Aged rats recover poorly after unilateral stroke, whereas young rats recover readily possibly with the help from the contralateral, healthy hemisphere. We asked whether anomalous, age-related changes in the transcriptional activity in the brains of aged rats could be one underlying factor contributing to reduced functional recovery. We analysed gene expression in the periinfarct and contralateral areas of 3-month- and 18-month-old Sprague Dawley rats. Our experimental end-points were cDNA arrays containing genes related to hypoxia signalling, DNA damage and apoptosis, cellular response to injury, axonal damage and regrowth, cell lineage differentiation, dendritogenesis and neurogenesis. The major transcriptional events observed were: (i) Early up-regulation of DNA damage and down-regulation of anti-apoptosis-related genes in the periinfarct region of aged rats after stroke; (ii) Impaired neurogenesis in the periinfarct area, especially in aged rats; (iii) Impaired neurogenesis in the contralateral (unlesioned) hemisphere of both young and aged rats at all times after stroke and (iv) Marked up-regulation, in aged rats, of genes associated with inflammation and scar formation. These results were confirmed with quantitative real-time PCR. Conclusion I: reduced transcriptional activity in the healthy, contralateral hemisphere of aged rats in conjunction with an early up-regulation of DNA damage-related genes and pro-apoptotic genes and down-regulation of axono- and neurogenesis in the periinfarct area are likely to account for poor neurorehabilitation after stroke in old rats. Efficient neuroprotection after stroke requires long-term, regulated lowering of whole body temperature. After stroke, exposure of aged rats to hydrogen sulfide resulted in sustained, deep hypothermia (30.8 ± 0.7°C) that led to a 50% reduction in infarct size with a concomitant reduction in the number of phagocytic cells. At the transcription level, we found an overall decrease in the transcriptional activity related to inflammation and apoptosis. Behaviorally, hypothermia was associated with better performance on tests that require complex sensorimotor skills, in the absence of obvious neurological deficits or physiological side-effects, in aged rats. Conclusion II: Prolonged hypothermia is a simple and efficacious method to limit damage inflicted by stroke in aged rats.
Die von uns erhobenen Daten belegen eine transiente Dysfunktion des angeborenen und des adaptiven Immunsystems nach chirurgischen Operationen. Übereinstimmend mit den Ergebnissen anderer Arbeitsgruppen beobachteten wir eine Suppression des angeborenen Immunsystems mit einer reduzierten monozytären HLA– DR Expression im postoperativen Verlauf. Diese war abhängig von der Größe des Eingriffes. Die Intensität und Länge dieser postoperativen Immundysfunktion korrelierte mit der Schwere des chirurgischen Traumas. Patienten mit einer stärkeren und länger andauernden Immunsuppression haben dementsprechend ein erhöhtes Risiko für postoperative, infektiöse Komplikationen. Schwer septische Patienten zeigten unabhängig vom Outcome im Verlauf keinen Anstieg der Expressionsniveaus und verblieben in einem immunparalytischen Zustand. Im Gegensatz dazu stieg bei Patienten ohne postoperative Komplikationen die HLA– DR Expression zwischen dem ersten und dritten postoperativen Tag signifikant an. Ein Zusammenhang mit dem Auftreten einer postoperativen schweren Sepsis, in unserem Patientenkollektiv meist am 6 Tag postoperativ auftretend wird durch unsere Daten nahe gelegt. Die Einteilung der Sepsis in Typ A und Typ B verbindet die postoperativ vorhandene Immunsuppression chirurgischer Patienten mit dem dadurch gesteigerten Risiko einer schweren Sepsis. Bei Patienten mit