In the present study, we investigated whether inter-individual differences in vagally mediated heart rate variability (vmHRV) would be associated with inter-individual differences in empathy and alexithymia. To this end, we determined resting state HF-HRV in 90 individuals that also completed questionnaires assessing inter-individual differences in empathy and alexithymia. Our categorical and dimensional analyses revealed that inter-individual differences in HF-HRV were differently associated with inter-individual differences in empathy and alexithymia. We found that individuals with high HF-HRV reported more empathy and less alexithymia than individuals with low HF-HRV. Moreover, we even found that an increase in HF-HRV was associated with an increase in empathy and a decrease in alexithymia across all participants. Taken together, these findings indicate that individuals with high HF-HRV are more empathetic and less alexithymic than individuals with low HF-HRV. These differences in empathy and alexithymia may explain why individuals with high HF-HRV are more successful in sharing and understanding the mental and emotional states of others than individuals with low HF-HRV.
3-Hydroxy-3-Methylglutaryl-Coenzym A-Reduktase Inhibitoren (Statine) bewirken hauptsächlich die Beeinflussung des Lipidstoffwechsels. Sie besitzen aber auch lipidunabhängige (pleiotrope) Effekte, wie z.B. die Verbesserung der Endothelfunktion. Aufgrund ihrer molekularen Struktur kann man zwischen hydrophilen (z.B. Pravastatin) und lipophilen (z.B. Atorvastatin) Statinen unterscheiden. Ziel dieser Arbeit war zu untersuchen, ob eine unterschiedliche Lipophilie die pleiotropen Effekte von Statinen beeinflussen kann. Hierzu wurden Statine mit unterschiedlicher Lipophilie hinsichtlich der endothelialen Proliferation und flussinduzierten Migration charakterisiert sowie deren unterschiedliches Verhalten mit möglichen Signaltransduktionswegen assoziiert.
Über Bioimpedanzmessungen (xCelligence) und BrdU-Assays wurde die Proliferation von HUVEC unter Statinen oder in Kombination mit Mevalonat über 48 h untersucht. Zur Quantifizierung der flussinduzierten Migration wurden mit einem Flusskammersystem Live Cell Imaging Versuche über 15 h durchgeführt. In den Versuchen wurden Atorvastatin (ATOR) und Pravastatin (PRA) in den Konzentrationen 0,1 und 1 µM verwendet. Nach Beendigung der Versuche wurden über RT-qPCR und Microarray (Migrationsversuche) Genexpressionsanalysen durchgeführt.
ATOR und PRA hatten keinen negativen Einfluss auf die Vitalität der EC. ATOR zeigte einen positiven Effekt auf die endotheliale Proliferation und führte zu einer Verbesserung der Endothelfunktion unter statischen Bedingungen. Beide Statine zeigten unter Scherstress eine gerichtetere Migration. Allerdings erhöhte nur ATOR die Migration unter pathologischem Scherstress. Weiterhin wurden auch auf Transkriptionsebene Unterschiede festgestellt. Über den Microarray konnte mit XBP1 möglicherweise ein neuer Regulator der pleiotropen Statineffekte identifiziert werden.
Zur Vermeidung von In-Stent-Restenosen nach Perkutaner Koronarangioplastie mit Stent-Implantation ist eine optimale Reendothealisierung von großer Bedeutung für die weitere Prognose des Patienten. Basierend auf den Daten dieser Arbeit scheinen lipophile Statine, wie ATOR, insgesamt besser geeignet zu sein, da sie Proliferation, Migration und Endothelzellfunktion positiv beeinflussen. Diese Erkenntnis könnte u.a. für die zukünftige Entwicklung von Statin-freisetzenden Stents von hoher Bedeutung sein.
Background: Biomarkers for gains of evidence based interventions for upper limb motor training in the subacute stage following stroke have rarely been described. Information about these parameters might help to identify patients who benefit from specific interventions and to determine individually expected behavioral gains for a certain period of therapy.
Objective: To evaluate predictors for hand motor outcome after arm ability training in the subacute stage after stroke selected from known potentially relevant parameters (initial motor strength, structural integrity of the pyramidal tract and functional motor cortex integrity).
