Peatlands are wetland ecosystems covering a relatively small area of the World (~3%), but at the same time storing excessive amounts of carbon for a very long time (equivalent to the four times global annual net primary production). As carbon sinks, peatlands work in spite of their slow growth, absorbing carbon dioxide (CO2) through the photosynthetic activity of the peatland plants and their low growth rates, and because high groundwater table removes oxygen from the soil and slows down the decomposition of the dead plant matter. Because of the relative lack of the oxygen in the peat, especially compared to the mineral soils, methanogen populations in the peatlands are abundant, and releasing methane (CH4), a potent greenhouse gas, to the atmosphere. Therefore, peatlands are generally at the same time significant carbon sinks and stores as well as the methane sources. The balance among the two peatland gass fluxes (CO2 and CH4) will dictate the impact of any given peatland on the global climate and primarily driven by hydrology, in the form of the groundwater table levels.
Because of the slow decomposition rates, and from radiocarbon dating of the peat as well as the subfossil records buried in it, carbon stored in peatlands is locked for a very long time (centuries to millennia). It is, therefore, crucial to gain insights into the development of peatlands and their gas balance through time. One way to get both is by studying peatland hydrology in the form of the groundwater table levels and their historical variations. Unfortunately, intensive monitoring of peatland groundwater table, when available, is an only a recent endeavor. Therefore, we need to employ proxies to reconstruct the past by leveraging the present. In statistics, proxy variables are often used when the observations of the variable of interest, are either missing or too difficult to obtain.
In this thesis, I tested whether we can use the radial growth of the Scots pines growing on peat as proxies to the peatland hydrology. To that end, I studied growth responses of the peatland Scots pines. Other proxies can and are used for the reconstructions of the groundwater table levels, but tree-growth is widely used as one of the proxies to reconstruct past environments which is at the same time annually resolved.
First, I examined the growth ecology of the peatland Scots pines by looking at their intra-annual development and trying to find relationships between it and environmental factors while at the same time comparing it with the Scots pines growing at the forest sites. I first tried with wood anatomy and found that, unfortunately, peatland Scots pines do not form enough wood cells, and consequently do not have high temporal resolution, necessary to investigate the intra-annual patterns of the radial growth. Initial results from wood anatomical investigations were interesting none-the-less, indicating that peatland Scots pines might have smaller cell features than the Scots pines from forests, but might at the same time maintain Early/Latewood ratios of those same features.
After I found that wood anatomical series were not resolved enough I decided to go with dendrometers, linear displacement sensors which constantly monitor the variations of stem radius, to get insights into the intra-annual growth patterns of the peatland Scots pines. Before using dendrometers for ecological investigations, I was involved in implementing routines commonly used in the analysis of the dendrometer signals and bringing them to R in the form of the dendrometeR package.
At one peatland complex, I installed dendrometers on ten trees in total at both peatland and forest sites and compared the pattern of the standardized signal. I inferred from the comparisons and classifications that the signal from two sites was indistinguishable for the dendrometer series shorter than five days. Furthermore, the most important environmental factor driving the radial variation at the peatland site was hydrological, daily relative humidity, indicating further that peatland hydrology might indeed be the driver behind peatland Scots pine growth.
Finally, I looked at the growth responses of peatland Scots pines from central Estonia using dendrochronological methods. Peatland hydrology, in the form of the groundwater table levels, was indeed the environmental factor with the strongest, and also stationary, correlations with the radial growth of the peatland Scots pine. That relationship indicated that peatland Scots pines are indeed possible proxies for reconstructing past levels of the peatland groundwater tables.
My study further indicated that the growth response of the peatland Scots pines was non-linear, further complicating the reconstructions of the past peatland hydrology. However, the strength of the growth response was proportional to the general hydrological regime, expressed as median groundwater table level. As the hydrological regime of the peatland does not vary considerably on the annual scales, but more on decadal it might be more appropriate to find another, independent, proxy to the hydrological regime first, and than use annually resolved radial growth of the peatland Scots pine to reconstruct past levels of the peatland groundwater table.
Helicobacter (H.) pylori is the most important cause for peptic ulcer disease and a risk factor for gastric carcinoma. How colonization with H. pylori affects the intestinal microbiota composition in humans is unknown. We investigated the association of H. pylori infection with intestinal microbiota composition in the population-based cohort Study-of-Health-in-pomerania (SHip)-tRenD. Anti-H. pylori serology and H. pylori stool antigen tests were used to determine the H. pylori infection status. the fecal microbiota composition of 212 H. pylori positive subjects and 212 matched negative control individuals was assessed using 16S rRNA gene sequencing. H. pylori infection was found to be significantly associated with fecal microbiota alterations and a general increase in fecal microbial diversity. in infected individuals, the H. pylori stool antigen load determined a larger portion of the microbial variation than age or sex. the highest H. pylori stool antigen loads were associated with a putatively harmful microbiota composition. this study demonstrates profound alterations in human fecal microbiota of H. pylori infected individuals. While the increased microbiota diversity associated with H. pylori infection as well as changes in abundance of specific genera could be considered to be beneficial, others may be associated with adverse health effects, reflecting the complex relationship between H. pylori and its human host.
Herstellung sicherer und wirksamer Lebendvakzine gegen die Koi Herpesvirus Infektion von Karpfen

Das Koi Herpesvirus (KHV, Cyprinid herpesvirus 3) verursacht eine tödliche Erkrankung bei Kois und Karpfen. Um sichere und wirksame Lebendvirusimpfstoffe zu erhalten, haben wir Einzel- und Doppeldeletionsmutanten von KHV erzeugt, aus deren Genom die für die beiden Nukleotidstoffwechselenzyme Thymidinkinase (TK, ORF55) und Desoxyuridin-Triphosphatase (DUT, ORF123) codierenden Leserahmen gezielt entfernt worden waren. Die Mutationen wurden durch homologe Rekombination in den zellkulturadaptierten aber noch virulenten Stamm KHV-T eingeführt. Umfangreiche in vitro Tests zeigten, dass die Deletion der TK- und DUT- Gene die KHV-Replikation in Zellkultur (CCB Zellen) nicht erkennbar beeinträchtigt. In vivo Tests an Jungkarpfen zeigten jedoch eine im Vergleich zum Ausgangsvirus signifikant reduzierte Virulenz der Einzelgen-Deletionsmutanten eine fast vollständige Attenuierung der Doppelmutante. Dennoch waren alle immunisierten Karpfen gegen eine letale Belastungsinfektion mit virulentem KHV geschützt. Mittels einer neu entwickelten Triplex-Real-Time-PCR und aus Kiementupferproben isolierter DNA war es möglich, mit TK-negativem KHV immunisierte und Wildtyp- infizierte Karpfen zu differenzieren. Daher könnte die Doppelmutante KHV- TΔDUT/TK als genetischer Marker-Impfstoff geeignet sein.
In einer zweiten Studie wurde die Funktion von vier immunogenen Hüllglykoproteinen der ORF25-Genfamilie (ORF25, ORF65, ORF148 und ORF149) von KHV untersucht. Hierbei wurde festgestellt, dass alle vier Gene für die Virusreplikation in Zellkultur entbehrlich sind. Während die Deletion von ORF65 keinen erkennbaren Einfluss auf die Virusvermehrung hatte, führte die Deletion von ORF148 sogar zu einer leicht erhöhten Replikationsrate. Im Gegensatz dazu bewirkten Deletionen von ORF25 oder ORF149 einen verzögerten Eintritt in die Wirtszellen und damit auch eine verlangsamte Vermehrung und Ausbreitung der Viren. Interessanterweise führte die gemeinsame Deletion der Gene ORF148 und
ORF149 zu einem wildtypähnlichen Wachstumsverhalten, das auf gegensätzlicher Funktionen der beiden Proteine hindeutete. Elektronenmikroskopische Untersuchungen von CCB-Zellen, die mit den verschiedenen Glykoproteindeletionsmutanten infiziert waren, zeigten keine Auswirkungen auf die Bildung und Reifung der Virionen im Zellkern oder im Zytoplasma, oder die Virusfreisetzung. Im Tierversuch erwiesen sich KHV-Mutanten mit Deletionen der Gene ORF148 und/oder ORF149 als geringfügig, aber für eine Verwendung als Lebendvirus-Impfstoff nicht ausreichend abgeschwächt. Überlebende Fische waren jedoch gegen Belastungsinfektionen ebenso gut geschützt wie Wildtyp-infizierte Karpfen, so dass die Deletion dieser antikörperinduzierenden Proteine zur Entwicklung von KHV-Markerimpfstoffen beitragen könnte, die eine serologische Differenzierung von Wildtyp-infizierten und geimpften Fischen erlauben (DIVA- Prinzip). In einer dritten Studie wurden durch serielle Zellkulturpassage von virulentem KHV und anschließende in vivo Infektionsversuche Hinweise darauf gefunden, dass das bislang nicht näher charakterisierte, neben dem ORF149 Gen lokalisierte ORF150 für einen weiteren Virulenzfaktor von KHV codiert. Möglicherweise könnte also durch eine kombinierte Deletion der im Rahmen dieser Arbeit untersuchten KHV-Gene ein sicherer und wirksamer, genetisch und serologisch differenzierbarer Markerimpfstoff hergestellt werden.
The deep-sea tubeworm Riftia pachyptila lacks a digestive system but completely relies on bacterial endosymbionts for nutrition. Although the symbiont has been studied in detail on the molecular level, such analyses were unavailable for the animal host, because sequence information was lacking. To identify host-symbiont interaction mechanisms, we therefore sequenced the Riftia transcriptome, which served as a basis for comparative metaproteomic analyses of symbiont-containing versus symbiont-free tissues, both under energy-rich and energy-limited conditions. Our results suggest that metabolic interactions include nutrient allocation from symbiont to host by symbiont digestion and substrate transfer to the symbiont by abundant host proteins. We furthermore propose that Riftia maintains its symbiont by protecting the bacteria from oxidative damage while also exerting symbiont population control. Eukaryote-like symbiont proteins might facilitate intracellular symbiont persistence. Energy limitation apparently leads to reduced symbiont biomass and increased symbiont digestion. Our study provides unprecedented insights into host-microbe interactions that shape this highly efficient symbiosis.
AbstractThis paper takes concepts from spatial theory and globalization discourse and uses them in order to analyze the narrative function of descriptions of nature in romantic Icelandic poetry from the beginning of the 19th century and an Icelandic TV-Series from 2015. In Iceland’s romantic poetry of the early 19th century, especially in poems written by Bjarni Thorarensen, sublime nature is described as a form of guardian against foreign influences that threaten the way of living on the peripheral island. This romantic concept of Icelandic nature is closely connected to narrative patterns in the process of the Icelandic Nation-Building, as it characterizes Icelanders as simultaneously defined and protected by the harsh conditions on the island. The paper takes a comparative look at the underlying narrative concepts of nature in two of Bjarni Thorarensen’s poems and a recent Icelandic TV series, Baltasar Kormákur’s Ófærð (2015), that presents a different concept of Icelandic nature in its relation to a (threatening) global influence. The series depicts a globalized world in which crime does not only affect remote communities as an evil from the outside but as a local evil connected to forces on global scale. Nature as a narrative device in the TV series thus does not protect Icelanders from global forces, as it did in Bjarni Thorarensens poems in the early 19th century, but instead functions a catalyst that reveals the evil from the outside and the evil from within.