einer Typ B Sepsis ist das Immunsystem aufgrund der vorhergehenden Operation wesentlich eingeschränkt. Insbesondere im Zusammenhang mit dem weitaus schlechteren Outcome bei Patienten mit Typ B Sepsis ist eine frühzeitige Bestimmung des Immunstatus postoperativ als präventive Maßnahme zur frühzeitigen Erkennung dieser Erkrankung sinnvoll. Eine präoperative Disposition für einen komplizierten Verlauf charakterisiert durch eine verminderte Oberflächenaktivität der Monozyten, wie sie von anderen Arbeitsgruppen bei Malignomen beobachtet wurde konnte nicht nachgewiesen werden. Ebenfalls veränderten neoadjuvante Therapieschemen weder die prä-, noch die postoperative Expression von HLADR Molekülen auf der Oberfläche von Monozyten. Neben dem angeborenen Immunsystem kam es nach großen chirurgischen Eingriffen zu einer Alteration der adaptiven Abwehr. Alle in unserer Studie eingeschlos- senen Patienten waren postoperativ von einer generalisierten Reduktion der transkriptionalen Aktivität der T– Zellen betroffen, diese war bei Patienten mit septischen Komplikationen stärker ausgeprägt. Die Ursachen für die schnelle Änderung des Phänotyps der Monozyten und T– Lymphozyten nach chirurgischen Eingriffen müssen in weiteren Untersuchungen genauer analysiert werden. Unsere Resultate belegen, dass die Determination von Monozyten und T– Zell Parametern zur frühzeitigen Identifikation von Hochri- sikopatienten nach chirurgischen Eingriffen sinnvoll ist. In weiteren prospektiven Studien muss geklärt werden, ob die Dynamik der HLA– DR Expression auf Monozyten auch bei größeren Patientenkollektiven eine Vorhersage für postoperative Komplikationen zulässt. Weiterhin ist fraglich ob mit der Microarray Technologie und der damit identifizierbaren Genexpression eine genaue Analyse der Abwehrlage des Patienten möglich ist. Der Vergleich der Komorbiditätsfaktoren Alter und BMI zeigte vermehrt Komplikationen und eine verminderte HLA– DR Expression in der Gruppe der übergewichtigen Patienten. Wir konnten in unserer Studie zeigen, dass Patienten mit einem erhöhten BMI ein höheres Risiko für postoperative Komplikationen haben. Insbesondere zeigte sich eine signifikant erhöhte Rate der schwereren Sepsis bei Patienten mit einem BMI >30. Die Nutzung des BMI zur Ergänzung bereits bestehender Risikostratifizierungssysteme scheint sinnvoll. Mit den unterschiedlichen Risikostratifizierungen konnten Patienten mit einer eingeschränkten postoperativen HLA- DR Expression identifiziert werden. Insbesondere bei Patienten, die mit der modifizierten Risikostratifizierung beurteilt wurden waren diese Unterschiede hochsignifikant.
Bisphenol A (BPA) ist ein Umweltschadstoff, der für die Produktion von Polykarbonatplastik und Epoxydharzen verwendet wird. Aufgrund seiner weltweiten Verbreitung, seiner Persistenz in der Umwelt und vor allen Dingen wegen seiner endokrinen Wirkung, stellt diese Verbindung eine große Gefahr für die Tier- und Pflanzenwelt sowie für den Menschen dar. Deshalb wurde aus einem bepflanzten Festbettreaktor, welcher mit BPA behandelt wurde, der Bakterienstamm RW4 isoliert, der fähig ist Bisphenol A als alleinige Kohlenstoff- und Energiequelle zu nutzen. Das Gram-negative Bakterium wurde mittels 16S rRNA-Analyse als Cupriavidus basilensis identifiziert und mittel der BIOLOG-Methode charakterisiert. Der Bakterienstamm kann ein sehr großes Spektrum an aliphatischen Stoffen wie Aceton, Ethanol, 1-Oktanol und aromatischen Verbindungen wie Phenol, Mono- und Dihydroxybenzoesäuren für das Wachstum nutzen. C. basilensis RW4 baut Bisphenol A nur sehr langsam ab. Es ist jedoch möglich den BPA-Abbau sowohl in Schüttelkulturen und als auch in kontinuierlich laufenden Sandsäulen durch Zugabe von Phenol als Co-Substrat und Wachstumsstimulanz