Methods: We applied the arm ability training (AAT) over 3 weeks to a subpopulation of stroke patients with mild arm paresis, i.e., in 14 patients on average 4 weeks after stroke. The following biomarkers were measured before therapy onset: grip strength on the affected hand, transcranial magnetic stimulation recruitment curve steepness over the primary motor hand area [slope ratio between the ipsilesional hemisphere (IH) and contralesional hemisphere (CH)], and diffusion weighted MRI fractional anisotropy (FA) in the posterior limb of the internal capsule (PLIC; determined as a lateralization index between IH and CH). Outcome was assessed as the AATgain (percentage improvement over training). The “Test d'Evaluation des Membres Supérieurs de Personnes Âgées” (TEMPA) was assessed before and after training to test for possible associations of AAT with activity of daily living.
Results: A stepwise linear regression identified the lateralization index of PLIC FA as the only significant predictor for AAT-gain (R2 = 0.519; P = 0.029). AAT-gain was positively associated (r = 0.59; P = 0.028) with improvement in arm function during daily activities (TEMPA).
Conclusions: While all mildly affected patients achieved a clinically relevant therapeutic effect, pyramidal tract integrity nevertheless had a modifying role for clinical benefit.
The present study seeks to determine potential associations between viral infections and neuropsychiatric diseases. To address this issue, we investigated the peptide commonalities between viruses that have been related to psychiatric and neurological disorders—such as rubella, human immunodeficiency virus, and herpesviruses—and human distal-less homeobox (DLX) proteins expressed in developing brain—namely, DLX1, DLX2, DLX5, and DLX6. Peptide matching analyses revealed a high degree of pentapeptide sharing. From an immunological perspective, this overlap is relevant because pentapeptides are endowed with immunogenicity and antigenicity—that is, they are immune determinants. Moreover, infection-induced immune cross-reactions might have functional, spatial, and temporal implications related to the functions and expression patterns of DLX1 and DLX5 in the fetal and adult human brain. In sum, our data support the hypothesis that viral infections may be linked to neuropsychiatric diseases through autoimmune cross-reactions caused by molecular mimicry between viral proteins and brain-specific DLX self-antigens.
Vordergründiges Ziel der Arbeit war eine Grundlagenforschung über die Expression der TRPCKanäle in Osteosarkomzellen unter basalen und stimulierten Bedingungen durchzuführen. Als thematisches Setting wurde zum einen die bisher unklare Wirkung des erfolgversprechenden Medikaments Mifamurtid gewählt sowie zum anderen ein möglicher Zusammenhang mit der alternativen Ang-(1-7)/Mas-Achse gesucht.
Die vorliegende Arbeit hat einerseits die Expression der TRPC-Kanäle in Osteosarkomzellen nachgewiesen und andererseits interessante Schnittpunkte zu der alternativen ACE2/Ang-(1-7)/Mas-Achse des RAS gezeigt. So wird in Zukunft möglicherweise ein Zusammenhang zwischen der Mas-Aktivität und den TRPC-Kanälen – insbesondere von TRPC6 – substantiviert werden können. Die massive Induktion der Transkription von TRPC5 unter Inhibierung der PI3-Kinase ist eine interessante Beobachtung, die Ansätze für weiterführende Untersuchungen eröffnet. Inwieweit die pharmakologische Modulation von Mas- (oder MrgD-) Rezeptoren eine therapeutisch relevante Option beim Osteosarkom darstellt und inwieweit die Modulation der TRPC-Kanäle dazu beitragen kann, ist mit dieser Arbeit nicht geklärt. Die vorgelegten Daten zeigen jedoch eindeutig, dass der Mas-Antagonist D-Ala die Expression von zumindest TRPC 5, 6 und 3 sowie von Mas selbst in Osteosarkom-Zellen beeinflusst; Effekte die auch durch die Modulation von PI3K und MAP-Kinase Signalwegen erreicht werden können.
Zielsetzung: Die antimikrobielle Wirksamkeit von kaltem Atmosphärendruckplasma(CAP), auch als gewebeverträgliches Plasma (TTP) bezeichnet,könnte eine aussichtsreiche Option zur Eradikation von Methicillinempfindlichen ebenso wie von Methicillin-resistenten Staphylococcus aureus-Stämmen sein, die oft chronische Wunden kolonisieren. Bisher wurde der Einfluss von CAP auf die Antibiotikaempfindlichkeit von S. aureus kaum untersucht. Da eine Veränderung der Antibiotikaempfindlichkeit für die Wundbehandlung relevant sein könnte, sollte der Einfluss von CAP auf die Empfindlichkeit verschiedener S. aureus-Stämme gegen unterschiedliche Antibiotika untersucht werden.
