Overexpression of polo-like kinase 1 (PLK1) has been found in many different types of cancers. With its essential role in cell proliferation, PLK1 has been determined to be a broad-spectrum anti-cancer target. In this study, 3D-QSAR, molecular docking, and molecular dynamics (MD) simulations were applied on a series of novel pteridinone derivatives as PLK1 inhibitors to discover anti-cancer drug candidates. In this work, three models—CoMFA (Q² = 0.67, R² = 0.992), CoMSIA/SHE (Q² = 0.69, R² = 0.974), and CoMSIA/SEAH (Q² = 0.66, R² = 0.975)—of pteridinone derivatives were established. The three models that were established gave R²(pred) = 0.683, R²(pred) = 0.758, and R²(pred) = 0.767, respectively. Thus, the predictive abilities of the three proposed models were successfully evaluated. The relations between the different champs and activities were well-demonstrated by the contour chart of the CoMFA and CoMSIA/SEAH models. The results of molecular docking indicated that residues R136, R57, Y133, L69, L82, and Y139 were the active sites of the PLK1 protein (PDB code: 2RKU), in which the more active ligands can inhibit the enzyme of PLK1. The results of the molecular dynamic MD simulation diagram were obtained to reinforce the previous molecular docking results, which showed that both inhibitors remained stable in the active sites of the PLK1 protein (PDB code: 2RKU) for 50 ns. Finally, a check of the ADME-Tox properties of the two most active molecules showed that molecular N° 28 could represent a good drug candidate for the therapy of prostate cancer diseases.
Cyanobakterien sind eine vielversprechende Quelle an strukturell diversen und biologisch hochaktiven Naturstoffen für die Entwicklung neuer Wirkstoffe. Bislang konnte die Strukturklasse der [7.7]Paracyclophane nur in fädigen Cyanobakterien der Gattungen Nostoc und Cylindrospermum nachgewiesen werden. Vorangegangene Arbeiten zeigten, dass gerade die Carbamidocyclophane chemisch und biologisch interessante Verbindungen darstellen. Im Rahmen dieser Arbeit wurden vor allem die Carbamidocyclophane produzierenden Cyanobakterien Nostoc sp. CAVN2 und Nostoc sp. CAVN10 unter besonderer Berücksichtigung der strukturellen Vielfalt an biosynthetisierten Metaboliten sowie deren antimikrobieller Aktivität umfassend charakterisiert. Um das biosynthetische Potenzial auf der metabolischen Ebene zu untersuchen, wurde im Vorfeld eine spezifische [7.7]Paracyclophan-Analytik etabliert, die skalierbare Methoden für alle Aufarbeitungsschritte beinhaltet. Die Optimierung endete in einem validierten sowie arbeits- und zeitsparenden einstufigen Extraktions- und Aufreinigungsverfahren mittels eines Zweiphasensystems und anschließender LC-UV-Analyse, um biologische Proben reproduzierbar zu analysieren und enthaltene Carbamidocyclophane zu quantifizieren. Kultivierungsstudien zum Einfluss der Temperatur an metabolisch aktiven und defizienten Nostoc-sp.-CAVN10-Kulturen ergaben einen direkten Zusammenhang zwischen der Biomassezunahme und der Temperaturerhöhung. Im Gegensatz dazu zeigten die einzelnen Carbamidocyclophan-Gehalte ein eher differenzierteres Bild über die verschiedenen Wachstumsphasen und Temperaturen hinweg. Da nur eine geringe Korrelation zwischen der spezifischen Wachstumsrate und der spezifischen Carbamidocyclophan-Produktionsrate ermittelt werden konnte, ist eine Relevanz dieser Verbindungen für den primären Zellstoffwechsel nicht ersichtlich. Bei Kultivierungsexperimenten an Nostoc sp. CAVN2 hatte der Zusatz von Chlorid- oder Bromid-Ionen eine drastische Erhöhung der Basalrate und Strukturdiversität der [7.7]Paracyclophane zur Folge. Das gleichzeitige Vorhandensein beider Halogenide im Medium zeigte kompetitive Effekte, wobei Chlorid als Substrat für den Halogenierungsprozess favorisiert wurde. Mit Hilfe eigens entwickelter Kultivierungsprozedere und Separierungsstrategien konnten insgesamt 25 Verbindungen aus Stamm CAVN2 isoliert und strukturell aufgeklärt werden. Dabei bilden die Carbamidocyclophane H–U neue chlorierte, bromierte und nicht halogenierte Naturstoffe. Zusätzlich konnten aus Stamm Cylindrospermum stagnale PCC 7417 neben den bekannten Cylindrocyclophanen A, B und D die drei neuen Cylindrofridine A–C erhalten werden. Diese stellen den Cylindrocyclophanen strukturell eng verwandte lineare Mono- und Dialkylresorcinole dar. Die vergleichende Evaluierung der Bioaktivität von 30 Reinsubstanzen ergab, dass viele Verbindungen sehr starke antimikrobielle Aktivität gegen grampositive Bakterien zeigen – besonders gegen Antibiotika-resistente Kokken mit minimalen Hemmkonzentrationen von oftmals deutlich unter 1 µM. Dabei bedingten die verschiedenen Substituenten (Carbamoyl- und Acetoxy-Reste sowie Hydroxygruppen oder Halogene) z.T. signifikante Aktivitätsunterschiede. Die Zytotoxizität der [7.7]Paracyclophane ist vor allem an das Vorhandensein des Makrozyklus gebunden, da lineare Derivate (Cylindrofridine B/C) kaum biologisch aktiv waren. Eine Ausnahme stellt dabei das nicht zytotoxische, aber antimikrobiell aktive Cylindrocyclophan-D-Monomer Cylindrofridin A dar. Die phylogenetische Analyse der 16S-rDNA-Daten bestätigte die morphologisch-taxonomische Identifizierung der Stämme CAVN2 und CAVN10 als Cyanobakterien der Gattung Nostoc und ergab weiterhin, dass alle Carbamido- und Cylindrocyclophane produzierenden Nostoc-Stämme Bestandteil einer monophyletischen Gruppe sind, die phylogenetisch distinkt zu anderen [7.7]Paracyclophan-Produzenten ist. Des Weiteren konnten keine Nukleotidunterschiede zwischen Stamm CAVN2 und CAVN10 auf den untersuchten Markergen-Sequenzen festgestellt werden, was beide auf der phylogenetischen Ebene als identisch erscheinen lässt und sie somit nur metabolisch aufgrund der strukturellen Diversität und Quantität an [7.7]Paracyclophanen differenzierbar sind. Mit Hilfe von molekulargenetischen Analyseverfahren und bioinformatorischer Auswertung konnte in Stamm CAVN2 das Carbamidocyclophan-Biosynthesegencluster mit einer Gesamtgröße von ca. 26,9 kbp identifiziert werden. Dieses beinhaltet 13 offene Leserahmen (cabA-cabM), wobei das Gen cabL für eine putative Carbamoyltransferase codiert. Ein neuer Halogenase-Typ in Verbindung mit einer Tandem-ACP-Domänen-Struktur in der Typ I Polyketidsynthase CabD könnte für die Ausbildung halogenierter Derivate verantwortlich sein. Der Nachweis eines codierenden Bereichs mit Rieske-Domäne (cabM) lässt eine direkte oxidative intermolekulare Makrozyklisierung bei der Assemblierung vermuten.
Pentathiepins are cyclic polysulfides that exert antiproliferative and cytotoxic activity in cancer cells, induce oxidative stress and apoptosis, and potently inhibit GPx1. These properties render this class of compounds promising candidates for the development of anticancer drugs. However, the biological effects and how they intertwine to promote high cytotoxicity have not been systematically assessed throughout a panel of cancer cell lines from distinct tissues of origin. In this thesis, six novel pentathiepins were analyzed and constitute the second generation of compounds with additional properties such as fluorescence or improved water solubility to facilitate cellular testing. All compounds underwent extensive biological evaluation in 14 human cancer cell lines. These studies included investigations of the inhibitory potential with regards to GPx1 and cell proliferation, examined the cytotoxicity in human cancer cell lines, as well as the induction of oxidative stress and DNA strand breaks. Furthermore, selected hallmarks of apoptosis, ferroptosis, and autophagy were studied. Experimental approaches regarding these cellular mechanisms included observing morphological changes, detecting phosphatidyl serine exposure and caspase activity, and quantifying cleaved PARP1 and levels of LC3B II. In addition, the analysis of the cell cycle aimed to identify aberrations or arrests in cell division.
Five of the six tested pentathiepins proved to be potent inhibitors of the GPx1, while all six exerted high cytotoxic and antiproliferative activity, although to different extents. There was a clear connection observed between the potential to provoke oxidative stress and damage to DNA in the form of single- and double-strand breaks both extra- and intracellularly. Furthermore, various experiments supported apoptosis but not ferroptosis as the mechanism of cell death in four different cell lines. In particular, the externalization of PS, the detection of activated caspases, and the cleavage of PARP1 corroborated this conclusion. Additionally, indications for autophagy were found, but more investigations are required to verify the current data. The findings of this dissertation are mainly in line with the postulated mechanism of action proposed for pentathiepins and a previous publication from our group that described their biological activity. However, the influence of modulators such as oxygen and GSH on the biological effects was ambiguous and dependent on the compound. The expression profile of the cell lines concerning GPx1 and CAT did not influence the cellular response toward the treatment, whereas the cell doubling time correlated with the cytotoxicity.
As the various pentathiepins give rise to different biological responses, modulation of the biological effects depends on the distinct chemical structures fused to the sulfur ring. This may allow for future optimization of the anticancer activity of pentathiepins. An analysis of the structure-activity relationships revealed that the piperazine scaffold was associated with superior biological activity compared to the pyrrolo-pyrazine backbone. Furthermore, substituents with electron-withdrawing properties or those providing a free electron pair, such as fluorine or morpholine, were advantageous. These findings should help design and synthesize the next generation of pentathiepins, thereby expanding the library of compounds, allowing for the further deduction of structure-activity relationships and an improved understanding of their mechanism of action.
For the characterization of Kv7.2/3 channel activators, several analytical methods are available that vary in effort and cost. In addition to the technically elaborate patch-clamp method, which serves as a reference method, there exist several medium to high-throughput screening methods including a rubidium efflux flame-atomic absorption spectrometry (F-AAS) assay and a commercial thallium uptake fluorescence-based assay. In this study, the general suitability of a graphite furnace atomic absorption spectrometry (GF-AAS)-based rubidium efflux assay as a screening method for Kv7.2/3 channel activators was demonstrated. With flupirtine serving as a reference compound, 16 newly synthesizedcompounds and the known Kv7.2/3 activator retigabine were first classified as either active or inactive by using the GF-AAS-based rubidium (Rb) efflux assay. Then, the results were compared with a thallium (Tl) uptake fluorescence-based fluorometric imaging plate reader (FLIPR) potassium assay. Overall, 16 of 17 compounds were classified by the GF-AAS-based assay in agreement with their channel-activating properties determined by the more expensive Tl uptake, fluorescence-based assay. Thus, the performance of the GF-AAS-based Rb assay for primary drug screening of Kv7.2/3-activating compounds was clearly demonstrated, as documented by the calculated Z’-factor of the GF-AAS-based method. Moreover, method development included optimization of the coating of the microtiter plates and the washing procedure, which extended the range of this assay to poorly adherent cells such as the HEK293 cells used in this study.
Target proteins in biotechnological applications are highly diverse. Therefore, versatile flexible expression systems for their functional overproduction are required. In order to find the right heterologous gene expression strategy, suitable host-vector systems, which combine different genetic circuits, are useful. In this study, we designed a novel Bacillus subtilis expression toolbox, which allows the overproduction and secretion of potentially toxic enzymes. This toolbox comprises a set of 60 expression vectors, which combine two promoter variants, four strong secretion signals, a translation-enhancing downstream box, and three plasmid backbones. This B. subtilis toolbox is based on a tailor-made, clean deletion mutant strain, which is protease and sporulation deficient and exhibits reduced autolysis and secondary metabolism. The appropriateness of this alternative expression platform was tested for the overproduction of two difficult-to-produce eukaryotic model proteins. These included the sulfhydryl oxidase Sox from Saccharomyces cerevisiae, which forms reactive hydrogen peroxide and undesired cross-linking of functional proteins, and the human interleukin-1β, a pro-inflammatory cytokine. For the best performing Sox and interleukin, overproducing and secreting variants of these new B. subtilis toolbox fermentation strategies were developed and tested. This study demonstrates the suitability of the prokaryotic B. subtilis host-vector system for the extracellular production of two eukaryotic proteins with biotechnological relevance.
The present study covers the synthesis, purification and evaluation of a novel aminomethacrylate-based copolymer in terms of its suitability for improving the solubility and in vitro release of poorly water-soluble drug compounds. The new copolymer was synthesized by solvent polymerization with radical initiation and by use of a chain transfer agent. Based on its composition, it can be considered as a modified type of dimethylaminoethyl methacrylate-butyl methacrylate-methyl methacrylate “EUDRAGIT® E PO” (ModE). ModE was specifically developed to provide a copolymer with processing and application properties that exceed those of commercially available (co-)polymers in solubility enhancement technologies where possible. By varying the concentration of the chain transfer agent in the radical polymerization process, the molecular weight of ModE was varied in a range of 173–305 kDa. To evaluate the solubility-enhancing properties of ModE, a series of drug-loaded extrudates were prepared by hot melt extrusion using the novel—as well as several commercially available—(co-)polymers. These extrudates were then subjected to comparative tests for amorphousness, solubility-enhancing properties, storage stability, and drug release. Celecoxib, efavirenz, and fenofibrate were used as model drugs in all experiments. Of all the (co-)polymers included in the study, ModE with a molecular weight of 173 kDa showed the best performance in terms of desired properties and was shown to be particularly suitable for preparing amorphous solid dispersions (ASDs) of the three model drugs, which in a first set of dissolution experiments showed better release behavior under pH conditions of the fasting stomach than higher molecular weight ModE types, as well as a variety of commercially available (co-)polymers. Therefore, the results demonstrate the successful synthesis of a new copolymer, which in future studies will be investigated in more detail for universal application in the field of solubility enhancement.
Abstract
Because isoenzymes of the experimentally and therapeutically extremely relevant sirtuin family show high similarity, addressing the unique selectivity pocket of sirtuin 2 is a promising strategy towards selective inhibitors. An unrelated approach towards selective inhibition of isoenzymes with varied tissue distribution is targeted drug delivery or spatiotemporal activation by photochemical activation. Azologization of two nicotinamide‐mimicking lead structures was undertaken to combine both approaches and yielded a set of 33 azobenzenes and azopyridines that have been evaluated for their photochemical behaviour and bioactivity. For some compounds, inhibitory activity reached the sub‐micromolar range in their thermodynamically favoured E form and could be decreased by photoisomerization to the metastable Z form. Besides, derivatization with long‐chain fatty acids yielded potent sirtuin 2 inhibitors, featuring another intriguing aspect of azo‐based photoswitches. In these compounds, switching to the Z isomer increased aqueous solubility and thereby enhanced biological activity by up to a factor of 21. The biological activity of two compounds was confirmed by hyperacetylation of sirtuin specific histone proteins in a cell‐based activity assay.