deutlich zu steigern. Die erfassten Abbauintermediate wie 4-Hydroxyacetophenon und 2-(4-Propanol)-phenol wiesen darauf hin, dass C. basilensis RW4 einen ähnlichen Abbauweg nutzt wie Sphingomonas sp. TTNP3 von Kolvenbach et al. (2007) nutzt. In weiteren Untersuchungen, wurde die toxische Wirkung von Bisphenol A auf den Standardmikroorganismus Pseudomonas putida P8 in Schüttelkulturen und in kontinuierlich laufenden Sandsäulen analysiert. Es zeigte sich, dass BPA das Wachstum von P. putida P8 hemmt und Modifikationen in der Zusammensetzung der Phospholipide in der Membran hervorruft. P. putida P8 reagierte unmittelbar auf das Einwirken von BPA auf der Membranebene durch Veränderung des Verhältnisses der trans-ungesättigten Fettsäuren zu den cis-ungesättigten Fettsäuren, was schon in früheren Untersuchungen für andere organische Lösungsmittel beschrieben wurde. Dementsprechend wurde eine Korrelation zwischen der BPA-Konzentration und dem Anstieg des trans-cis-Verhältnisses ermittelt. Weil diese Anpassungsreaktion eine kurzfristige Antwort auf den einwirkenden Stressor ist, konnte der Anstieg des trans-cis-Verhältnisses nur kurz nach BPA-Zugabe erfasst werden. Somit kann das trans-cis-Verhältnis nur als temporärer Biomarker für die Toxizität von BPA in Schüttelkulturen genutzt werden, jedoch nicht in langfristig laufenden Sandsäulen.
We present classical and hybrid modeling approaches for genetic regulatory networks focusing on promoter analysis for negatively and positively autoregulated networks. The main aim of this thesis is to introduce an alternative mathematical approach to model gene regulatory networks based on piecewise deterministic Markov processes (PDMP). During somitogenesis, a process describing the early segmentation in vertebrates, molecular oscillators play a crucial role as part of a segmentation clock. In mice, these oscillators are called Hes1 and Hes7 and are commonly modeled by a system of two delay differential equations including a Hill function, which describes gene repression by their own gene products. The Hill coefficient, which is a measure of nonlinearity of the binding processes in the promoter, is assumed to be equal to two, based on the fact that Hes1 and Hes7 form dimers.However, by standard arguments applied to binding analysis, we show that a higher Hill coefficient is reasonable. This leads to results different from those in literature which requires a more sophisticated model. For the Hes7 oscillator we present a system of ordinary differential equations including a Michaelis-Menten term describing a nonlinear degradation of the proteins by the ubiquitinpathway. As demonstrated by the Hes1 and Hes7 oscillator, promoter behavior can have strong influence on the dynamical behavior of genetic networks. Since purely deterministic systems cannot reveal phenomenons caused by the inherent random fluctuations, we propose a novel approach based on PDMPs. Such models allow to model binding processes of transcription factors to binding sites in a promoter as random processes, where all other processes like synthesis, degradation or dimerization of the gene products are modeled in deterministic manner. We present and discuss a simulation algorithm for PDMPs and apply it to three types of genetic networks: an unregulated gene, a toggle switch, and a positively autoregulated network. The different regulation characteristics are analyzed and compared by numerical means. Furthermore, we determine analytical solutions of the stationary distributions of one negatively, and three positively autoregulated networks. Based on these results, we analyze attenuation of noise in a negative feedback loop, and the question of graded or binary response in autocatalytic networks.