Phenolics and its derivatives are aromatic compounds with a wide range of industrial applications. Gallic acid, protocatechuic acid, catechol or pyrogallol are only a few examples of industrially relevant aromatics. The production of bulk fine chemicals primarily for chemical and pharmaceutical industry has put a strong emphasis on optimizing manufacturing conditions. Commercial production of many chemicals is still based on organic chemical synthesis using petroleum derivatives as starting material. Since these processes are considered environmentally unfriendly and posing an irresponsible strain on limited fossil resources, much attention is paid to the development of new microbial factories for the bioproduction of industrially relevant chemicals using renewable sources or organic pollutants as starting material. Arxula adeninivoras is a non-conventional yeast possessing attractive properties for industrial application such as thermo- and osmotolerance. Another major advantage of this organism is its broad substrate spectrum with tannin at the forefront. The present project is dedicated to the study of the tannic acid degradation pathway in A. adeninivorans. Two genes encoding enzymes annotated as gallic acid decarboxylase (AGDC1) and catechol-1,2-dioxygenase (ACDO1) have been selected and investigated. Both enzymes were characterized and their function in tannin catabolism analyzed.
Hepeviruses are small viruses with a RNA-genome of positive polarity that form the family Hepeviridae. The family includes two genera: members of the genus Piscihepevirus were detected in fish species and members of the genus Orthohepevirus were found in different mammal and bird species. The genus Orthohepevirus contains four different species, namely Orthohepevirus A, B, C and D. The species Orthohepevirus A contains five human pathogenic genotypes, with three of them being zoonotic. The species Orthohepevirus C contains mammal-associated pathogens, which were identified in rats and carnivores. The human pathogenic genotypes are responsible for a self-limiting acute hepatitis in humans, which could become chronically in immunocompromised individuals. The main route of transmission is the consumption of undercooked meat and direct contact with HEV-positive excreta or blood. In Germany, hepatitis E is a notifiable disease since 2001 with an increased number of cases per year. Rats are the reservoir of rat-associated HEV (ratHEV), but also the zoonotic HEV-3 genotype was detected in rats. The European rabbit (Oryctolagus cuniculus) was identified as a reservoir host of a subgenotype of human pathogenic HEV-3 (HEV-3ra).
For the development of small mammal animal models, the objective of this study was to evaluate different small mammal populations for novel hepeviruses and to study the presence of HEV and sequence divergence of ratHEV and rabbitHEV in rat and rabbit populations from Europe.
Approximately 3000 rodents from Germany and the Czech Republic were screened by broad spectrum HEV-RT-PCR. As a result, 13 common voles (Microtus arvalis) and one bank vole (Myodes glareolus) were detected to be HEV-RNA positive. Comparison of the obtained sequences, complete genome determination and phylogenetic analysis indicated the finding of a novel common vole-associated HEV (cvHEV), which shows a high sequence divergence towards other members of the species Orthohepevirus C, but shares a high sequence similarity to a HEV-genome derived from a kestrel (Falco tinnunculus). The finding of cvHEV-RNA in a bank vole might be caused by a spillover infection. The cvHEV genome shares the hepevirus-typical open reading frames, but also has unique cvHEV-specific attributes in its genome.
The investigation of 420 Norway rats (Rattus norvegicus) and 88 Black rats (Rattus rattus) identified HEV-RNA in Norway rats from eight of nine and Black rats from two of four European countries. In a single Norway rat from Belgium, a HEV-3-strain with high sequence similarities to rabbitHEV (HEV-3ra), was detected. The investigation of zoo animals revealed a ratHEV spillover infection in a Syrian brown bear (Ursus arctos syriacus). This infection was most likely caused by ratHEV-infected free-living, wild rats from the same zoo.
Investigation of wild rabbit populations trapped in and around Frankfurt am Main, Germany, showed anti-HEV antibodies (34.7%) and rabbitHEV-RNA (25%). A high sequence similarity of rabbitHEV in the animals trapped at the urban site was observed, whereas a high sequence divergence was seen for the animals trapped at the rural trapping sites.
In conclusion, hepeviruses are widespread among different small mammal populations in Europe. The broad geographical distribution of these hepeviruses should be taken into account in further public health risk assessments. Further investigations are needed to characterize the presence of cvHEV in more detail, especially by taking the population dynamics of common voles into account. The detected HEV-strains could be taken as basis for the establishment of novel HEV-animal models, which might replace the so far used swine and non-human primate models.
Die degenerativ-kalzifizierende Aortenklappenstenose ist das häufigste Herzklappenvitium in der Altersgruppe der über 65-jährigen und tritt mit zunehmendem Alter vermehrt auf. (Vahanian et al. 2012; Stewart et al. 1997). Lange Zeit galt der offen-chirurgische Aortenklappenersatz als einziges kuratives Verfahren in der Behandlung der Aortenklappenstenose. Bei vielen Patienten wird jedoch aufgrund eines sehr hohen Lebensalter oder zahlreicher Komorbiditäten dieser Eingriff abgelehnt (Grube et al. 2006, Iung et al. 2003). Für diese Menschen steht seit wenigen Jahren die perkutane kathetergestützte Aortenklappenimplantation (TAVI) als neue Therapieoption zur Verfügung, welche sich gleichfalls in den letzten Jahren zu einer Therapieoption für Patienten mit niedrigerem Operationsrisiko zu entwickeln scheint (Gaede 2017).
Um die derzeit bestehenden Mortalitäts- und Komplikationsraten nach TAVI weiter zu senken, ist die präoperative Ermittlung von prognoserelevanten Einflussfaktoren von großer Bedeutung.
Für die vorliegende Dissertationsschrift wurden die Ergebnisse von 193 Patienten, die am Klinikum Karlsburg von September 2009 bis Juli 2014 einen transapikalen oder transfemoralen kathetergestützten Aortenklappenersatz (TAVI) erhielten, analysiert und anhand aktueller Literatur diskutiert.
Alle untersuchten Patienten wiesen eine hochgradige Aortenklappenstenose (AÖF ≤ 1,0 cm²) auf und konnten als Hochrisikopatienten angesehen werden. Sie waren im Durchschnitt 79,3 ± 6,0 Jahre alt und besaßen einen logistsichen EuroSCORE von 22,24 ± 16,7%, einen EuroSCORE II von 5,79 ± 5,8% und einen STS Score von 5,50 ± 3,9%. Das präinterventionelle Risikoprofil, sowie die vorab erhobenen echokardiografischen Daten waren mit denen bereits veröffentlichter Studien vergleichbar. In 98% der Interventionen erfolgte eine technisch erfolgreiche Prozedur. Die mittlere OP-Dauer betrug 72,2 ± 21,9 min, die Durchleuchtungszeit 9,2 ± 5,1 min und die verbrauchten Kontrastmittelmenge 94,4 ± 38,8. Die stationäre Aufenthaltsdauer betrug durchschnittlich 13,8 ± 9,1 Tage.
Insgesamt konnte ein sehr gutes 30-Tage-Outcome erzielt werden und damit die TAVI, als vergleichsweise komplikationsarmes und bezogen auf die Kurz- und Langzeitergebnisse gut funktionierendes Verfahren bestätigt werden. Die 30-Tage-Mortalität betrug 5,2% und die 30-Tage-MACCE-Rate 9,3% und ähnelt damit den Ergebnissen der Vergleichsliteratur.
Anhand einer umfangreichen Prädiktoranalyse, die sowohl univariat, als auch multivariat erfolgte, konnten eine periphere arterielle Verschlusskrankheit, eine ACI-Stenose ≥70 %, eine

zerebrovaskuläre Insuffizienz, der additive und logistische EuroSCORE, der STS-Score, die OP-Dauer, die Kontrastmittelmenge als Einflussfaktoren auf die 30-Tage-Mortalität ermittelt werden. In der multivariaten Analyse verblieben eine periphere arterielle Verschlusskrankheit, eine ACI-Stenose ≥70 %, eine pulmonale Hypertonie >60 mmHg, ein Diabetes mellitus, die OP-Dauer und die chirurgische Leistenrevision als unabhängige Prädiktoren. Bei Betrachtung der 30-Tage-MACCE-Rate stellten sich in der univariaten Analyse zusätzlich zu den die 30-Tage-Mortalität beeinflussenden Parametern, eine pulmonale Hypertonie >60 mmHg und der EuroSCORE II als signifikante Faktoren heraus. Multivariat konnten eine ACI-Stenose ≥70%, die eingeschränkte linksventrikuläre Ejektionsfraktion, der logistische EuroSCORE und die OP-Dauer als unabhängige Einflussfaktoren der 30-Tage-MACCE-Rate analysiert werden. Durch den Vergleich mit den Ergebnissen bereits veröffentlichter Arbeiten konnten dabei prognoserelevante Faktoren der genannten Endpunkte bestätigt, beziehungsweise neue Einflussfaktoren ermittelt werden.
Immunogenicity and protectivity of surface-localized lipoproteins of Streptococcus pneumoniae

Steptococcus pneumoniae (pneumococcus) represents a common colonizer of the human upper respiratory tract (URT). However, under certain conditions, for example following viral infections, or in indiciduals with a weakened immune system, including young children, elderly and immunocompromised persons, it can cause a wide range of life-threatening diseases, such as pneumonia, meningitis or sepsis. Based on the polysaccharide capsule that surrounds the bacterium, pneumococci are classified into so far 98 different serotypes. Prevention of S. pneumoniae infections was achieved by the development of pneumococcal polysaccharide-based (PPSV) vaccines. However, these vaccines have important limitations, including high manufacturing costs and restricted serotype coverage facilitating replacement by non-vaccine serotypes. Aiming for the development of a serotype-independent vaccine, the potential of surface-exposed and highly conserved pneumococcal lipoproteins was evaluated for being targeted as a future protein-based vaccine. Therefore, selected lipoproteins were examined i) for their surface abundance and accessibility, ii) for their presence in clinically relevant S. pneumoniae strains, and iii) for their immunogenicity. Finally, based on these initial screenings, the most promising candidates were selected to analyze their protective efficacy in a moude model of colonization. DacB and PnrA were identified as highly abundant lipoproteins on the pneumococcal surface. They showed to be immunogenic both during natural infection using convalescent patient sera and when given to mice as a subunit vaccine formulation. Following intranasal immunization and challenge of mice with two heterologous S. pneumoniae strains, both proteins reduced the pneumococcal load in the nasopharynx. The protection correlated with increased production of IL-17A indicative for a Th17-mediated immunity, which is strongly suggested to play a critical role in preventing pneumococcal colonization and infection. Lipoproteins are triggering innate receptors on antigen-presenting cells, thereby linking innate with adaptive immune responses. Therefore, lipidated proteins were evaluated for their potential to be used as an adjuvant for vaccination. Lipidation clearly enhanced humoral immune responses to DacB and PnrA without the need of an additional adjuvant. However, an additional adjuvant was required to confer protection against pneumococcal colonization. In conclusion, Lipoproteins are interesting candidates for future protein-based vaccine strategies because they are highly conserved, abundant and immunogenic. PnrA and DacB were identified as potential candidates, since they induced protection against pneumococcal colonization, which in turn may lead to a decline in infections and transmission.