In recent years, the colon has become a hot topic in biopharmaceutical research as several in vitro models of the human colon have been presented. A major focus is on the characterization of the microbiota and its capabilities. The aim of the present study was to further develop the MimiCol, preserving its properties and accelerating data acquisition. Emphasis was placed on the simplicity of its design and easy scalability. To prove the viability of the concept, degradation of sulfasalazine was investigated, and the bacterial composition during the experiment was assessed by 16S rRNA sequencing. The transfer of the experimental conditions to the new model was successful. Commercially available components were implemented in the setup. The model MimiCol3 represented the colon ascendens satisfactorily in its properties regarding volume, pH value, and redox potential. 16S rRNA sequencing led to further insights into the bacterial composition in the vessels. Degradation of sulfasalazine was in good agreement with in vivo data. The new model of the colon ascendens MimiCol3 enabled us to collect more reliable data, as three experiments were conducted simultaneously under the same conditions.
Im Rahmen der vorliegenden Arbeit wurde ein auf dem Promotor Pdes basierendes Kälte-induzierbares Expressionssystem für B. subtilis konstruiert und sukzessive optimiert. Dazu wurden verschiedene Kälte-regulatorische DNA-Sequenzen aus B. subtilis an das entsprechende Zielgen fusioniert, was neben der Kälte-Induzierbarkeit in einem positiven Einfluss auf die Expressionsstärke durch eine effizientere Translation bzw. Stabilisierung der mRNA resultierte. Vorausgehend wurde in vergleichenden Versuchen die Eignung unterschiedlicher Galaktosidasen zur Verwendung als Reporterenzyme für B. subtilis untersucht. Hierbei wurde erstmals die heterologe Expression einer Kälte-angepassten β-Galaktosidase aus P. haloplanktis TAE79 in B. subtilis durchgeführt und diese durch die Integration der DB-Sequenz sowie einer stem-loop-Struktur aus der 5‘-UTR des B. subtilis cspB-Gens gesteigert. Somit konnte nachgewiesen werden dass sowohl die additiven Sequenzen der cspB-DB und der cspB-sl-UTR als auch des bkdB-Terminators zu einer deutlich erhöhten Synthese der entsprechenden Zielproteine führt. Anhand der Überexpression einer Xylanase aus B. subtilis sowie einer α-Glucosidase aus S. cerevisiae wurde abschließend die Eignung des konstruierten Systems für die sekretorische und intrazelluläre Proteinsynthese in B. subtilis demonstriert. Diese Ergebnisse bestätigen die Eignung von B. subtilis als Wirtsorganismus auch für die Überproduktion kritischer, schwer zu faltender Proteine.
Analysis and Reduction of Cellular Heterogeneity in Strain Optimization of Bacillus licheniformis

Bacillus species invest substantial resources in inherent cellular processes for pre-adaptation to environmental changes, many of which are dispensable in the controlled environment of industrial bioprocesses. The underlying physiological mechanisms are well characterized in B. subtilis, but only little is known about these processes in the closely related B. licheniformis. Moreover, experimental conditions in previous studies differ from industrial settings in most parameters, foremost in batch cultures or plate-based analysis over fed-batch processes. In this thesis, cellular heterogeneity was analyzed in B. licheniformis in optimized, nutrient-rich media in batch and fed-batch cultivations. Systematic inactivation of genes involved in biofilm formation and synthesis of the flagellar apparatus or global regulators thereof resulted in higher protein production and provided new insights into biofilm formation and cellular heterogeneity in this strain.
Species of the genus Drosera, known for carnivorous plants, such as sundew, have been traditionally used for centuries as medicinal plants. Efficacy-determining compounds are naphthoquinones and flavonoids. Flavonoids possess a broad spectrum of bioactive properties, including biofilm inhibitory activity. Biofilms render antibiotics ineffective, contributing to the current rise in antimicrobial resistance. In this study, the biofilm inhibitory activity of two European sundew species (Drosera rotundifolia and Drosera intermedia) grown agriculturally in Germany and four commercial sundew products (declared as Drosera longifolia, Drosera sp. and Drosera planta trit.) against three multidrug-resistant Escherichia coli strains was tested. The aim of the study was to comparatively investigate the biofilm inhibitory potential of sundew species extracts grown locally in northern Germany and commercial sundew products. The minimum biofilm inhibitory concentration of the European sundew species was approx. 35 µg mL−1. In comparison, commercial sundew products ranged in concentration from 75 to 140 µg mL−1. Additionally, individual compounds isolated from European sundew were tested. Among these compounds, biofilm inhibitory activity was determined for four of the eight substances, with 2″-O-galloyl hyperoside standing out for its activity (38 µg mL−1). The whole plant extracts of Drosera rotundifolia and Drosera intermedia proved to be more effective than the commercial products and the single compounds in its biofilm inhibition activity against Escherichia coli strains. Sundew extracts may serve as a potential therapeutic approach for targeting biofilm production.
Anwendung additiver Fertigungstechniken zur Herstellung und Applikation oraler Darreichungsformen

Der 3D-Druck hat in den vergangenen Jahren einen enormen Interessenzuwachs erzeugt. Aufgrund der einfachen Zugänglichkeit und Bedienbarkeit und aufgrund seiner enormen Möglichkeiten als additive Fertigungstechnologie ist das große Potenzial dieser Technologie auch im pharmazeutischen Bereich nicht verborgen geblieben. Daraus hat sich ein Hype ergeben, welcher 2015 sogar in der Zulassung des ersten 3D-gedruckten Fertigarzneimittels Spritam® durch die FDA resultierte. Jedoch ist dieses Präparates gleichzeitig auch ein gutes Beispiel für die Limitationen des 3D-Druckes. Die Herstellung einer 3D-gedruckten Spritam®-Tablette kostet ein Vielfaches im Vergleich zur Herstellung mit konventioneller Tablettierung. Daraus ergibt sich fast schon zwangsläufig, dass der 3D-Druck sich nicht als Technologie zu Herstellung klassischer oraler Darreichungsformen eignet. Seine Möglichkeiten liegen viel mehr im Gebiet der Individualisierung von Arzneiformen und der Entwicklung von Arzneiformen und Medizinprodukten. Hier kann das Potenzial dieser Technologie wesentlich effizienter ausgeschöpft werden. Daher war es das Ziel dieser Arbeit, den 3D-Druck zu nutzen, um innovative und individualisierbare Darreichungsformen zu entwickeln. Außerdem sollte mittels 3D-Druck eine Applikationshilfe für eine innovative Darreichungsform entwickelt werden. Auch die Herstellung von Minitabletten für die pädiatrische Anwendung wurde untersucht.
Die durchgeführten Untersuchungen zeigten, dass der 3D-Druck vielfältige Anwendung in der Pharmazie finden kann. So konnten unter Nutzung des 3D-Druckes lebensmittelechte Applikatoren hergestellt werden, die die In vivo-Testung der innovativen Darreichungsform EsoCap erst ermöglichte und so im Rahmen einer Machbarkeitsstudie die prinzipielle Funktionalität dieser Darreichungsform zeigte. Dabei bietet der 3D-Druck im Rapid Prototyping den Vorteil, dass es sich um eine vergleichsweise günstige und schnell zu realisierende Technik handelt. Weiterhin bieten die leichte Bedienbarkeit, die realisierbare Formenvielfalt, eine Vielzahl zur Verfügung stehender Filamente mit unterschiedlichsten Eigenschaften und der niederschwellige Zugang zu dieser Technologie auch Personen ohne entsprechenden Hintergrund die Möglichkeit zur Prototypenentwicklung. Dies ermöglicht eine schnelle und angepasste Entwicklung ohne weitere Beteiligung externer Institute oder Dienstleiter. Darüber hinaus kann der 3D-Druck auch Anwendung in der Modifizierung von Freisetzungstestern finden oder zur Ersatzteilproduktion dienen. Auch zur Herstellung von Darreichungsformen mit neuartigen Freisetzungsmechanismen kann der 3D-Druck genutzt werden. Dabei zeigte sich in der durchgeführten In vivo-Studie an Hunden, dass eine prinzipielle Herstellung mittels 3D-Druck möglich ist, aber auch Limitationen, speziell des FDM-basierten 3D-Druckes, aufweist. So zeigte sich eine mangelnde Reproduzierbarkeit bei der Herstellung der drucksensitiven Arzneiform, die wahrscheinlich dazu führte, dass bei der Hälfte der Applikationen bei den Versuchstieren die Kapsel nicht durch die physiologischen Kräfte zerstört wurde und so keinen Wirkstoff freisetzte. Hierbei bleibt anzumerken, dass durch Nutzung alternativer additiver Herstellungsverfahren im Rahmen weiterer Untersuchungen diese Problematik weiter erforscht werden kann. Nichtsdestotrotz ermöglicht der 3D-Druck die Herstellung von Darreichungsformen mit neuartigen Freisetzungsmechanismen, die mittels konventionellen Herstellungstechniken schwer beziehungsweise gar nicht hergestellt werden können. Im letzten Punkt dieser Arbeit wurde der 3D-Druck als alternatives Herstellungsverfahren für pädiatrische Arzneimittel untersucht. Dazu wurden Minitabletten mit unterschiedlichen Durchmessern unter Variation der Wirkstoffe und Polymere hergestellt. Dabei zeigte sich, dass der 3D-Druck sowohl die Möglichkeit bietet, über die Variation der Anzahl applizierter Minitabletten als auch über die Variation der Größe eine Dosisindividualisierung vorzunehmen. Die so hergestellten Arzneimittel entsprachen dabei auch den Vorgaben des europäischen Arzneibuches hinsichtlich der Massegleichförmigkeit. Damit konnte gezeigt werden, dass die Herstellung von Minitabletten mittels 3D-Druck eine Alternative zu der konventionellen Kapselherstellung darstellt. Dabei bleibt anzumerken, dass weitere Untersuchungen notwendig sind, um weitere, im Bereich der Pädiatrie eingesetzten Arzneistoffe auf ihre Eignung für die Schmelzextrusion und den 3D-Druck zu untersuchen.
Zusammenfassend konnte in der vorliegenden Arbeit gezeigt werden, dass die Verwendung additiver Herstellungsverfahren wie der FDM-basierte 3D-Druck in der Pharmazie auf unterschiedlichste Weise Anwendung finden kann. So können zum einen schnell, einfach und auch kostengünstig Prototypen und auch Ersatzteile hergestellt werden. Dieser Aspekt und die Tatsache, dass auch ohne umfangreiche Schulungen der 3D-Druck nutzbar ist, zeigen auf, dass der 3D-Druck ein nützliches Werkzeug im Bereich des Rapid Prototypings von Applikationshilfen, aber auch im Bereich der Entwicklung von biorelevanten Testmodellen darstellt. Auch können durch die Nutzung des 3D-Druckes neuartige, innovative Freisetzungssysteme hergestellt und getestet werden. Darüber hinaus schaffen additive Fertigungstechniken weiterhin die Möglichkeit, mögliche Alternativen im Bereich der Herstellung pädiatrischer Arzneimittel herzustellen.
In der vorliegenden Arbeit sollten zwei verschiedene Wirkstoffklassen auf ihre Fähigkeit Apoptose und Autophagozytose zu aktivieren analysiert werden. Dabei wurden 39 Sigma-Rezeptor-Liganden, die an der Universität Münster (Arbeitsgruppe von Prof. Bernhard Wünsch) synthetisiert wurden, zunächst auf ihre antiproliferativen Eigenschaften in acht humanen Krebszelllinien untersucht. Anhand der Struktur-Wirkungs-Beziehungen konnte gezeigt werden, dass sich große, raumfüllende Substituenten an beiden N-Atomen des Piperazin-Grundgerüstes positiv auf die Hemmung des Zellwachstums auswirken. Da die Multiple Myelom Zellinie RPMI 8226 eine hohe Dichte an Sigma-Rezeptoren exprimiert, wurde sie für weitere Versuche herangezogen. Als repräsentative Liganden wurde das Enantiomerenpaar (S)-11 und (R)-11 für weitere Versuche ausgewählt, da es neben einer guten Affinität zu beiden Rezeptor-Subtypen auch antiproliferierende Eigenschaften in der RPMI 8226-Zelllinie zeigte. Die Behandlung führte zur zeitabhängigen Induktion der Apoptose, die mit den typischen Charakteristika wie morphologische Membranausstülpungen, Annexin-V positive Zellen und der Chromatinkondensation einherging. Die Detektion von aktivierter Caspase-3, -8 und -9 im Durchflusszytometer deutete zunächst auf eine Beteiligung beider Signalwege der Apoptose hin. Da im Western Blot keine Caspase-9-Spaltprodukte detektiert werden konnten und auch die Vorinkubation mit den Caspase-Inhibitoren z-VAD-FMK und M50054 keinen Effekt auf die Apoptoserate bewirkte, scheinen die Caspasen im Sigma-Ligand-vermittelten Zelltod eine untergeordnete Rolle einzunehmen. Die frühe Abnahme des mitochondrialen Membranpotentials verbunden mit der zunehmenden Konzentration des Apoptose-induzierenden Faktors (AIF) im Zytosol der RPMI 8226-Zellen deuten ebenfalls auf eine Caspase-unabhängige Form der Apoptose hin, die besonders für Sigma(2)-Agonisten bereits beschrieben ist. Und auch in diesem Projekt zeigen die Sigma-Liganden zytotoxische Aktivitäten, wenn sie eine Affinität zum Sigma(2)-Rezeptor aufweisen. Neben der Apoptose konnten (S)-11 und (R)-11 auch die Autophagozytose induzieren. Dieser Prozess scheint jedoch der Apoptose nachgeschaltet zu sein und weist darauf hin, dass es sich nicht um einen Schutzmechanismus der Zelle handelt, sondern eher in einer direkten Form des programmierten Zelltodes vom Typ II begründet ist. Dieser ist bisher noch nicht für Sigma-Rezeptor-Liganden beschrieben und könnte einen Vorteil gegenüber anderen Vertretern dieser Wirkstoffklasse darstellen. Eine Gemeinsamkeit von Sigma-Ligand-induzierter Apoptose und Autophagozytose liegt in ihrer Aktivierung durch die Lipidperoxidation. Es wurde ein sehr früher Anstieg von LPO-Produkten detektiert, der sich nur durch das lipophile Antioxidans alpha-Tocopherol beeinflussen ließ. Desweiteren konnte die antiproliferierende Wirkung von (S)- 11 und (R)-11 im MTT-Assay komplett blockiert werden, und auch die Apoptoseraten wurden durch den Radikalfänger signifikant erniedrigt. Die Hemmung der LC3-II-Expression im Western Blot durch alpha-Tocopherol verdeutlicht auch hier die Beteiligung der LPO an der Autophagozytose. Als Vertreter einer weiteren Wirkstoffklasse wurde der phtoaktivierbare Pt(IV)-Komplex FM 165, der in der Arbeitsgruppe von Prof. Dr. Peter Sadler (Universität Warwick, UK) synthetisiert wurde, untersucht. Die antiproliferierenden Eigenschaften der photoaktivierbaren Pt(IV)-Verbindung können nicht mit der Apoptose als Zelltodmechanismus begründet werden. In diesem Fall ließen sich keine morphologischen Charakteristika nachweisen und auch die Annexin-V Anfärbung fiel negativ aus. Die weiteren Untersuchungen zeigten jedoch auch die Aktivierung der Autophagozytose durch Expression von LC3-II und p62 im Western Blot. Eine Hemmung lysosomaler Enzyme durch die Protease-Inhibitoren E64d und Pepstatin A führte zu erhöhten LC3-II-Konzentrationen sowie einer p62-Akkumulation. Im Gegensatz zu den Sigma-Rezeptor-Liganden deutet dies eher auf eine vollständige Verlaufsform hin. Da die Autophagozytose bereits nach 6 h-Behandlung mit FM 165 detektiert wurde, ist es nicht ausgeschlossen, dass es sich zunächst um eine zellschützende Funktion handeln könnte. Auch beim FM 165 scheinen oxidative Vorgänge eine wichtige Rolle hinsichtlich der Autophagozytose-Aktivierung zu übernehmen. Nach erfolgter Photoaktivierung der Zellen ließen sich ansteigende ROS Konzentrationen verzeichnen, die nach 2 h deutlich wurden und somit noch vor der Autophagozytose entstehen.
