Background: Men and women with valvular heart disease have different risk profiles for clinical endpoints. Non-esterified fatty acids (NEFA) are possibly involved in cardio-metabolic disease. However, it is unclear whether NEFA concentrations are associated with physical performance in patients undergoing transcatheter aortic valve implantation (TAVI) and whether there are sex-specific effects. Methods: To test the hypothesis that NEFA concentration is associated with sex-specific physical performance, we prospectively analysed data from one hundred adult patients undergoing TAVI. NEFA concentrations, physical performance and anthropometric parameters were measured before and 6 and 12 months after TAVI. Physical performance was determined by a six-minute walking test (6-MWT) and self-reported weekly bicycle riding time. Results: Before TAVI, NEFA concentrations were higher in patients (44 women, 56 men) compared to the normal population. Median NEFA concentrations at 6 and 12 months after TAVI were within the reference range reported in the normal population in men but not women. Men but not women presented with an increased performance in the 6-MWT over time (p = 0.026, p = 0.142, respectively). Additionally, men showed an increased ability to ride a bicycle after TAVI compared to before TAVI (p = 0.034). NEFA concentrations before TAVI correlated with the 6-MWT before TAVI in women (Spearman’s rho −0.552; p = 0.001) but not in men (Spearman’s rho −0.007; p = 0.964). No association was found between NEFA concentrations and physical performance 6 and 12 months after TAVI. Conclusions: NEFA concentrations improved into the reference range in men but not women after TAVI. Men but not women have an increased physical performance after TAVI. No association between NEFA and physical performance was observed in men and women after TAVI.
The content of this thesis can be summarized as follows: (i) The deposition processes of SiOx and SiOxCyHz coatings were investigated in a low-pressure, low temperature HMDSO-O2-N2 plasmas. Infrared laser absorption spectroscopy (IRLAS) and optical emission spectroscopy (OES) were combined to measure the gas temperatures in the hot and colder zones of the plasma as well as to monitor the concentration of the methyl radical, CH3, and of seven stable molecules, HMDSO, CH4, C2H2, C2H4, C2H6, CO and CO2. Tunable lead salt diode lasers (TDLs) and an external-cavity quantum cascade laser (EC-QCL) were simultaneously employed as radiation sources to perform the IRLAS measurements. They were found to be in the range between 10^{11} to 10^{15} cm^{−3}. The influence of the discharge parameters of power, pressure and gas mixture on the molecular concentrations was studied. The plasma generation is characterized by a certain degree of inhomogeneity with different temperature zones, i.e., hottest, hot and colder zones depending on the construction of the reactor. This complexity is characterized by the multiple molecular species including the HMDSO precursor and products in ground and excited states existing in the plasma. (ii) Employing similarly IRLAS and OES techniques, the deposition of nanocrystalline diamond at relatively low temperature in low-pressure MW H2 plasmas with small ad-mixtures of methane and carbon dioxide was investigated. Five methods were applied for an extensive temperature analysis, providing new insights into energetic aspects of the multi-component non-equilibrium plasma. The OES method provided information about the gas temperature of H2 inside the MW plasma. Using lead salt diode lasers, the rotational temperature of the methyl radical, CH3 , and gas temperature of methane molecule, CH4 , was measured. A variety of CO lines in the ground and in three excited states have been analysed using an EC-QCL with a relatively wide spectral range. These methods have shown that based on the construction of the DAA reactor using 16 single plasma sources the plasma generation is characterized by a variety of hottest, hot and colder zones. Extensive measurement of these various species temperatures in the complex plasma enabled the concentration determination of the various stable and unstable plasma species, which were found to be in the range between 10 11 to 10 15 cm −3 . The influence of the discharge parameters, power and pressure, on the molecular concentrations has been studied. To achieve insight into general plasma chemical aspects, the dissociation of the carbon precursor gases including their fragmentation and conversion to the reaction products was analysed in detail. The evolution of the concentration of the methyl radical, CH 3 , of five stable molecules, CH4, CO2, CO, C2H2 and C2H4, and of vibrationally excited CO in the first and second hot band was monitored in the plasma processes by in situ infrared laser absorption spectroscopy using lead salt diode lasers (TDL) and an external-cavity quantum cascade laser (EC-QCL) as radiation sources. OES was applied simultaneously to obtain complementary information about the degree of dissociation of the H2 precursor gas. The analysis of the carbon and oxygen mass balances shows clearly, that the deposition on the reactor walls and the production of other hydrocarbons species may act as sinks for carbon and oxygen. (iii) The absolute line strengths of many P-branch transitions of the ν3 fundamental of {28}^SiH4 were determined using the wide tuning range and the narrow line width of a cw EC-QCL between 2096 and 2178 cm^{−1}. The line positions and line strengths of transitions of the stretching dyad within the P-branch of {28}^SiH4 were determined with an estimated experimental measurement accuracy of 10%. The high spectral resolution available has enabled us to resolve and measure representative examples of the tetrahedral splittings associated with each component of the P-branch. The positions of these components are in excellent agreement with spherical top data system (STDS) predictions and theoretical transitions from the TDS spectroscopic database for spherical top molecules. To our knowledge, this is the first reported measurement of these line strengths in this band and is an example of the applicability of high-powered, widely tunable EC-QCLs to high resolution spectroscopy in the MIR. (iv) Similarly, the determination of the silyl radicals, ν3 band, line strengths is ongoing using the same cw EC-QCL. This effort was impaired by silane and other unknown species lines overlap; however, the silyl radicals was successfully detected in a SiH4/H2 plasma. A method to determine the silyl line strengths has been presented through its iterative decay measurements which relied on the value of the silyl radical self reaction constant. There was a consensus of its value in the literature.
Physics-regularized Machine Learning To Approximate 3D Ideal-MHD Equilibria At Wendelstein 7-X

The magnetohydrodynamic (MHD) equilibrium model is one of the fundamental building blocks in the description of a magnetically confined plasma. The computational cost of constructing solutions to the 3D ideal-MHD equilibrium problem is one of the limiting factors in stellarator research and design; in particular, it limits the extent to which we can perform sample-intensive applications, applications which require many samples to be evaluated to yield meaningful results. Sample-intensive applications in stellarator research and design include, for example, equilibrium reconstruction, stellarator optimization, and flight simulators. In this thesis, we investigate how faithfully artificial neural networks (NNs) can quickly approximate ideal-MHD equilibria in stellarator geometries, starting with Wendelstein 7-X (W7-X), the world’s most advanced stellarator. In particular, we investigate (see section 1.7):
RQI: to what extent can NN models approximate the MHD equilibrium solution for different W7-X configurations and plasma profiles? What
is the speed-accuracy trade-off offered by NN models?
RQII: to what degree the NN model faithfully reproduces equilibrium quantities of interest (e. g., MHD stability)? To what extent can NN models meet the requirements of downstream applications (e. g., Bayesian
inference, stellarator optimization) in terms of equilibrium quantities
accuracy?
RQIII: whether we can exploit the implicit representation of a MHD equilibrium, i. e., the equilibrium solution should satisfy the ideal-MHD force
balance equation, to improve the NN approximation’s accuracy;
RQIV: the reconstruction of the full posterior istribution of plasma parameters and equilibrium quantities with self-consistent MHD equilibria; moreover, how does the adoption of MHD equilibria approximated by NN models affect the inferred plasma parameters?
A deep NN model is developed to learn the ideal-MHD solution operator in W7-X operational subspace, yielding 3D equilibria up to six orders of magnitude faster than currently available MHD equilibrium codes. Physics domain knowledge is embeded into the NN model: equilibrium solution symmetries are satisfied by construction, and the MHD force balance regularizes the NN model to satisfy the ideal-MHD equations. The model accurately predicts the equilibrium solution and it faithfully reproduces global equilibrium quantities and proxy functions used in stellarator optimization. Finally, the developed fast NN equilibrium model has been applied in downstream applications to obtain W7-X configurations with improved fast-particle confinement and to infer plasma parameters with self-consistent MHD equilibria at W7-X.
