ZusammenfassungTOPORAZ ist eine virtuelle Forschungsumgebung, die ein wissenschaftlich fundiertes 3D-Modell des Hauptmarktes der Stadt Nürnberg in vier Zeitstufen mit einer Datenbank verknüpft. Diese Plattform für die objekt- und raumbezogene geisteswissenschaftliche Forschung verbindet vielfältige Quellen und Literatur mit den Gebäuden des 3D-Modells über Verknüpfungspunkte (Hotspots). TOPORAZ unterstützt übergreifende Forschungsansätze und vernetztes, transdisziplinäres Arbeiten.
Today the process of improving technology and software allows to create, save and explore massive data sets in little time. "Big Data" are everywhere such as in social networks, meteorology, customers’ behaviour – and in biology. The Omics research field, standing for the organism-wide data exploration and analysis, is an example of biological research that has to deal with "Big Data" challenges. Possible challenges are for instance effcient storage and cataloguing of the data sets and finally the qualitative analysis and exploration of the information. In the last decade largescale genome-wide association studies and high-throughput techniques became more effcient, more profitable and less expensive. As a consequence of this rapid development, it is easier to gather massive amounts of genomic and proteomic data. However, these data need to get evaluated, analysed and explored. Typical questions that arise in this context include: which genes are active under sever al physical states, which proteins and metabolites are available, which organisms or cell types are similar or different in their enzymes’or genes’ behaviour. For this reason and because a scientist of any "Big Data" research field wants to see the data, there is an increasing need of clear, intuitively understandable and recognizable visualization to explore the data and confirm thesis. One way to get an overview of the data sets is to cluster it. Taxonomic trees and functional classification schemes are hierarchical structures used by biologists to organize the available biological knowledge in a systematic and computer readable way (such as KEGG, GO and FUNCAT). For example, proteins and genes could be clustered according to their function in an organism. These hierarchies tend to be rather complex, and many comprise thousands of biological entities. One approach for a space-filling visualization of these hierarchical structured data sets is a treemap. Existing algorithms for producing treemaps struggle with large data sets and have several other problems. This thesis addresses some of these problems and is structured as follows. After a short review of the basic concepts from graph theory some commonly used types of treemaps and a classification of treemaps according to information visualization aspects is presented in the first chapter of this thesis. The second chapter of this thesis provides several methods to improve treemap constructions. In certain applications the researcher wants to know, how the entities in a hierarchical structure are related to each other (such as enzymes in a metabolic pathway). Therefore in the 3 third chapter of this thesis, the focus is on the construction of a suitable layout overlaying an existing treemap. This gives rise to optimization problems on geometric graphs. In addition, from a practical point of view, options for enhancing the display of the computed layout are explored to help the user perform typical tasks in this context more effciently. One important aspect of the problems on geometric graphs considered in the third chapter of the thesis is that crossings of edges in a network structure are to be minimized while certain other properties such as connectedness are maintained. Motivated by this, in the fourth chapter of this thesis, related combinatorial and computational problems are explored from a more theoretical point of view. In particular some light is shed on properties of crossing-free spanning trees in geometric graphs.
Zur Eignung von Gd-EOB-DTPA zur Visualisierung des Transportes von Arzneimitteln zum Ort der Wirkung