Application of In Vivo Imaging Techniques and Diagnostic Tools in Oral Drug Delivery Research

Drug absorption following oral administration is determined by complex and dynamic interactions between gastrointestinal (GI) physiology, the drug, and its formulation. Since many of these interactions are not fully understood, the COST action on “Understanding Gastrointestinal Absorption-related Processes (UNGAP)” was initiated in 2017, with the aim to improve the current comprehension of intestinal drug absorption and foster future developments in this field. In this regard, in vivo techniques used for the characterization of human GI physiology and the intraluminal behavior of orally administered dosage forms in the GI tract are fundamental to gaining deeper mechanistic understanding of the interplay between human GI physiology and drug product performance. In this review, the potential applications, advantages, and limitations of the most important in vivo techniques relevant to oral biopharmaceutics are presented from the perspectives of different research fields.
The goal of this study was to assess the anticancer efficacy of chlorojanerin against various cancer cells. The effects of chlorojanerin on cell cytotoxicity, cell cycle arrest, and cell apoptosis were examined using MTT assay, propidium iodide staining, and FITC Annexin V assay. RT-PCR was employed to determine the expression levels of apoptosis-related genes. Furthermore, docking simulations were utilized to further elucidate the binding preferences of chlorojanerin with Bcl-2. According to MTT assay, chlorojanerin inhibited the proliferation of all tested cells in a dose-dependent manner with a promising effect against A549 lung cancer cells with an IC50 of 10 µM. Cell growth inhibition by chlorojanerin was linked with G2/M phase cell cycle arrest in A549 treated cells. Flow cytometry analysis indicated that the proliferation inhibition effect of chlorojanerin was associated with apoptosis induction in A549 cells. Remarkably, chlorojanerin altered the expression of many genes involved in apoptosis initiation. Moreover, we determined that chlorojanerin fit into the active site of Bcl-2 according to the molecular docking study. Collectively, our results demonstrate that chlorojanerin mediated an anticancer effect involving cell cycle arrest and apoptotic cell death and, therefore, could potentially serve as a therapeutic agent in lung cancer treatment.
Der Naturstoff Splitomicin war als selektiver Inhibitor (der Aktivität) der NAD+- abhängigen Histon-Desacetylase des Sir2-Proteins Ausgangspunkt der Untersuchungen, ist aber an humanen Enzymen inaktiv und aufgrund der im menschlichen Körper zu erwartenden Hydrolyse der Verbindung weder als Arzneistoff noch als Grundgerüst für Analoga geeignet. Da die Stabilität der Verbindung Voraussetzung dafür ist, dass die Verbindungen ihre Wirkung entfalten können, und eine Verschiebung der Selektivität hin zu humanen Enzymen beabsichtigt war, wurde im Rahmen dieser Arbeit Splitomicin zwar als Leitstruktur übernommen aber das heterocyclische Ringsystem auf der Grundlage von Struktur-Aktivitätsbeziehungen modifiziert. Der in Splitomicin enthaltene Sechsring des Pyranons wurde durch einen Fünfring ersetzt. Neben anellierten Imidazolonen wurden Imidazolthione und Chinolinimidazolthione dargestellt, um die Stabilität der Verbindungen gegenüber Hydrolyseeinflüssen zu erhöhen.
Koronare Drug-eluting Stents sind röhrenförmige, netzartig aufgebaute, medizinische Implantate aus meist metallischen Grundkörpern, die zur Vermeidung eines erneuten Gefäßverschlusses mit Wirkstoff beschichtet sind. Derzeit finden die Beschichtungen der Stents typischerweise über speziell entwickelte Einzelbeschichtungsverfahren statt, die es ermöglichen, die feingliedrige und fragile Struktur der Implantate zu beschichten. Wesentliche Nachteile dieser Verfahren liegen allerdings in hohen Materialverlusten und geringer Fertigungs¬menge, die zu hohen Produktionskosten führen. Im Rahmen dieser Arbeit sollte untersucht werden, inwieweit die Wirbelschichttechnologie zur Beschichtung von Stents eingesetzt werden kann. Die Wirbelschichttechnologie stellt ein etabliertes Verfahren zur Beschichtung von Partikeln im Großmaßstab mit geringen Verlusten dar. Obwohl in der Literatur das Verhalten verschiedener Partikel (Größe, Form, Dichte) in der Wirbelschicht beschrieben ist, wurde das Verhalten von netzartig aufgebauten Hohlkörpern noch nicht untersucht. Inwiefern sich die Wirbelschichttechnologie zur Beschichtung der fragilen Stents eignet, ist daher nicht vorhersagbar. Im Rahmen dieser Arbeit sollte eine Beurteilung hinsichtlich der Anwendbarkeit des Wirbelschichtverfahrens zur Beschichtung von Stents erfolgen. Dazu wurde das Fluidisierungs-verhalten der Körper unter Verwendung verschiedener Anströmböden untersucht. Darüber hinaus wurde der Einfluss der Prozessparameter, verschiedener Beschichtungsrezepturen und Stenttypen auf die Qualität der Beschichtung untersucht. Dazu wurden die beschichteten Körper hinsichtlich Homogenität und Defekte der Oberfläche, ihrer Robustheit gegenüber mechanischen Einflüssen, der Schichtverteilung und Gehaltsgleichförmigkeit und des Wirkstofffreisetzungsverhaltens untersucht. Im Rahmen dieser Arbeit konnten die Stents den Partikel der Geldart- Klasse D zugeordnet werden und die Beschichtungen der Stents erfolgte unter Verwendung eines speziell angefertigten konisch ausgeformten Anströmbodens. In Versuchen mit verschiedenen Beschichtungsmaterialien war es möglich typischer Weise auftretende Beschichtungsdefekte und präventive Gegenmaßnahmen zu identifizieren. So zeigte sich, dass es unter Verwendung von wässrig dispergierten Beschichtungsmaterialien die Bildung von Brücken oder Schwimmhäuten ausblieb, während diese typischer Weise unter Verwendung von organisch gelösten Beschichtungsrezepturen auftraten. Darüber hinaus konnte gezeigt werden, dass es durch Anpassung der Prozessparameter möglich war Beschichtungen mit hoher Qualität zu erzeugen. Dabei wurden unabhängig von der Lackrezeptur leichter Abrieb, Risse oder Deformationen der Stentgrundkörper vor allem an den Stentenden beobachtet, da die Stents während des Beschichtungsprozesses einer starken mechanischen Belastung ausgesetzt waren. In Versuchen mit unterschiedlichen Auftragsmengen beziehungsweise Prozesszeiten und unterschiedlich langen Stents konnte gezeigt werden, dass die Defekte unter gleichbleibenden Bedingungen mit zunehmender Prozesszeit und abnehmender Stentlänge zunehmen. Zudem zeigte sich, dass auch die Prozessparameter und Flexibilität der Beschichtung Einfluss auf den Anteil defekter Stents nimmt. Durch Adaptierung des Beschichtungsprozesses ist es entsprechend möglich die Defekte zu reduzieren. In der Regel wurde ein etwas höherer abluminaler Schichtauftrag erzielt, wobei die Schicht homogen über die gesamte Stentlänge verteilt war. Es resultierten üblicherweise Abscheideraten des Beschichtungslackes von 40 - 50 % und darüber hinaus konnten durch die gleichzeitige Beschichtung einer hohen Stückzahl von Grundkörpern Fertigungsmengen von umgerechnet bis zu 25 Stents pro Minute erreicht werden. Im Rahmen dieser Arbeit konnte gezeigt werden, dass die Wirbelschichttechnologie ein vielversprechendes Verfahren zur Beschichtung von Stents im Großmaßstab darstellt, das in Bezug auf Ausbeute und Fertigungsmenge den derzeit herkömmlich verwendeten Beschichtungstechnologien überlegen ist, wobei die Qualität der erzeugten Beschichtungen vergleichbar mit der Beschichtungsqualität herkömmlich beschichteter Stents ist.
Neu isolierte und synthetisierte Wirkstoffe müssen neben ihrer biologischen Wirksamkeit auch auf ihre Unbedenklichkeit für den Menschen hin untersucht werden. Ein Bestandteil der Untersuchungen zur Unbedenklichkeit ist die Prüfung auf mögliche Immuntoxizität. Die Risikobewertung und -klassifizierung von (immun-)toxischen Substanzen erfolgt derzeit in Tierversuchen, die, abgesehen von ethischen Bedenken, zeit- und kostenintensiv sind und deren Übertragbarkeit auf den Menschen nicht vollständig gewährleistet ist.
Im Fokus dieser Arbeit stand die Etablierung und Anwendung eines Methodensets basierend auf funktionalen in vitro Methoden zur Charakterisierung immunologischer Wirkungen ausgewählter Naturstoffe. Dieses sollte der Beurteilung der immuntoxischen Wirkungen der getesteten Naturstoffe und der Entwicklung eines Entscheidungsbaums, der die Vorhersage des immuntoxischen Potentials mithilfe von in vitro Versuchen gestattet, dienen. Dazu wurden zwei humane Immunzelllinien (Jurkat-Zellen als Beispiel für T-Zellen, THP-1-Zellen als Beispiel für Monozyten) und für einige Versuche vergleichsweise primäre Blutzellen eingesetzt. Es wurden Methoden zur Untersuchung folgender Parameter etabliert und angewendet: Vitalität, Zellzyklusverteilung, Induktion von Apoptose, iROS, DNA-Schäden (Genotoxizität), Zytokinfreisetzung und mitochondriale Funktion. Folgende Naturstoffe wurden für die Untersuchungen ausgewählt: Mannitol und Urethan als Negativkontrollen, Cyclosporin A, Deoxynivalenol und Mycophenolsäure als Positivkontrollen, ausgehend von Hinweisen auf Wirkungen im Immunsystem Tulipalin A, Helenalin, Vincristin, Cannabidiol, Agaritin und p-Tolylhydrazin als Testsubstanzen.
Es zeigten sich nur geringfügige Unterschiede der Substanzwirkungen zwischen den Immunzelllinien, welche v.a. auf Zytokinebene nachweisbar waren. Die Substanzen besaßen zeit- und konzentrationsabhängige Effekte. Die Negativkontrolle Mannitol hatte eine geringe Wirkung auf die Immunzelllinien, während Urethan die Zytokinfreisetzung supprimierte/¬stimulierte. Die untersuchten Positivkontrollen zeigten einen Einfluss auf die Zytokinfreisetzung und führten weiterhin zu immuntoxischen Effekten durch eine konzentrationsabhängige Apoptoseinduktion. Die Testsubstanzen Vincristin, Agaritin und p-Tolylhydrazin besaßen nur eine geringe toxische Wirkung auf die Immunzellen. Weitere Substanzen wie Cannabidiol, Helenalin und Tulipalin A wiesen immunspezifisch und - unspezifisch vermittelte Immuntoxizität durch einen Einfluss auf die Zytokinfreisetzung, Apoptose und iROS auf.
Funktionale in vitro Untersuchungen zur Vitalität, Zellzyklusverteilung, Apoptose und Zytokinfreisetzung waren zum Nachweis bzw. Ausschluss von Immuntoxizität geeignet und neben Proteom- und Metabolomanalysen wesentlicher Bestandteil eines Entscheidungsbaums zur Klassifizierung von direkt immuntoxischen Substanzen. Es zeigte sich, dass die Zytokinmessung der wichtigste Parameter zur Klassifizierung von immuntoxischen Substanzen im subtoxischen Bereich ist. Es konnte sowohl Cyclosporin A als Positivkontrolle als auch Mannitol als Negativkontrolle in beiden Zelllinien bestätigt werden. Von den hinreichend untersuchten Testsubstanzen wurde Cannabidiol, Helenalin und Tulipalin A in Jurkat-Zellen sowie Cannabidiol und Tulipalin A in THP-1-Zellen unter Verwendung des Entscheidungsbaums als immuntoxisch klassifiziert.
Darüber hinaus konnte die hautsensibilisierende Wirkung von Farnesol und Tulipalin A durch Anwendung von weiteren in vitro Methoden bestätigt werden.
Eine Validierung der Ergebnisse mit weiteren bekannten immuntoxischen und nicht-immuntoxischen Substanzen würde eine Anwendung als Vorscreening Testung neuer Substanzen ermöglichen und nicht nur zu einer Reduktion von Tierversuchen führen, sondern auch eine Zeit- und Kostenersparnis bedeuten.
Ziel der vorliegenden Arbeit war es, das Herstellungsverfahren der wässrig-fermentativen Frischpflanzenextraktion nach HAB, Vs. 33 und 34 sowie die dabei ablaufenden biochemischen und mikrobiologischen Reaktionen zu betrachten. Neben der Extraktion findet begleitend eine Fermentation statt und die daraus entstehende Urtinktur wird bis zur Weiterverarbeitung mindestens 6 Monate gelagert. Diese drei Prozessschritte -Extraktion, Fermentation, Lagerung- können Einfluss auf die Qualität der Urtinktur nehmen. Es sollte daher geklärt werden, welche bio- und phytochemischen Reaktionen bei der Herstellung und anschließenden Lagerung einer wässrig-fermentierten Urtinktur ablaufen und welche Mikroorganismen daran maßgeblich beteiligt sind.
In diesem Zusammenhang wurden Extrakte aus Atropa belladonna, blühendes Kraut hergestellt und durch Variation bestimmter Herstellungsparameter die Robustheit des Verfahrens überprüft.

