusverteilung ergab, dass nach kombinierter Behandlung sub-G1-Populationen mit fragmentierter DNA, die für eine späte Phase der Apoptose charakteristisch sind, gebildet werden, und zudem S-Phase und G2/M-Arreste vorliegen.
Die Ergebnisse der vorliegenden Dissertation zeigen, dass eine Behandlung mit einer Temoporfin-basierten PDT das Potential besitzt, einige Typen von Tumorzellen gegenüber Pt(II)-Komplexen, vor allem 1-OHP, zu sensibilisieren und so besser für die Tumortherapie zugänglich zu machen. Das Erreichen synergistischer Effekte hängt dabei jedoch in hohem Maße vom Ursprungsgewebe und tumorspezifischen Eigenschaften der Zellen ab und es können sogar antagonistische Effekte auftreten.
Die Glutathionperoxidase 1 (GPX1) kann bei einer Überexpression in Tumorzellen zu einer Inhibition von Apoptose führen, was zum Überleben der Tumorzellen beiträgt. Zudem kann eine erhöhte GPX-Aktivität zu einer Resistenzentwicklung gegenüber oxidativen Schäden beitragen, was den Tumor z.B. vor Chemotherapeutika schützen kann. Potential in der Tumortherapie besitzen deshalb GPX-Inhibitoren, die einer Überexpression der GPX1 durch Verringerung der Gesamtaktivität entgegen¬wirken. In Kombination mit Chemotherapeutika führten GPX-Inhibitoren so bereits zu einer Re-Sensibilisierung Zytostatika-resistenter Zelllinien. In der vorliegenden Dissertation wurde eine Kombination der GPX-Inhibitoren 9-Chlor-6-ethyl-6H-[1,2,3,4,5]pentathiepino[6,7-b]indol (CEPI), ein kürzlich synthetisiertes trizyklisches Pentathiepin, oder Mercaptobernsteinsäure (MBS), der klassische Inhibitor der GPX in vielen veröffentlichten Studien, mit einer Temoporfin-PDT eingesetzt, um durch die Blockierung eines effektiven Abbauweges für H2O2 eine vermehrte Akkumulation von ROS zu erzeugen und so den phototoxischen Effekt des Temoporfins zu verstärken. Synergismus wurde dabei nach Kombination mit beiden GPX-Inhibitoren, jedoch nicht in allen fünf Zelllinien, beobachtet. Auf der anderen Seite wurden mit beiden Inhibitoren auch antagonistische Effekte in bestimmten Zelllinien detektiert. Die ROS-Level konnten nach kombinierter Behandlung mit CEPI, nicht jedoch mit MBS, in einigen Zelllinien gesteigert werden, was darauf hindeutet, dass die Inhibition der GPX durch CEPI tatsächlich zu einer zusätzlichen Akkumulation von ROS führt. Beide Inhibitoren erzeugen allein keine Steigerung der ROS-Spiegel. Überraschenderweise wurde eine vermehrte Apoptoseauslösung anschließend jedoch nach Kombination mit beiden GPX-Inhibitoren in allen untersuchten Zelllinien erreicht, was darauf schließen lässt, dass weitere Faktoren die Induktion von Apoptose fördern, z.B. eine Autophagie-assoziierter Zelltodmechanismus oder eine Ferroptose, die durch LPO nach Verlust der GPX4-Aktivität eingeleitet wird. Die erhöhte Apoptoseauslösung wurde zudem in der Zellzyklusanalyse bestätigt, in der nach kombinierter Behandlung mit beiden GPX1-Inhibitoren erhöhte Anteile apoptotischer sub-G1-Frak¬tionen mit fragmentierter DNA nachweisbar waren. Zudem wurde die DNA-Fragmentierung nach Kombination mit MBS von einem gesteigerten G2/M-Arrest begleitet.
Die Ergebnisse der vorliegenden Dissertation zeigen, dass eine Kombination von Inhibitoren der GPX mit einer Temoporfin-basierten PDT zu synergistischen Effekten führen kann, wodurch eine Verstärkung der Phototoxizität möglich ist. Da die Inhibitoren in nicht-toxischen Konzentrationen eingesetzt werden, werden zudem keine zusätzlichen Nebenwirkungen erwartet. Der Erfolg des kombinierten Ansatzes hängt dabei jedoch in hohem Maße vom Ursprungsgewebe und tumorspezifischen Eigenschaften der Zellen ab, aber auch antagonistische Effekte können auftreten.