Neuroblastoma (NB) is an aggressive, poorly immunogenic tumor in childhood. Therapy for high-risk NB remains challenging. Immunotherapy with anti-GD 2 antibody ch14.18/CHO effectively prolongs the survival of NB patients.
Killer-immunoglobulin-like receptor (KIR)/human leucocyte antigen (HLA) mismatch and Fc gamma receptor (FCGR) polymorphisms are reported to affect antibody-dependent cellular cytotoxicity (ADCC) induced by monoclonal antibodies. To determine whether FCGR polymorphisms and KIR/HLA mismatch are associated with the survival following ch14.18-based immunotherapy, genotyping methods that allow for genotype determination of FCGR2A, -3A, -3B, KIR2DL1, 2DL2, 2DL3, and 3DL1 have been established and applied to the analysis of 53 NB patients treated with ch14.18/CHO.
High-affinity polymorphisms of FCGR2A (H131) and FCGR3A (V158) were associated with improved survival. Importantly, patients displaying both the FCGR3A-V158 and FCGR2A-H131 alleles exhibited significantly improved event-free survival. No association was found between KIR/HLA genotypes or FCGR3B alleles and patients’ survival in our patient cohort.
In conclusion, impact of FCGR2A and -3A genotypes in response to ch14.18/CHO immunotherapy in combination with IL2 was demonstrated. FCGR2A and -3A might therefore provide a prognostic marker when conducting ch14.18/CHO-based immunotherapy.
The advances in high-throughput sequencing technologies have revolutionized the possibilities for pathogen identification in cases of unknown disease origin. Diagnostic metagenomics allows the unbiased and simultaneous detection of almost all nucleic acids in a clinical sample, with the potential to provide pivotal insights into otherwise undeterminable causes of human or animal disease.
In this thesis, possibilities, pitfalls and the suitability of Ion Torrent and Illumina sequencing platforms for comprehensive use in diagnostic metagenomics were assessed and optimized procedures developed. Clinical field samples, undiagnosable by standard diagnostics, were taken as real-life examples for the investigations. The results show that cross-contamination due to index swapping and run-to-run-carryover constitute a major issue on Illumina platforms, severely compromising the correct interpretation of results for clinical specimens. In contrast, Ion Torrent platforms did not display any form of cross-contamination, however, the commercial library preparation method is less efficient. Combining the advantages of both platforms, customized Y adapters, facilitating highly efficient library preparation, were developed for Ion Torrent sequencing and applied in further experiments. The obstacles of strongly degraded RNA in formalin-fixed paraffin-embedded samples were identified and the workflow adapted to meet the requirements of smaller fragments. Additionally, it was shown that adequate sampling is a very important step, if not the most important step, in the workflow, as well as subsequent validation of the obtained results in terms of causation. The achievements in this study allow other researchers the application of a sensitive and optimized diagnostic metagenomics workflow.
Furthermore, the investigations on the clinical samples resulted in the discovery of a novel respirovirus with putative zoonotic potential, the first description of Borna disease virus 1 in human organ transplant recipients, and the discovery of a very distantly related novel ovine picornavirus. These discoveries build a basis for further research and expand the knowledge regarding new and emerging viruses.
Abstract
Proteome analyses are often hampered by the low amount of available starting material like a low bacterial cell number obtained from in vivo settings. Here, the single pot solid‐phase enhanced sample preparation (SP3) protocol is adapted and combined with effective cell disruption using detergents for the proteome analysis of bacteria available in limited numbers only. Using this optimized protocol, identification of peptides and proteins for different Gram‐positive and Gram‐negative species can be dramatically increased and, reliable quantification can also be ensured. This adapted method is compared to already established strain‐specific sample processing protocols for Staphylococcus aureus, Streptococcus suis, and Legionella pneumophila. The highest species‐specific increase in identifications is observed using the adapted method with L. pneumophila samples by increasing protein and peptide identifications up to 300% and 620%, respectively. This increase is accompanied by an improvement in reproducibility of protein quantification and data completeness between replicates. Thus, this protocol is of interest for performing comprehensive proteomics analyses of low bacterial cell numbers from different settings ranging from infection assays to environmental samples.
Objectives: Performing proper toothbrushing is a complicated process for children. Therefore, the aim of this study was to investigate the effect of a smartphone app for improving manual toothbrushing via a gravitation sensor. Methods: In this prospective, controlled, single-blinded, randomized clinical trial, 49 children (mean age 5.1 ± 0.6 years, 27 female) were randomly assigned to test (n = 26) and control (n = 23) groups. All children were provided with manual toothbrushes with an integrated gravitation sensor and they received oral health instructions. Only the children of the test group got an additional smartphone app to visualize and reward proper brushing in form and time. At baseline and recalls after 6 and 12 weeks, plaque and gingival indices (QHI, PBI) were recorded for analysis between the two groups. Results: At baseline, there were no significant differences between the test and control group regarding plaque and gingival indices (QHI: 2.36 ± 0.7 and 2.42 ± 0.8; p = 0.94; PBI: 0.42 ± 0.2 and 0.47 ± 0.3; p = 0.59). At the 6- and 12-week recalls, the test group showed statistically significantly better oral health indices than the controls (6-week recall, QHI: 0.8 ±0.5 and 1.88 ± 0.9; p < 0.001; PBI: 0.08 ± 0.1 and 0.26 ± 0.2; p < 0.001; 12-week recall, QHI: 0.44 ± 0.5 and 1.49 ± 0.7; p < 0.001; PBI: 0.05 ± 0.18 and 0.21 ± 0.1; p < 0.001). Conclusion: The results highlight the enormous possibilities of a toothbrushing application via the smartphone, at least for medium-term oral hygiene improvement in preschool children and even after excluding the app. The long-term effect should also be investigated to exclude the expected novelty effect.
Die chronische Herzinsuffizienz ist aktuell eine der häufigsten Todesursachen in Deutschland und verursacht jährlich Gesamtkosten von ca. 3,2 Millionen Euro (Stand: 2008, Quelle: Statistisches Bundesamt). Eine kausale Therapie existiert bisher nicht und viele Patienten lassen sich auf Grund mangelnder Compliance oder starker Progredienz der Erkrankung nur schwer therapieren. Adulte kardiale Progenitorzellen sind erstmals vor ca. 15 Jahren beschrieben worden und stehen seitdem im Mittelpunkt vieler Untersuchungen. Progenitorzellen, die durch das Vorhandensein des Oberflächenmarkers Sca-1+ charakterisiert sind, wird zugeschrieben, dass sie in der Lage sind, unter anderem zu Kardiomyozyten zu differenzieren. Sca-1+ Zellen sind zusätzlich zur Synthese parakriner Faktoren fähig, die u.a. mit der Hemmung von Apoptose, Fibrose, kardialem Remodeling und einer Stimulation der Angiogenese in Verbindung gebracht werden. Im Rahmen dieser Arbeit wurde untersucht, wie sich die Gabe des Sekretoms von Sca-1+ und Sca-1- Zellen auf die Progression der HI auswirkt und welchen Einfluss das Herzinsuffizienz-milieu auf die parakrinen Effekte der Zellen hat.
Zur Untersuchung dieser Fragestellung wurde ein transgenes Herzinsuffizienzmodell der Maus, das Cyclin T1/Gαq Modell, genutzt. Das Herz der 5 Wochen alten Versuchstiere wurde an Tag 0 mit Hilfe eines Kleintier-MRTs untersucht. An Tag 1 erhielten die Mäuse das Sekretom unbehandelter oder aldosteronvorbehandelter Sca-1+ oder Sca-1- Zellen. Die abschließende MRT-Untersuchung fand am 14. Tag statt, um die Änderung der kardialen Parameter während des Untersuchungszeitraumes zu bestimmen. Zur Beurteilung wurden dabei die linksventrikuläre Ejektionsfraktion (LVEF), das endsystolische Volumen (ESV), das enddiastolische Volumen (EDV), die linksventrikuläre Masse (LVM) und das fractional shortening (FS) herangezogen. Anschließend wurden anhand der extrahierten Herzen und des gewonnenen Plasmas qRT-PCR-, ELISA- und Zytokinuntersuchungen durchgeführt.
Das Sekretom aldosteronvorbehandelter Sca-1+ Zellen wies während des untersuchten Zeitraumes eine signifikant geringere Verschlechterung der LVEF und des LVFS im Vergleich zu den unbehandelten Tieren auf. Die qRT-PCR Untersuchungen zeigten, dass sowohl die Tiere, die das Sekretom von Sca-1+ Zellen, als auch die Gruppe, die das Sekretom von Sca-1- Zellen erhalten hatte eine signifikant niedrigere Expression fibroseassoziierter Proteine im Vergleich zu unbehandelten Versuchstieren aufwiesen. Anhand der ELISA-Untersuchungen konnte nachgewiesen werden, dass die Sekretomtherapie, unabhängig von dem verwendeten Zelltyp, bei allen Therapiegruppen zu einer signifikanten Senkung des N-terminalen Propeptid BNP (NT-proBNP) und zur Steigerung der Galektin-1-Plasmakonzentration führt.
Zusammenfassend lässt sich feststellen, dass das Sekretom von Sca-1+ und Sca-1- Zellen einen positiven Effekt auf den Krankheitsverlauf herzinsuffizienter Mäuse hat. Die Steigerung der EF und FS, die Senkung der NT-proBNP Plasmakonzentration, die Steigerung der Galektin-1 Plasmakonzentration und die Senkung fibroseassoziierter Proteine deuten auf einen kardioprotektiven Effekt des Sekretoms beider Zellreihen hin. Somit sind möglicherweise verschiedene kardiale Zellpopulationen in der Lage, nach einer Schädigung des Herzens über parakrine Mechanismen protektiv wirksam zu sein. Welche langfristigen Effekte die Sekretomtherapie hat und ob sich dadurch eine Mortalitätsreduktion und ein verbesserter Outcome erreichen lässt, muss mittels weiterer Untersuchungen geklärt werden.
Ziel:
Aussagen zur Scherhaftfestigkeit und Torsionsstabilität 3D-gedruckter Polymethacrylat-Brackets mit makroretentiver bzw. mikroretentiver Basis im Vergleich zu konventionell
hergestellten Kunststoff-Brackets.
Material und Methode:
Die mit der Software FreeCad 0,16 (freecadweb.org) konstruierten
Brackets (Standard Edgewise-Bracket mit 22 inch Slotgröße, Torque 0°, und Tip 0°) wurden im DLP-Verfahren aus einem medizinisch zugelassenen Polymethacrylat (SHERAprint ortho Plus, SHERA, Lemförde) 3D-gedruckt.