Weiterhin erfolgte ein Vergleich zwischen diesem Extraktionsverfahren mit der verbreiteten ethanolischen Frischpflanzenextraktion. In den Jahren 2006-2009 wurden insgesamt 106 wässrig-fermentierte Urtinkturen und 4 ethanolische Auszüge hergestellt.
Da im Verlauf der Lagerung in einigen wässrig-fermentierten Urtinkturen Abnahmen des Atropingehaltes beobachtet wurden, wurde zur Klärung das Verhalten von Milchsäurebakterien in atropinhaltigen Lösungen untersucht.
Die Ergebnisse der Untersuchungen zeigten, dass an der Fermentation in erster Linie Milchsäurebakterien beteiligt sind, die durch Zuckerabbau Milchsäure, daneben auch Acetat und Ethanol, bilden. Als verantwortliche Milchsäurebakterien konnten vor allem der homofermentative Lactobacillus plantarum und der heterofermentative Lactobacillus brevis isoliert und identifiziert werden. Hierbei war in der Hauptfermentationsphase eine Dominanz von Lactobacillus plantarum erkennbar. Die Kultivierung von Milchsäurebakterien in atropinhaltigen Lösungen zeigte, dass das Wachstums- und Fermentationsverhalten der gewünschten Milchsäurebakterien durch Atropin nicht negativ beeinflusst wird.
Bei dem betrachteten Herstellungsverfahren kann ein Verlust des Wirkstoffs Atropin unter Einhaltung einer ausreichenden Säuerung ausgeschlossen werden. Damit ist die wässrig-fermentative Urtinkturherstellung hinsichtlich der Atropinextraktion unter Einhaltung bestimmter Herstellungsregeln als ebenso effektiv und robust anzusehen wie die ethanolische Frischpflanzenextraktion. Unterschiedliche Gehalte zwischen ethanolischen und wässrig-fermentierten Extrakten ließen sich bei Scopoletin, Flavonoiden und einer im Rahmen der Arbeit nachgewiesenen Substanz X feststellen.
Das untersuchte Herstellungsverfahren führte in den meisten Fällen zu einem stabilen Extrakt, wobei sich Mikroflora und Fermentationsverläufe trotz Variation der Herstellungsparameter ähnelten. Auf Grund der Variabilität der mikrobiologischen Flora des Pflanzenmaterials unterliegt die Fermentation unter den Bedingungen des Homöopathischen Arzneibuchs allerdings natürlichen Schwankungen. Dies führte in einigen Fällen zu einer nicht spezifikationskonformen Urtinktur. Deswegen wurden für eine optimierte Herstellung die Zugabe einer Starterkultur, der Zusatz von Molke, eine durchgängige Mazeration bei 37 °C, eine flexible Anpassung des Kohlenhydratbedarfs, Sauerstoffausschluss während der Mazerationswoche, eine kürzere, für den Fermentationsverlauf individuelle Lagerungszeit und die Berücksichtigung des Wassergehaltes des Pflanzenmaterials empfohlen. Da die Ergebnisse der Arbeit mit den Untersuchungen anderer pflanzlicher Materialien Sauerkraut, Sauerteig, Silage) in Einklang stehen, ist davon auszugehen, dass diese Empfehlungen auch bei anderen Pflanzen die Herstellsicherheit erhöhen und zu einer reproduzierbaren Extraktqualität führen.
In the search for bioactive compounds, 32 fungal strains were isolated from Indonesian marine habitats. Ethyl acetate extracts of their culture broth were tested for cytotoxic activity against a urinary bladder carcinoma cell line and for antifungal and antibacterial activities against fish and human pathogenic bacteria as well as against plant and human pathogenic fungi. Bioassay-guided fractionation led to the isolation of bioactive compounds. Altogether 14 compounds were isolated and further elucidated. The compounds were obtained from the ethyl acetate and dichloromethane extracts of six fungal strains. They included 9 polyketides, 2 terpenes, 1 alkaloid and 2 till now undefined structures.
Für die orale Pharmakotherapie stellen die gastrointestinalen Flüssigkeiten einen der relevantesten Einflussfaktoren dar, da es sich bei ihnen um das Medium für Zerfallsprozesse der Arzneiform und zur Auflösung des Arzneistoffs handelt. Neben den physikochemischen Eigenschaften wie dem pH-Wert ist es vor allem auch die vorliegende Medienmenge, die für die konzentrationsabhängigen Auflösungs- und Absorptionsprozesse oraler Arzneistoffe von entscheidender Bedeutung ist. Auch der Transport der Flüssigkeiten im Gastrointestinaltrakt ist aufgrund des Cotransports von Arzneiform und Arzneistoff für das Anflutungsverhalten oral eingenommener Arzneimittel relevant. Insbesondere die Magenentleerung stellt hier bekanntermaßen einen essenziellen Parameter dar. Das Wissen zu welchem Zeitpunkt, an welchem Ort im Gastrointestinaltrakt, welches Volumen mit den bestimmten Eigenschaften vorliegt ist aber in vielen Fällen nur relevant, wenn man zum selben Zeitpunkt auch die Position und das Zerfalls- oder Freisetzungsverhalten der oralen Arzneiform kennt. Das Verhalten oraler Arzneiformen in vivo kann sich unter Umständen deutlich von dem Verhalten in vitro unterscheiden. Zudem sind die Transitbedingungen im Gastrointestinaltrakt vielfältigen Abhängigkeiten, sowie Schwankungen unterworfen und unterliegen daher einer relevanten Variabilität. Die Verbindung der Untersuchungen von gastrointestinalen Flüssigkeitsmengen und ihren Verteilungskinetiken zusammen mit der Untersuchung von Transit, Freisetzung und Zerfall oraler Arzneiformen ist umfangreich und komplex, daher wurden im Rahmen dieser Arbeit verschiedene Einzelfragestellungen zu dem übergeordneten Themenbereich bearbeitet.
So wurden relevante Aspekte der Interaktionen von gastrointestinalen Volumina, dem Transport und oral verabreichten Arzneimitteln durch Mitarbeit an einem Review umfassend ausgearbeitet und einige unzureichend geklärte Zusammenhänge identifiziert. Zusätzlich wurden verschiedene in vivo Studien unter Zuhilfenahme der MRT Bildgebung durchgeführt. Um die Vergleichbarkeit und Übertragbarkeit sicherzustellen, entsprachen die Protokolle und Designs dieser Studien weitgehend den Bedingungen Klinischer Studien der Phase I.
Eine wichtige Fragestellung im Bereich der oralen Biopharmazie ist dabei immer die Interaktion mit Nahrung. So wurde in der ersten Studie die spezifische Nahrungsmittel-interaktion mit Grapefruitsaft untersucht. So weisen viele oral verabreichte Arzneistoffe gleichgerichtete Effekte nach Einnahme von Nahrung und nach Einnahme von Grapefruitsaft auf, sodass neben der bekannten Enzym- und Transporterwirkung des Grapefruitsaftes auch ein Volumeneinfluss vermutet wurde. Für einige Arzneimittel ist dieser Volumeneffekt möglicherwiese sogar maßgeblich. In der durchgeführten Studie wurden jeweils 240 mL Grapefruitsaft, isokalorische Fruktose- und Glukoselösung, sowie Wasser von 6 Probanden eingenommen und die gastrointestinalen Volumina über mehr als 90 min quantifiziert. Neben einer verlangsamten Magenentleerung ergab sich durch den Grapefruitsaft dabei vor allem ein vergrößertes Flüssigkeitsvolumen im Dünndarm. So war 80 min nach Einnahme des Grapefruitsafts das Dünndarmvolumen im Mittel doppelt so hoch wie nach Einnahme von Wasser. Die Verlangsamung der Magenentleerung des Grapefruitsafts war dabei vor allem durch Glukose vermittelt, während die Veränderungen im Dünndarmvolumen vor allem auf den Fruktosegehalt zurückzuführen sind. Insbesondere die Volumenvergrößerung kann über den Effekt auf die Arzneistoffkonzentration sowohl zu verbesserter als auch verschlechterter Bioverfügbarkeit führen. Auflösungsprozesse insbesondere von BCS II Stoffen werden zwar verbessert, die Absorption von BCS III Stoffen wird durch den verringerten Konzentrationsgradienten allerdings verschlechtert. Sowohl die positiven als auch die negativen Effekte von Grapefruitsaft auf die Bioverfügbarkeit verschiedener Arzneistoffe können demnach auch kalorische Effekte der enthaltenen Kohlenhydrate sein und lassen sich damit auch auf andere Fruchtsäfte übertragen. Inwieweit insbesondere intestinale Volumina wirklich eine relevante Rolle für die Arzneistoffabsorption spielen bleibt jedoch unklar.
Ein weiteres ausgesprochen relevantes Phänomen bei der postprandialen Arzneimitteleinnahme stellt die Magenstraße dar, die eine schnelle Wasserentleerung am Speisebrei vorbei beschreibt und auch Arzneistoffe aus dem Magen mitreißen kann. In einer weiteren Studie wurde daher die Entleerung von 240 mL Wasser nach Einnahme verschiedener Mahlzeiten mit der Wasserentleerung im nüchternen Zustand verglichen. Bei den Mahlzeiten handelte es sich um ein feststoffbasiertes fettreiches Standardfrühstück, ein feststoffbasiertes fettarmes leichtes Frühstück und eine homogene fettreiche Cremespeise mit je 500 kcal. Der ursprünglich vermutete Zusammenhang zwischen Fettgehalt und Auftreten der Magenstraße konnte widerlegt werden. Bei beiden festen Mahlzeiten kam es unabhängig vom Fettgehalt und im Gegensatz zur fettreichen Cremespeise auch 60 min nach der ersten Wassergabe zu einer ausgeprägten Magenstraße. Dieses Auftreten zu späteren Zeitpunkten nach Nahrungseinnahme ist so relevant, da es durch das Ausspülen von Arzneistoff aus dem postprandialen Magen Dose Dumping-ähnliche Effekte herbeiführen kann. Dies geschieht nur mit einer gewissen Wahrscheinlichkeit, was zu einer kritischen Erhöhung der Variabilität im Real Life, aber auch in Studien führt. Dieser Effekt ließe sich auch für die schnelle Anflutung im postprandialen Zustand zum Beispiel mithilfe von Arzneistofflösungen ausnutzen. Für klinische Studien erscheint vor allem die Standardisierung und Dokumentation der Flüssigkeitsgabe über längere Zeiträume als bisher zur Vermeidung unerwünschter Variabilitäten essenziell.
Die Variabilität der Volumina und ihrer Transportvorgänge stellt bekanntermaßen eine Quelle für die pharmakokinetische Variabilität dar und sollte daher ebenfalls charakterisiert werden. Dazu wurden die pseudonymisierten Daten eigens durchgeführter und vergleichbarer Studien gepoolt und die nüchternen Residualvolumina des Magens, sowie die Entleerung von 240 mL Wasser nach nüchterner Applikation untersucht. Dabei wurde die interindividuelle Variabilität zwischen Probanden mit der intraindividuellen Variabilität einzelner Probanden verglichen. Die Magenentleerung stellt unbestritten einen Kernparameter dar und die Menge des Residualvolumens bestimmt maßgeblich die physikochemischen Eigenschaften des Freisetzungsmediums nach nüchterner Applikation. Selbst unter den hochstandardisierten Bedingungen klinischer Studien mit 10 stündigem Fasten über Nacht, ergaben sich dabei deutliche Unterschiede im nüchternen Residualvolumen des Magens. Im interindividuellen Mittel innerhalb der Studienkollektive betrug das nüchterne Residualvolumen 49 ± 19 mL mit einer Spannbreite von 17 mL bis 93 mL. Dies unterschied sich kaum vom intraindividuellen Mittel innerhalb der wiederholten Messungen eines Probanden mit 44 ± 18 mL und einer Spannbreite von 22 mL bis 77 mL. Auch die Magenentleerung war sowohl interindividuell als auch intraindividuell vergleichbaren Schwankungen unterworfen. So stellte sich sowohl bei den nüchternen Residualvolumen als auch bei der Magenentleerung heraus, dass die intraindividuelle Variabilität vergleichbar mit der interindividuellen Variabilität ist. Diesem Fakt wurde bisher zu wenig Bedeutung beigemessen, denn das bedeutet, dass ein Proband nach Mehrfachgabe die gleiche Variabilität zeigen kann, wie ein ganzes Studienkollektiv aus mehreren Probanden. Bei oraler Gabe stellt die gleiche Person demnach keineswegs eine reproduzierbare Referenz für sich selbst dar. Es zeigt auch, dass der Einnahmezeitpunkt in Relation zum MMC eine große Bedeutung für die Menge und physikochemische Zusammensetzung des gastrischen Freisetzungsmediums hat.
Um über die Untersuchung der gastrointestinalen Flüssigkeiten hinaus auch den Arzneiformentransit und -zerfall und das Zusammenspiel mit dem Flüssigkeitstransport zu untersuchen, wurden bekannte Kontrastierungsmethoden für die MRT adaptiert und neue Methoden unter Nutzung von Lebensmitteln entwickelt. Mithilfe einer auf getrockneten Ananasstücken basierenden Methode konnte unter anderem der Transit und Zerfall von DRcapsTM als Cap in-Cap Formulierung im Zusammenspiel mit der Magenentleerung des eingenommenen Wassers untersucht werden. Der Produktclaim einer verzögerten Desintegration, die zeitgesteuert die Freisetzung im Magen verhindert und erst im Dünndarm zu einer Freisetzung führen soll wurde ebenfalls überprüft. Dazu wurden im nüchternen Zustand eine schwimmende und sinkende Formulierung getestet. Hartkapseln haben aufgrund ihrer Pulverfüllung in der Regel eine relativ niedrige Dichte. Dies könnte die Vergleichbarkeit des Transits mit etablierten magensaftresistenten Tabletten einschränken. Bekanntermaßen stellen schwimmende Monolithen auch ein Konzept der Gastroretention dar, was für die Funktion der DRcapsTM aufgrund der Gefahr einer Desintegration im Magen nachteilig wäre und daher in dieser Studie ebenfalls überprüft wurde. Die Magenentleerungszeitpunkte lagen im Mittel bei 45 ± 35 min für die schwimmenden Kapseln und 36 ± 18 min für die sinkenden Kapseln. Demnach sind schwimmende Monolithen entgegen verbreiteter Meinung nach nüchterner Applikation nicht gastroretentiv. Für die Vergleichbarkeit des Transits schwimmender modifiziert freisetzender Kapseln ist dies jedoch vorteilhaft. Die Zerfallszeiten konnten ebenfalls bestimmt werden und die Desintegration fand ausschließlich im Dünndarm statt. Des Weiteren wurde unabhängig von der Dichte in mehreren Fällen eine Magenentleerung der Kapseln lange vor abgeschlossener Wasserentleerung beobachtet. Dieser schnellen Entleerung wird in Modellen bisher möglicherweise zu wenig Bedeutung beigemessen.