Physiological and neural synchrony in emotional and neutral stimulus processing: A study protocol

Background: As psychotherapy involves at least two individuals, it is essential to include the interaction perspective research. During interaction, synchrony, i.e., the occurrence of simultaneous responses, can be observed at the physiological, neural, and behavioral level. Physiological responses include heart rate and electrodermal activity; neural markers can be measured using electroencephalogram. Emotionally arousing stimuli are allocated more attentional resources (motivated attention), which is reflected in physiological activation and brain potentials. Here we present a protocol for a pilot study implementing a new research methodology, and replication of the motivated attention to emotion effect in in dyads. There is evidence that higher synchrony is associated with more positive (therapeutic) relationships. Thus, the secondary outcome will be the association between physiological and neural synchrony and subjective ratings.
Methods and design: Individuals (18−30 years) will participate in same-sex pairs in two experiments. In the first experiment (triadic interaction), both participants attentively watch unpleasant, neutral and pleasant pictures, and read/listen to standardized scripts (unpleasant, neutral, and pleasant, respectively) for the imagination task. In the second experiment, participants will read out three scripts (unpleasant, neutral, pleasant) to each other, followed by a joint imagination period. Stimuli will be presented in counterbalanced orders. After each picture and imagination, participants rate their subjective arousal and valence. In the beginning and in the end of the procedure, dyads rate their relationship, sympathy, and bonds (Working Alliance Inventory subscale). Heart rate, electrodermal activity and electroencephalogram will be continuously measured during both experiments using portable devices (EcgMove4 and EdaMove4, nine-channel B-Alert X-Series mobile-wireless EEG). Synchrony analyses will include the dual electroencephalography analysis pipeline, correlational analyses and Actor–Partner Interdependence Models.
Discussion: The present study protocol provides an experimental approach to investigate interpersonal synchrony during emotion processing, allowing for the establishment of research methods in a pilot study, which can later be translated into real-life psychotherapy research. In the future, fundamental understanding of such mechanisms in dyadic interactions is essential in order to promote therapeutic relationships, and thus, treatment effectiveness and efficiency.
Global climate change is omnipresent all over the world and is affecting and challenging organisms in various ways. Species either have to adapt to the changing environmental conditions or move to new habitats in order to avoid extinction.
Possible ways for an organism to react can be dispersal, phenotypic plasticity, genetic adaptation or a combination of these factors. Among the various consequences of climate change, especially changes in temperature affect plenty of species. In ectotherms, the body temperature and associated mechanisms are strongly dependent on environmental conditions.
The aim of this work was to investigate the mechanisms underlying adaptation to thermal variation and heat stress in the widespread butterfly species <i>Pieris napi<i>.
Focusing on indicators of individual condition, including morphology, physiology and life history traits, the purpose was to specify whether the species’ responses to temperature variation have a plastic or genetic basis. In the first experiment, phenotypic variation along a latitudinal and altitudinal cline was investigated. Yellow reflectance of wings was negatively correlated with wing melanisation, providing evidence for a trade-off between a sexually selected trait (yellow color) and thermoregulation (black color). Body size decreased with increasing latitude and led to the assumption that warmer conditions are more beneficial for <i>P. napi<i> than cooler ones. An increased flight performance at higher altitudes but not latitudes may
indicate stronger challenges for flight activity in high-altitude environments.
The second experiment focused on clinal variation and plasticity in morphology, physiology and life history in F1-generation individuals reared in captivity at different temperatures. It could be shown that individuals from cooler environments were less heat-tolerant, had a longer development but were nevertheless smaller, and had more melanised wings. These differences were genetically-based. Furthermore, it could be shown that a higher developmental temperature speeded up development, reduced body size, potential metabolic activity, and wing melanisation but increased heat tolerance, documenting plastic responses.
In a third experiment, we examined physiological responses to heat stress. A transcriptome analysis revealed an upregulation in molecular chaperones under hot conditions, whereas antioxidant responses and oxidative damage remained unaffected. The antioxidant glutathione (GSH) though was reduced under both cold and hot conditions. Interestingly, Swedish individuals were characterized by higher levels of GSH, lower early fecundity, and lower larval growth rates compared with German or Italian populations, suggesting a ‘pace-of-life’ syndrome. Thus, the individuals from warmer regions show the opposite pattern with a lower investment into maintenance but a faster lifestyle.
In summary, we found clinal variation in body size, growth rates and concomitant development time, wing aspect ratio, wing melanisation and heat tolerance. The effects of high developmental temperature very likely reflect adaptive phenotypic plasticity. When speeding up development; heat tolerance is increasing while body size, potential metabolic activity and wing melanisation are decreasing. Overall, body size of <i>P. napi<i> individuals decreased from south to north while the melanisation of the wings increased. Furthermore, we found a connection between increased wing melanisation and decreased yellow reflectance, most likely caused by a trade-off between the two. We could confirm that <i>P. napi<i> individuals from warmer environments were more heat-tolerant and larger than individuals from colder environments. Due to increasing temperatures and heat waves becoming more frequent in the future, being able to cope with such conditions will be advantageous. As warmer conditions had positive effects on individual development, <i>P. napi<i> may benefit from global warming, but its association with moist habitats suggests negative consequences of climate change. We could also reveal pronounced plastic and genetic responses in <i>P. napi<i>, which may indicate high adaptive capacities. Thus, increasing temperature may not be too problematic for the species, as it seems to be rather well equipped to deal with such challenges. However, as climate change entails changes in precipitation / humidity along with temperature changes, such issues need further investigation.
The six extraocular muscles (EOMs) are arranged around the eyeball as agonist-antagonist pairs performing the eye movements. The EOMs comprise a distinct muscle group that is fundamentally different from other skeletal muscle, which is reflected on many levels, such as functionality, anatomy as well as in their molecular make-up. Physiologically EOMs are considered superfast, high endurance muscles that are continuously active. In addition, EOMs contain unusual slow-tonic fibers that share features with amphibian and avian slow-tonic fibers. EOMs also express slow/cardiac isoforms of proteins and genes along with the typical isoforms of fast muscle fibers. Another striking hallmark of EOM is their differential involvement in a number of diseases. For instance, EOMs are preferentially spared in Duchenne Muscular Dystrophy (DMD). DMD is the most common fatal, genetic disease in males clinically characterized by progressive muscle wasting. Mutations in the dystrophin gene result in a destabilization of the muscle membrane causing muscle fiber damage. While all other skeletal muscles deteriorate the EOMs remain morphologically and functionally healthy. In the pathogenesis of DMD elevated Ca2+ levels are believed to be an early event and it has been shown that EOMs are protected from pharmacologically induced Ca2+ damage. The goal of this study was to characterize the spared EOMs, in particular their Ca2+ homeostasis, in the context of DMD pathology to reveal new potential therapeutic targets for the disease. A combination of physiological, molecular and biochemical methods was used to investigate the Ca2+ homeostasis of EOMs to demonstrate clear differences compared with the fast limb muscle tibialis anterior (TA). Ca2+ handling of stimulated cultured EOM myotubes suggested more efficient Ca2+ removal from the cytoplasm after induced Ca2+ influx compared with cultured myoblasts from TA. Subsequent mRNA and protein expression analyses of myoblasts and adult muscle tissue revealed high expression levels of many key Ca2+ regulating and buffering proteins in rodent EOMs compared with TA. Among these Ca2+ proteins were slow/cardiac proteins, which normally are not found in fast muscles. For instance, the sarcoplasmic Ca2+ ATPase SERCA2 was elevated along with its regulator phospholamban (PLN). Further, PLN was preferentially endogenously phosphorylated at Thr17 suggesting continuous activation of SERCA2 and possibly the fast isoform SERCA1, the main Ca2+ pumps responsible for removing Ca2+ from the cytoplasm after muscle contraction. Furthermore, Ca2+ buffers, such as calsequestrin (CASQ2) and parvalbumin (PARV) were elevated. These results suggest that EOMs are endowed with a unique and superior Ca2+ homeostasis that facilitates efficient Ca2+ buffering and removal from the cytoplasm. This is in agreement with their continuous and fast activation cycles, as well as with a potential protective mechanism in prevention of Ca2+ overload in DMD. The extreme activity patterns of EOM suggested that a high activity of store-operated Ca2+ entry (SOCE) plays a critical part to replenish Ca2+ for rapid and continuous cycles of contractions. To extend the data on general Ca2+ homeostasis and because of possible implications of store-operated Ca2+ influx and other Ca2+ influx pathways in DMD, the expression patterns of group 1 transient receptor potential (TRP) channels and the proteins Orai1 and STIM1 were studied. The TRP channels, TRPC1, TRPC6 and TRPV4 channel proteins in addition to STIM1 showed higher expression in EOM compared with TA. High TRPC1, TRPV4 and STIM1 levels could play a significant role in the high fatigue resistance, muscle differentiation and SOCE in EOM. In addition, tissue from the mdx mouse model of DMD was investigated. The only channels differentially expressed in mdx EOM compared with normal EOM were TRPM4 and TRPM7 (decreased in mdx EOM) and TRPV4 (increased in mdx EOM). Although, these changes in mdx EOM were of small magnitude, they could point toward subtle compensatory changes related to the disease process. In general, EOMs seem to be unaffected by the disease and inherently protected. In conclusion, the results in this thesis have improved the understanding of the Ca2+ homeostasis in EOMs and suggest that EOM may be better able to prevent prolonged elevation of cytoplasmic Ca2+ levels. These data may help to design new therapeutic approaches targeting Ca2+ handling proteins to ameliorate muscular dystrophy.