Die Nachhärtung erfolgte in zwei Zyklen unter Stickstoff Schutzatmosphäre (1,5bar, je 3000 Blitze, 10/sec, 200 Watt, 280 bis 580nm) (Otoflash G171, NK Optik, Baierbrunn) nach einer
Reinigung im Ultraschallbad (2 x 2min, 90% Ethanol).
Zur Untersuchung der Scherhaftfestigkeit wurden bei 40 dieser Brackets Retentionsrillen in die Basis konstruiert (Makroretention). Weitere 40 Brackets erhielten eine glatte Bracketbasis, die 3 Sekunden lang mit Aluminiumoxidkristallen sandgestrahlt wurde (Mikroretention). Als Kontrollgruppe dienten konventionelle Kunststoffbrackets vergleichbarer Größe (Brilliant, Forestadent, Pforzheim). Die Brackets aller drei Versuchsgruppen wurden im Standard-etch- Verfahren mit lichtpolymerisierendem Adhäsiv auf bovinen Zahnprüfkörpern befestigt und einem Thermocycling unterzogen (1000 Zyklen, je 30sec, 5/55°C). Die Ermittlung der Scherhaftfestigkeit erfolgte gemäß DIN 13990-246 in einer Universalprüfmaschine (Zwick BZ050/TH3A, Zwick Roell, Ulm). Zur statistischen Auswertung wurden eine einfaktorielle Varianzanalyse (ANOVA), der t-Test und der Mann-Whitney-Test durchgeführt. Zur Durchführung der Torsionsstabilitätversuche wurden die bereits in den Scherversuchen verwendeten Bracketgrundkörper um einen Lastarm erweitert. Eine in die Universalprüfmaschine eingespannte Haltekonstruktion diente der Fixierung eines senkrecht zur Druckrichtung ausgerichteten geraden Stangendrahts (Querschnitt von 0,019 inch x 0,025 inch), an dem der Bracketanteil der Prüfkörper mit einer kieferorthopädischen Stahlligatur befestigt wurde. Die Torque- und Tipbewegung entstand durch Kraftapplikation des Druckscherbügels auf den Lastarm des Prüfkörpers.
Ergebnisse:
3D-gedruckte Brackets mit makroretentiver Basis zeigten eine Scherhaftfestigkeit von 3,89 ± 1,0663MPa. Bei 3D-gedruckten Brackets mit mikroretentiver Basis wurde eine signifikant geringere Scherhaftfestigkeit von 3,09 ± 1,0454MPa ermittelt.
Die Kontrollgruppe mit konventionell hergestellten Kunststoffbrackets zeigte eine vergleichbare Scherhaftfestigkeit von 3,84 ± 0,995MPa. Die in der Literatur beschriebene minimale Scherhaftfestigkeit von 6MPa wurde von keiner der Versuchsgruppen erreicht. Die Torsionsstabilität 3D-gedruckter Bracket zeigte beim maximalen Torque einen Mittelwert von 51,10 ± 9,40Nmm, das mittlere maximale Tipdrehmoment betrug 32,42 ± 5,43Nmm.
Schlussfolgerungen:
Die Scherhaftfestigkeitswerte 3D-gedruckte Brackets mit Makroretention ähneln denen von konventionell hergestellten Kunststoffbrackets. Um den klinischen Anforderungen besser gerecht zu werden, bedarf es hier allerdings noch einer Steigerung. Die Bruchfestigkeit 3D-gedruckter Brackets in Bezug auf die Torsionsbelastung, sowohl unter Torque- als auch unter Tipbewegung, scheint für den klinischen Gebrauch hingegen angemessen zu sein. In-office verwendbare 3D-Drucker könnten zukünftig für die Herstellung individualisierter Multibracket-Apparaturen genutzt werden.
The Madden–Julian oscillation (MJO) is a major
source of intraseasonal variability in the troposphere. Recently, studies have indicated that also the solar 27-day variability could cause variability in the troposphere. Furthermore, it has been indicated that both sources could be linked, and particularly that the occurrence of strong MJO events could be modulated by the solar 27-day cycle. In this paper, we analyze whether the temporal evolution of the MJO phases could also be linked to the solar 27-day cycle. We basically count the occurrences of particular MJO phases as a function of time lag after the solar 27-day extrema in about 38 years of MJO data. Furthermore, we develop a quantification approach to measure the strength of such a possible relationship and use this to compare the behavior
for different atmospheric conditions and different datasets, among others. The significance of the results is estimated based on different variants of the Monte Carlo approach, which are also compared. We find indications for a synchronization between the MJO phase evolution and the solar 27-day cycle, which are most notable under certain conditions: MJO events with a strength greater than 0.5, during the easterly phase of the quasi-biennial oscillation, and during boreal winter. The MJO appears to cycle through its eight phases within two solar 27-day cycles. The phase relation between the MJO and the solar variation appears to be such that the MJO predominantly transitions from phase 8 to 1 or from phase 4 and 5 during the solar 27-day minimum. These results strongly depend on the MJO index used such that the synchronization is most clearly seen when using univariate indices like the OLR-based MJO index (OMI) in the analysis but can hardly be seen with multivariate indices like the real-time multivariate MJO index (RMM). One possible explanation could be that the synchronization pattern is encoded particularly in the underlying outgoing longwave radiation (OLR) data. A weaker dependence of the results on the underlying solar proxy is also observed but not further investigated. Although we think that these initial indications are already worth noting, we do not claim to unambiguously prove this relationship in the present study, neither in a statistical nor in a causal sense. Instead, we challenge these initial findings
ourselves in detail by varying underlying datasets and methods and critically discuss resulting open questions to lay a solid foundation for further research.
Using validated analytical tools and optimized sampling procedures, it was possible to detect a vast number of metabolites from the extracellular space but also from the cytosol of B. subtilis. The results indicate that the complement of the analytical methods was suitable in the monitoring of the metabolome since it allowed a great coverage of physicochemical diverse metabolites. However, a wide number of unknown metabolites/features were also detected. Although broad databases exist that can help in the annotation of metabolites, further investigation is needed in their identification. In unpredictable changing conditions, bacterial cells possess appropriate adaptation strategies for a successful bacterial growth. These rely on sensing mechanisms that globally adjust gene expression via transcription and feedback regulations. The metabolic sensing mechanisms have emerged as key roles in the nutritional information and regulation of cell cycle processes. In this work, a new quality of information regarding the metabolism and adaptation to the absence of key signal mechanisms in B. subtilis was provided. Investigations of cells lacking Pyk uncovered alterations in the import of glucose and pyruvate from the nutritional media. These results gives insights to the pyruvate homeostasis mechanism but also brought new questions concerning the regulation of the CCR. Pyruvate wasn't susceptible to the glucose dependent CCR in Δpyk. The earlier in ux of pyruvate in these cells is in accordance to the newly discovered pyruvate transport mechanism. Also, it was speculated that the lower consumption of external glucose could be a consequence of the impairment of the PTS system in the mutant cells due to the accumulation of glycolytic metabolite FBP. In future studies, insights of the PTS system mechanism should be done in these conditions, that could comprise the determination of HPr phosphorylation and the HPrK activity. This study also arose new questions that should be address, that include the higher secretion of acetoin and 2,3-butanediol, and the lower accumulation of shikimate 3-phosphate by the mutant cells. In an untargeted metabolomic analysis, a vast number of altered features were suggested to be fatty acids metabolites, precursors of phospholipids and LTA. Complementary approaches should be done for the confirmation of these metabolites and the inspection of possible alterations in the membrane structure. In the study of LTA mutants, the accumulation of PG precursors provided a hint of altered cell wall assembly. Although by uorescence microscopy no clear changes were detected, the metabolic results emphasized the previous assumption of the affected hydrolytic activity occurring in the PG. For comprehensive knowledge of the cell wall it would be important to detect and identify more metabolites of the LTA anchor using optimized cromatographic method. These results could be complemented with other omics data sets studies which would help in the elucidation of these key regulatory systems mechanisms.

With the caries decline, particularly for populations with low caries levels, these indices reach their statistical limits. This paper aims to introduce a specific term, the Specific affected Caries Index (SaC) for the risk groups in populations with low caries prevalence and to illustrate its use based on the consecutive German National Oral Health Survey (GNOHS) in children. In groups with a caries prevalence less than one-third of the population, many caries-free children (DMFT = 0) are included in the SiC (risk group), which calls for a new way of illustration. Mean caries experience (DMFT), caries prevalence, the SiC and SaC were portrayed for 12-year-olds in the GNOHS from 1994/95 to 2016. The SaC describes the mean caries experience (DMFT) in the group presenting caries experience (DMFT > 0). In 12-year-old 6th graders in Germany, the mean caries experience decreased from 2.4 (1994/95) to 0.4 DMFT (2016), with a recent prevalence of 21.2% (DMFT > 0, 2016). In 2016, the mean number of affected teeth in children with DMFT > 0 (SaC) was 2.1, while the SiC including 12% DMFT-free children in the risk group was 1.3. The SiC fails to reflect the caries severity in children in a population with low caries prevalence. Therefore, the newly introduced term Specific affected Caries Index (SaC) may be used to describe accurately caries experience in caries risk children in populations presenting low caries prevalence.

The goal of this thesis was to characterize the properties of tetramyristoyl cardiolipin (TMCL) and several environmental influences on it. This included investigating the pH and temperature dependency of TMCL as well as the influences of ROS on TMCL and exam-ining the lipid-protein interactions between TMCL and cytc. Furthermore, I extended the research to the analysis of binary mixtures composed of TMCL and dimyristoyl phosphati-dylcholine (DMPC). To this end, I investigated the samples with the aid of the Langmuir monolayer technique. This method allowed me to mimic interactions occurring at the membrane surface as it represents one membrane layer. The recording of π-A isotherms was also coupled with further other techniques like Brewster angle microscopy (BAM), Infrared Reflection-Absorption Spectroscopy (IRRAS), Grazing Incidence X-Ray Diffraction (GIXD) and Total Reflection X-Ray Fluorescence (TRXF) to enable a more comprehensive monolayer study. In addition, some systems were analyzed using Thin-layer Chromatography (TLC) and/or Differential Scanning Calorimetry (DSC) to be able to draw conclusions about sample composition or characteristic temperatures, respectively.
Streptococcus pneumoniae is one of the leading human pathogen causing morbidity and mortality worldwide. The pneumococcus can cause a variety of different diseases ranging from mild illnesses like otitis media and sinusitis to life-threatening diseases such as pneumonia, meningitis and sepsis. Mostly affected are infants, elderly and immune-suppressed patients. Although, there are vaccines against pneumococci available, still hundreds of thousands of people got infected each year. These vaccines are targeting the pneumococcal polysaccharide capsule. Because of the high number of different serotypes, it is not possible to generate a vaccine against all present serotypes. In the last years a shift to non-vaccine serotypes was noticed. This strengthens the need for the development of vaccines which do not target polysaccharides. Thus, proteins came into focus as potential new vaccine candidates or targets for drug treatment, because several proteins are highly conserved among different strains or even genera. Proteome analyses can give insights into the protein composition in a certain state of a bacterium. So, targets can be identified, which are especially expressed under infection-relevant conditions. Iron limitation is one of these conditions and the knowledge on iron acquisition in pneumococci is still limited. Iron is an essential trace element and as redox-active catalyst or as cofactor involved in various key metabolic pathway in nearly all living organisms and thus also in bacteria. For instance, iron is necessary during biosynthesis of amino acids and in electron transport as well as in DNA replication. Within the human host iron is extremely limited due to its high insolubility under physiological conditions, which is part of the nutritional immunity of its human host. Hence, bacteria had to evolve mechanism to overcome iron starvation. In this thesis the adaptation process triggered by iron limitation in the S. pneumoniae serotype 2 strain D39 was investigated in a global mass spectrometry-based proteome analysis.