The Syilx Okanagan environmental ethic is a philosophy expressed in the practice of Indigeneity as a social (cultural) paradigm and is identified by an inter-reliant experience in the land, as demonstrated in land-use practice which is shaped by the land’s realities as observed, learned and communicated to succeeding generations. Syilx Okanagan Indigeneity reflects an epistemology that optimum human wellbeing cannot be achieved through a human centered ethic but must focus on the optimum ability for the environment to regenerate itself. Syilx Okanagan stories convey the social experience and act as a records system to preserve, maintain and transfer the knowledge of the land. On one level, captikwl contain essential specific environmental knowledge as an oral documentation method, while on another level, as literature, captikwl reconstructs the ethos of interdependency specific to the ecology of the Syilx Okanagan territory through reenactment of nature’s interactions. captikwl in the Nsyilxcen language mimics the dynamic aspects of nature’s required regenerative principles to each succeeding generation, and acts as a feed-back loop reconstructing the social paradigm as an environmental ethic. captikwl might be seen as a distinctly Indigenous human adaptive response scheme within a natural system as it constructs the Syilx Okanagan world and results in behavior with sustainable outcome in the environment. captikwl is a distinct oral artistry that must be read through a literary framework cognizant of oral memory device, structure and Syilx Okanagan context. Okanagan author Morning Dove’s collection of Okanagan stories, as well as, the Mattina and DeSautel bilingual collections and other original version captikwl were consulted. Captikwl demonstrates the concept of tmixw which better translates as a life-force. The word for land is tmxwulaxw, which translates better as a life-force-place, rather than land as location or ecology type. Syilx society demonstrates an “ecological conscience” as the common text through captikwl which is enacted in their social institutions in the manner theorized by respected American conservationist Aldo Leopold, as desirable to achieve within society. The Syilx environmental ethic, rather than a sustainable human ethic of utility, is a willingness to live within a strict imperative to continuously sustain a unity of existence through societal knowledge and reverent practice of respect toward all life-forms. The Syilx environmental ethic diverges from ecocentrism, as articulated by Callicott, in recognizing a fundamental distinction between non-life forms and life forms, in their ability for self-regeneration through inter-reliance, as the focus for delineating moral considerability. The Syilx environmental ethic differs from biocentrism, as articulated by Taylor, in recognizing moral considerability as resting with the on-going life form which is capable of regeneration within its ecology, rather than the singular biological unit. The Syilx environmental ethic also differs from the concept of the ethics of place, as articulated by Berthold-Bond and characterized as an ethical bioregional human utility of a location. Tmixw is the life-force which makes up the tmxwulaxw or life-force-place and the humans are only “placed” as a life-force themselves through Indigeneity as a social paradigm within a criteria of full reciprocity in the regeneration of all life forms of a place. The Syilx environmental ethic also differs from the ethic of sustainability proposed by Daly as a steady state economic model of human utilitarianism, in the positioning of nature treated as capital, to be prudently developed in a way that off-sets depletions of renewable and non-renewable resources in meeting human requirements. The Syilx Okanagan view of economy, while structurally a sustainability model, does not construct value based on human utility as a defining line in decision-making as to which life forms are to be devalued and displaced. Syilx Indigeneity is guided through a societal dialogue practice of Enowkinwixw, a process of decision-making structured to include living within the requirements of the land to fully regenerate. Syilx Okanagan Indigeneity frames an environmental ethic from a tmixwcentric position and offers a model proposing an ethic of re-indigenization as a path to sustainability. The thesis proposes a common text for human society in the form of such literatures, since literature is widely accessible, which demonstrate, imbed and advocate a regenerative land ethic towards the re-indigenization of place.
Der Biokomplex “Reaktionsmuster in Vertebratenzellen” (RiV) besteht aus elektronenmikroskopisch nachweisbaren Partikeln, die stressinduziert sezerniert werden. Der Transkriptionsfaktor NF-kappa B ist bei inflammatorischen Prozessen und insbesondere auch bei der endothelialen Dysfunktion von Bedeutung. Ich habe die Hypothese untersucht, dass RiV-Partikel die zytokin-induzierte Expression von zellulären Adhäsionsmolekülen (ICAM-1, VCAM-1, E-Selektin) und des „Lectin-like oxidized“-LDL-Rezeptors-1 (LOX-1) in humanen umbilikalen venösen Endothelzellen (HUVEC) reduziert. Nach einer Inkubationszeit von vier Stunden wurde die TNF-alpha-induzierte mRNA-Expression von ICAM-1, VCAM-1 und LOX-1 unter dem Einfluss von RiV über eine reduzierte Translokation von NF-kappa B signifikant vermindert. Nach einer Inkubationszeit von 16 Stunden zeigte sich eine gesteigerte mRNA-Expression bei Exposition mit RiV durch eine Neusynthese der mRNA im Vergleich zu TNF-alpha-behandelten Endothelzellen. Die Oberflächenexpression blieb unverändert. Unter dem Einfluss von RiV auf TNF-alpha-stimulierte HUVEC wurde das Shedding von sICAM-1 und sE-Selektin induziert. RiV hatte keinen relevanten Einfluss auf die Expression der endothelialen NO-Synthase, eNOS. Der Biokomplex RiV enthält thermosensitive und thermostabile Komponenten.