Gadolinium-Ethoxybenzyl-Diethylentriaminpentaessigsäure (Gd-EOB-DTPA) ist ein leberspezifisches Magnetresonanztomographie (MRT)-Kontrastmittel. Es ist ein häufig in der Klinik eingesetztes Diagnostikum bei fokalen Leberläsionen. Im Vergleich zu anderen Gadolinium-haltigen Kontrastmitteln wird Gd-EOB-DTPA spezifisch von gesunden Hepatozyten aufgenommen. Somit ist es bei der Erkennung von hepatischen Tumoren von großer Bedeutung. Nach einer Bolusinjektion wird Gd-EOB-DTPA bis zu 50% über die Galle und 50% über die Nieren ausgeschieden. Die Mechanismen der hepatischen Aufnahme und der biliären Elimination sind bisher nur unzureichend verstanden. Ein weiterführendes Verständnis ist aber auch nötig, um die großen interindividuellen Unterschiede der Leberanreicherung von Gd-EOB-DTPA bei Patienten zu erklären und auch mögliche Arzneimittelinteraktionen vorhersagen zu können. Deswegen war das Ziel der vorliegenden Dissertation, erstens die Transportmechanismen des Kontrastmittels in in vitro Experimenten und dabei sowohl die Aufnahme- also auch die Effluxtransporterproteinen zu untersuchen. Zweitens wurde ein Tierexperiment in Wildtyp- und Abcc2-definzienten Ratten durchgeführt, um die Mechanismen der hepatobiliären Elimination von Gd-EOB-DTPA zu untersuchen und den intestinalen Arzneimitteltransportweg mit Hilfe des bildgegebenden Verfahrens MRT zu visualisieren. Diese in vitro-Untersuchungen zeigten, dass Gd-EOB-DTPA ein Substrat der leberspezifischen Transporter OATP1B1, OATP1B3 und NTCP, aber nicht des ubiquitären OATP2B1 ist. Hiermit kann die hohe Leberspezifität des Kontrastmittels erklärt werden. In vitro wurde Gd-EOB-DTPA von allen genetischen Varianten des OATP1B1 mit unterschiedlichen Km-Werten aufgenommen, wobei die *1b Variante eine signifikant erhöhte Transportkapazität im Vergleich zum Wildtyp (WT) zeigte. Allerdings sieht man bei OATP1B1*5 (nicht signifikant) und *15 (signifikant) eine verringerte Aufnahme von Gd-EOB-DTPA. Bei OATP1B3, zeigte die Variante p.233Ile eine signifikant niedrigere Substrat-Affinität in vitro. Gd-EOB-DTPA wurde über den Polymorphismus p.112Ala/233Ile mit einer signifikant niedrigeren Transportaktivität aufgenommen im Vergleich zum WT. Nach intravenöser Applikation von Gd-EOB-DTPA in Wildtyp- und Abcc2-defizienten Ratten wurde gezeigt, dass Abcc2 eine zentrale Rolle für die hepatobiliäre Ausscheidung von Gd-EOB-DTPA spielt. Gleichzeitig wurde gezeigt, dass bei Abcc2-defizienten Ratten die Ausscheidung des Kontrastmittels vollständig über die Nieren kompensiert wird. In Abcc2-defizienten Ratten ist die Expression des in der basolateralen Membran der Hepatozyten exprimierten Effluxtransporters Abcc3 signifikant erhöht. In vitro konnte eine konzentrationsabhängige Aufnahme von Gd-EOB-DTPA in ABCC2- bzw. ABCC3-enthaltende inside-out Vesikel dargestellt werden. Abcc3 könnte ein Kandidat für den Rücktransport von Gd-EOB-DTPA aus den Hepatozyten ins Blut sein. ABCC2 und ABCC3 sind auch im Darm exprimiert. Da Gd-EOB-DTPA ein Substrat von ABCC2 und ABCC3 ist, wurde untersucht, ob das Kontrastmittel auch im Darm absorbiert wird und somit die Absorption von Arzneimitteln visualisiert werden könnte. Nach oraler Gabe wurde Gd-EOB-DTPA im Darm der Ratten hinreichend absorbiert (Bioverfügbarkeit 17%). Für den intestinalen Efflux scheint ABCC2 von großer Bedeutung zu sein. Denn nach oraler Applikation stieg die Bioverfügbarkeit von Gd-EOB-DTPA in Abcc2-defizienten Ratten von 17% auf 25%. Zusammenfassend konnte die vorliegende Dissertation verdeutlichen, dass Gd-EOB-DTPA ein in vitro-Substrat der leberspezifischen Transporter OATP1B1, OATP1B3 und NTCP ist und auch von OATP1A2, ABCC2 und ABCC3 prozessiert wird. Der Arzneistoff ist somit ein gutes Beispiel für das komplexe Wechselspiel von intestinalem und hepatischem Aufnahme- und Effluxtransport, welcher für zahlreiche Stoffe bereits gut etabliert ist (z.B. Statine, Ezetmib). Es konnte gezeigt werden, dass die leberspezifische Aufnahme von Gd-EOB-DTPA über OATP1B1 und OATP1B3 realisiert wird, wohingegen ABCC2 den wesentlichen Effluxtransporter der hepoatobiliären Elimination darstellt. Die Arbeit konnte des Weiteren zeigen, dass genetische Varianten eine plausible Erklärung für die in der klinischen Praxis beobachtete hohe interindividuelle Variabilität der Leberanreicherung wären. Basierend auf den in vitro- und in vivo-Ergebnissen erweist sich, Gd-EOB-DTPA als neues „probe drug“, um den Absorptionsweg von Arzneimitteln zu visualisieren und die Funktion der Transporter zu charakterisieren.