Um auch schnell freisetzende Hartkapseln besser untersuchen zu können, wurde die Methodik angepasst und der Lebensmittelfarbstoff Eisenoxid als MRT-Kontrastmittel und Koffein als pharmakokinetischer Marker im Speichel verwendet. In einer Pilotstudie ergab sich eine deutliche Abhängigkeit vom Zerfallsort der Kapseln und der Geschwindigkeit der pharmakokinetischen Anflutung. Vom visuellen Zerfall der Kapsel vergingen nach Desintegration im Fundus 6,5 ± 2,5 min, im Antrum 3 ± 2 min, wohingegen in dem Einzelfall des Zerfalls im Dünndarm ein unmittelbares Auftauchen des Koffeins beobachtet wurde. Die Studie lieferte wertvolle erste Einblicke und bestätigte den Nutzen der Kombination von MRT Bildgebung und der Untersuchung der Pharmakokinetik von Koffein im Speichel. Es liegen nun mehrere etablierte Methodiken vor, um zu untersuchen wann und wo konventionelle, aber auch magensaftresistente oder zeitlich verzögert freisetzende Kapseln freisetzen und welchen pharmakokinetischen Effekt dies gegebenenfalls hat.
Über die Kernthemen der Arbeit hinaus wurden diverse andere Projekte insbesondere mithilfe Telemetrischer Kapseln in Human- und Tierstudien erfolgreich mitbearbeitet. Darüber hinaus fanden die im Rahmen dieser Arbeit erhobenen Daten bereits Eingang in verschiedene biorelevante Freisetzungsapparaturen und PBPK Modelle. Bisher sind jedoch weder der Einfluss der gastrointestinalen Volumina noch des Transits abschließend geklärt. Die hoch wirkstoffabhängigen Einflüsse, werden in weiteren Studien weiter wirkstoffspezifisch zu untersuchen sein, oder können mithilfe der Implementierung der gewonnenen Daten in in vitro und in silico Tools simuliert werden.
Abstract
Neutrophils are the most abundant leukocytes in circulation playing a key role in acute inflammation during microbial infections. Phagocytosis, one of the crucial defence mechanisms of neutrophils against pathogens, is amplified by chemotactic leukotriene (LT)B4, which is biosynthesized via 5‐lipoxygenase (5‐LOX). However, extensive liberation of LTB4 can be destructive by over‐intensifying the inflammatory process. While enzymatic biosynthesis of LTB4 is well characterized, less is known about molecular mechanisms that activate 5‐LOX and lead to LTB4 formation during host–pathogen interactions. Here, we investigated the ability of the common opportunistic fungal pathogen Candida albicans to induce LTB4 formation in neutrophils, and elucidated pathogen‐mediated drivers and cellular processes that activate this pathway. We revealed that C. albicans‐induced LTB4 biosynthesis requires both the morphological transition from yeast cells to hyphae and the expression of hyphae‐associated genes, as exclusively viable hyphae or yeast‐locked mutant cells expressing hyphae‐associated genes stimulated 5‐LOX by [Ca2+]i mobilization and p38 MAPK activation. LTB4 biosynthesis was orchestrated by synergistic activation of dectin‐1 and Toll‐like receptor 2, and corresponding signaling via SYK and MYD88, respectively. Conclusively, we report hyphae‐specific induction of LTB4 biosynthesis in human neutrophils. This highlights an expanding role of neutrophils during inflammatory processes in the response to C. albicans infections.
Abstract
The KV7 potassium channel openers flupirtine and retigabine have been valuable options in the therapy of pain and epilepsy. However, as a result of adverse reactions, both drugs are currently no longer in therapeutic use. The flupirtine‐induced liver injury and the retigabine linked tissue discolouration do not appear related at first glance; nevertheless, both events can be attributed to the triaminoaryl scaffold, which is affected by oxidation leading to elusive reactive quinone diimine or azaquinone diimine metabolites. Since the mechanism of action, i. e. KV7 channel opening, seems not to be involved in toxicity, this study aimed to further develop safer replacements for flupirtine and retigabine. In a ligand‐based design strategy, replacing amino substituents of the triaminoaryl core with alkyl substituents led to carba analogues with improved oxidation resistance and negligible risk of quinoid metabolite formation. In addition to these improved safety features, some of the novel analogues exhibited significantly improved KV7.2/3 channel opening activity, indicated by an up to 13‐fold increase in potency and an efficacy of up to 176 % compared to flupirtine, thus being attractive candidates for further development.
Transmucosal drug delivery systems can be an attractive alternative to conventional oral dosage forms such as tablets. There are numerous in vitro methods to estimate the behavior of mucoadhesive dosage forms in vivo. In this work, a tensile test system was used to measure the mucoadhesion of polyvinyl alcohol films. An in vitro screening of potential influencing variables was performed on biomimetic agar/mucin gels. Among the test device-specific factors, contact time and withdrawal speed were identified as influencing parameters. In addition, influencing factors such as the sample area, which showed a linear relationship in relation to the resulting work, and the liquid addition, which led to an abrupt decrease in adhesion, could be identified. The influence of tissue preparation was investigated in ex vivo experiments on porcine small intestinal tissue. It was found that lower values of Fmax and Wad were obtained on processed and fresh tissue than on processed and thawed tissue. Film adhesion on fresh, unprocessed tissue was lowest in most of the animals tested. Comparison of ex vivo measurements on porcine small intestinal tissue with in vitro measurements on agar/mucin gels illustrates the inter- and intra-individual variability of biological tissue.
Heparin is an anticoagulant drug. It is important in the treatment of deep vein thrombosis,pulmonary embolism and during surgeries. Heparin-induced thrombocytopenia (HIT) is a severe adverse reaction caused by the formation of ultralarge complexes of platelet factor 4 (PF4) with unfractionated heparin (UFH). It can lead to limb loss or fatal events like stroke, myocardial infarction or pulmonary embolism. HIT has an incidence of about 3% in patients receiving anticoagulative heparin treatment. PF4 is a tetrameric protein, released from the α-granules of platelets upon activation. PF4 is known to form antigenic complexes with UFH accompanied by structural changes of PF4. In this thesis, the size and size distribution of PF4 and PF4/heparin complexes were analyzed using asymmetrical flow field-flow-fractionation (AF4), photon correlation spectroscopy (PCS) and atomic force microscopy (AFM). PF4 tends to form auto-aggregates and to adsorb to different surfaces, including regenerated cellulose, polyethersulfone, quartz and glass. The aggregates are less pronounced in solutions at isotonic NaCl concentration. Arginine and Tween 20 were identified as possible ingredients to hinder the auto-aggregation of PF4. Also, it is shown by combining circular dichroism (CD) spectroscopy, atomic force microscopy (AFM) and isothermal titration calorimetry (ITC) with UFH and defined chain length (16-, 8-, 6-, 5-mer) heparins that structural changes (i.e., increase in β-sheets) alone are not sufficient to induce antigenicity. While UFH, 16-, 8-, and 6-mer heparins all induced an increase in the antiparallel β-sheet content to > 30% (as determined by CD spectroscopy), complex antigenicity as measured by anti-PF4/heparin antibody binding in an enzyme-linked immunosorbent assay (EIA) was only induced by UFH and 16-mer heparin. Fondaparinux (5-mer heparin), which forms in vitro non-antigenic complexes with PF4, did not induce structural changes of PF4. Interestingly, the structural changes induced by antigenic UFH and 16-mer heparin but not by non-antigenic shorter heparins were reversible at higher heparin concentrations. Furthermore, the complexes formed by PF4 with longer heparins were larger than those formed with shorter heparins as shown by atomic force microscopy (AFM). UFH, HO16 and HO08 are able to form ultralarge multimolecular complexes with PF4. ITC data indicated strong electrostatic interactions and energetically unfavorable conformational changes of PF4 with longer heparins, while for the short heparins, favorable conformational changes in the structure of PF4 are induced. This explains the reversibility of the structural changes seen for UFH and HO16 upon addition of an over-saturating amount of heparin. Finally, using differential scanning calorimetry (DSC) the thermal stability of PF4 and PF4/heparin complexes was assessed. Despite its tendency to form auto-aggregates, PF4 is a heat-stable protein. This stability is, length dependently, even increased in complex with heparins. This work shows important differences in the binding between PF4 and heparins of different chain length and might be relevant for the understanding of other biological functions of heparins (e.g., involvement in allergic and inflammatory reactions).
The biodiversity of marine microorganisms opens a promising potential for the discovery of new technical enzymes. During this study a characterization of marine microorganisms, isolated from Arctic or Antarctic ice, sea water or sediment from the ocean was performed based on a comprehensive strain collection at the Alfred-Wegener-Institut für Polar- und Meeresforschung. These marine psychrophilic bacteria indicated a wide spectrum of extracellular cold-active enzymes. 16S rRNA sequencing revealed that many of these psychrophilic bacteria represent new species. Characterization of selected isolates by means of transmission electron or raster electron microscopy showed remarkably pleomorphic cellular structures throughout their growth. The major part of this thesis focuses on a marine Antarctic, psychrophilic bacterium (strain ANT/505) isolated from sea ice covered surface water from the Southern Ocean, which was identified to express a very uncommon enzymatic activity for the marine environment, namely a pectinolytic activity. The sequencing of the 16S rRNA of isolate ANT/505 and biochemical tests indicated a taxonomical affiliation to the specie Pseudoalteromonas haloplanktis. The supernatant of this bacterial isolate showed after growth on citrus pectin three different pectinolytic activities. By activity screening of a genomic DNA library of isolate ANT/505 in Escherichia coli, two different pectinolytic clones could be isolated. Subcloning and sequencing revealed two open reading frames of 1671 and 1968 nt corresponding to proteins of 68 and 75 kDa. The deduced amino acid sequence of the two orfs showed homology to pectate lyases from Erwinia chrysanthemi and Aspergillus nidulans. The pectate lyases contain signal peptides of 17 and 26 amino acids length that were correctly processed after overexpression in E. coli BL21. Both enzymes were purified by anionic exchange chromatography. Maximal enzymatic activities for both pectate lyases were observed at a temperature of 30°C and a pH range of 9-10. The Km values of both lyases for pectate and citrus pectin were 1 g⋅l-1 and 5 g⋅l-1, respectively. Calcium was required for activity on pectic substrates, while the addition of 1 mM ethylenediaminetetraacetic acid (EDTA) resulted in complete inhibition of the enzymes. These two cold-adapted enzymes represent the first pectate lyases isolated and characterized from a marine bacterium. Further cloning and sequence analyses revealed that PelA from P. haloplanktis is an exceptionally big bifunctional enzyme featuring pectate lyase and pectin methylesterase activity. The deduced amino acid sequence of the pectin methylesterase domain showed homology to group I pectin methylesterases from Erwinia chrysanthemi and Erwinia carotovora. The pectin methylesterase domain of PelA was found to show highest homology to a potential pectin methylesterase from Saccharophagus degradans strain MD2-40. Maximum pectin methylesterase activity of PelA was detected at a pH of 7.5 and a maximum temperature of 30°C. This cold-adapted enzyme revealed high remaining pectin methylesterase activity at low temperatures around 5°C and was quickly unstabilized at temperatures above 45°C. The analysis of the localization of the two pectinolytic genes on the genome of P. haloplanktis ANT/505 revelaed that these pectinase genes are expressed from independent cistrons, which are not clustered but located at distant positions on chromosome I of the P. haloplanktis genome. It was found that the transcription of both pectinase genes is induced by the presence of pectin. By means of primer extension the promoter regions of both cistrons were detected.
Characterization, Chemical Compounds and Biological Activities of Marrubium vulgare L. Essential Oil

As consumer trends shift towards more natural and ecological consumption patterns, industrialists are actively working towards substituting synthetic chemicals with natural and vegan products that contain bioactive properties. Thus, considering the shifts in customer demand and the growing concern around vegetable sourced productions, this work aims to contribute to the valorization of aromatic and medicinal Moroccan plants. By focusing on the Marrubium vulgare L. species, our objective is to carry out a physicochemical characterization to determine its chemical composition and biological activities. The volatile fraction collected by hydrodistillation (0.61%) and analyzed by GC-MS (gas chromatography coupled to mass spectrometry) contains five main compounds: 3-Thujanone, Eugenol, Topanol, Menthone and Piperitone. The antioxidant activity has been estimated by applying the DPPH (1,1-diphenyl-2-picrylhydrazyl) free radical scavenging test and the ferric reducing antioxidant power (FRAP). The values of inhibitory concentration prove that our oil is a good antioxidant, with values of IC50 = 1.136 mg/mL and IC50 = 2.998 mg/mL, respectively, for the DPPH and FRAP tests. The results of the antifungal activity indicate a significant inhibition of mycelial growth for both tested molds, as well as a total inhibition of spore production at a concentration of 0.25 µL/mL.
Die orale Einnahme stellt für Patienten die einfachste und unkomplizierteste Möglichkeit dar, ein Arzneimittel zu applizieren und ist das angestrebte Ziel der Arzneimittelentwicklung. Dem entgegen stehen jedoch die evolutionär entstandenen Möglichkeiten des Körpers, aufgenommene Fremdstoffe zu inaktivieren und zu eliminieren. Ein Zusammenspiel aus anatomischen Gegebenheiten und den Enzymen des Fremdstoffmetabolismus sorgt dafür, dass ein Teil der oral applizierten Dosis bereits verstoffwechselt wird, bevor er über das arterielle System an den Wirkort gelangen kann (first-pass-Effekt). Als Ort dieses Metabolismus wurde, neben der Leber, auch der Darm identifiziert. Um das Ausmaß des first- pass-Effektes abschätzen zu können, werden Daten über den Gehalt der arzneistoffmetabolisierenden Enzyme in diesen Organen benötigt. Als Methode der Wahl bietet sich dazu die LC-MS/MS an, da mit ihr verschiedene Enzyme in einem analytischen Lauf bestimmt werden können und sie sich durch eine hohe Empfindlichkeit, Reproduzierbarkeit und Spezifität auszeichnet.
Mit der vorliegenden Arbeit wurde das analytische Spektrum der bisher publizierten Methoden zur Bestimmung von CYP- und UGT-Enzymen erweitert. Mit der neuen Methode können nun zwei Carboxylesterasen, 17 CYP-Enzyme und fünf UGT-Enzyme quantifiziert werden. Weiterhin wurde die Methode anhand von Richtlinien für bioanalytische Methoden umfassend validiert. Durch die Verwendung von rekombinant hergestellten arzneistoffmetabolisierenden Enzymen konnte der gesamte analytische Prozess, von der Probe bis zum Endergebnis, erstmalig umfassend charakterisiert werden. Dabei zeigte sich eine, für einen derart komplexen Prozess bemerkenswerte Präzision von maximal 15,5% Variation nach sechsmaliger Durchführung.