In preceding growth experiments the pneumococcal growth was adapted to the needs of proteomic workflows. In order to investigate the pneumococcal response to iron limitation, the organic iron-chelating agent 2,2’-bipyridine (BIP) was applied. For the quantification of changes in protein abundances comparing stress to control conditions the very reliable and robust metabolic labeling technique Stable Isotope Labeling with Amino Acids in Cell Culture (SILAC) was used. This method requires the bacterial cultivation in a chemically defined medium, for which reason modified RPMI 1640 medium was chosen. A pooled protein extract with heavy labeled amino acids was applied as an internal standard, which included proteins expressed under control and stress condition, to control, BIP and BIP-iron-complex (BIP control experiment) samples. Samples were analyzed by liquid chromatography coupled directly to a tandem mass spectrometer. It is described that under iron-restricted conditions proteins associated to pathogenesis are higher abundant in pathogenic bacteria like Staphylococcus aureus. Hence, similar observations were expected also for the proteomic adaptation of S. pneumoniae, but the first results showed a reduction in protein abundance of virulence factors. In order to explain these results inductively-coupled-plasma mass spectrometry was executed to determine the iron concentration of chemically defined medium (CDM) used in this experiment. The analysis revealed a relatively low iron concentration of approximately 190 µg l-1. Therefore, the iron concentration of the complex medium THY, in which pneumococci are usually grown, was investigated. THY contains four-fold (740 µg l-1) more iron than the CDM. Subsequently, an additional iron limitation approach was carried out in THY. As SILAC is not applicable in complex media like THY, MaxLFQ was applied as quantification method in this case. Because two different media were used, an additional comparative proteome analysis with regard to the two investigated media was executed.
Comparing the protein composition in both cultivation media it became clear that pneumococci exhibit a totally different proteome depending on the medium. Major differences were found in metabolisms of amino acids, vitamins and cofactors as well as in pathogenesis-associated proteins. These differences have to be taken into account during the analyses of both iron limitation approaches. Overall, more proteins were identified and quantified in CDM samples. The pneumococcal adaptation to iron limitation in both media was different; especially, the alterations in protein abundances of virulence factors. In contrast to the iron limitation in CDM, proteins involved in pathogenesis were higher abundant under iron limitation in THY, which was the expected result. Because of proteomic changes of cell division and lipid metabolism involved proteins in iron-limited pneumococci in CDM, electron microscopic pictures were taken in order to proof cell morphology. The pictures showed an impaired cell division in iron-limited CDM, but not in THY medium. However, both datasets have similarities as well. Thus, the iron uptake protein PiuA is strongly increased in iron-restricted conditions and the abundance of the iron storage protein Dpr is significantly decreased in both datasets. Notably, PiuA and Dpr seem to have important roles during the pneumococcal adaptation to iron-restricted environments.
One the basis of these results, it could be shown that the proteomic response of pneumococci to iron limitation is strongly dependent to the initial iron concentration of the environment. Hence, pneumococci will adapt differently to varying niches and thus potential vaccine candidates should be expressed independently of the localization within the human host.
Formation of singly and doubly charged Arq+ and Tiq+ (q = 1,2) and of molecular Ar 2 +, ArTi+, and Ti 2 + ions in a direct current magnetron sputtering discharge with a Ti cathode and argon as working gas was investigated with the help of energy-resolved mass spectrometry. Measured ion energy distributions consist of low-energy and high-energy components resembling different formation processes. Intensities of Ar 2 + and ArTi+ dimer ions strongly increase with increasing gas pressure. Addition of oxygen gas leads to the formation of positively charged O+, O2 +, and TiO+ and of negatively charged O− and O2 - ions.
BK polyomavirus-associated haemorrhagic cystitis (BKHC) is a complication after allogeneic stem cell transplantation, which can occur in 5–60% of the cases. BK viruria alone can also occur in up to 100%. BKHC can lead to severe morbidity in stem cell-transplanted patients, but data about this disease is limited. Consequently, we conducted a prospective unicentric non-interventional trial on BKHC as well as BK viruria after first adult allogeneic stem cell transplantation with a follow-up time of 1 year after inpatient treatment. Between November 2013 and December 2015, we were able to include 40 adult patients with a mean age of 52.8 years. Twenty-seven (67.5%) of these patients were male and 13 (32.5%) were female. Acute myeloid leukaemia was the most frequent underlying disease (n = 15; 37.5%). Only 1 patient developed BKHC during inpatient treatment (n = 1; 2.5%), but BK viruria was frequent (n = 11; 27.5%) during inpatient treatment as well as in the follow-up time (n = 14; 35%). Interestingly, BK viruria was significantly associated with mucositis (p = 0.038) and number of transfused platelet concentrates (p = 0.001). This unexpected association will be discussed and needs further investigation.

Parodontitis wird als Risikofaktor für z.B. Diabetes mellitus Typ 2, kardiovaskuläre oder respiratorische Erkrankungen diskutiert. Auch ein Zusammenhang mit einer chronischen Nierenerkrankung (CKD) bzw. einer geringeren renalen Leistung ist beschrieben worden. Um die Evidenz letzterer, kaum mit longitudinalen Daten belegten These zu unterfüttern, untersucht diese Arbeit den Einfluss von Parodontitis auf eine erniedrigte Nierenfunktion anhand eines Untersuchungzeitraums von 11 Jahren. In einer populationsbasierten Kohortenstudie konnten dazu Daten (Baselinedaten (n=2297) und follow-up-Daten (n=1512) nach 11 Jahren der Study of Health in Pomerania (SHIP) in volladjustierten, linear und logistischen gemischten Modellen analysiert werden.
Parodontitis wurde über Sondierungstiefe (ST) und Attachmentverlust (AV) erfasst. Beide Parameter wurden jeweils als mittlerer Wert und als Prozentangabe bei Zahnflächen mit ≥3mm angegeben, um sowohl Schweregrad als auch Ausmaß der Entzündung Rechnung zu tragen. Die ST (≥4 mm) wurde zusätzlich als kumulativer Parameter angegeben.
Die Nierenfunktion wurde mit der geschätzten glomerulären Filtrationsrate (eGFR) und der Albumin/Kreatinin-Ratio des Urins (uACR) definiert. Dabei wurde die eGFR mit Hilfe des Kreatinins und des Cystatin C im Serum geschätzt (eGFR Kreatinin bzw. eGFR Kreatinin-CystatinC ) und eine Mikroalbuminurie mit uACR > 30 mg/g festgelegt. Im Ergebnis sank die eGFR Kreatinin-CystatinC in den 11 Jahren des Follow-ups durchschnittlich um 13,3 ml/min/1,73 m2 von 118,3 auf 105,0 ml/min/1,73 m2. Dies lässt auf eine insgesamt schlechtere Nierenfunktion innerhalb der Kohorte schließen. Jedoch zeigten longitudinale Analysen unter Verwendung von gemischten Modellen bei keiner der Parodontitisdefinitionen einen konsistenten signifikanten Zusammenhang mit der eGFR Kreatinin-CystatinC . Bezüglich der logtransformierten uACR traten einzelne signifikante Zusammenhänge auf. Diese konnten allerdings (multiple Imputationen) nicht bestätigt werden. Auch ein Zusammenhang mit Mikroalbuminurie konnte nicht festgestellt werden.
In dieser Studie konnte kein konsistent signifikanter Zusammenhang zwischen chronischer Parodontitis und einer verminderten Nierenfunktion festgestellt werden. Somit kann durch unsere Ergebnisse die Hypothese, dass Parodontitis ein Risikofaktor für CKD ist, nicht bestätigt werden.
Zusammenfassung
Zielsetzung: Ziel der vorliegenden experimentellen Studie war es, die Kontamination von Kurzinfusionssystemen an Zentralen Venenkathetern prospektiv zu untersuchen.
Methode: Die Probensammlung der Kurzinfusionssysteme erfolgte auf Intensivtherapiestationen. Es wurden ausschließlich Infusionssysteme zur Gabe von Kurzinfusionen verwendet. Infusionssysteme zur alleinigen Volumengabe wie E153 wurden von der Studie ausgeschlossen.
Beim Wechsel des Kurzinfusionssystems nach einer 72-stündigen Verwendungsdauer wurde die im System verbliebene Restlösung für 48 h bei 36 °C auf Blutagar zum Nachweis einer Kontamination kultiviert.
Ergebnisse: In einer von 616 Proben wurden 10 KbE Coagulase-negative Staphylokokken (CoNS) pro ml nachgewiesen.
Eine Kontrolltestung war bei dieser Probe zu Beginn der Studie nicht vorgesehen, sodass das getestete Kurzinfusionssystem als kontaminiert galt.
2 weitere Proben in der Anschlussuntersuchung wiesen wiederum 10 KbE CoNS pro ml auf, was bei einer Kontrolltestung mittels erneuter Kultivierung der Infusionslösung nicht bestätigt wurde, sodass der initiale Befund als Laborkontamination gewertet wurde. Damit liegt auch bei der kontaminierten Probe eine Laborkontamination (z.B. Hautflora) nahe, ohne die Durchführung einer Kontrolltestung konnte dies jedoch nicht bewiesen werden.
Schlussfolgerung: Infusionssysteme werden bei intermittierender Gabe von Kurzinfusionen mit Zwischenspülungen und einer Verwendungsdauer von 72h mikrobiell nicht kontaminiert. Obwohl das nur für aerob wachsende Bakterien nachgewiesen wurde, ist die Art der Entstehung einer Kontamination beim Umgang mit ISs für Aerobier und Anaerobier analog, so dass bei fehlendem Nachweis von Aerobiern kein Risiko einer Kontamination mit Anaerobiern anzunehmen ist. Damit können ISs bei intermittierender Gabe von Kurzinfusionen mit Zwischenspülungen bis zu 72 h verwendet werden, wodurch können Kosten eingespart werden.
Ziel der Arbeit war eine Analyse des Auftretens von Risikoclustern unter der Berücksichtigung der Risikoausprägung der Risikoverhaltensweisen Tabakrauchen, Alkoholkonsum, Übergewicht und geringe körperliche Aktivität in einer bevölkerungsrelevanten Stichprobe. Dafür wurde ausgehend von Literaturrecherchen ein Risikoscore entwickelt, anhand dessen Prävalenzdarstellungen sowie multinomiale logistische Regressionsanalysen für als besonders relevant erachtete Risikoscoreausprägungen erfolgten. Die Ergebnisse zeigten ein Dominieren von nicht und wenig riskanten Verhaltenskombinationen in der Gesamtbevölkerung sowie eine Häufung von Risikoverhaltensweisen bei Jüngeren und kürzer Ausgebildeten. Zudem bestand bei den Befragten mit geringerem Bildungsgrad eine hohe Varianz der Risikoverhaltenskombinationen. Als wichtigste Risikoverhaltenskombination mit erhöhter Risikoausprägung wurde das Vorliegen eines Bewegungsmangels kombiniert mit dem Konsum von Tabak festgestellt.