Eine häufige Ursache von Hypoglykämien bei insulinbehandelten Diabetespatienten ist die fehlende oder unzureichende vorausschauende Anpassung der Insulindosierung an erhöhte körperliche Aktivitäten. Ziel der vorliegenden Arbeit war es, die erforderliche Reduktion der Insulindosierung vorhersagbar zu ermitteln, um dem einzelnen Patienten geeignete Empfehlungen geben zu können. Grundlage der durchgeführten Untersuchungen war eine vorausgegangene klinisch-experimentelle Studie im Institut für Diabetes „Gerhardt Katsch“ Karlsburg, die gezeigt hatte, dass es im aeroben Belastungsbereich bezüglich der Blutglukosekontrolle ein Insulindosisäquivalent für körperliche Aktivitäten (IDE) gibt. Dieses kann in IE sc. applizierten Kurzzeitinsulins pro Wh ausgedrückt werden und ist eine belastungsabhängige Funktion der Steady-state-Herzfrequenz (SSHF). Mittels eines aus den dabei angefallenen Daten entwickelten, publizierten und zur Anwendung patentierten Nomogramms können die IDE definierter Aktivitäten anhand der im Ergometerstufentest gemessenen Belastungsherzfrequenz bestimmt werden. Die hier beschriebene nichtinterventionelle klinisch-diagnostische Beobachtungsstudie wurde an 63 Kindern mit Typ 1 Diabetes (Alter 8-18 J., Diabetesdauer 0,5 - 14 J.) während einer 3-wöchigen stationären Reha-Maßnahme durchgeführt. Zunächst wurden in Fahrradergometerstufentests die IDE als reproduzierbare leistungsbezogene individuelle Kennziffer in IE Kurzzeitinsulinäquivalent pro 50 Wh identifiziert. Die SSHF ist dabei auch der Ausweis der Fitness (körperlicher Trainingszustand), wenn sie mit einem, ebenfalls im Rahmen der Studie ermittelten, geschlechts- und altersbezogenen Erwartungswert folgendermaßen verglichen wird: (gemessene SSHF – Erwartungswert) ± SD entspricht durchschnittlicher Fitness, darunter liegende Werte weisen überdurchschnittliche Fitness aus, darüber liegende Werte repräsentieren eine unterdurchschnittliche Fitness. Insgesamt betragen die IDE im Mittel 2,6 IE/50 Wh (4,3 IE/50 Wh bei überdurchschnittlicher, 2,7 IE/50 Wh bei durchschnittlicher und 0,6 IE/50 Wh bei unterdurchschnittlicher Fitness). Der Hauptteil der Studie war ein Feldtest mit 24 h - Herzfrequenz(HF)-Monitoring bei Protokollierung der körperlichen Aktivitäten in 15 min-Intervallen. Aus den HF-Profilen wurden mittels der zuvor im Ergometertest ermittelten leistungsbezogenen SSHF individuell die scheinbaren zirkadianen körperlichen Belastungsprofile „Wh = f(t)“ und daraus anhand des Nomogramms die entsprechenden IDE- Profile „IE Kurzzeitinsulinäquivalente = f(t)“ ermittelt. Patienten mit durchschnittlicher Fitness entwickeln bei mäßig belastenden körperlichen Aktivitäten (z.B. Wandern, Gymnastik, Hausarbeit) IDE-Aktivitäten von 0,5–1,0 IE/h, bei mittlerer Belastung (z.B. Fitnesstraining, Reiten, Gelände- und Ballspiele) 1,0–2,0 IE/h und bei starker Belastung (z.B. Jogging, Radfahren, Schwimmen, Tanzen) 1,5-2,5 IE/h. Für Patienten mit überdurchschnittlicher Fitness betragen diese Werte 1,0–1,5, 1,5–2,5 und 2,5–4,0 IE/h. Für Patienten mit unterdurchschnittlicher Fitness liegen sie durchschnittlich bei 0,1, 0,2 bzw. 0,3 IE/h und sind somit bei klinischen Empfehlungen im Allgemeinen zu vernachlässigen. Durchschnittlich entfallen unter den betrachteten Tagesabläufen in diesen 3 Fitnessgruppen 30%, 25% und 5% der täglichen Gesamtinsulinwirkung in der Blutglukosekontrolle auf die Auswirkung körperlicher Aktivitäten. Die vorgelegten Daten können bereits jetzt als Beratungs- und Schulungsgrundlage zur vorbeugenden Insulindosisreduktion vor überdurchschnittlichen körperlichen Belastungen dienen. Vorläufige Studien zur Anwendung der IDE im Blutglukosemanagement, welche die Machbarkeit, Akzeptanz und den möglichen Nutzen zeigen, sind zu vertiefen.