Methode: Im Agardiffusionstest wurden Antibiotikatestplättchen mit Cefuroxim, Gentamicin, Oxacillin, Vancomycin, Ciprofloxacin, Co-Trimoxazol, Clindamycin und Erythromycin eingesetzt. Die Teststämme wurden auf Agar ausplattiert und mit CAP exponiert, bevor die Testplättchen aufgelegt wurden. Nach 24 h Bebrütung wurden die Inhibitionszonen gemessen und statistisch auf Unterschiede geprüft.
Ergebnisse: In den meisten Fällen war die Einfluss von CAP auf die Antibiotikaempfindlichkeit zu vernachlässigen. Für zwei Stämme wurde die Empfindlichkeit gegenüber β-Lactam-Antibiotika signifikant herabgesetzt.
Schlussfolgerung: Da CAP die Antibiotikaempfindlichkeit beeinflussen kann, sollten vor beabsichtigter kombinierter lokaler CAP-Behandlung und gleichzeitiger systemischer Antibiotikagabe Interaktionen in vitro untersucht werden, um unerwünschte Kombinationseffekte auszuschließen.
Manipulating and utilizing plasmas becomes a more and more important task in various research fields of physics and in industrial developments. Especially in nowadays spacerelevant applications there are different ideas to modify plasmas concerning particular tasks.
One major point of interest is the ability to influence plasmas using magnetic fields. To study the underlying physical effects that were achieved by these magnetic fields for both scenarios Particle-in-Cell simulations were done. Two examples are discussed in this thesis.
The first example originates from an experiment performed by the European Space Agency ESA in collaboration with the German Space Agency DLR. To verify the possibility of heat-flux reduction by magnetic fields onto the thermal protection system of a space vehicle a simplified experiment on earth was developed. Most of the heat that is created during re-entry comes from compression of the air ahead of the hypersonic vehicle, as a result of the basic thermodynamic relation between temperature and pressure. The shock front, which builds up in front of the vehicle deflects most of the heat and prohibits the surface of the space vehicle from direct contact with the maximum flux. State of the art spacecrafts use highly developed materials like ceramics to handle the enormous heat. An attractive approach to reduce costs is to use magnetic fields for heat-flux reduction. This would allow the use of cheaper materials and thus reduce costs for the whole space mission. A partially-ionized Argon beam was used to create a certain heat-flux onto a target. The main finding of the experimental campaign was a large mitigation of heat-flux by applying a dipole-like magnetic field. The Particle-in-Cell method was able to reproduce experimental observations like the heat-flux reduction. An additionally implemented optical diagnostics module allowed to confirm the results of the spectroscopy done during the experiment. The underlying effect that is responsible for the heat-flux reduction was identified as a coupling between the modified plasma and the dominating neutral flux component. The plasma, that is guided towards the target, act as a shield in front of the target surface for arriving neutrals. These neutrals are slowed down by charge-exchange collisions. Furthermore the magnetic field induces an increased turbulent transport that is also needed to reach a reduction in heat-ux. The turbulent transport was also obtained by three-dimensional Direct Simulation Monte Carlo simulations. Unfortunately, such source driven turbulence can not be expected in space, so that a heat flux reduction in real space applications is questionable. Nevertheless, other effects like the induced turbulence by the rotating vehicle can compensate the missing source driven effect.
The second scenario in which a magnetic field is used to modify the heat flux of a plasma is the operation of the pulsed cathodic arc thruster. The same Particle-in-Cell code was used to simulate a typical pulse of this newly developed thruster of Neumann Space Pty Ltd. The typical behavior of the thruster could be reproduced numerically. The thrust is mainly produced by fast electrons. These electrons are accelerated by electric fields as a result of a plasma-beam instability. This plasma-beam instability was verified by a phase space diagnostics for the electrons. To demonstrate the influence of the magnetic field a simulation of the cathodic arc thruster without magnetic field and one with magnetic field were compared. It was shown that the use of a magnetic field leads to a ten times larger thrust by directing the heat ux. The resulting narrow plume is an additional Advantage of the particle guiding magnetic field. This narrowness of the plume reduces the danger of interaction with other components of the space vehicle.