Kieferorthopädische Behandlungen mit individell gefertigten aktiven Elementen führen zu besseren Behandlungsergebnissen in kürzerer Behandlungszeit bei gleichzeitig geringeren Nebenwirkungen. Mit CAD-Programmen und FE-Simulationen werden individuelle kieferorthopädische Elemente entworfen, die schnell und kostengünstig durch die RapidPrototyping-Technologie und speziell den 3D-Druck produziert werden können. Diese Studie zeigt dabei, dass es möglich ist, Objekte z. B. in Form von Federn und Bögen mit vorberechneter Kraftentfaltung zu planen und zu drucken. Die Druckparameter und die Parameter der gewählten Simulation sind entscheidend für die Qualität und die Eigenschaften des gedruckten Objektes. In dieser Studie wurden zwei verschiedene Versuchsaufbauten entwickelt, welche die Kraftentfaltung der Elemente mit einem Gelsensor bzw. mit einer Wägezelle gemessen hat. Es wurden Versuchsobjekte im Design eines Expansionsbogens aus den Materialien PLA und PETG sowie Zugfedern/Elastikketten aus den Materialen TPU und Filaflex mit 3D-Druckern produziert.
Es wurde festgestellt, das Gelsensoren infolge fehlender Konstanz zur Messung von Kräften in der Zahnmedizin ungeeignet sind, während sich Wägezellen als geeignet erwiesen haben. Geplante/simulierte Kraftentwicklungen korrelierten mit r=0,995 bzw r=0,998 nach Pearson signifikant zu den Kraftentwicklungen von gedruckten Expansionsbögen aus den Materialien PLA und PETG. Weiterhin wurde für elastische Ketten/Federn aus TPU eine signifikante Korrelation mit r=0,980 von simulierter zu gemessener Kraft nachgewiesen. Das Material TPU war geeignet zur individuellen Produktion von elastischen Zugfedern als Ergänzung zu konventionellen elastischen Ketten, deren Kräfte nicht exakt planbar sind. Die Experimente zeigten, dass Langzeitbelastung und Aufbewahrung in Wasser zu geringen Kraftverlusten der TPU-Federn führten. Es wurde das Potential der Rapid-Prototyping-Technologie kombiniert mit FE-Simulationen veranschaulicht. Die ständige Weiterentwicklung dieser Technologie ermöglicht schnell und kostengünstig individuelle Behandlungselemente zu produzieren bei geringerer Nebenwirkungsrate, so dass diese Technik in der Kieferorthopädie zunehmend wichtig wird.
Vor dem Hintergrund des noch wenig erforschten RNA-Ladungstransfers, lag der Fokus der Arbeit auf die Etablierung eines Ladungstranfers innerhalb einer funktionellen RNA. Als Modellsystem diente dazu das HPAR2, ein FMN-abhängiges Aptazym, dessen strukturdynamische Funktionsweise noch nicht komplett verstanden ist. Dabei galt es zum einen innerhalb der funktionellen Aptamerdomäne einen Ladungstransport zu etablieren. Zum anderen musste eine geeignete Position innerhalb der Aptamerstruktur für die Einführung eines nukleophilen Linkers identifiziert und verifiziert werden, um postsynthetisch die Verknüpfung mit dem FMN zu ermöglichen. Zusätzlich wurde durch die Synthese verschiedener nukleosidischer Sonden die Anwendung spektroskopischer Methoden zur Untersuchung dynamische RNA-Funktionen ermöglicht. Dabei gelang es eine neue Strategie zur Einführung einer Spin-Sonde in eine RNA zu entwickeln. Des Weiteren gelang die Darstellung einer nukleosidischen PHIP-Sonde, die eine außergewöhnlich hohe Signalverstärkung zeigte. Um die Funktionskontrolle des Modellsystems über einen intramolekularen Elektronentransport zu ermöglichen, musste zunächst die Synthese eines dementsprechend Linker-modifizierten Adenosins erfolgen. Der Einbau dieses Linker-modifizierten Adenosins durch chemische Festphasensynthese lieferte zwei RNAs, die durch Hybridisierung mit entsprechenden Gegensträngen das FMN-Aptamer und das FMN-abhängige Modellsystem HPAR2 bilden. Der zweite Teil dieser Arbeit, der Vorbereitung eines Elektronentransfer-sensitiven Aptazyms, setzte die Bereitstellung eines nukleosidischen Elektronendonors voraus. Dafür erfolgte die Synthese und Charakterisierung zweier Pyren-modifizierte Uridinderivate. Die Charakterisierung beider Elektronendonoren durch optische Spektroskopie (Fluoreszenz und UV/Vis) resultierte in vielversprechenden Hinweisen, dass die Erzeugung eines Überschusselektrons nach Anregung mit Licht einer bestimmten Wellenlänge gelang. Der erfolgreiche Einbau des substituierten Pyren-Nukleosidderivates in fünf verschiedene Duplex- und sechs verschiedene Aptamerstrukturen und deren spektroskopische Charakterisierung erlaubte die Untersuchung des RNA-Ladungstransports. Der Nachweis eines Ladungstransfers gelang für beide Systeme über zwei unterschiedliche Methoden. Einerseits konnte der Ladungstransfer über Fluoreszenzspektroskopie nachgewiesen werden und andererseits gelang der Nachweis über die Degradierung des eingebauten Akzeptors. Diese Ergebnisse stellen den ersten Ladungstransfers durch eine nicht Watson-Crick gepaarte Nukleinsäurestruktur dar. Zudem ist dies die erste Demonstration eines Sequenz-abhängigen Ladungstransportes innerhalb einer RNA.
Hintergrund
Die chronisch venöse Insuffizienz (CVI) ist eines der häufigsten Krankheitsbilder weltweit.
Die Prävalenz der CVI steigt mit zunehmendem Alter und ist aufgrund von Schmerzen,
reduzierter körperlicher Funktion und Mobilität mit einer reduzierten Lebensqualität
verbunden. Symptomatische CVI mit Ulcus cruris venosum als Endstadium indiziert einen
phlebochirurgischen Eingriff um den venösen Reflux auszuschalten und somit eine
Ulcusabheilung zu fördern sowie ein Ulcusrezidiv zu vermeiden. Nach unserem
Kenntnisstand gibt es bisher keine Studie, die in einer Patientenkohorte offen-chirurgische
Eingriffe in Bezug auf die Komplikationsentwicklung, venöse Hämodynamik und
perioperative Beschwerdeentwicklung untersucht. Dies erfolgte in einem retrospektivem,
monozentrischem Studiendesign, an unserer Universitätsklinik in Greifswald.
Methode
Daten von n= 429 Patienten (467 Extremitäten), die einen operativen Eingriff am
Venensystem erhielten, wurden von 2009-2013 eingeschlossen. Art und Anzahl an
Komplikationen wurden auf einen möglichen Einfluss von demographischen Parametern, Co-
Morbiditäten und Medikation und therapieabhängigen Faktoren untersucht. Komplikationen
wurden in die Gruppen: no events (NE) = keine Komplikationen; neglectable adverse events
(NAE) = leichte Komplikationen; non-neglectable adverse events (NNAE) = mittelschwere
Komplikationen und serious adverse events (SAE) = schwere/ lebensbedrohliche
Komplikationen unterteilt.
CVI-bedingte Beschwerden wurden mit Hilfe eines Fragebogens sowie der Dokumentation
zu den Verlaufskontrollen analysiert.
Bei allen Patienten wurden die prä- sowie postoperative Venenfunktion evaluiert, hierbei
wurde zusätzlich eine Subgruppenanalyse hinsichtlich der Komplikationen durchgeführt.
Ergebnisse
In 130 (27,84%) Extremitäten von 467 (von 429 Patienten) wurden postoperative
Komplikationen observiert, hierunter 64 (13,7%) NAE, 66 (14,14%) NNAE and 0 SAE. 29
(6,76%) Patienten entwickelten eine postoperative Wundinfektion (SSI), wobei 4 dieser Patienten (0,9%) eine Sepsis erlitten. Abgesehen von einem Fall mit intraoperativer
Nervenschädigung, die über den Beobachtungszeitraum hinaus bestehen blieb, waren alle
Komplikationen innerhalb von 12 Monaten regredient. Patienten mit mittelschweren
Komplikationen (NAE) hatten einen höheren BMI (p = 0.003), eine höhere
Wahrscheinlichkeit an einem Diabetes mellitus Typ II zu leiden (p <0.001) und häufiger Co-
Morbiditäten, die zur Einnahme von thrombozytenhemmender oder gerinnungshemmender
Medikation führen (p < 0.001). Das metabolische Syndrom als auch die Einnahme von
thrombozytenhemmender oder gerinnungshemmender Medikation konnte als unabhängiger
Risikofaktor für die Komplikationsentwicklung analysiert werden. Patienten, die eine
Stripping-Operation der Vena saphena magna erhielten, hatten ein 8-fach höheres Risiko eine
Muskelvenenthrombose zu entwickeln. Eine Stripping-Operation bis zum distalen
Insuffizenzpunkt war mit einem erhöhten Risiko für postoperative Sensibilitätsstörungen
verbunden. Postoperativ zeigten sich Beschwerden wie Schmerzen (p < 0.001), Schwellung (p
< 0.001) und Juckreiz (p = 0.003) signifikant gebessert. Ungeachtet der
Komplikationsentwicklung zeigte sich eine signifikante Besserung der Venenfunktion nach 6-
Wochen und einem Jahr postoperativ (p < 0.05). Eine Besserung der Venenfunktion ging
einher mit einem Beschwerderückgang.
Schlussfolgerung
Patienten mit metabolischem Syndrom oder einer thrombozytenehmmenden bzw.
gerinnungshemmenden Therapie sollten einen operativen Eingriff unter stationären
Bedingungen erhalten und regelmäßig verlaufskontrolliert werden. Insbesondere Patienten mit
Stripping-Operation der Vena saphena parva sollten eine postoperative Heparinprophylaxe
erhalten. Eine Stripping-Operation bis zum distalen Insuffizienzpunkt war mit einer höheren
Rate an postoperativen Sensibilitätsstörungen verbunden, diese zeigte sich in unseren
Patienten in fast allen Fällen innerhalb von 12 Monaten rückläufig. Ungeachtet der
Komplikationsentwicklung profitierten die Patienten von einem Eingriff da sich sowohl die
Venenfunktion als auch Beschwerden besserten.