Wird nach einer ischämischen Phase, in der Herzmuskelgewebe vorübergehend mit Sauerstoff und weiteren Nährstoffen pathologisch bedingt unterversorgt wird, die Durchblutung des Gewebes wiederhergestellt (Reperfusion), finden auf zellulärer Ebene komplexe Signaltransduktionsmechanismen statt, die entweder für das Gewebe schädlich sein können („Ischämie-Reperfusionsschaden“) oder das Herzmuskelgewebe vor weiteren Schädigungen schützen (Kardioprotektion). Felix et al. gaben Hinweise auf während der frühen Reperfusion freigesetzte Substanzen in post-ischämischem Effluat, die den Calciummetabolismus von isolierten adulten Rattenkardiomyozyten beeinflussen und in Folge die Kontraktilität reduzieren (Felix et al. 2001). Ziel der Arbeit war es, die Signalwege zu identifizieren, die negativ-inotrop auf isolierte Kardiomyozyten wirken, um letztendlich Rückschlüsse auf die Identität der Mediatoren im Effluat ziehen zu können. Experimentelle Vorarbeiten gaben Hinweise darauf, dass die negativ-inotropen Mediatoren des Effluates aus dem Arachidonsäure-Stoffwechsel freigesetzt werden. Daher wurde in der Arbeit die Rolle des Cyclooxygenase- und Prostaglandin-Metabolismus bei der Vermittlung des negativ-inotropen Effekts von post-ischämischem Effluat auf isolierte Kardiomyozyten untersucht. Nach 10 min globaler „stop-flow“-Ischämie wurden Herzen adulter Ratten reperfundiert und das Koronareffluat über 30 s gesammelt. Die Effekte dieses post-ischämischen Effluates auf die Zellkontraktion (systolische Zellverkürzung) und den Calciumstoffwechsel (Calciumtransienten) wurden mit Hilfe der Fluoreszenzmikroskopie an elektrisch stimulierten, laminierten und mit Fura-2AM gefärbten adulten Rattenkardiomyozyten analysiert. Als Kontrolle wurde Effluat aus der Perfusion vor der Ischämiephase verwendet. Die vorliegende Untersuchung analysierte die Rolle der Cyclooxygenase und von Prostaglandin-Rezeptoren in der Signaltransduktion der negativ-inotropen Faktoren durch Anwendung verschiedener Inhibitoren und Antagonisten. Die Expression der Cyclooxygenase-Isoformen und der verschiedenen Prostaglandin-Rezeptoren wurde mittels Western Blot und Immunfluoreszenzfärbungen untersucht. Laminierte und mit Fura-2AM gefärbte adulte Kardiomyozyten exprimierten beide Isoformen der Cyclooxygenase. Die Expression der Stress-induzierten Cyclooxygenase-2 war in laminierten Kardiomyozyten im Vergleich zu nicht-laminierten Kardiomyozyten erhöht. Adulte Rattenherzen exprimierten auf niedrigem Level basal die Cyclooxygenase-2. Eine 10-minütige Ischämiephase führte nicht zu einer Verstärkung der Expression. Durch Vorinkubation der Zellen mit dem Cyclooxygenase-Inhibitor Indomethacin und den Cyclooxygenase-2-Inhibitoren NS-398 und Lumiracoxib wurde der negativ-inotrope Effekt von post-ischämischem Effluat auf Calciummetabolismus und Kontraktilität gehemmt. Der Cyclooxygenase-1-Inhibitor SC-560 hingegen beeinflusste die Effekte des post-ischämischen Effluates nicht. Post-ischämisches Effluat reduzierte in autonom kontrahierenden neonatalen Kardiomyozyten wie in adulten Zellen den Calciumtransienten und wirkte außerdem reprimierend auf die Schlagfrequenz. Die reduzierende Wirkung auf beide Parameter konnte durch Anwendung von Indomethacin, NS-398 und Lumiracoxib blockiert werden. Im Western Blot konnten in isolierten Kardiomyozyten Rezeptoren für Prostaglandin D (DP1 und DP2), E (EP1-EP4), I und Thromboxane nachgewiesen werden. Durch die Anwendung selektiver Prostaglandin-Rezeptor-Antagonisten konnte der Signaltransduktionsmechanismus „downstream“ der Cyclooxygenase-2 auf die Rezeptoren EP2 und EP4 eingegrenzt werden. Der Effekt von post-ischämischem Effluat ist außerdem Proteinkinase A-abhängig. Die Ergebnisse der Experimente mit dem Proteinkinase A-Inhibitor Rp-cAMPS stehen im Einklang mit den cAMP-Analysen von Lysaten adulter Kardiomyozyten, die mit post-ischämischem Effluat vorbehandelt wurden und im Vergleich zur Kontrolle eine erhöhte cAMP-Konzentration aufwiesen. Die negativ-inotrope Wirkung von post-ischämischem Effluat zeigt eine kardioprotektive Funktion der post-ischämisch freigesetzten Substanzen an. Die Wirkung des Effluates konnte durch Anwendung des Inhibitors für sarkolemmale und mitochondriale Kalium(ATP)-Kanäle Glibenclamid und des für sarkolemmale Kalium(ATP)-Kanäle selektiven Inhibitors HMR-1098 aufgehoben werden. Außerdem wurde die Wirkung des post-ischämischen Effluates auf Kardiomyozyten in extrazellulärem Milieu mit erhöhter Calciumkonzentration untersucht. Im Unterschied zur Kontrolle verminderte das post-ischämische Effluat den intrazellulären diastolischen und systolischen Calciumanstieg. Bei den Mediatoren im post-ischämischen Effluat handelt es sich vermutlich um Substanzen aus dem Arachidonsäuere-Stoffwechsel, die in isolierten Kardiomyozyten durch die Cyclooxygenase-2 umgesetzt werden und über EP2-/EP4-Rezeptoren durch die Aktivierung von sarkolemmalen Kalium(ATP)-Kanälen negativ-inotrop und kardioprotektiv wirken.