Both scenarios demonstrate the different capabilities for electromagnetic fields to manipulate plasmas and especially the corresponding heat-flux with respect to certain tasks. The possibilities range from reducing the heat-flux onto a target to maximizing the thrust by directing the heat-ux. This thesis demonstrates that simulations are a great tool to support experiments and to deliver an improved physics understanding. They help to identify the basic physics principles in the different systems, because they can deliver information not accessible to experiments.
In particular, a better understanding of the influence of electromagnetic fields on the heat-flux distribution in space-relevant applications was obtained. This can be the basis for further simulation-guided optimization, e.g. for the design of more effective cathodic arc thrusters. Here, the goal is to minimize costs for prototypes by replacing the hardware by virtual prototypes in the simulations. This allows to test basic design ideas in advance and get more highly-optimized designs at a fraction of time and costs.
Das klinische Bild der Sepsis, des septischen Schockes und des Multiorganversagens sind nach wie vor Krankheitsbilder, wie sie auf Intensivstationen vorzufinden sind und häufig mit einer hohen Letalität assoziiert sind. Die Störung bzw. das Versagen der Mikrozirkulation ist und bleibt einer der Hauptmechanismen als Ursache dafür [1]. Die Zielsetzung der vorliegenden Arbeit war es, einen auf Zellebene beschriebenen Weg auf ein in vivo Tiermodell zu übertragen. Der bereits beschriebene Weg besagt, dass die Hemmung der Src-Tyrosinkinase durch das synthetische PP2, eine Reduktion der proinflammatorischen Zytokine zur Folge hat. Dieses sollte die Mikrozirkulation verbessern. In der vorliegenden Versuchsreihe wurde dafür ein bereits gut etablierter Versuchsablauf der experimentellen Endotoxinämie mit anschließender Intravitalmikroskopie am Tiermodell Ratte verwendet
In der vorliegenden Arbeit hat sich gezeigt, dass sich unter experimenteller Endotoxinämie sowie unter der Verwendung von PP2 bei einer Dosierung von 0,2mg/kg KG, die Mikrozirkulation signifikant verschlechtert. Dies wird bei Betrachtung der funktionellen Kapillardichten besonders deutlich. Im Vergleich der beiden endotoxiämischen Gruppen kann bei der Gruppe, die PP2 erhalten hat, ein Abfall der FCD von 47,8% für die Lamina longitudinalis und 25,5% für die Lamina circularis zu der Gruppe, die kein PP2 erhalten hat, festgestellt werden. Folglich ist es umgekehrt bei der Betrachtung der DFCD. Hierbei kann bei der Gruppe, welche LPS und PP2 erhalten hat, ein Anstieg von 30,27% für die Lamina longitudinalis bzw. 28,2% für die Lamina circularis, im Vergleich zu der Gruppe, der nur LPS appliziert wurde, beobachtet werden. In Bezug auf die NFCD zeigt sich ebenfalls ein Anstieg bei der Gruppe, die LPS und PP2 erhalten hat, jedoch ist dieser nicht signifikant.
Zudem zeigt sich eine Steigerung der Leukozyteninteraktion bei Endotoxinämie und zusätzlicher Verwendung von PP2 in den postkapillären Venolen 1. und 3. Ordnung. Dabei zeigt sich ein Anstieg der Leukozyten mit temporärer Adhäsion von 112,9% bei Venolen 1. Ordnung und 95% bei Venolen 3. Ordnung. Allerdings befindet sich der Mittelwert in jeder Gruppe bei <10 Leukozyten im Beobachtungszeitraum. Bei der permanenten Leukozytenadhäsion hingegen können keine signifikanten Unterschiede bei der Verwendung von PP2 beobachtet werden. Weiterhin zeigte sich bei der Verwendung von PP2 ein signifikanter Anstieg der Lactatkonzentration von 16,35%, wenn zuvor LPS gegeben worden ist. In der Untersuchung der Zytokine zeigten sich Veränderungen zwischen den Gruppen mit und ohne Endotoxinämie. Die Verwendung von PP2 ist dabei irrelevant.