Zweck: Bestimmung Langzeitüberleben nach LITT hepatischer Metastasen kolorektaler Karzinome und das Outcome beeinflussende Faktoren. Besonders hervorzuheben ist die einzigartige Vergleichbarkeit auf Grund eines durchführenden Interventionalisten. Abschließend wurde hinterfragt, ob eine LITT weiterhin eine palliative Therapieoption ist, oder eventuell parallel zum derzeitigen Goldstandard der operativen Resektion eingesetzt werden könnte.
Material und Methoden: Diese retrospektive Studie umfasste 119 Patienten. Die Behandlungen via LITT fanden von 07/97 bis 10/13 statt. 95 Patienten waren männlich, 24 Patienten weiblich. Der Hauptanteil der Patienten war zwischen 61 und 70 Jahre alt. Betrachtet wurden das Langzeitüberleben nach LITT in Monaten, sowie beeinflussende Faktoren darauf. Hierbei wurden in beeinflussbare Faktoren (Applikatorenzahl, Leistung, Zeit der Leistung und Gesamtmenge der LITT), sowie unbeeinflussbare Faktoren (Alter, Geschlecht, Komorbiditäten nach ASA und synchrone/metachrone Metastasen) unterschieden. Zum Zeitpunkt der Datenabfrage waren 110 Patienten (92,4%) verstorben. Zur statistischen Auswertung der Überlebenszeitanalyse wurde das kaplan- Meier- Verfahren genutzt.
Ergebnisse: Die Mittlere Überlebenszeit betrug 36 Monate (Median 27 Monate), die 1-, 3- und 5- Jahresüberlebensrate (JÜR) betrug 80,67%, 33,61% und 11,76%. Statistisch signifikant waren Einflüsse auf die Mittlere Überlebenszeit bei den unterschiedlichen Komorbiditäten nach ASA, zwischen Patienten mit und ohne extrahepatische Metastasen, zwischen den unterschiedlichen Wattzahlen und hinsichtlich der Gesamtmenge der Behandlungen via LITT.
Die höchste Mittlere Überlebenszeit zeigten Patienten der ASA- Klasse 1 (66,3 Monate), gefolgt von ASA 2 (38,3 Monate), ASA 3 (219 Monate) und ASA 4 (19,3 Monate). Die höchste 1 JÜR zeigte sich bei Patienten mit ASA 2 (87,5%), gefolgt von ASA 1 (76,5%), ASA 3 (66,7%) und ASA 4 (33,33%). Die 3- JÜR zeigte sich wie folgt: ASA 1 58,8%, ASA 2(31,3%, ASA 3 19% und ASA 4 33,33%). Die 5- JÜR zeigte sich in ASA 1 mit 35,3%, ASA 2 mit 17,9%, ASA 3 mit 4,76% und ASA 4 mit 0%.
Patienten ohne extrahepatische Metastasen überlebten im Mittel 56,55 Monate nach LIT (Median 33 Monate). Der Anteil Überlebender bei Abfrage betrug 24%. Die 1-, 3- und 5- JÜR waren 80%, 48% und 24%. Patienten mit extrahepatischen Metastasen überlebten im Mittel nach LITT 32,67 Monate (Median 24 Monate). Der Anteil der Überlebenden betrug 6,7%. Die 1-, 3- und 5- JÜR waren 75,5%, 24,4% und 14,8%.
Unterschiede im Outcome bestanden auch hinsichtlich der Wattzahl. Patienten, bei denen Leistungen unter 14 Watt eingesetzt wurden lebten nach LITT im Mittel 13,36 Monate. Die 1-, 3- und 5- JÜR betrug 50%, 7,1% und 0%.
Patienten bei denen Leistungen von 14 Watt verwendet wurden lebten nach LITT im Mittel 40,26 Monate (Median 28 Monate). Hier betrug die 1-, 3- und 5- JÜR 77,8%, 28,9% und 15,5%. Patienten mit Leistungen über 14 Watt lebten im Mittel 37,33 Monate (Median 30 Monate). Hier lagen die 1-, 3- und 5- JÜR bei 88,7%, 41,5% und 127%.
Patienten mit über 5 LITT´s lebten mit 56 Monaten Überlebenszeit im Median am längsten nach LITT. Die 1-, 3- und 5- JÜR betrug 75%, 75% und 50%. Patienten mit 5 LITT´s lebten im Median nach LITT 37 Monate. Hier lag die 1-, 3- und 5- JÜR bei 85,7%, 57,1% und 0%. Patienten mit 4 Ablationen lebten im Median 16 Monate nach LITT und die 1-, 3-und 5- JÜR lag bei 53,8%, 23,1% und 0%. Bei 3 Interventionen lebten Behandelte nach Therapie im Median 29 Monate und die 1-, 3- und 5- JÜR lag bei 92,9%, 28,6% und 7,1%. Bei 2 LITT´s überlebten Patienten im Median26 Monate und die 1-, 3- und 5- JÜR betrug 85,3%, 26,5% und 13,7%. Bei 1 LITT betrug die mediane Überlebenszeit 27 Monate, die 1-, 3-und 5- JÜR 78,7%, 34% und 16,8%. Die anderen untersuchten Ergebnisse zeigen Trends, waren jedoch nicht statistisch signifikant.
Schlussfolgerung: Weniger schwere Komorbiditäten bedingen eine längere Überlebenszeit nach LITT. Sind zusätzliche extrahepatische Metastasen vorhanden, so verkürzt sich die Überlebenszeit nach LITT. Angewendete Leistungen unter 14 Watt führen ebenfalls zu kürzeren Überlebenszeiten. Es sollten also nur Leistungen von größer oder gleich 14 Watt angewendet werden. Eine multifokale hepatische Metastasierung ist keine Kontraindikation für eine LITT, Re-Ablationen sind möglich- auch ohne Verkürzung der Überlebenszeit. Die Durchführung durch nur einen Behandler stellt keinen Überlebenszeitvorteil dar, generiert jedoch eine einzigartige Vergleichbarkeit der Ergebnisse. Das Outcome der LITT in dieser Studie ist vergleichbar mit in der Literaturrecherche zu findenden Vorstudien. Sie kann als Alternative zur operativen Therapie angewendet werden.
The site at the southern shore of Krakower See shows the Quaternary geology of the surrounding
area. The local Quaternary sequence comprises a thickness of 50–100m of Quaternary deposits while
the surface morphology is dominated by the ice marginal position of the Pomeranian moraine, which
passes through the area. The bathymetry of the lake basin of Krakower See indicates a predominant
genesis by glaciofluvial erosion in combination with glacial exaration. Past research in this area has focussed
on the reconstruction of Pleniglacial to Holocene environmental changes, including lake-level
fluctuations, aeolian dynamics, and pedological processes and their modification by anthropogenic
land use.
Lipohypertrophien (LHT) sind Veränderungen des subkutanen Fettgewebes, die durch die Langzeitanwendung von Insulin bei ca. 40 % der Patienten entstehen. Sie waren bisher von fraglicher klinischer Relevanz und teilweise schwierig zu diagnostizieren. Die Veröffentlichungen im Rahmen dieser kumulativen Dissertation untersuchten die pharmakotherapeutische Bedeutung von Lipohypertrophien und untersuchten erstmals die Thermographie als diagnostische Option für LHT. Dabei zeigte sich im euglykämischen Clamp-Versuch eine signifikant reduzierte Bioverfügbarkeit des Insulins nach Injektion in LHT von 78-80 % im Vergleich zur Applikation in gesundes Gewebe. Zwei der dreizehn Probanden zeigten keine Insulinwirkung. Die intraindividuelle Variabilität war signifikant höher in LHT als im gesunden Gewebe. Bei der erstmaligen systematischen Untersuchung von LHT mit einer hochwertigen Wärmebildkamera zeigte sich eine mäßige diagnostische Güte, verglichen mit den ebenfalls teilweise schwierigen Methoden der Inspektion, Palpation und des Ultraschalls. Insbesondere in der Kontrollgruppe war eine klare Abgrenzung von gesundem Gewebe schwierig. Aufgrund der Beeinflussung der Sicherheit, Effektivität und der Effizienz der Pharmakotherapie durch LHT, sollten diese als Injektionsort vermieden werden. Diese Arbeit konnte den Nutzen der Thermographie als diagnostische Option bei LHT zwar nicht klar beweisen, jedoch diesen auch nicht ausschließen.

This thesis describes mass measurements at ISOLTRAP/ISOLDE/CERN in the region of the neutron-rich calcium isotopes. For the less exotic and more abundantly produced isotopes 51Ca and 52Ca the Penning trap based ToF-ICR technique could be used to validate the available mass data and to improve their precision. For the isotopes 53Ca and 54Ca, a Multi-Reflection Time-of-Flight Mass Spectrometer (MR-ToF MS) was used to determine the mass of these exotic isotopes for the first time experimentally. This also represents the first time an MR-ToF MS was applied to derive the masses of previously unknown radioactive ions from the high precision time-of-flight data that can be gathered with the device. The mass data was then used to benchmark the strength of the N=32 neutron subshell closure and at the same time to compare to state-of-the-art shell-model calculations.
Furthermore, the capability of the MR-ToF device to deliver isobarically pure beams to a subsequent experiment was developed further and studied in detail. The new technique is based on the in-trap lift, which is normally used to in- and eject ions into and from the device. With this new selective ejection technique after separation of the ion ensemble in the MR-ToF trap, no external components are required.
Additionally, a new stabilization system for voltages supplies, based on a PI-algorithm, was developed and thoroughly tested. The stabilized voltage supply was then used to supply the most sensitive mirror voltage of the MR-ToF MS to significantly increase the short term and long-term mass resolving power of the apparatus.
Bats (Chiroptera) form the second largest order of mammals and with over 1,250 species, they represent about 20% of all mammalian species worldwide. They are the only mammals with true and sustained flight and distributed all over the world except the arctic regions. Moreover, bats entered specific ecological niches and with their food spectra, they reduce different arthropod populations as well as disperse seeds and pollen of plant species in various regions and habitats.
Bats also have a crucial role in spreading high-pathogenic and zoonotic viruses, harbor in general more viruses (zoonotic and non-zoonotic), and, related to the species, number even more than rodents. However, clinical symptoms of viral diseases are rarely reported in bat communities. Also seroconversions after infection were not reported for a variety of viruses found in bats. Since the incidence of virus-positive bats estimated in passive surveillance studies is usually very low, it is a question how such viruses can use bats as reservoir hosts. There is obviously a special evolutionary relationship between the pathogens and bats as hosts, which are based on possibly physiologic adaptations also in resistance and immunity.
In this thesis, the two lyssaviruses, European Bat Lyssavirus 1 and 2 (EBLV-1 and -2) were chosen as a model to investigate the immune response of European bats against viral infection in vitro. Lyssaviruses are the causative agents of rabies, a fatal zoonotic disease with neurotropic characteristics.
One main question to investigate was in which way bats act as reservoir host and developed a high disease resistance. The present thesis is based on three hypotheses about innate immune response against lyssavirus infection:
A) In bats specific peripheral resistance mechanisms evolved which reduce the risk of systemic viral infection after a hypothesized airborne transmission and infection via nasal epithelium supported by the social structure of and communication within bat communities.