Auf den inneren und äußeren Oberflächen des Menschen existieren zahlreiche Mikrohabitate mit limitiertem Sauerstoffangebot. Vor allem während infektiöser Vorgänge kann aufgrund einwandernder Neutrophile die Sauerstoffkonzentration im menschlichen Gewebe auf unter 1% sinken. Eine rasche Anpassung an das vorherrschende Sauerstofflevel und die Nutzung effizienter alternativer Atmungsformen oder des Gärungsstoffwechsels sind deshalb entscheidend für das mikrobielle Überleben im menschlichen Wirt. In der vorliegenden Dissertationsarbeit wurde die anaerobe Genexpression von Staphylococcus aureus sowie die zugrundeliegenden regulatorischen Mechanismen näher untersucht. Die sich in vier Teile gliedernde Arbeit befasst sich zunächst mit einer eingehenden Beschreibung der anaeroben Adaptation und Physiologie von S. aureus auf Ebene des Transkriptoms, der Proteinsynthese und des extrazellulären Metaboloms. Die Identifikation eines konservierten Sequenzmotivs (inverted repeat) vor zahlreichen anaerob induzierten Genen war Ausgangspunkt für die Untersuchung der entsprechenden regulatorischen Vorgänge im zweiten Teil dieser Arbeit. Diese führten letztlich in Kooperation mit Arbeitsgruppen aus den USA, Schweden und Deutschland (AG R. Proctor, Universität Wisconsin; AG C. von Wachenfeldt, Universität Lund; AG C. von Eiff, Universität Münster; AG M. Lalk, Universität Greifswald) zu der Identifikation des Rex Proteins (SACOL2035) als zentraler Regulator der anaeroben Genexpression in S. aureus. Neben der Rex-abhängigen Expressionskontrolle wurde in Kooperation mit der Arbeitsgruppe von Friedrich Götz (Universität Tübingen) auch der Einfluss des Zwei-Komponenten¬systems NreBC auf die Genexpression in S. aureus näher untersucht. Auf Ebene des Transkriptoms, Proteoms und Metaboloms konnte so die essentielle Bedeutung des NreBC-Systems für die Expression der dissimilatorischen Nitrat- und Nitritreduktasen in S. aureus nachgewiesen werden. Der dritte Teil dieser Arbeit beschäftigt sich mit der Einordnung des anaeroben Proteinsynthese¬musters (Proteomsignatur) in den Kontext zahlreicher anderer stressinduzierter Proteomsignaturen von S. aureus. Die aus diesem komplexen Vergleich gewonnenen Ergebnisse geben detaillierte Einblicke in die Spezifitäten und Gemeinsam¬keiten der Proteinsynthese von S. aureus als Reaktion auf oxidativen Stress (H2O2, Diamid und Paraquat), nitrosativen Stress (NO), Sauerstofflimitation in An- und Abwesenheit von Nitrat, Hitzestress (48°C) sowie subinhibitorische Antibiotikakonzentrationen (Puromycin, Mupirocin). Für die Bereitstellung der entsprechenden Daten wurde im Rahmen dieser Arbeit zudem ein mySQL-basiertes System entwickelt, das die Visualisierung der Daten mit komplexen Abfrage- und Filtermöglichkeiten verknüpft (http://www.aureolib.de). Im letzten Teil gibt diese Arbeit schließlich einen Überblick über die Leistungen und Möglichkeiten der Proteomanalyse hinsichtlich physiologischer und infektionsrelevanter Fragestellungen. Besondere Beachtung findet hier die Aufklärung und Struktur des bereits erwähnten Rex Modulons.
In a changing world, phytoplankton communities face a large variety of challenges including altered light regimes. These alterations are caused by more pronounced stratification due to rising temperatures, enhanced eutrophication, and browning of lakes. Community responses toward these effects can emerge as alterations in physiology, biomass, biochemical composition, or diversity. In this study, we addressed the combined effects of changes in light and nutrient conditions on community responses. In particular, we investigated how light intensity and variability under two nutrient conditions influence (1) fast responses such as adjustments in photosynthesis, (2) intermediate responses such as pigment adaptation and (3) slow responses such as changes in community biomass and species composition. Therefore, we exposed communities consisting of five phytoplankton species belonging to different taxonomic groups to two constant and two variable light intensity treatments combined with two levels of phosphorus supply. The tested phytoplankton communities exhibited increased fast reactions of photosynthetic processes to light variability and light intensity. The adjustment of their light harvesting mechanisms via community pigment composition was not affected by light intensity, variability, or nutrient supply. However, pigment specific effects of light intensity, light variability, and nutrient supply on the proportion of the respective pigments were detected. Biomass was positively affected by higher light intensity and nutrient concentrations while the direction of the effect of variability was modulated by light intensity. Light variability had a negative impact on biomass at low, but a positive impact at high light intensity. The effects on community composition were species specific. Generally, the proportion of green algae was higher under high light intensity, whereas the cyanobacterium performed better under low light conditions. In addition to that, the diatom and the cryptophyte performed better with high nutrient supply while the green algae as well as the cyanobacterium performed better at low nutrient conditions. This shows that light intensity, light variability, and nutrient supply interactively affect communities. Furthermore, the responses are highly species and pigment specific, thus to clarify the effects of climate change a deeper understanding of the effects of light variability and species interactions within communities is important.
Die vorliegende Arbeit beschäftigt sich mit der genetischen Grundlage der essentiellen Hypertonie. Viele Experimente konnten Genorte finden, die an der Blutdruckregulation beteiligt sind und bei Mutation zu Anstiegen des Blutdruckes führen. Bisher wurden jedoch keine Gene gefunden, die eine essentielle Hypertonie verursachen. Mittels der phänotypischen Selektion auf Bluthochdruck wurden Rückkreuzungshybriden aus normotensiven BB/OK Ratten und spontanhypertensiven SHR/Mol Ratten gezüchtet und im Anschluss mit Mikrosatellitenmarkern auf Veränderungen im Genom untersucht. Der Tierexperimentelle Teil dieser Arbeit und eine erste Untersuchung des Genoms mit 259 Mikrosatellitenmarkern wurde bereits 1997 durchgeführt [Klöting I. et al. 1997]. Die genaue Beschreibung des Rattengenoms und daraus resultierend die Vervielfachung der Mikrosatellitenmarker zur Analyse des Genoms machten eine Weiterführung der Arbeit sinnvoll. Im Rahmen dieser Arbeit wurde das Genom der Rückkreuzungshybriden mithilfe von weiteren 509 Mikrosatellitenmarkern auf Unterschiede im Vergleich zu normotensiven BB/OK Ratten untersucht. Wie bereits 1998 konnten auf Chromosom 14 Genorte ausgemacht werden, die nicht ausschließlich BB/OK zuzuordnen waren, sondern stattdessen Eigenschaften der spontanhypertensiven SHR-Mol Rattenlinie zeigten. Mithilfe der DNA-Sequenzierung konnten auf Chromosom 3 Mutationen entdeckt werden, die im kodierenden Bereich für das Gen Rnd3 liegen. Dieses Gen spielt eine Rolle bei der Hemmung der Ausbildung von Stressfasern. Diese Proteine verankern Zellen über Fokalkontakte an der Plasmamembran und sind unter anderem in den großen Gefäßen und dem Myokard zu finden. Hieraus lässt sich eine Relevanz für die Ausprägung von Hypertonie ableiten. Weitere Veränderungen im Genom der Rückkreuzungstiere konnten nicht gefunden werden. Hieraus kann man schlussfolgern, dass bei guter Abdeckung des Genoms der Ratten nur wenige bestimmende Gene eine Rolle bei der Ausprägung der Hypertonie spielen. Die Annahme, dass die essentielle Hypertonie über eine Vielzahl von Genen vererbt wird, erscheint anhand der gefundenen Ergebnisse unwahrscheinlich und bestätigt Ergebnisse der Entwicklung einer hypertonen Wistarratte von Okamato et al. 1966 [Udenfriend S. Et al. 1976]. Ob es sich bei den Veränderungen, die entdeckt wurden, um hauptursächliche Gene der essentiellen Hypertonie handelt, kann mit dieser Arbeit nicht geklärt werden. In der vorliegenden Arbeit wurden Typ 2 Diabetiker hinsichtlich ihrer Zugehörigkeit zum LADA analysiert, außerdem wurde nach genetischen Zusammenhängen von SNPs auf und zwischen den Genen REPIN1 und RARRES2 in Bezug auf BMI und WHR als Facetten des Metabolischen Syndroms untersucht. Dazu wurden 98 Typ 2 Diabetiker einer allgemeinmedizinischen Praxis in Lühmannsdorf, Ostvorpommern sowie eine Kontrollgruppe von 104 normgewichtigen nichtdiabetischen Probanden (BMI<25) geschlechtsspezifisch erfasst, ausgewertet und statistisch verglichen. Für die genetischen Untersuchungen wurde als Vergleich noch eine Gruppe mit ähnlichem Charakter aus Leipzig hinzugezogen. Bei 3,4 Prozent der insgesamt 98 untersuchten Typ 2 Diabetiker aus OVP wurden GAD-Antikörper als Hinweis für eine Zuordnung zum LADA nachgewiesen. Das waren weniger Typ 2 Diabetiker als nach publizierten Studien erwartet (5-10 Prozent). Übereinstimmend zeigte sich, dass die Diabetiker beider Populationen mit einem BMI >30 und einer WHR >1 übergewichtig bzw. adipös waren. Signifikante Unterschiede wurden jeweils bei einem der fünf untersuchten SNPs in der Genotypenverteilung zwischen diabetischen und nichtdiabetischen Probanden beider Populationen nachgewiesen. Signifikante Unterschiede zeigten sich auch zwischen den Genotypen und dem BMI bei nichtdiabetischen Männern und Frauen aus Leipzig sowie der WHR bei diabetischen Frauen aus Leipzig. Keine signifikanten Unterschiede fanden sich bei den Probanden aus OVP.