Die entwickelte Methode wurde dann auf gepaarte Proben aus Leber und Jejunum von elf gesunden Organspendern angewendet. Im Jejunum wurden CES1, CES2, CYP2C9, CYP2C18, CYP2C19, CYP2D6, CYP2J2, CYPA4, CYP3A5, CYP4F2, CYP4F12, UGT1A1, UGT1A3, UGT2B7 und UGT2B17 gefunden. In der Leber konnten alle untersuchten Enzyme (CES1, CES2, CYP1A1, CYP1A2, CYP2A6, CYP2B6, CYP2C8, CYP2C9, CYP2C18, CYP2C19, CYP2D6, CYP2E1, CYP2J2, CYP3A4, CYP3A5, CYP3A7, CYP4F2, CYPF12, UGT1A1, UGT1A3, UGT2B7, UGT2B15 und UGT2B17), bis auf CYP4A11 nachgewiesen werden. Für einige Enzyme (CES2, CYP2C18, CYP2C19, CYP2J2, CYP3A4, CYP4F2, CYP4F12) wurden im Jejunum Enzymgehalte gemessen, die mit denen in der Leber vergleichbar sind, was noch einmal unterstreicht, dass der Darm auch als klinisch relevanter Ort des Arzneistoffmetabolismus betrachtet werden muss. Auffällig war hier zudem die deutlich höhere Variabilität in den Darmproben, verglichen mit den Leberproben, die ihre Ursache in Umwelteinflüssen oder dem Mikrobiom des Darms haben könnten. Außerdem wurde die Expression der zugehörigen Gene mittels quantitativer real-time PCR untersucht. Hier bestand nur in einigen Fällen eine signifikante Korrelation zwischen Genexpression und Proteingehalt, was für zwischengeschaltete regulatorische Mechanismen spricht.
Weiterhin wurden mit dieser Methode Leberproben einer Kohorte von Patienten mit Krankheitsbildern, die mit einer Einschränkung der Leberfunktion einhergehen, untersucht. Dazu wurden die Patienten nach der verbleibenden Leberfunktion (Child-Pugh-Score) und nach der zugrundeliegenden Erkrankung eingeteilt. Es zeigt sich eine generelle Abnahme des Gehaltes an arzneistoffmetabolisierenden Enzymen mit fortschreitender Verschlechterung der Leberfunktion, wobei sich CYP2E1 als besonders anfällig erwiesen hat und bereits in Child- Pugh-Klasse A signifikant erniedrigt war. Bei den verschiedenen Erkrankungen zeigt sich ein uneinheitliches Bild, die prozentuale Verteilung der Enzyme ist jedoch bei allen Erkrankungen gegenüber den gesunden Kontrollproben verändert.
Über die Regulation der Expression von arzneistoffmetabolisierenden Enzymen ist bisher noch wenig bekannt. Es gibt aber Hinweise aus der Literatur, dass bestimmte nukleäre Rezeptoren an der Regulation der Enzyme beteiligt sein können. Deshalb wurde eine LC-MS/MS-basierte targeted-proteomics-Methode zur Quantifizierung von nukleären Rezeptoren in Darm- und Lebergewebe entwickelt und validiert. Im Gewebe konnten nur AhR und HNF4α nachgewiesen werden, da die Empfindlichkeit des verwendeten experimentellen Ansatzes vermutlich nicht ausreichend ist. Dabei war HNF4α in Darmgewebe deutlich höher exprimiert als AhR. Außerdem wurde die Expression der nukleären Rezeptoren auf Genebene durch quantitative real-time PCR untersucht. Dabei wurde eine höhere Expression von CAR in der Leber gefunden, während PXR in Darm stärker exprimiert wird. Dies entspricht den Erkenntnissen aus der Literatur, nach denen CAR einen regulatorischen Effekt auf arzneistoffmetabolisierende Enzyme in der Leber hat, während dies für PXR in Darm zutrifft. Diese Arbeit kann einen Beitrag zum weitergehenden Verständnis der Regulation von arzneistoffmetabolisierenden Enzymen durch nukleäre Rezeptoren beitragen.
Bei allen diesen Arbeiten gilt es zu beachten, dass das Vorhandensein eines Proteins nicht zwangsläufig mit seiner Aktivität gleichzusetzen ist. Jedoch zeigen zahlreiche Beispiele aus der Literatur, dass sich mit den Daten aus Proteomics-Studien PBPK-Modelle aufstellen lassen, die die in klinischen Studien erhobenen Daten mit beeindruckender Genauigkeit reproduzieren können.
Die Verlängerung des Magenaufenthalts von oralen Arzneiformen steht seit mehr als 30 Jahren im Fokus internationaler Forschungsgruppen. Trotz der Vermarktung diverser Systeme gelang es bislang nicht, eine sichere und reproduzierbare Gastroretention von Arzneiformen zu realisieren. Dies würde jedoch enorme Möglichkeiten für die Therapie mit oral applizierten Arzneimitteln mit sich bringen. Die Reduktion der Einnahmefrequenz, das Vermeiden von Plasmaspiegelspitzen sowie die gesteigerte Patientenadhärenz sind nur einige der denkbaren Vorteile. Die größte Hürde gastroretentiver Systeme ist dabei die Motilität des menschlichen Magens. Starke Kontraktionswellen sind für eine rasche Entleerung insbesondere unter Nüchternbedingungen verantwortlich. Daneben kommt es zu höchsten Belastungen auf Arzneiformen, was wiederum die Wirkstofffreisetzung beschleunigen kann, mit drastischen Folgen für den Patienten. In der präklinischen Testung neu entwickelter Systeme fehlt häufig der Bezug zur Physiologie des Magens und die Vorhersagekraft von Freisetzungstests ist dementsprechend gering. Ziel der Arbeit war daher die Charakterisierung der relevanten Parameter im Magen im Rahmen einer Humanstudie. Die aus dieser Humanstudie gewonnenen Daten zu pH-Werten, Temperaturen und insbesondere Drücken im Magen sollten anschließend genutzt werden, um die im Arbeitskreis verfügbaren, biorelevanten Freisetzungsmodelle weiterzuentwickeln. Abschließend sollten verschiedene, kommerziell erhältliche gastroretentive Arzneiformen unter Berücksichtigung der Magenphysiologie auf ihr Freisetzungsverhalten getestet werden. Die Ergebnisse der Humanstudie zeigten die enorme Abhängigkeit der Magenaufenthaltszeit einer telemetrischen Kapsel vom prandialen Status der Probanden. Nach Einnahme der Standardmahlzeit, wie sie in klinischen Studien zu Nahrungsmitteleffekten Verwendung findet, kam es zu Magentransitzeiten von über 20 h. Dagegen wurde die Kapsel unter Nüchternbedingungen spätestens nach 2,7 h aus dem Magen entleert. Die intragastralen Drücke nach postprandialer Einnahme der Kapsel betrugen mindestens 240 mbar und waren aufgrund des verlängerten Magenaufenthalts deutlich zahlreicher im Vergleich zur Nüchterneinnahme. Die Ergebnisse der In vitro-Untersuchungen zeigten, dass die herkömmlich verwendeten Freisetzungstestgeräte nicht in der Lage sind, biorelevante Belastungen auf eine telemetrische Kapsel auszuüben. Maximale Drücke von 14 mbar waren im eintauchenden Zylinder zu beobachten, welche wir jedoch auf den hydrostatischen Druck beim Eintauchen zurückführen konnten. Im Gegensatz dazu waren wir mit Hilfe unserer neuartigen In vitro-Freisetzungsmodelle in der Lage, vollständige Druckprofile nachzustellen, wie sie auch in vivo beobachtbar waren. Die Freisetzungsuntersuchungen der gastroretentiven Präparate Glumetza® 1000 und Madopar® Depot unter biorelevanten Bedingungen offenbarten die extreme Drucksensitivität dieser Systeme. Hierfür definierten wir auf Basis der In vivo-Daten drei realistische Druckprofile und stellten diese in vitro nach. Früh auftretende, leichte Belastungen während der Freisetzungstests führten bei der flotierenden Arzneiform Madopar® Depot bereits zur vollständigen Wirkstofffreisetzung. Glumetza® 1000 schien abhängig vom Quellungszustand auf die Belastungen zu reagieren, wobei spätestens stärkere Belastungen nach 6 h zur vollständigen Freisetzung des Wirkstoffs führten. Auf Basis dieser Ergebnisse ist anzuzweifeln, dass die bislang erhältlichen gastroretentiven Systeme über einen längeren Zeitraum im Magen intakt bleiben und kontrolliert ihren Wirkstoff freisetzen. Daneben können die entwickelten Testmethoden dazu genutzt werden, um die Entwicklung neuartiger gastroretentiver Systeme voranzutreiben.
Summary Cyanobacteria are a diverse and ancient group of photosynthetic prokaryotic organisms that can inhabit a wide range of environments including extreme conditions such as hot springs, desert soils and the Antarctic. They are abundant producers of natural products well recognized for their bioactivity and utility in drug discovery and biotechnology applications. Novel intracellular and extracellular compounds from various cultured and field cyanobacteria with diverse biological activities and a wide range of chemical classes have considerable potential for development of pharmaceuticals and other biomedical applications. However, cyanobacteria are still viewed as unexplored source of potential drugs. Especially the collections of cyanobacterial strains from South East Asia where biodiversity is high are still largely unexplored. Thus, we investigated twelve soil cyanobacterial strains isolated from soil samples collected from rice, cotton, and coffee fields in Dak Lak province of Vietnam and one marine strain, Lyngbya majuscula collected from Khanh Hoa province of Vietnam for the search for new compounds with antimicrobial and cytotoxic activities. From the 12 soil cyanobacterial strains, 48 extracts prepared with n-hexane, methanol, and water for biomasses and ethyl acetate for growth media were screened for antibacterial activity against Gram-positive bacteria (Bacillus subtilis ATCC 6051 and Staphylococcus aureus ATCC 6538) and Gram-negative bacteria (Escherichia coli ATCC 11229, Pseudomonas aeruginosa ATCC 27853). Of 48 extracts, 47.92% and 45.83% showed activity against Bacillus subtilis and Staphylococcus aureus, respectively, while 22.92% and 6.25% exhibited activity against Escherichia coli and Pseudomonas aeruginosa, respectively. All investigated cyanobacteria (12/12) showed antibacterial activity to at least one of the test organisms applied. Among the active extracts, extracts obtained from 5 cyanobacterial strains, Westiellopsis sp. VN, Calothrix javanica, Scytonema ocellatum, Anabaena sp. and Nostoc sp. showed the highest strength and range of antibacterial activity and therefore were selected for chemical investigation with an emphasis on the isolation and structure elucidation of antimicrobial compounds. Bioassay-guided fractionation of the methanol extract prepared from biomass of Westiellopsis sp. VN by silica gel chromatography, followed by sephadex LH-20 chromatography and reversed-phase HPLC led to isolation and identification of 6 compounds as ambiguine D isonitrile, ambiguine B isonitrile, dechloro-ambiguine B isonitrile, fischerellin A, hydroxy-eicosatetraenoic acid and methoxy-nonadecadienoic acid. Identification of these active compounds was established by direct comparison of our spectroscopic data, including 1H NMR and HR-ESI-MS with those reported in the literature. All these compounds showed biological activity. The identification of fatty acids and other volatile components by GS-MS in the active MeOH fraction obtained from EtOAc extract of growth medium was done before commencing further fractionation processes. Culture optimization of Westiellopsis sp.VN showed that NaNO3 deficiency increased accumulation of antimicrobial compounds. Biosynthesis of antimicrobial compounds increased over cultivation time resulting in increased diameter of inhibition zone of the methanol extract towards the end of the 7-to 8- week growth period, but the most clear inhibition zone of this extract was detected after cultivation time of 8 weeks. Bioassay-guided fractionation of the methanol extract prepared from biomass of either Calothrix javanica by C18 chromatography followed by reversed-phase HPLC or Scytonema ocellatum by C18 chromatography followed by silica gel chromatography and reversed-phase HPLC led to isolation and structure elucidation of new cyclic peptide named daklakapeptin. Structure of daklakapeptin was elucidated by exhaustive 1D (1H) and 2D (COSY, TOCSY, NOESY, HMQC, HMBC) NMR spectroscopy in combination with HR-ESI-MS. Daklakapeptin was found to have totally 12 residues including 6 proteinogenic amino acids (Pro, Tyr, Ile, Leu, Gln, Thr), 4 complexes (X,Y,T,Z) and the methyl derivative of Ile. The exact sequence of daklakapeptin is shown in following figure with X: (CH3)2CHCH2CH2CH(NH-)CH2CO-, Y:(CH3)2CHCH(OH)CH(NH-)CO-, T: HOCH2CH2CH(NH-)CO-, Z: HOCH2CHOHCH(NH-)CO- This new cyclic peptide exhibited antibacterial activity against Staphylococcus aureus with diameter of inhibition zone of 12.5 mm in concentration of 200 mg/disc. Further test for activity to other bacteria and for cytotoxic activity are in progress. Using reversed-phase HPLC to separate compounds in the crude ethyl acetate extract obtained from culture medium of Anabaena sp. led to isolation and structure elucidation of flourensadiol. The structure of flourensadiol was established using an extensive array of 1D (1H, 13C, DEPT-135) and 2D (HMQC, COSY, HMBC) NMR and HR-ESI-MS experiments. Flourensadiol was isolated previously from the common western shrub Flourensia cernua. However, only MS, IR, and proton NMR data but no reports on biological activity were available. In this study, we report the complete NMR data of flourensadiol for the first time. Flourensadiol was found to be very strong antibacterial active against Escherichia coli with diameter of inhibition zone of 20.0 mm in concentration of 200 mg/disc. Further test for activity to other bacteria and cytotoxic activity are in progress. Bioassay-guided fractionation of the methanol extract from biomass of Nostoc sp. by silica gel chromatography followed by C18 chromatography and reversed phase HPLC led to isolation of the active fraction NsF2 which exhibited antibacterial activity against Staphylococcus aureus with diameter of inhibition zone of 10.0 mm in concentration of 500 mg/disc. The low resolution ESI-MS of fraction NsF2 showed signal at m/z 426 [M+H]+. The NMR and MS characterization of compounds in fraction NsF2 is in progress. Bioassay-guided fractionation of the methanol extract prepared from biomass of marine cyanobacterium Lyngbya majuscula collected from Khanh Hoa province of Vietnam by various chromatographic methods (CC, PTLC, HPLC) afforded 3 cytotoxic compounds anhydrodebromoaplysiatoxin, debromoaplysiatoxin, and anhydroaplysiatoxin. Identification of these cytotoxic compounds was established by direct comparison of our spectroscopic data, including (1H, 13C) NMR and HR-ESI-MS with those reported in the literature. In our study, debromoaplysiatoxin and anhydroaplysiatoxin exhibited cytotoxic activity against bladder cancer cell line 5637 with IC50 of 86 ng/ml and 40 ng/ml, respectively but anhydrodebromoaplysiatoxin was not yet tested for cytotoxic activity. The identification of fatty acids by GS-MS technique in the n-hexane extract obtained from biomass of this marine cyanobacterium was undertaken before commencing further fractionation processes. The presented results prove that soil cyanobacteria are a promising source to yield chemical and pharmaceutical interesting compounds.