B) The co-evolution of EBLV and the innate resistance of bats resulted in a very effective type I interferon response to inhibit a systemic lyssavirus infection.
C) The specific physiology of body temperature of bats with daily torpor depresses the viral replication but favours the type I interferon response.
To analyze the interferon-based resistance mechanisms, the type I interferon (IFN) genes of two European bats species (Eptesicus serotinus and Myotis myotis) were cloned and sequenced. Using established cell lines from the respiratory nasal epithelium (MmNep), olfactory nasal epithelium (MmNol), and Bulbus olfactorius brain (MmBr), the type I IFN response along a possible airborne infection route was investigated. The anti-viral effects and induction of IFNs/interferon stimulated genes (ISGs) in each cell line were also investigated in detail after infection in vitro. Finally, the influence of different temperatures on lyssavirus replication was analyzed in cell culture experiments.
The results indicated that (a) along the hypothesized airborne infection route the susceptibility for lyssavirus infections is decreased, (b) the type I IFN activity in contrast is increased contributing to a limitation of lyssavirus replication and (c) an obvious influences of varying cultivation temperatures on the resistance against lyssavirus infections, which favor the IFN response and repressing lyssavirus replication.
The result from these in vitro studies supports the hypothesis of a special co-evolution between lyssaviruses and bats. However, in vivo studies on the relevance in infected animals are missing so far. This model could also explain the generally limited pathogenicity of bat-associated viruses.
Die Dissertation leistet einen Beitrag zur Analyse des intensionalen semantischen Vokabulars von Gebrauchssprachen und formalen Sprachen. Die Frage danach, wann Ausdrücke sinnvoll sind bzw. eine Bedeutung haben, ist eine altehrwürdige. Vor allem in den letzten beiden Jahrhunderten haben Philosophen wie Frege, Russell, Wittgenstein, Carnap und Quine versucht, Antworten auf sie zu finden. In der vorliegenden Schrift wird eine konsistente Menge von Prinzipien des Sinnvollseins herausgearbeitet, die in semantischen Theorien eingesetzt werden könnte. Dafür werden die folgenden Schritte unternommen:
• Bewertung und kritische Abwägung von einschlägigen Intuitionen,
• Präzisierung und Kritik der Vorschläge von logischen Empiristen,
• Würdigung von Vorschlägen jüngeren Datums,
• Begründung aller zentralen Behauptungen in einem formalen Rahmen,
• Untersuchung von Sinnlosigkeitsbehauptungen anhand von Beispielen.
Salivary glands provide secretory functions, including secretion of xenobiotics and among
them drugs. However, there is no published information about protein abundance of drug transporters
measured using reliable protein quantification methods. Therefore, mRNA expression and absolute
protein content of clinically relevant ABC (n = 6) and SLC (n = 15) family member transporters in the
human parotid gland, using the qRT-PCR and liquid chromatography-tandem mass spectrometry
(LC−MS/MS) method, were studied. The abundance of nearly all measured proteins ranged between
0.04 and 0.45 pmol/mg (OCT3 > MRP1 > PEPT2 > MRP4 > MATE1 > BCRP). mRNAs of ABCB1,
ABCC2, ABCC3, SLC10A1, SLC10A2, SLC22A1, SLC22A5, SLC22A6, SLC22A7, SLC22A8, SLCO1A2,
SLCO1B1, SLCO1B3 and SLCO2B1 were not detected. The present study provides, for the first time,
information about the protein abundance of membrane transporters in the human parotid gland,
which could further be used to define salivary bidirectional transport (absorption and secretion)
mechanisms of endogenous compounds and xenobiotics.
Patient-reported outcomes (PROs) refer to any report coming directly from patients about how they function or feel in relation to a health condition or its therapy. PROs have been applied in medicine for the assessment of the impact of clinical phenomena. Self-report scales and procedures for assessing physical pain in adults have been developed and used in clinical trials. However, insufficient attention has been dedicated to the assessment of mental pain. The aim of this paper is to outline the implications that assessment of mental pain may entail in psychiatry and medicine, with particular reference to a clinimetric index. A simple 10-item self-rating questionnaire, the Mental Pain Questionnaire (MPQ), encompasses the specific clinical features of mental pain and shows good clinimetric properties (i.e., sensitivity, discriminant and incremental validity). The preliminary data suggest that the MPQ may qualify as a PRO measure to be included in clinical trials. Assessment of mental pain may have important clinical implications in intervention research, both in psychopharmacology and psychotherapy. The transdiagnostic features of mental pain are supported by its association with a number of psychiatric disorders, such as depression, anxiety, eating disorders, as well as borderline personality disorder. Further, addressing mental pain may be an important pathway to prevent and diminish the opioid epidemic. The data summarized here indicate that mental pain can be incorporated into current psychiatric assessment and included as a PRO measure in treatment outcome studies.
Abstract
Metabolomics studies now approach large sample sizes and the health characterization of the study population often include complete blood count (CBC) results. Upon careful interpretation the CBC aids diagnosis and provides insight into the health status of the patient within a clinical setting. Uncovering metabolic signatures associated with parameters of the CBC in apparently healthy individuals may facilitate interpretation of metabolomics studies in general and related to diseases. For this purpose 879 subjects from the population‐based Study of Health in Pomerania (SHIP)‐TREND were included. Using metabolomics data resulting from mass‐spectrometry based measurements in plasma samples associations of specific CBC parameters with metabolites were determined by linear regression models. In total, 118 metabolites significantly associated with at least one of the CBC parameters. Strongest associations were observed with metabolites of heme degradation and energy production/consumption. Inverse association seen with mean corpuscular volume and mean corpuscular haemoglobin comprised metabolites potentially related to kidney function. The presently identified metabolic signatures are likely derived from the general function and formation/elimination of blood cells. The wealth of associated metabolites strongly argues to consider CBC in the interpretation of metabolomics studies, in particular if mutual effects on those parameters by the disease of interest are known.
Die vorliegende Studie ist die erste, die den breiten Effekt von IGF-I auf den menschlichen Metabolismus abbildet. Es zeigt sich ein facettenreiches Bild aus IGF-I assoziierten Metaboliten in Plasma und Urin, das die vielfältigen biologischen Effekte von IGF-I repräsentiert. Ein besonderer Befund ist die große Diskrepanz zwischen Frauen und Männern in den metabolischen Profilen von IGF-I, die eine Verbindung zu geschlechtsspezifischen Assoziationen zwischen IGF-I und bevölkerungsrelevanten Erkrankungen darstellen könnte. Zudem konnte der Zusammenhang von IGF-I mit dem Lipidstoffwechsel sowie Peptiden und Aminosäuren bestätigt werden. Für einige dieser Assoziationen gibt es nach aktuellem Forschungsstand bereits molekulare zellbiologische Erklärungsansätze. Viele der detektierten Metaboliten lassen sich in den Zusammenhang zu IGF-I assoziierten Erkrankungen einordnen: beispielsweise Betaine und Cortisol mit kardiovaskulären Erkrankungen, Diabetes, Dyslipiämie und dem Metabolischen Syndrom. Bradykinin und einige Fettsäurederivate sowie Cortisol verbinden IGF-I mit inflammatorischen Prozessen, ihre inverse Assoziation mit IGF-I kann unter anderem zur Erklärung des Zusammenhangs mit endothelialen Entzündungsprozessen wie der Atherosklerose beitragen. Für andere Metabolite ergeben sich Assoziationen zu Wachstum und Zelldifferenzierung, darunter Phospholipide sowie Aminosäure- bzw. Peptidabkömmlinge. Unsere Daten bestätigen den vielfältigen Einfluss von IGF-I auf den menschlichen Metabolismus, wie aus vorherigen Experimentalstudien beschrieben. Die vorliegenden Ergebnisse aus relativ gesunden Probanden erlaubt die Identifikation von IGF-I assoziierten Biomarkern. Warum sich nur für einige spezifische Repräsentanten pro Stoffgruppe signifikante Assoziationen ergeben und welche spezifischen molekularen zellbiologischen Prozesse dem zugrunde liegen, kann hier nicht allumfassend beantwortet werden. Zur weiteren Überprüfung der generierten Hypothesen, zur Klärung der genauen pathophysiologischen Auswirkungen von IGF-I auf den Metabolismus und auch zur Findung neuer Diagnose- und Therapiekonzepte für IGF-I assoziierte Erkrankungen, sind weitere unabhängige, interventionelle und experimentelle Studien erforderlich.
Beim Endometriumkarzinom (EC) bieten sich gemäß aktueller Leitlinie (AWMF) wenige Möglichkeiten einer Chemotherapie. Tumormarker und targeted therapy rücken daher in den Fokus. In diesem Zuge erlangen molekulare Signalkaskaden und deren Modulatoren einen wachsenden Stellenwert. Die PI3-Kinase/AKT-Signalkaskade mit ihrem Regulator MikroRNA 21 (miR-21) ist in vielen Tumorentitäten von Mutationen betroffen und bietet dabei verschiedene Angriffspunkte. Ziel dieser Arbeit war es, die Signalkaskade im EC darzustellen und die regulatorische Rolle von miR-21 näher zu charakterisieren.
Um eine möglichst umfassende Aussage bezüglich der Tumorentität EC zu treffen, wurde je eine Zelllinie vom prognostisch günstigen Typ I und ungünstigen Typ II nach der Klassifikation von Bokhman (1983) ausgewählt. Die Zelllinien wurden hinsichtlich der Morphologie und ihrer molekularen Eigenschaften charakterisiert. Mittels Transfektion von pmiR-21 wurde in den EC-Zellen miR 21 überexprimiert und die Proteinexpressionen der PI3K/AKT-Signalkaskade nach 6 96 h mittels Gelelektrophorese und anschließendem Western Blot analysiert. In der Zelllinie MFE 296 kam es zu den erwartenden Veränderungen der Signalkaskade. Die Expression der Phosphatase PTEN wurde inhibiert und der Zellzyklus enthemmt. Die Zelllinie MFE 280 zeigte keine Veränderungen nach Transfektion. Ursachen sind vermutlich die unterschiedliche PTEN-Aktivität aufgrund von Mutationen und andere Regulationsmechanismen. miR-21 greift neben der direkten Inhibition von PTEN posttranskriptionell indirekt in die Stabilität des Proteins ein. p85α, die regulatorische Untereinheit der PI3K, ist ein weiteres Target von miR-21. Neben der Interaktion mit den katalytischen Untereinheiten kann p85α Dimere mit PTEN bilden und dessen Ubiquitinierung reduzieren. Die Wirkmechanismen von miR-21 sind vielfältig und die genauen Zusammenhänge bisher nicht komplett verstanden.
Diese Arbeit stellt die unterschiedliche Gewichtung von miR-21-Modulationen auf die PI3K/AKT-Signalkaskade in den beiden EC-Typen dar. miR-21 kann im EC-Typ-I als Marker fungieren. Außerdem wurden einige Möglichkeiten der targeted therapy in der PI3K/AKT-Signalkaskade angedeutet. Um diese Aussage und die therapeutischen Konsequenzen in den klinischen Alltag zu integrieren, bedarf es weiterführender Arbeiten.