Self-affine tiles and fractals are known as examples in analysis and topology, as models of quasicrystals and biological growth, as unit intervals of generalized number systems, and as attractors of dynamical systems. The author has implemented a software which can find new examples and handle big databases of self-affine fractals. This thesis establishes the algebraic foundation of the algorithms of the IFStile package. Lifting and projection of algebraic and rational iterated function systems and many properties of the resulting attractors are discussed.
Einleitung
Die Heilung einer Wunde unterscheidet sich in der Narbenbildung, dem Einfluss von Risikofaktoren, dem funktionellen und ästhetischen Ergebnis und in der Wundheilungsdauer. Eine Vielzahl wissenschaftlicher Studien beschäftigt sich mit Möglichkeiten, die Dauer der Wundheilung zu verkürzen. Dabei fällt auf, dass bisher keine klinischen Daten zur Wundheilungsdauer existieren, insbesondere keine, die die Dauer der Re-Epithelialisierung ohne spezifische Wundbehandlung wiedergeben. Ziel dieser Studie ist daher die Ermittlung der Re-Epithelialisierungszeit einer klinischen Wunde in Tagen und wie diese durch individuelle Faktoren beeinflusst wird. Die vorliegende Arbeit umfasst Ergebnisse einer klinischen Studie von Spalthautentnahmestellen hinsichtlich der durchschnittlichen Dauer des epithelialen Wundverschlusses in Abhängigkeit verschiedener individueller Einflussfaktoren.
Material und Methoden
Die verwendeten Daten entstammen zweier multizentrischer Phase-III-Studien zur Untersuchung der klinischen Wirksamkeit und Sicherheit eines topischen BetulinGels bezüglich der Heilung von Spalthautentnahmestellen (EudraCT no. 2012-003390-26, EudraCT no. 2012-000777-23). Die Spalthautentnahmestellen wurden halbiert und nach Randomisierung eine Hälfte mit Betulin Gel behandelt und die andere Hälfte als Kontrollseite mit einem Standardwundverband verschlossen. Gegenstand dieser Studie waren ausschließlich die Kontrollseiten. Die Beurteilung der Wundheilung erfolgte mittels Auswertung von Fotos durch drei unabhängige und verblindete Wundexperten via eines Online-Tools. Das Studienprotokoll schloss 198 Patienten ein. Die Beobachtung endete nach Erreichen einer Re-Epithelialisierung von über 95% der Wundfläche oder spätestens 28 Tagen. Die statistische Auswertung erfolgte mittels Kaplan-Meier-Überlebenszeitanalysen mit einem Konfidenzintervall von 95%. Dabei wurde die kumulative Ereignisrate für verschiedene Einflussfaktoren aufgetragen und mittels Logrank Test statistisch analysiert.
Ergebnisse
Die Re-Epithelialisierung der Wunden dauerte mindestens sieben Tage. Die schnellsten 25% der Patienten („fast healers“) zeigten einen epithelialen Wundverschluss innerhalb von elf Tagen, während die Re-Epithelialisierung im Median 14 Tage andauerte und die langsamsten 25% („slow healers“) der Studienpopulation 18 Tage benötigten. Patienten unter 40 Jahren zeigten einen signifikant schnelleren epithelialen Wundverschluss von zwölf Tagen während die Re-Epithelialisierung bei Patienten über 60 Jahren 18 Tage dauerte (p=0). Gebärfähigkeit reduziert die Dauer signifikant um vier Tage (p=0,002), während Krebserkrankungen und begleitende Glucocorticoid-Therapien die oberflächliche Wundheilungsdauer um zwölf beziehungsweise zehn Tage verlängern (p=0).
Zusammenfassung
Eine oberflächliche, artifiziell durch eine Spalthautentnahme entstandene Wunde zeigt ohne gezielte Wundbehandlung einen Schluss der Epitheldecke nach frühestens sieben Tagen, in der Gruppe der „fast healers“ nach elf Tagen, im Median nach 14 Tagen und benötigt 18 Tage in der Gruppe der „slow healers“. Die Wundheilung wird um vier bis zwölf Tage verlängert durch ein Patientenalter über 60 Jahren, eine maligne Grunderkrankung und eine Begleitmedikation mit Glucocorticoiden. Die Wundheilung wird durch ein Patientenalter unter 40 Jahren und die Gebärfähigkeit von Frauen um zwei bis vier Tage beschleunigt. Diese ergab die Analyse von 198 Patientendaten aus 32 europäischen Krankenhäusern basierend auf einer verblindeten Beurteilung standardisierter fotografischer Wunddokumentation.