The aim of the present dissertation was to investigate the biological and chemical potential of two European mushroom species: Fomitopsis betulina and Calvatia gigantea. For this purpose, different extracts of both fungi were tested for: antimicrobial, antifungal, cytotoxic, in vitro wound healing, and anti-adhesive properties. Bioassay-guided fractionation led to the isolation of bioactive compounds, altogether 20 compounds were isolated and identified. The compounds were obtained from the ethyl acetate extracts, they included triterpenes, sterols and aromatic compounds. The separated substances from both fungi were proved for biological activities, some of them showed antimicrobial and cytotoxic activities.
Chemistry and biology of Phenolics isolated from Myricaria germanica (L.) Desv. (Tamaricaceae)

In accordance with the recent worldwide interest in plant phenolics, which emerges from their broad range of biological activities, particular emphasis has been focused, in the present thesis, on the constitutive phenolics of the extract of Myricaria germanica (L.) Desv. (Tamaricaceae). During the current thesis twenty phenolics (1 – 20) were isolated and identified from the aqueous/ethanol extract of the whole Myricaria germanica plant. The isolates include four hitherto unknown natural phenolics (2, 10, 12 and 20). Also, the cytotoxic activities of M. germanica extract, column fractions, and one new natural isolate against three different solid tumor cell lines, namely, breast cancer (MCF-7), prostate (PC-3), and liver (Huh-7) cancer cell using SRB viability assay have been investigated and first insights into mode of action have been obtained.
Chemosymbiosis in marine bivalves – unravelling host-symbiont interactions and symbiotic adaptions

Symbiosis essentially forms the cornerstone of complex life on earth. Spearheading
symbiosis research in the last few decades include the exploration of diverse mutualistic
animal-bacterial associations from marine habitats. Yet, many facets of symbiotic
associations remain under-examined. Here we investigated marine bivalves of the genera
Bathymodiolus and Codakia, inhabiting hydrothermal vents and shallow water
ecosystems, respectively, and their bacterial symbionts. The symbionts reside
intracellularly within gill epithelia and supply their host with chemoautotrophically fixed
carbon. They oxidize reduced substrates like sulfide (thiotrophic symbionts) and methane
(methanotrophic symbionts) from surrounding fluids for energy generation. The nature of
interactions between host and symbiont at the metabolic and physical level, as well as
between the holobiont and its environment remain poorly understood. In vitro cultivations
of both symbiont and host are difficult till date, hampering the feasibility of targeted
molecular investigations.
We bypassed culture-based experiments by proteogenomically investigating physically
separated fractions of host and symbiont cell components for the bivalves Bathymodiolus
azoricus, Bathymodiolus thermophilus and Codakia orbicularis. Using these
enrichments, we sequenced the symbionts’ genomes and established semi-quantitative
host-symbiont (meta-) proteomic profiles. This combined approach enabled us to resolve
symbiosis-relevant metabolic pathways and adaptations, detect molecular factors
mediating physical interactions amongst partners and to understand the association of
symbiotic traits with the environmental factors prevailing within habitats of the respective
bivalve.
Our results revealed intricate metabolic interdependence between the symbiotic partners.
In Bathymodiolus, these metabolic interactions included (1) the concentration of essential
substrates like CO2 and thiosulfate by the host for the thiotrophic symbiont, and (2) the
host’s replenishment of essential TCA cycle intermediates for the thiotroph that lacks
biosynthetic enzymes for these metabolites. In exchange (3), the thiotroph compensates
the host’s putative deficiency in amino acid and cofactor biosynthesis by cycling aminoacids
derived from imported precursors back to the host. In case of Codakia orbicularis,
the symbionts may metabolically supplement their host with N-compounds derived from
fixation of molecular nitrogen, a trait that was hitherto unknown in chemosynthetic
thiotrophic symbionts.
Individual proteogenomic investigations of the bivalves Bathymodiolus azoricus and
Bathymodiolus thermophilus showed that their symbionts are able to exploit a multitude
of energy sources like sulfide, thiosulfate, methane and hydrogen to fuel chemosynthesis.
The bivalves and their thiotrophic symbionts, however, are particularly adapted to
thiosulfate-utilization, as indicated by mitochondrial production and concentration of
thiosulfate by host and dominant expression of thiosulfate oxidation enzymes in the
symbiont. This may be advantageous, because thiosulfate is less toxic to the host than
sulfide. The central metabolic pathways for energy generation, carbon and nitrogen
assimilation and amino acid biosynthesis in thiotrophic symbionts of both Bathymodiolus
host species are highly conserved. Expression levels of these pathways do, however, vary
between symbionts of both species, indicating differential regulation of enzyme synthesis,
possibly to accommodate differences in host morphology and environmental factors.
Systematic comparison of symbiont-containing and symbiont-free sample types within
and between B. azoricus and B. thermophilus revealed the presence of ‘symbiosisspecific’
features allowing direct host-symbiont physical interactions. Host proteins
engaged in symbiosis-specific functions include 1) a large repertoire of host digestive
enzymes predominant in the gill, possibly facilitating symbiont population control and
carbon acquisition via direct enzymatic digestion of symbiont cells and 2) a set of host
pattern-recognition receptors, which may enable the host to selectively recognize
pathogens or even symbionts “ripe” for consumption. Symbiont proteins engaged in
symbiosis-specific interactions included 3) an enormous set of adhesins and toxins,
putatively involved in symbiont colonization, persistence and host-feeding.
Bathymodiolus symbionts also possess repertoires of CRISPR-Cas and restrictionmodification
genes for phage defense that are unusually large for intracellular symbionts.
Genomic and proteomic comparisons of thiotrophic symbionts of distinct Bathymodiolus
host species from different vent sites revealed a conserved core genome but divergent
accessory genomes. The B. thermophilus thiotroph’s accessory genome was notably more
enriched in genes encoding adhesins, toxins and phage defense proteins than that of other
Bathymodiolus symbionts. Phylogenetic analyses suggest that this enrichment possibly
resulted from horizontal gene acquisition followed by multiple internal gene duplication
events. In others symbionts, these gene functions may be substituted by alternate
mechanisms or may not be required at all: The methanotrophic symbionts of B. azoricus,
for example, has the genetic potential to supplement phage defense functions. Thus, the
accessory genomes of Bathymodiolus symbionts are species- or habitat-associated,
possibly facilitating adaptation of the bivalves to their respective micro- and macroenvironments.
In support of this, we show that symbiont biomass in B. thermophilus,
which hosts only one thiotrophic symbiont phylotype, is considerably higher than in B.
azoricus that hosts thiotrophic and methanotrophic symbionts. This suggests that different
symbiont compositions in each species produce distinct microenvironments within the
holobiont.
Our study presents an exhaustive assessment of the genes and proteins involved in this
bivalve-microbe interaction, hinting at intimate host-symbiont interdependencies and
symbiotic crosstalk between partners. The findings open novel prospects for
microbiologists with regard to mechanisms of host-symbiont interplay within highly
specialized niches, origin and distribution of prokaryote-eukaryote interaction factors
across both mutualistic and pathogenic associations.
Multi-drug resistant (MDR), gram-negative Enterobacteriaceae, such as Escherichia coli (E. coli) limit therapeutic options and increase morbidity, mortality, and treatment costs worldwide. They pose a serious burden on healthcare systems, especially in developing countries like Rwanda. Several studies have shown the effects caused by the global spread of extended-spectrum beta-lactamase (ESBL)-producing E. coli. However, limited data is available on transmission dynamics of these pathogens and the mobile elements they carry in the context of clinical and community locations in Sub-Saharan Africa. Here, we examined 120 ESBL-producing E. coli strains from patients hospitalized in the University Teaching Hospital of Butare (Rwanda), their attending caregivers as well as associated community members and livestock. Based on whole-genome analysis, the genetic diversification and phylogenetics were assessed. Moreover, the content of carried plasmids was characterized and investigated for putative transmission among strains, and for their potential role as drivers for the spread of antibiotic resistance. We show that among the 30 different sequence types (ST) detected were the pandemic clonal lineages ST131, ST648 and ST410, which combine high-level antimicrobial resistance with virulence. In addition to the frequently found resistance genes blaCTX–M–15, tet(34), and aph(6)-Id, we identified csg genes, which are required for curli fiber synthesis and thus biofilm formation. Numerous strains harbored multiple virulence-associated genes (VAGs) including pap (P fimbriae adhesion cluster), fim (type I fimbriae) and chu (Chu heme uptake system). Furthermore, we found phylogenetic relationships among strains from patients and their caregivers or related community members and animals, which indicates transmission of pathogens. Also, we demonstrated the presence and potential transfer of identical/similar ESBL-plasmids in different strains from the Rwandan setting and when compared to an external plasmid. This study highlights the circulation of clinically relevant, pathogenic ESBL-producing E. coli among patients, caregivers and the community in Rwanda. Combining antimicrobial resistance with virulence in addition to the putative exchange of mobile genetic elements among bacterial pathogens poses a significant risk around the world.
In the search for new antifungal agents, this study dealt with the antimicrobial screening, extraction, isolation, structural elucidation as well as selective biological investigations of the isolated compounds. In addition, the impact of the culture conditions on growth and on biosynthesis of bioactive compounds was also studied. Besides, selective cyanobacteria were axenized and the taxonomy as well as the genetic relationship of axenic cyanobacteria that produced bioactive compounds with some other cyanobacteria was identified basing on the 16S rRNA gene sequences. 22 Vietnamese and 6 German cyanobacterial strains were screened for their antifungal activity using the agar diffusion assay. Among them, the MeOH/water extract from the biomass obtained from a laboratory culture of strain Bio 33, isolated from the Baltic Sea near Rügen Island, exhibited a specific antifungal activity against Candida maltosa and others human pathogenous fungi such as Candida albicans, Candida krusei, Aspergillus fumigatus, Microsporum gypseum, Trichophyton rubrum and Mucor sp. Besides, it was very impressed that extracts of strain Bio 33 showed no antibacterial activity against Escherichia coli, Pseudomonas aeruginosa, Bacillus subtilis, and Staphylococcus aureus. The taxonomy basing on 16S rRNA gene sequence of the axenic Bio 33 identified this strain as Anabaena cylindrica species. As a result of the bioassay-guided fractionation of the crude MeOH/water extract, four new lipopeptides, named balticidins A – D, were isolated. These lipopeptides represent a new structural type with the co-occurrence of a glycosylated cyclic peptide, a fatty acid containing chlorine and a disaccharide moiety. The main active fraction isolated from the MeOH/water extract of the biomass of Bio 33 which contains the four lipopeptides exhibited only marginal cytotoxic activity against the human bladder carcinoma cell line 5637 (IC50 = 93 μg/ml). The weak cytotoxic activity and the absence of antibacterial effects in the used in vitro test systems opens a promising future for further investigations to clarify the antifungal mechanism and for in vivo applications of the new lipopeptides. Different media, temperature, light intensity and period of irradiance, the depletion of nitrate and the trace element cobalt were investigated to figure out conditions at which Bio 33 produces maximum of balticidins under laboratory conditions. Temperature was the most apparent factor influencing the growth of Bio 33 and the production of balticidins. Bio 33 grew best in BG 11 medium plus 0.5% NaCl at 26°C, under white fluorescent continuous light and a light intensity of 20 μmol photons m-2 s-1. Nevertheless, under the same conditions, 22.5°C was the best temperature for the production of balticidins. Besides, harvesting of Bio 33 during the logarithmic growth phase, particularly at 20th day, should supply approximately maximum quantity of balticidins. At 22.5°C and 20 μmol photons m-2 s-1 under 24 h continuous irradiance, the depletion of nitrate had no negative effect on the growth and concentration of balticidin A but increased balticidin B and decreased balticidin C; the absence of cobalt slightly decreased the growth but had no clear effect on the production of balticidins. On the other hand, extracts of the culture medium of the Vietnamese cyanobacterium TVN40, exhibited antifungal activity against Candida maltosa and weak antibacterial activity. Extraction of the culture medium with XAD-16 and elution of the XAD-bounded compounds by different solvents resulted in five fractions (water, 80% MeOH, 100% MeOH, acetone, dichloromethan). Four compounds have been isolated from the 80% MeOH fraction and one was identified as a dioxindole derivative. Structural elucidation of the other three compounds is still in progress. TVN40 was formerly identified as an Anabaena sp. according to the morphological properties, but the 16S rRNA gene sequence confirms that the strain belongs to the genus Nostoc. Microscopic examination of TVN40 revealed that the filamentous strain was not a unialgal but a mixed culture with strange round cells (SRCs) - a unicellular cyanobacterium belonging to the order Chroococcales. Laboratory cultures of the pure filamentous strain TVN40, the isolated SRCs and the mixed culture of both strains were established. Both TVN40 and SRC culture media were responsible for the antibacterial activity against B. subtilis, S. aureus and E. coli. However, only the extract of the culture medium of TVN40 was active against C. maltosa. The supplement of cobalt enhanced the antimicrobial activity of the culture medium. Pure strains showed higher activity in comparison to the mixed culture of TVN40 and SRC.