Die Körpergröße kann als ein Marker der Entwicklungsgeschichte von Individuen angesehen werden und ist damit auch durch eine besondere Krankheitsempfindlichkeit beeinflusst. Wenn schon im frühen Lebensalter eine Disposition zu entzündlichen Erkrankungen vorliegt, kann diese eine lebenslange inflammatorische Belastung mit sich bringen und zu einem verzögerten Längenwachstum führen. Auswirkungen der frühen Entwicklung können durch die Körpergröße auch im späteren Leben noch zu einem gewissen Maße nachvollzogen werden. Die Basisstudie SHIP-0 (Study of Health in Pomerania) zeigte einen Zusammenhang zwischen der Körpergröße und Entzündungen des Parodonts. Die 5-Jahres-Folgestudie (SHIP-1) sollte diesen Zusammenhang bestätigen. Folgende Ergebnisse wurden erzielt: 1. Die Körpergröße hatte nur einen geringen Einfluss auf die Veränderung der parodontalen Situation in den fünf Folgejahren. 2. Es zeigte sich eine Tendenz des Zusammenhangs des Attachmentverlustes mit der Körpergröße. 3. Es gab Hinweise darauf, dass größere Probanden in Zusammenhang mit den Risikofaktoren einen geringeren Attachmentverlust aufwiesen als die Kleineren. 4. Die Risikofaktoren (Rauchverhalten, Bildungsniveau, Diabetes mellitus, Alter und Geschlecht) zeigten einen größeren Einfluss auf die Schwere und das Ausmaß der Parodontitis. 5. Der Zusammenhang der Körpergröße mit der parodontalen Erkrankung im Zusammenwirken mit den Risikofaktoren konnte nicht vollständig bestätigt werden. 6. Die Entzündungsparameter WBC und CRP wiesen bei den größten Probanden die geringsten Werte auf im Vergleich zu den kleinsten Probanden. 7. Von den 4290 Probanden der SHIP-0-Studie nahmen nur noch 3300 Probanden an der SHIP-1-Studie teil. Dadurch hat sich die Verteilung der Risikofaktoren verändert. Daher ergibt die Folgestudie nicht mehr bevölkerungs-repräsentative Aussagen, weil besonders die risikobelasteten Patienten dem follow-up ferngeblieben sind. Mit dieser Studie konnte der Zusammenhang der Körpergröße und Entzündungen mit der Parodontitis nicht vollständig bestätigt werden. Der Zeitraum von fünf Jahren ist möglicherweise nicht ausreichend, so dass die 10-Jahres-follow-up-Studie eine Klärung bringen sollte.