Auf der Grundlage der populationsbezogenen Daten der Study of Health in Pomerania wurden in dieser Studie a) die Häufigkeit einer dynamischen Überblähung und b) prädiktive Faktoren für das Auftreten dieses Phänomens analysiert.
Es wurden Daten von 6753 Probanden (3510 weiblich) aus 2 Kohorten der Study of Health in Pomerania (SHIP-2 und SHIP-TREND) untersucht.
Eine dynamische Überblähung wurde mit Hilfe der IC-Manöver, welche im Rahmen der Spiroergometrie durchgeführt wurden, als Konstanz oder Anstieg des EELV unter Belastung definiert.
Analysiert wurden die Ergebnisse anhand von 2148 Probanden (1052 weiblich) mit einem vollständigen kardiopulmonalen Untersuchungskomplexes ohne gröbere Fehler, mindestens 3 IC-Manövern der Spiroergometrie und einer Variabilität des EELV von weniger als 200 ml.
Aus dieser Gesamtgruppe wurde durch Ausschluss von aktiven Rauchern und Probanden mit kardiopulmonalen Vorerkrankungen und Vormedikation eine gesunde Studienpopulation extrahiert. Mit Hilfe einer detaillierten Qualitätsanalyse ergab sich aus der Gesamtpopulation zusätzlich eine Studienpopulation mit fehlerfreien dynamischen Fluss-Volumen-Kurven.
Alle Ergebnisse wurden als Absolut- und Relativwerte angegeben. Die Relativwerte wurden anhand der vorgestellten Normwertformeln berechnet.
Bei einem mittleren Alter von 54 und 52 Jahren und einem mittleren BMI von 27 kg/m2 wiesen die untersuchten Studienpopulationen lungenfunktionelle Werte im Normbereich auf. Zudem war eine normale Belastbarkeit bei normwertiger peakVO2 und VO2@AT gegeben.
Die Datenlage zu Häufigkeiten und Prädiktoren der dynamischen Überblähung war bisher noch gering. So konnten wir mit Hilfe unserer Studie zeigen, dass eine dynamische Überblähung bei Probanden einer epidemiologischen Studie mit einer Häufigkeit von bis zu 23 % vorkommt. Zusätzlich war es möglich, die Prävalenz der dynamischen Überblähung über alle Altersgruppen getrennt für Männer und Frauen anzugeben.
Signifikante Unterschiede zwischen den Gruppen der Probanden mit und ohne dynamische Überblähung zeigten sich in allen drei Studienpopulationen hinsichtlich Alter (Jahren), BMI (kg/m2) und Größe (cm) sowie bezüglich der spirometrischen Parameter FEV (l), PEF (l/s) und MEF 75 (l/s) und der bodyplethysmografischen Parameter Rtot (kPa*s/l und %pred) und ITGV (l).
Weitere signifikante Unterschiede waren in den spiroergometrischen Parametern peakVO2 (ml/min), Vtin_max (l) und peakVt (l und %pred) zu verzeichnen.
Als bester Prädiktor einer dynamischen Überblähung erwies sich in unserer Studie der Atemwegswiderstand Rtot in %pred, gefolgt von Alter, Größe und Gewicht.
Unsere Studie stellt die Bedeutung der dynamischen Fluss-Volumen-Kurven als ein wichtiges diagnostisches Verfahren im Rahmen der Spiroergometrie heraus und zeigt Möglichkeiten einer qualitativen Bewertung dieser Untersuchungsmethode auf. Sie ist zudem eine der ersten Studien, welche die lungenfunktionellen und spiroergometrischen Parameter hinsichtlich ihrer prädiktiven Potenz zur Feststellung einer dynamischen Überblähung untersuchte.
Um die Bedeutung für den klinischen Alltag herauszuarbeiten, werden weitere Studien notwendig sein.