Ein neuer vielversprechender Ansatz, die Resistenz von Tumorzellen gegen Zytostatika zu umgehen, stellte die Hemmung von selenhaltigen Redoxenzymen, Glutathionperoxidase (GPx) und Thioredoxinreduktase (TrxR), dar. Das Ziel dieser Arbeit war, neuartige GPx-Inhibitoren zu entdecken und zu entwickeln. Der erste Ansatz war die Synthese und biologische Testung einer Zuckeracetal-Struktur, die als GPx-1-Inhibitor postuliert wurde. Synthetische Abwandlungen ergaben vier Acetale aus Glucosederivaten und Benzaldehyden. Jedoch zeigten diese ersten hergestellten Verbindungen keinerlei Hemmwirkung auf die bovine GPx-1. Der zweite Ansatz war, bereits bekannte, schwach aktive Leitstrukturen durch Koordinierung von Pt(II) an den Heterozyklus (2-Methylimidazol, Imidazol oder Pyrazol) zu stärkeren Hemmstoffen der GPx abzuwandeln, wobei das Platin(II)-atom an das Selen des Enzyms koordiniert und dies irreversibel hemmt. Es wurde eine Substanzbibliothek aus 15 heterozyklischen Liganden synthetisiert, welche anschließend mit Cis- oder Transplatin zu den cis- bzw. trans-Monochlorido-Platin(II)-Komplexen umgesetzt wurden. So wurden 28 Platinverbindungen in guter Reinheit erhalten und verschiedenen biologischen Testungen unterzogen. Diese Testungen umfassten Versuche zur Hemmung der bovinen GPx-1, wobei jedoch festgestellt wurde, dass dieses Enzym nicht gehemmt wurde. Weiterhin wurde untersucht, ob die TrxR, ein weiteres Selen-abhängiges Redoxenzym, durch diese Verbindungen inhibiert wird, was teilweise der Fall war, auch wenn keine eindeutigen Struktur-Wirkungs-Beziehungen aufgestellt werden konnten. Die Pt(II) enthaltenden Substanzen wurden auch auf ihre Fähigkeit, die Proliferation von humanen Krebszelllinien zu hemmen, getestet. Es wurde festgestellt, dass einige der Verbindungen in der Lage sind, die Zellteilung mit IC50-Werten unterhalb 1 µM zu unterbinden. Dabei handelte es sich meist um cis-konfigurierte Platin(II)-Verbindungen, aber auch manche trans-Pt-Komplexe waren aktiv. Auch konnte gezeigt werden, dass der Zelluntergang durch Apoptose herbeigeführt wird Diese ermutigenden Ergebnisse belegen, dass ein Platinkomplex nicht zwangsläufig bifunktionell sein muss, um das Zellwachstum effektiv zu hemmen. Weiterhin wurde eine starke Kreuzresistenz zu Cisplatin nur bei ein paar Substanzen beobachtet, manche Verbindungen konnten die Kreuzresistenz sogar vollständig umgehen. Ein paar Struktur-Wirkungs-Beziehungen konnten beschrieben werden. Zwei cis-trans-Paare der Platin(II)-Komplexe wurden dann für weitergehende Bindungsstudien an DNA ausgewählt. Hierzu wurde die Bindung an Kalbsthymus-DNA untersucht, ebenso wie die Bindung an zelluläre DNA, das Aufwinden von supercoiled DNA, die Veränderung des DNA-Schmelzverhaltens und des Circulardichroismus (CD) von DNA durch Bindung der Platinkomplexe, sowie die Veränderung der Ethidiumbromid-Fluoreszenz durch Hinzugabe der Verbindungen. Es stellte sich heraus, dass die strukturell ähnlichen Verbindungen sehr unterschiedliche Einflüsse auf die DNA haben, was auch in Zusammenhang mit ihrem hydrolytischen Zerfall während der Assays stehen könnte. Da Stabilitätsprobleme bemerkt wurden, wurden die hergestellten Verbindungen mittels HPLC und UV/Vis-Spektroskopie genauer untersucht um ihre Stabilität in wässrigen Medien beurteilen zu können. Es wurde festgestellt, dass die verwendeten Hydrazone in wässrigen Medien zu Benzaldehyden bzw. Acetophenonen und wahrscheinlich zu den korrespondierenden Hydraziden zerfallen. Die Stabilität der Verbindungen könnte starke Einflüsse auf die Ergebnisse der biologischen Testungen haben, sodass dies immer mit berücksichtigt werden sollte, wenn die Ergebnisse interpretiert werden. Es kann nicht aus-geschlossen werden, dass (i) die dargestellten Ergebnisse in Wirklichkeit die Ergebnisse der Zerfallsprodukte sind, und dass (ii) es einen gemeinsamen Grund für den Zerfall und die Auswirkungen im DNA- und Zellmodell gibt. Trotzdem oder auch deswegen handelt es sich um eine vielversprechende Substanzklasse, bei der sich eine nähere Untersuchung hinsichtlich Aufnahme in die Zelle, DNA-Bindung und -Reparatur lohnt, um zu ergründen, ob Resistenzen umgangen werden können. Auch die Zerfallsprodukte sollten hinsichtlich ihrer biologischen Aktivität untersucht werden. Eine weitere Verbindungsklasse, die im Rahmen dieser Arbeit untersucht wurde, waren N-heterozyklische Carben (NHC)-Au(I)-Komplexe. Sie basierten hauptsächlich ebenfalls wie die Platinverbindungen auf Hydrazonen, auch hier sollte durch die Koordinierung des Metallatoms an den Heterozyklus Imidazol eine Wirkungssteigerung erreicht werden. Die untersuchten Verbindungen zeigten eine starke Inhibition der GPx, der Glutathionreduktase (GR) sowie der TrxR. Eine Selektivität für ein bestimmtes Redoxenzym ist somit mit diesen Verbindungen nicht gegeben, weshalb sie keine guten Kandidaten für weitere Wirkstoffentwicklungen darstellen.
Der Einfluss von kaltem Atmosphärendruckplasma auf die Melaninsynthese in humanen Melanozyten

Das zur Behandlung von chronischen Wunden zugelassene Plasmagerät kINPen®MED wurde in der vorliegenden Arbeit hinsichtlich eines möglichen Effektes auf die Pigmentierung der Haut in vitro untersucht. Dazu wurden zunächst Untersuchungen an Melanomzellen durchgeführt. Verschieden stark pigmentierte Melanomzelllinien wurden einer indirekten Plasmabehandlung ausgesetzt, das heißt mit plasmabehandeltem Zellkulturmedium inkubiert, und der Melaningehalt wurde vor und nach der Behandlung durch eine UVspektroskopische Analyse evaluiert. Im Vergleich zu den unbehandelten Zellen war der Melaningehalt aller getesteten Melanomzelllinien erhöht. Auch im Vergleich zu Zellen, die mit den Positivkontrollen IBMX beziehungsweise NH4Cl / Tyrosin behandelt wurden, zeigte sich ein verstärkter Effekt der Plasmabehandlung auf die Pigmentierung der Melanomzellen. Da Melanomzellen nicht in erster Linie das Ziel von therapeutisch indizierten Beinflussungen der Pigmentierung sind, wurde im nächsten Schritt die Bestimmung des Melaningehaltes von primären Melanozyten nach in vitro-Plasmabehandlung durchgeführt. Dazu wurde zunächst die Methode zur relativen Melaninquantifizierung grundlegend überarbeitet. Es wurden zwei verschiedene Methoden zur verbesserten Melaninanalytik entwickelt, die hinsichtlich der benötigten Probenmenge im Vergleich mit der UV- spektroskopischen Melaningehaltsbestimmung im Melanomzellmodell eine höhere Sensitivität aufweisen. Die erste Methode basiert, wie in der Literatur beschrieben, auf der Oxidation des Melanins in alkalischer Lösung durch Wasserstoffperoxid und anschließender Analyse der melaninspezifischen Abbauprodukte. Die bestehende Methode wurde um eine massenspektrometrische Detektion der Abbauprodukte erweitert, was eine Reduktion der benötigten Probenmenge um den Faktor 10 ermöglichte bei Erhalt der Differenzierungsmöglichkeit zwischen Eumelanin und Phäomelanin. Die Analyse von primären Melanozyten nach indirekter Plasmabehandlung im Vergleich mit unbehandelten
Melanozyten ergab eine geringe Steigerung des Eumelaningehaltes in Melanozyten eines stark pigmentierten Spenders, der Phäomelaningehalt blieb unverändert. Der als
Positivkontrolle verwendete cAMP-Abbau-Inhibitor IBMX erhöhte sowohl den Eu- als auch den Phäomelaningehalt. Die zweite Methode zur Bestimmung des relativen Melaningehaltes beruht auf der Autofluoreszenz des Melanins, die bei einer Anregungswellenlänge von 785 nm im NIRBereich zu beobachten ist. Unter Verwendung der Durchflusszytometrie konnte eine Einzelzellanalyse der primären Melanozyten hinsichtlich ihrer Autofluoreszenzintensität durchgeführt werden. Dabei korrelierten die erzielten Ergebnisse der relativen Intensitätsänderung mit denen der massenspektrometrischen Quantifizierung der Melaninabbauprodukte. Gegenüber der unbehandelten Kontrolle war der Melaningehalt nach indirekter Plasmabehandlung geringfügig gesteigert. Nach Behandlung der Zellen mit IBMX als pharmakologischer Positivkontrolle sowie infolge einer Behandlung mit direkter UVStrahlung als physikochemischer Positivkontrolle war jedoch ein stärkerer Effekt auf die Steigerung der Melaninproduktion als bei der indirekten Plasmabehandlung zu beobachten. Die in vitro erhaltenen Ergebnisse konnten auf ein komplexeres Modell übertragen werden, indem eine direkte Plasmabehandlung von Hautproben ex vivo durchgeführt wurde. Außerdem wurde auf diese Weise ein Vergleich zwischen direkter Plasmabehandlung und UVStrahlung neben der unbehandelten Kontrolle möglich. Dazu wurden aus den Hautproben nach Behandlung und anschließender Inkubationszeit Kryoschnitte angefertigt. Um das enthaltene Melanin sichtbar zu machen, wurden die Schnitte einer Silbernitratfärbung unterzogen. Die an einem konfokalen Laser-Scanning-Mikroskop aufgenommenen Bilder wurden anschließend mit Hilfe zweier Softwares ausgewertet und die Ergebnisse verglichen. Insgesamt lässt sich trotz starker Schwankung der Ergebnisse beobachten, dass eine wiederholte direkte Plasmabehandlung eine Steigerung der Pigmentierung bewirkt, wenn auch in schwächerem Ausmaß, als dies nach UV-Bestrahlung zu beobachten ist. Insgesamt konnte durch Analyse des Melaningehaltes mit grundlegend unterschiedlichen Methoden wiederholt beobachtet werden, dass eine Plasmabehandlung einen geringen Effekt auf die Pigmentierung humaner Melanozyten ausübt. Dieser Effekt ist bei Behandlungszeiten, wie sie für den kINPen®MED zur Wundbehandlung empfohlen sind, im Vergleich zu einer UVBestrahlung beziehungsweise im Vergleich mit einer pharmakologischen Induktion der Melanogenese deutlich niedriger. Eine Intensivierung der Pigmentierung im Rahmen einer Wundbehandlung ist demnach nicht zu erwarten. Nichtsdestotrotz übt die Plasmabehandlung einen, wenn auch geringen, Effekt auf die Melaninproduktion in der humanen Haut aus. Um etwaige beteilige Enzyme an der gesteigerten Melaninproduktion zu erkennen, wurden Genexpressionsanalysen durchgeführt. Durch die Untersuchung der differenziellen Expression mittels Microarray konnte für 287 annotierte Gene eine Änderungen der Expression festgestellt werden, davon konnten 2 Gene identifiziert werden, für die ein Zusammenhang mit der Pigmentierung beschrieben ist. Veränderte Expressionslevel der hauptsächlich an der Melaninsynthese beteiligten Enzyme wurden durch die Plasmabehandlung der Melanozyten nicht detektiert. Die Untersuchung einiger ausgewählter, an der Melanogenese beteiligter Enzyme mittels qPCR führte zum selben Ergebnis: durch die getesteten Parameter der indirekten Plasmabehandlung wurden keine biologisch relevanten Änderungen des Transkriptionslevels hervorgerufen. In der vorliegenden Arbeit wurde umfassend gezeigt, dass das Medizinprodukt kINPen®MED in den durchgeführten in vitro Tests einen vergleichsweise geringen Effekt auf die Pigmentierung ausübt, der den Positivkontrollen unterlegen ist. Diese Ergebnisse zeigen, dass der kINPen®MED keinen geeigneten Modulator der Pigmentierung darstellt. Gleichzeitig unterstreichen die Resultate die Unbedenklichkeit der sachgemäßen Anwendung des kINPen®MED an der humanen Haut hinsichtlich etwaiger Melanin-bedingter Veränderungen. Es ist weder eine sichtbare Verstärkung der Pigmentierung noch eine Depigmentierung zu erwarten. Zudem wurde durch die Weiterentwicklung der Methodik im Bereich der Melaninanalytik ein Beitrag zur zukünftigen Testung von Pigmentierungsmodulatoren in Zellkulturmodellen geleistet.
Oral drug delivery is the preferred route of administration for the majority of drugs. Solid dosage forms arewell-accepted because of ease of administration, accurate dosing and high degree of patient compliance. The orodispersible technology platform has attracted increasing interest. Fast disintegrating in the mouth before swallowing, orodispersible dosage forms like orodispersible tablets (ODTs) address the need for patient-compliant medicines. ODTs represent a convenient alternative to conventional tablets or capsules. ODTs are an interesting approach when a rapid onset of therapeutic action is important. So far, ODTs have often been considered as an innovative variant of conventional oral solid dosage forms. Still, the development of ODT formulations is typically assisted by compendial in vitro test methods. However, the techniques described in international pharmacopoeias are non-specific for ODTs. After administration, the dispersion of an ODT in the mouth may provide effects which might influence the absorption of the drug. The performance of ODTs is more comparable to solutions/suspensions than to traditional tablets. To better guide the development of a new ODT formulation, this lack needs to be addressed. It is the aim of this work to design more specific in vitro test methods helping to improve understanding ODT formulations. To reflect the physiological conditions experienced by an ODT after administration, particular attention was given to the mouth where the ODT disperses and releases the drug before swallowing. In vitro biorelevant test setups simulating in vivo conditions were designed. An electronic tongue system was used to assess taste properties of ODTs. These test methods were applied in different stages of the ODT formulation development. Diclofenac being a poorly soluble and weakly acidic NSAID which is a standard medication for acute painful inflammatory conditions was used as a drug model. Three forms, i.e. the free acid and its sodium/potassium salt, were investigated for the formulation of palatable and fast acting ODTs. In Chapter 1, the development of biorelevant test setup reflecting the physiological conditions experienced by ODTs is described in detail. The newly-designed in vitro models successfully discriminated the different diclofenac forms in successive in vitro compartments simulating the mouth, the stomach and the small intestine. It was possible to identify peculiar dissolution profiles with diclofenac salts. Characterizing in-depth the diclofenac free acid and salt particles provided a better understanding of the peculiar dissolution profiles. Critical behaviors of diclofenac salts on their way from the mouth to the stomach and passing different pH conditions were extensively evaluated. Reasons for pH-dependent API precipitation and particle agglomeration were studied in detail. In pre-formulation studies, the proposed biorelevant test setups succeeded in helping to early identify critical pharmaceutical properties for diclofenac salts and to select diclofenac free acid as the most appropriate drug form providing the most stable in vitro performance. In Chapter 2, the electronic tongue method as an in vitro taste assessment tool for ODTs is proposed. Using the TS-5000Z taste sensing system (Insent Inc., Japan), the method was able to differentiate between the taste/aftertaste qualities and intensities of the three diclofenac candidates. The electronic tongue was also successfully used to differentiate different ODT formulations. The results obtained proved that valuable information can be gained. By this means, the taste perception of the diclofenac drug candidates were classified and rank against each other. For manufacturing taste-masked ODTs, diclofenac free acid, could be selected easily. The electronic tongue found out to be a precious tool in assisting the development of a new ODT product and finding the most appropriate multi-component formulation. Both proposed methods successfully showed their discriminative ability and also their utility in pre-formulation studies of ODTs. In the previous chapters, it was indeed possible to early select diclofenac free acid as the most suitable drug candidate for the targeted product profile. In Chapter 3, said methods were further used to guide the development of the taste masked diclofenac ODT formulation. This study highlights the importance of considering in vitro the physiological aspects which may have an impact on the in vivo performance of ODT dosage forms. The contact of ODTs with the mouth should be simulated in vitro for a better understanding of the in vivo behavior. With feasible biorelevant in vitro dissolution methods, an optimized correlation of in vitro and in vivo results may be achieved. The proposed in vitro test methods may provide data of predictive value and may support the rational development of ODT formulations.