Die Sepsis stellt ein hoch komplexes, klinisch heterogenes Krankheitsbild dar, das die Medizin und Wissenschaft auf der ganzen Welt vor große Herausforderungen stellt. Obwohl nahezu jeder erfahrene Intensivmediziner Unterschiede zwischen einem Patienten mit abdomineller Sepsis und einem Patienten mit Urosepsis benennen könnte, haben nur sehr wenige Forschungsgruppen diese Unterschiede
weiter untersucht und beschrieben. Jede Sepsis beginnt in einem anatomischen Fokus der Infektion, der sich fast überall im menschlichen Körper befinden kann. Existieren Unterschiede zwischen Patienten mit unterschiedlichem Sepsisfokus? Ist Sepsis vielleicht nicht gleich Sepsis? Um diese Fragen zu beantworten, hat unsere Arbeitsgruppe über einen Zeitraum von acht Jahren die Daten von 816 Sepsispatienten ausgewertet, deren Fokus ausschließlich im Abdomen, Respirationstrakt, Urogenitaltrakt oder den Knochen und Weichteilen lag. Die Patienten wurden über den gesamten Studienzeitraum konsekutiv eingeschlossen und prospektiv auf das Vorliegen einer Sepsis gescreent. Wir verglichen klinische, mikrobiologische und laborchemische Daten zum Sepsiszeitpunkt zwischen den Fokusgruppen und konnten signifikante Unterschiede in allen drei Bereichen beobachten. Beispielsweise war die Inzidenz für das Auftreten eines septischen Schocks bei Patienten mit abdominellem und urogenitalem Fokus signifikant höher als bei den anderen Gruppen. In Bezug auf die mikrobiologischen Unterschiede waren die Urosepsis und die Knochen- und Weichteilsepsis mit deutlich höheren Blutkulturpositivitäten assoziiert. Außerdem unterschied sich das Erregerspektrum zwischen den Gruppen. Des Weiteren konnten wir Unterschiede in den Laborergebnissen der Patienten zum Sepsiszeitpunkt feststellen. Fast die Hälfte der Patienten mit Urosepsis zeigte pathologisch erhöhte PCT-Werte im Vergleich zu nur 15 % der respiratorischen Gruppe. Weiterhin hatten dreimal so viele Patienten mit Fokus im Bereich Knochen/Weichteile eine Hypoalbuminämie wie Patienten der urogenitalen und pulmonalen Gruppe. Darüber hinaus waren die verschiedenen Sepsisfokusse mit Unterschieden in den SOFA-Scores, SIRS-Kriterien und dem Auftreten von Organdysfunktionen zum Sepsisbeginn assoziiert. Die vorliegende Dissertation liefert eindeutige Hinweise auf klinische, laborchemische und mikrobiologische Unterschiede zwischen Sepsispatienten mit verschiedenen Sepsisfokussen zum Beginn der Sepsis. In großen randomisierten Studien sollte den deutlichen Unterschieden zwischen den verschiedenen Sepsisfokussen möglicherweise mehr Aufmerksamkeit geschenkt werden, damit therapeutische Ansätze, welche nur einer Subgruppe helfen, mit ausreichender Fallzahl herausgearbeitet werden können.
Particle-in-Cell (PIC) simulations are used to model the MS4 test thruster of Thales Deutschland. Given as input the geometric shape, material components, magnetic field and the operating parameters of the experiment, the model is able to reproduce the experimentally observed emission pattern in the plume. This is determined by the magnetic field line structure and the resulting plasma dynamics in the near-field region close to the exit.
Durch die Entstehung neuer Pilgerwege erhoffen sich die Initiatoren eine touristische Erschließung peripherer Räume. Ähnlich beim Projekt des Pommerschen Jakobsweges, welches das Küstenhinterland des südlichen Ostseeraumes im Zuge der Revitalisierung einer kulturhistorischen Pilgerroute touristisch aufwerten soll. Ziel dieser tourismusgeographischen Studie ist es, das Potential des Pommerschen Jakobsweges im Hinblick auf die Nachfrageseite zu untersuchen und Handlungsmöglichkeiten aufzuzeigen. Die Arbeit dient zudem als Konzeption und Leitlinie für zukünftige Projekte und setzt sich kritisch mit gegenwärtigen Forschungsergebnissen auseinander. Der Fokus der Untersuchung wird dabei auf die reisespezifischen Aspekte des Pilgerns sowie die Funktion des Pilgerweges als touristisches Produkt gelegt. Als Ergebnis wurden fünf Reiseformen herausgearbeitet, die inmitten des alternativen Pilgertourismus eine besondere Rolle übernehmen und die nachgewiesene, starke Nachfrage an peripheren Pilgerwegen mitgestalten.
PIM1 Inhibition Affects Glioblastoma Stem Cell Behavior and Kills Glioblastoma Stem-like Cells

Despite comprehensive therapy and extensive research, glioblastoma (GBM) still represents the most aggressive brain tumor in adults. Glioma stem cells (GSCs) are thought to play a major role in tumor progression and resistance of GBM cells to radiochemotherapy. The PIM1 kinase has become a focus in cancer research. We have previously demonstrated that PIM1 is involved in survival of GBM cells and in GBM growth in a mouse model. However, little is known about the importance of PIM1 in cancer stem cells. Here, we report on the role of PIM1 in GBM stem cell behavior and killing. PIM1 inhibition negatively regulates the protein expression of the stem cell markers CD133 and Nestin in GBM cells (LN-18, U-87 MG). In contrast, CD44 and the astrocytic differentiation marker GFAP were up-regulated. Furthermore, PIM1 expression was increased in neurospheres as a model of GBM stem-like cells. Treatment of neurospheres with PIM1 inhibitors (TCS PIM1-1, Quercetagetin, and LY294002) diminished the cell viability associated with reduced DNA synthesis rate, increased caspase 3 activity, decreased PCNA protein expression, and reduced neurosphere formation. Our results indicate that PIM1 affects the glioblastoma stem cell behavior, and its inhibition kills glioblastoma stem-like cells, pointing to PIM1 targeting as a potential anti-glioblastoma therapy.
