Makrophagen stellen einen wesentlichen Bestandteil des leukozytären Infiltrates des Kolonkarzinoms und anderer maligner Tumoren dar. Für eine Reihe anderer Tumorentitäten konnte bereits nachgewiesen werden, dass tumor-assoziierte Makrophagen das Wachstum, die Neoangiogenese und die Metastasierung fördern. Bisherige Studien zur Rolle der Makrophagen im Kolonkarzinom zeigten hingegen widersprüchliche Ergebnisse.
Um den Einfluss von Makrophagen auf das Wachstums- und Metastasierungsverhalten im Kolonkarzinom genauer zu untersuchen, wurde zum einen ein orthotopes, syngenes Kolonkarzinommodell, zum anderen ein Lebermetastasenmodell unter Verwendung der murinen Kolonkarzinomzelllinie CT-26 in der immunkompetenten Balb/c-Maus verwendet und die Makrophagen bzw. Kupffer-Zellen mittels intraperitonealer Injektion von Clodronat-Liposomen selektiv depletiert. Die Tumorvolumina bzw. die Metastasenanzahl wurden mittels 7-Tesla Kleintier-MRT bestimmt und das Tumorgewicht per Feinwaage ermittelt. Die Makrophagen- und Gefäßdichte in den orthotopen Tumoren sowie die Kupffer-Zelldichte im Lebermetastasenmodell wurden mit Hilfe der Immunhistochemie ausgewertet.
Die Makrophagendepletion führte zu einer signifikanten Verringerung des Tumorwachstums im orthotopen Kolonkarzinommodell und verminderte hier gleichzeitig die Anzahl an Tieren, die eine Peritonealkarzinose bzw. Lebermetastasen entwickelten. Darüber hinaus führte die Depletion zu einer signifikanten Hemmung der Neoangiogenese im Kolonkarzinom. Im Lebermetastasenmodell führte die Kupffer-Zelldepletion zu einer signifikant geringeren Anzahl an Lebermetastasen, wobei die Kupffer-Zelldichte direkt mit der Metastasenanzahl korrelierte.
Die Ergebnisse unterstützen die Vermutung, dass tumor-assoziierte Makrophagen im Kolonkarzinom, wie auch in einigen anderen malignen Erkrankungen, eine tumor-unterstützende Rolle ausüben. Darüber hinaus scheinen Kupffer-Zellen einen entscheidenden Promotor bei der Ausbildung von Lebermetastasen darzustellen.
Zukünftige Therapien könnten darauf abzielen, selektiv bestimmte Makrophagenpopulationen wie die tumor-assoziierten Makrophagen oder Kupffer-Zellen zu depletieren. Eine weitere Möglichkeit bestünde in der Umdifferenzierung von tumor-assoziierten Makrophagen (M2) in proinflammatorische Makrophagen (M1), die dann suppressiv oder zytotoxisch auf den Tumor wirken.
Des Weiteren wurden zwei Untersuchungsmethoden zur Darmbildgebung in der Maus mittels MRT entwickelt, die eine deutliche Steigerung der Bildqualität und einen zusätzlichen Informationsgewinn in Tierversuchen ermöglichen. Beide Untersuchungstechniken erwiesen sich als gut reproduzierbar und erlauben eine longitudinale Beurteilung von Darmerkrankungen über einen längeren Zeitraum.
In weiteren Versuchen müsste nun die Sensitivität und Spezifität der beiden beschriebenen Methoden bei der Untersuchung pathologischer Läsionen des Darmes beurteilt werden.
We aimed to evaluate the outcome of different treatment modalities for extremity venous thrombosis (VT) in neonates and infants, highlighting the current debate on their best tool of management. This retrospective study took place over a 9-year period from January 2009 to December 2017. All treated patients were referred to the vascular and pediatric surgery departments from the neonatal intensive care unit. All patients underwent a thorough history-taking as well as general clinical and local examination of the affected limb. Patients were divided into 2 groups: group I included those who underwent a conservative treated with the sole administration of unfractionated heparin (UFH), whereas group II included those who were treated with UFH plus warfarin. Sixty-three patients were included in this study. They were 36 males and 27 females. Their age ranged from 3 to 302 days. Forty-one (65%) patients had VT in the upper limb, whereas the remaining 22 (35%) had lower extremity VT. The success rate of the nonsurgical treatment was accomplished in 81% of patients. The remaining 19% underwent limb severing, due to established gangrene. The Kaplan-Meier survival method revealed a highly significant increase in both mean and median survival times in those groups treated with heparin and warfarin compared to heparin-only group (P < .001). Nonoperative treatment with anticoagulation or observation (ie, wait-and-see policy) alone may be an easily applicable, effective, and a safe modality for management of VT in neonates and infants, especially in developing countries with poor or highly challenged resource settings.
Sphingosine-1-phosphate (S1P) regulates the migration of follicular B cells
(B2 cells) and directs the positioning of Marginal zone B cells (MZ B cells) within the spleen. The
function of S1P signalling in the third B cell lineage, B1 B cells, mainly present in the pleural and
peritoneal cavity, has not yet been determined. Methods: S1P receptor expression was analysed
in peritoneal B cells by real-time polymerase chain reaction (qPCR). The chemotactic response to
S1P was studied in vitro. The role of S1P signalling was further explored in a s1p4
−/− mouse
strain. Results: Peritoneal B cells expressed considerable amounts of the S1P receptors 1 and 4
(S1P1 and S1P4, respectively). S1P1 showed differential expression between the distinct peritoneal B
cell lineages. While B2 cells showed no chemotactic response to S1P, B1 B cells showed a migration
response to S1P. s1p4
−/− mice displayed significant alterations in the composition of peritoneal
B cell populations, as well as a significant reduction of mucosal immunoglobulin A (IgA) in the
gut. Discussion: S1P signalling influences peritoneal B1 B cell migration. S1P4 deficiency alters the
composition of peritoneal B cell populations and reduces secretory IgA levels. These findings suggest
that S1P signalling may be a target to modulate B cell function in inflammatory intestinal pathologies.