Asymmetrical capacitively coupled RF discharges in oxygen, argon and hydrogen have been experimentally investigated with the innovative technique of the phase resolved optical emission spectroscopy. This diagnostic tool allows to measure spatio-temporally resolved emission intensities of electronically excited species with a high resolution. The spatial (axial) resolution was better than 1 mm and a temporal resolution of about 1.5 ns has been achieved. Therefore the plasma induced optical emission within the RF cycle (TRF = 73.75 ns) from the RF sheath region with a typical mean sheath thickness of about 5mm has been studied. Spatio-temporally resolved optical emission patterns of the following optical transitions have been measured for a total gas pressure in the range of 20 to 100 Pa and self-bias voltages between -50 and -550 V: Oxygen plasma Emission at 777.4 nm and 844.6 nm (atomic oxygen) Argon plasma Emission at about 751 nm and 841 nm (argon) Hydrogen plasma Emission at 656.3nm (atomic hydrogen, H alpha-line) These transitions are the most prominent ones of the investigated excited species in these plasmas as could be shown from overview spectra of the plasma induced optical emission in the range from 350 to 850 nm. For the first time such extensive PROES measurements in oxygen CCRF plasmas are presented in this work. The additional investigations of argon and hydrogen plasmas serve as a reference and for a direct comparison with results from the literature. The temporal behavior of the emission intensity is influenced by the effective lifetime of the emitting states which is on the order of the nanosecond time scale of the RF cycle. Therefore, it does not represent the real temporal behavior of the excitation. A simple method has been applied to calculate relative excitation rates from the measured emission intensities to distinguish different excitation mechanisms and their correct relative temporal behavior. In a close collaboration within the framework of the Sonderforschungsbereich Transregio 24 'Fundamentals of Complex Plasmas' a newly 1d3v PIC-MCC code for simulations of capacitive RF discharges in oxygen has been developed by Matyash et al. The very close coupling of experiment and modeling allowed a really detailed and microscopic understanding of the processes and dynamics from the sheath to the bulk plasma in CCRF discharges. The spatio-temporally resolved excitation rate profiles show four different excitation structures (I-IV). Excitation processes due to the following mechanisms in CCPs could be identified and characterized: I Electrons expelled from growing sheath II Electrons detached from negative ions (collisions with neutrals) + secondary electrons from the electrode surface (ion bombardment) III Field-reversal effect, reduced mobility of electrons (electron-neutral collisions) IV Heavy-particle collisions These excitation mechanisms are characterized by different temporal and spatial behaviors of the excitation rate within the RF cycle. Additionally it has been shown that the excitation by electron impact in the investigated oxygen plasmas results mainly from dissociative electron impact excitation (O2 + e -> O + O* + e) and not from direct electron impact excitation (O + e -> O* + e). Actinometry measurements show that the results are not really credible. Thus actinometry is not applicable on the investigated oxygen RF plasma. A challenge in interpretation is the observed excitation pattern IV. Pattern IV has to be caused in connection with heavy particle collisions nearby the electrode surface and could be observed in all the three plasmas oxygen, argon and hydrogen. It is located directly in front of the powered electrode and appears during almost the whole RF cycle. The temporal modulation is nearly sinusoidal and weak in comparison to the first three patterns. This is due to the weak RF modulation of the ion flux towards the electrode surface which has been proven by a PIC simulation. It could be shown that the modulation degree of pattern IV depends on the transition time of the corresponding positive ions through the RF sheath which is influenced by the ion mass. In oxygen as well as in argon CCRF plasmas pattern IV is less modulated than in hydrogen CCRF plasmas due to the heavier ions in oxygen and argon. Additionally the modulation degree increases with increasing pressure due to the more confined plasma at higher pressures which is yielding in a stronger modulated ion current towards the powered electrode.
The experimental determination of the electron energy distribution of a low pressure glow discharge in neon from emission spectroscopic data has been demonstrated. The method extends an approach by Fischer and Dose [5]. The spectral data were obtained with a simple overview spectrometer and analyzed using a strict probabilistic, Bayesian data analysis. It is this Integrated Data Analysis (IDA) approach, which allows the significant extraction of non-thermal properties of the electron energy distribution function (EEDF). The results bear potential as a non-invasive alternative to probe measurements. This allows the investigation of spatially inhomogeneous plasmas (gradient length smaller than typical probe sheath dimensions) and plasmas with reactive constituents. The diagnostic of reactive plasmas is an important practical application, needed e.g. for the monitoring and control of process plasmas. Moreover, the experimental validation of probe theories for magnetized plasmas as a long-standing topic in plasma diagnostics could be addressed by the spectroscopic method.