Indications for surgery of pineal cysts without ventriculomegaly are still under debate. In view of the limited data for pineal cyst resection in the absence of hydrocephalus, and the potential risk of this approach, we have analyzed our patient cohort focusing on strategies to avoid complications according to our experience in a series of 73 pineal cyst patients. From 2003 to 2015, we reviewed our database retrospectively for all patients operated on a pineal cyst. Furthermore, we prospectively collected patients from 2016 to 2020. In summary, 73 patients with a pineal cyst were treated surgically between 2003 and 2020. All patients were operated on via a microscopic supracerebellar-infratentorial (SCIT) approach. The mean follow-up period was 26.6 months (range: 6–139 months). Seventy-three patients underwent surgery for a pineal cyst. An absence of enlarged ventricles was documented in 62 patients (51 female, 11 male, mean age 28.1 (range 4–59) years). Main presenting symptoms included headache, visual disturbances, dizziness/vertigo, nausea/emesis, and sleep disturbances. Complete cyst resection was achieved in 59/62 patients. Fifty-five of 62 (89%) patients improved after surgery with good or even excellent results according to the Chicago Chiari Outcome Scale, with complete or partial resolution of the leading symptoms. Pineal cysts resection might be an indication in certain patients for surgery even in the absence of ventriculomegaly. The high percentage of postoperative resolution of quality-of-life impairing symptoms in our series seems to justify surgery. Preoperatively, other causes of the leading symptoms have to be excluded.
Hepatocellular carcinoma (HCC) is a deadly form of liver malignancy with limited treatment
options. Amplification and/or overexpression of c-MYC is one of the most frequent genetic events
in human HCC. The mammalian target of Rapamycin Complex 1 (mTORC1) is a major functional
axis regulating various aspects of cellular growth and metabolism. Recently, we demonstrated that
mTORC1 is necessary for c-Myc driven hepatocarcinogenesis as well as for HCC cell growth in vitro.
Among the pivotal downstream effectors of mTORC1, upregulation of Fatty Acid Synthase (FASN) and
its mediated de novo lipogenesis is a hallmark of human HCC. Here, we investigated the importance
of FASN on c-Myc-dependent hepatocarcinogenesis using in vitro and in vivo approaches. In mouse
and human HCC cells, we found that FASN suppression by either gene silencing or soluble inhibitors
more effectively suppressed proliferation and induced apoptosis in the presence of high c-MYC
expression. In c-Myc/Myeloid cell leukemia 1 (MCL1) mouse liver tumor lesions, FASN expression
was markedly upregulated. Most importantly, genetic ablation of Fasn profoundly delayed (without
abolishing) c-Myc/MCL1 induced HCC formation. Liver tumors developing in c-Myc/MCL1 mice
depleted of Fasn showed a reduction in proliferation and an increase in apoptosis when compared
with corresponding lesions from c-Myc/MCL1 mice with an intact Fasn gene. In human HCC samples,
a significant correlation between the levels of c-MYC transcriptional activity and the expression
of FASN mRNA was detected. Altogether, our study indicates that FASN is an important effector
downstream of mTORC1 in c-MYC induced HCC. Targeting FASN may be helpful for the treatment
of human HCC, at least in the tumor subset displaying c-MYC amplification or activation.
Planning Modes for Major Transportation Infrastructure Projects (MTIPs): Comparing China and Germany

Seventeen bacterial strains able to suppress plant pathogens have been isolated from healthy Vietnamese crop plants and taxonomically assigned as members of the Bacillus cereus group. In order to prove their potential as biocontrol agents, we perform a comprehensive analysis that included the whole-genome sequencing of selected strains and the mining for genes and gene clusters involved in the synthesis of endo- and exotoxins and secondary metabolites, such as antimicrobial peptides (AMPs). Kurstakin, thumolycin, and other AMPs were detected and characterized by different mass spectrometric methods, such as MALDI-TOF-MS and LIFT-MALDI-TOF/TOF fragment analysis. Based on their whole-genome sequences, the plant-associated isolates were assigned to the following species and subspecies: B. cereus subsp. cereus (6), B. cereus subsp. bombysepticus (5), Bacillus tropicus (2), and Bacillus pacificus. These three isolates represent novel genomospecies. Genes encoding entomopathogenic crystal and vegetative proteins were detected in B. cereus subsp. bombysepticus TK1. The in vitro assays revealed that many plant-associated isolates enhanced plant growth and suppressed plant pathogens. Our findings indicate that the plant-associated representatives of the B. cereus group are a rich source of putative antimicrobial compounds with potential in sustainable agriculture. However, the presence of virulence genes might restrict their application as biologicals in agriculture.
Recent climate change and its consequences for living organisms constitute one of the greatest problems of our century. Global warming entails an increase in mean temperature and the frequencies of extreme weather events. Those changes in environmental conditions affect both plants and animals. Because of their inability to escape from unsuitable environments, plants have evolved a wide spectrum of molecular programs to protect themselves against changing conditions. Responding on altered environmental conditions will change plants chemical composition and therefore also affect plants interaction with other species (e.g., predator-prey or symbiotic relationships). For instance, changes in the chemical composition of plants may influence the survival of associated herbivores. In other words, these herbivores will be affected indirectly by climate change due to changes in the suitability / quality of their food. The aim of this doctoral thesis was to discover the effects of climate change within the relationship of the butterfly Pieris napi and its host plant (Sinapis alba used here as host plant), including individual conditions (e.g. chemical compositions of plants; morphology, physiology of the butterfly) and behavior of female butterflies and larvae. In the first experiment, the influence of simulated climate change on the chemical composition of the plant Sinapis alba was investigated. The second experiment aimed to examine the influence of changes in plant composition on the butterfly P. napi. Glucosinolates (secondary compound of plants) are known to have an important effect on the preference and performance of herbivores. Therefore, in the third experiment, the impact of glucosinolates on the preference and performance of P. napi was investigated in order to see if these plant compounds had the most important influence on this butterfly. Furthermore, in the fourth experiment, it was explored whether there is a latitudinal gradient within the species´ responses to changes in its host plant. The fifth and last experiment aimed to examine, if there are general principles across species regarding indirect effects of climate change.
Climate change, simulated by different combinations of temperature and water regimes, had an effect on the plant chemistry. The combination of temperature and water availability changed plant composition substantially. Especially the amount of carbon and glucosinolates (here above all sinalbin) in S. alba plants varies between the different treatments and therefore between the different combinations of temperature and water regimes. Regarding glucosinolates, elevated temperatures increased their concentration in leaves, whereas water deficit in combination with higher temperature reversed this pattern. For carbon content, all plants, except those of the control group, showed a decreased amount of total carbon. However, simulated heat waves had no effect on plants, leading to the assumption that the plants were able to recover from heat stress sufficiently during the control phases. Changes in plant composition affected both larvae and females of the butterfly P. napi. Therefore, changed host-plant chemistry alters the plant quality for this herbivore, meaning that plants of different treatments represent different plant qualities defined by their composition. Females of P. napi may be able to differentiate between plant qualities and even show a direct preference. Therefore, glucosinolates seem to act as oviposition stimulants. However, preferring another plant quality with lower amount of glucosinolates suggest that females of this butterfly species were attracted by more than high levels of glucosinolates alone. Larvae fed with different plant qualities performed differently, indicated by smaller wings (lighter bodies) and prolonged development when fed with plants contained higher amount of the glucosinolate sinalbin. It can be assumed that a higher amount of sinalbin decreases the quality of the host plant and therefore lead to these responses. Probably larvae need to shift their resources from growth to detoxification and therewith survival. Furthermore, drought conditions during plant growth seem to reduce the overall negative effects of higher temperatures, lead to an increase of host plant quality. Larvae seem to benefit from feeding on these “double-stressed” plants. Comparison between the results of the preference and performance tests suggests that there might be a mismatch between female preference and larval performance. It seems that the stimulating effect of high concentration of glucosinolates, in this case sinalbin, misdirects females´ decision to less suitable host plants, meaning that the advantage of less competition for larvae come at costs through detoxification. Using Brassica napus plants with genetically fixed glucosinolate levels, it could be demonstrate that there must be other plant components influencing females´ oviposition behavior been seen in the choice experiment with S. alba. The comparison of German and Italian populations to changes in host-plant quality showed fewer differences between countries as expected. However, German and Italian individuals differed in their reaction to altered plant quality, at least in developmental time and larval growth rate. It seems that Italian larvae benefitted from plants grown under higher temperatures, whereas drought-stressed plants affected them negatively. German individuals in contrast seem to benefit only from water stress during plant growth. With regard to the sexes of P. napi, it seems that females respond differently than males to changes in plant quality. Furthermore, the results of the performance test on Bicyclus anynana showed that there might be some general principles for the respond of butterflies to changes of its host plant. B. anynana responded in a similar way to different host plant qualities as P. napi did, meaning that plants grown under higher temperatures and drought conditions seem to be beneficial for the larval performance.
