- TNF-alpha and TRAIL represent interesting candidates for treatment of malignant tumors. several cancers display a resistance to the apoptotic effects of TNF-alpha and TRAIL application, suggesting that treatment with TRAIL and TNF-alpha alone can be insufficient for tumor treatment. in this study we explored the apoptotic response of A549 lung cancer cells to TRAIL and TNF-alpha application in combination with suppressing the antiapoptotic proteins Bcl-xl and PKC-eta. transfection of A549 cells with chimeric antisense-ODN resulted in a time and dose dependent knockdown of Bcl-xl and PKC-eta as shown by RT-PCR and western-blot-analysis. knockdown of Bcl-xl and PKC-eta displayed an enhanced cytotoxic effect of TRAIL in caspase-activity tests and cell cycle analysis. we did not see the induction of apoptosis by TNF-alpha in the cell line A549 however. our conclusion is that Bcl-xl and PKC-eta act as significant resistance factors in regard to TRAIL application and represent exploitable targets to enhance the cytotoxic effects of apoptosis inducing agens like TRAIL.