In summary, these findings may have important implications for the indirect effects of climate change on this butterfly in natural environments. First, climate change seems to have an impact on the chemical composition of plants. Second, changes in plants caused by increasing temperature and droughts seem to influence the preference and performance of this butterfly. However, there are differences between populations, which seem to be induced by former adaptation. And third, there might be some general principles for the respond of butterflies to changes in their host plants. This thesis focuses only on possible indirect effects of climate change. However, there are direct effects, which may alter the responses of herbivores to changes in their host plant as well. Therefore, further investigations in this linkage and in other plant-herbivore relationships will be necessary to explore how climate change may alter the relationship between herbivores and their hosts.
Abstract
Relatively little is known about how plant–soil feedbacks (PSFs) may affect plant growth in field conditions where factors such as herbivory may be important. Using a potted experiment in a grassland, we measured PSFs with and without aboveground insect herbivory for 20 plant species. We then compared PSF values to plant landscape abundance. Aboveground herbivory had a large negative effect on PSF values. For 15 of 20 species, PSFs were more negative with herbivory than without. This occurred because plant biomass on “home” soils was smaller with herbivory than without. PSF values with herbivory were correlated with plant landscape abundance, whereas PSF values without herbivory were not. Shoot nitrogen concentrations suggested that plants create soils that increase nitrogen uptake, but that greater shoot nitrogen values increase herbivory and that the net effect of positive PSF and greater aboveground herbivory is less aboveground biomass. Results provided clear evidence that PSFs alone have limited power in explaining species abundances and that herbivory has stronger effects on plant biomass and growth on the landscape. Our results provide a potential explanation for observed differences between greenhouse and field PSF experiments and suggest that PSF experiments need to consider important biotic interactions, like aboveground herbivory, particularly when the goal of PSF research is to understand plant growth in field conditions.
The aim of this thesis is to concentrate on the investigation of these ROS&RNS composition distribution and their production pathways in the gas phase produced by a plasma jet. By understanding the physical mechanisms behind the generation of the ROS&RNS a precise tuning and design of the composition distribution in the gas phase can be achieved. One crucial physical parameter is the dissipated power inside the plasma. Only if this parameter is known a meaningful comparison of different feed gas settings is possible. Therefore, a concept for measuring the dissipated power inside the plasma for the modified micro-scaled atmospheric pressure plasma jet( µAPPJ) is designed. Additionally, due to achievements within this thesis it is now possible to ignite a homogeneous discharge in argon and helium within the geometry of the µAPPJ. The used feed gas is a determining factor concerning the electron energy distribution function and consequently influencing the production mechanism of the ROS&RNS. First of all, the electrical characterisation of the modified µAPPJ was performed including the alpha-to-gamma transition. It is shown that the alpha-to-gamma transition power is increasing with increasing frequency. For the first time it is now feasible to investigate the influence of the dissipated power on the neutral gas temperature, the metastable atom densities and the ROS&RNS production for the modified µAPPJ with argon and helium as feed gas. Due to the possibility of changing the feed gas and controlling the dissipated power a fundamental insight into the production mechanism of the ROS&RNS generated by the plasma jet is achieved. With rising dissipated power the temperature and the metastable densities as well as the ozone and nitrogen dioxide concentrations are increasing. By adding molecular oxygen and nitrogen to the feed gas of a plasma jet the ROS&RNS composition can be tuned. However, also the dissipated power is changed by the small amount of admixtures. Due to the developed dissipated power measurements within this thesis it was possible to disentangle the influence of the admixture on the power and on the ROS&RNS production. If the dissipated power is fixed for the µAPPJ with argon and helium feed gas, respectively, the highest amount of ozone was measured with oxygen admixture in an argon discharge, the highest amount of dinitrogen pentoxide with nitrogen admixture in an argon discharge and the highest amount of nitrogen dioxide with nitrogen admixture in a helium discharge. Beyond the influence of the dissipated power and the molecular admixture on the ROS&RNS production the feed gas temperature is a crucial parameter for the corresponding chemical reactions. By changing this parameter the distribution of ozone and nitrogen dioxide can be tuned precisely in such a way that with increasing temperature the ozone density goes down and the nitrogen dioxide density rises. Another determinant for the ROS&RNS composition produced by an atmospheric pressure plasma jet is the influence of ambient air. If the ambient air is changing from pure nitrogen to pure oxygen atmosphere the ozone density produced by the plasma jet is increasing. For the same conditions the nitrogen dioxide has a maximum at an oxygen-to-nitrogen ratio of 1:1. To avoid the influence of the ambient air on the reactive species production the afterglow of the µAPPJ was prolonged with a glass tube. By increasing the amount of molecular admixtures to the feed gas with each in equal quantities a totally different ROS&RNS composition can be obtained compared without the glass tube. It figures out that for small molecular admixtures the reactive species composition is nitrogen dominated and for higher admixtures it is oxygen dominated. Consequently, by shielding the ambient air from the active effluent and by admixing molecular oxygen and nitrogen the ROS&RNS composition can be designed.
Plasma medical oncology: Immunological interpretation in head and neck squamous cell carcinoma

Over the past several years, various important articles focusing on cancer therapy
approaches in head and neck squamous cell carcinomas (HNSCCs) using cold
atmospheric plasma (CAP) have been published (SEMMLER et al. 2020 [53],
METELMANN et al. 2018 [44], KEIDAR et al. 2011 [33]). This doctoral thesis presents
selected results from a prospective observational clinical study in CAP therapy of
palliative HNSCC patients, carried out at the Department of Oral and Maxillofacial
Surgery/Plastic Surgery of the Greifswald University Medicine. For oral and
maxillofacial surgeons, ulcerated surfaces of locally advanced head and neck squamous
cell carcinomas (UICC IV) offer a challenging treatment assignment with microbial
contamination and tumour progression. The clinical attempt appears to eradicate
microbial contamination and to initiate tumour regression. This doctoral thesis will
describe the processes of human tumour biology and tumour immunology in HNSCCs
and the extent of present knowledge concerning plasma medical oncology as an
anticancer modality. In the introduction of the doctoral thesis clinical results of plasma
therapy in locally advanced HNSCCs (UICC IV) are set out. This mainly includes the
investigation of a therapeutic concept, the treatment phases, the tumour size development
and the morphological changes of the infected tumour surface following cold atmospheric
plasma therapy. In the main part, a detailed immunological interpretation is proposed on
the basis of present preclinical and clinical immunological knowledge. Finally,
unexplored questions in plasma medical oncology are highlighted. This is highly
significant for future plasma research and clinical anticancer therapy.
Plasma Metabolome Profiling Identifies Metabolic Subtypes of Pancreatic Ductal Adenocarcinoma

The development of innovative coatings with multifunctional properties is an ambitious task in modification of material surfaces. A novel approach is a hybrid method combining the non-thermal plasma processing with nanotechnology for the development of multifunctional surface coatings. The conception of the hybrid coating process is based on three steps: the preparation of a suspension consisting of an organic liquid and functional nanoparticles, the deposition of the suspension as a thin liquid film on the material surface, and the plasma modification of the liquid organic film to achieve a thin solid composite film with embedded nanoparticles demonstrating multifunctional properties and good adherence on the substrate material. In this work the liquid polydimethylsiloxane (PDMS) was applied as a model system, and the experimental investigations were focused on the PDMS plasma modification. In particular, the specific role of the different plasma components and the influence of the plasma and processing parameters on the PDMS modification were studied. The applied capacitively coupled radio frequency (CCRF) plasma was analyzed by electric probe measurements and optical emission spectroscopy, whereas the molecular changes in PDMS due to plasma-induced chemical reactions were studied by the Fourier transform infrared reflection absorption spectroscopy. Additionally, the photocatalytic activity of thin composite films consisting of plasma cross-linked PDMS with embedded TiO2 nanoparticles was demonstrated. During the investigation it was found that the CCRF discharge modifies efficiently thin liquid PDMS films to solid coatings. The samples were positioned in the plasma bulk at floating potential. The penetration depth of particles like neutrals, ions, electrons and radicals in the film is strongly limited. The heating of samples in the CCRF discharge is weak to modify PDMS by itself and only the plasma radiation is able to transform the liquid bulk to solid one. It is known that the absorption onset of PDMS lies in the VUV region (below 200 nm). The energetic VUV radiation penetrates into the PDMS film on a thickness from several hundred nanometers to few micrometers and initiates photochemical reactions there. Thus, different gases like Ar, Xe, O2, H2O, air and H2 were tested to provide the strongest VUV emission intensity of the CCRF discharge. Discharge pressure and power were varied for all these gases and it was found that at all conditions the H2 plasma demonstrates drastically stronger emission. Thus, H2 gas was selected for the plasma treatment of liquid PDMS films. The IRRAS analysis revealed the transformation process of PDMS with the degradation of CH3 groups, the formation of new groups like SiOH, CH2 and SiH, the formation of the SiOx material and crosslinking. It was found that the modification effect is not uniform across the film thickness. The top region with an initial thickness up to 100 nm loses all CH3 groups, in the underlying region the CH3 concentration increases gradually from zero to the value for PDMS, if the film was thick enough. The methyl-free SiOx top layer contains also SiOH and SiH groups. Furthermore, the SiH groups are concentrated only in a very thin layer with a thickness below 10 nm. The presence of the unscreened polar SiOSi and SiOH groups on the surface causes the adsorption of H2O from the atmosphere, which was also observed by IRRAS. By means of the spectroscopic ellipsometry it was found out that all above described regions experience a shrinking. The reason is the crosslinking and loss of material. The most shrunken layer is the top SiOx layer with the shrinking ratio (final thickness/initial thickness) of 0.55 - 0.60. Further, this ratio gradually rise up to the value of 0.95 in the deeper region, which has the concentration of CH3 groups of about that for PDMS. After the analysis of all results the depth of effective modification was estimated at 300 400 nm for the most optimal conditions. The optimization of the plasma VUV intensity was realized by variation of discharge pressure and power. The strongest plasma emission at studied conditions provided the irradiance of the sample of ca. 13 mW/cm2. However, such strong radiation causes very strong production rate of the gases. These products leave the modifying film slower as they are produced, what causes their accumulation in there. Their pressure grows up leading to formation of bubbles, which later explode. Finally, the film becomes heavily damaged. To avoid this effect the pressure and the RF power were changed to reduce the irradiance to 6 - 7 mW/cm2. This resulted in the absence of any damages.
The thyroid as the largest endocrine gland mainly produces and secretes the thyroid hormones (TH): 3,3’,5-triiodo-L-thyronine (T3) and its pro-hormone L-thyroxine (T4). Besides the impact on growth, normal development, bone marrow structure, the cardiovascular system, body weight and thermogenesis, TH play a vivid role in many metabolic regulatory mechanisms in almost all tissues. Thyroid diseases are relatively prevalent and cause, due to the resulting TH imbalances, a broad spectrum of effects. Many of them manifest in pathologically increased or decreased TH levels defined as hyperthyroidism or hypothyroidism, respectively. Routinely, determination of the thyroid state is based on the assessment of the classical markers TSH and free T4. However, this practice has several drawbacks. Moreover, elucidation of the pleiotropic effects of TH on multiple molecular pathways is mostly based on cell culture, tissue and rodent models. Analysis of animal biofluids like serum and urine using metabolomics approaches demonstrated the extensive impact of TH on other body compartments. In contrast, proteome profiling has not been exploited for the comprehensive characterization of the general metabolic effects of TH. Plasma as a large and diverse compartment of the human proteome provides a great opportunity to identify novel protein markers of thyroid function as well as to characterize metabolic effects of TH in humans.
Therefore, a study of experimental thyrotoxicosis was performed with 16 male volunteers treated with 0.25 mg/d levothyroxine (L-T4) for 8 weeks to induce a hyperthyroid state. Plasma samples were collected before the L-T4 application started, two times during the treatment and additionally two times after withdrawal. Proteome analysis revealed remarkable alterations including increased levels of two known proteins known to correlate with TH levels (sex hormone-binding globulin and cystatin C). The correlation with free T4 levels revealed 76 out of 437 detected proteins with a Pearson correlation coefficient of r ≥ |0.9|. One prominent signature included 10 coagulation cascade proteins exhibiting significantly increased plasma levels during thyrotoxicosis, thereby revealing a trend towards a hypercoagulative state in hyperthyroidism. To overcome the statistical drawbacks of the Pearson correlation analysis, additionally a mixed-effect linear regression model using serum free T4 concentrations as exposure and protein abundances as outcome while controlling for age, BMI, and batch was implemented. Application of this model resulted in the detection of 63 proteins with significant associations to free T4 levels. Besides the already mentioned augmented coagulation, a significant drop in the amounts of three apolipoproteins (ApoD, ApoB-100 and ApoC3) was observed. Furthermore, an increased abundance of proteins assigned to the complement system was detected.
Experimental studies in humans were complemented by corresponding analyses in murine models. In the current work, plasma samples of two murine studies including male C57BL/6 wildtype mice were analyzed to elucidate the impact of thyroid dysfunction on the plasma proteome. The first study was similarly designed as the human model of experimentally induced thyrotoxicosis and assigned the animals to three groups: a control group, a T4 treatment group, and a T4 recovery group, whereupon the latter first received T4 followed by a subsequent TH normalization period. A high proportion of plasma proteins exhibited significantly different protein levels during T4 application (n = 120), where 90 of these also showed a corresponding reverse trend after T4 withdrawal (T4 recovery vs. T4), thereby displaying transient alterations. The molecular pattern of hyperthyroidism in the murine model indicated, as in the human study, a pronounced decrease in apolipoproteins. However, in clear contrast to the human data, the levels of proteins related to the coagulation cascade and complement system were also transiently decreased in mice, while being increased in humans.
The second murine analysis focused on the impact of hyper- and hypothyroidism caused by T3 or T4 treatment and MMI/KClO4 application, respectively. In general, compared to the first murine study less clear alterations of protein levels were detected. Proteins related to the complement system revealed fewer changes in the T3 group and only marginal changes after T4 induction. Unexpectedly, the MMI/KClO4-induced hypothyroidism caused a reduction of the levels of several proteins assigned to the complement system, although different components and factors were affected.
Generally, rodent studies partially provided a divergent picture of TH action as compared to human studies. However, in spite of inconsistent results in studies regarding the effects of TH that are possibly due to species-specific differences, an important role of TH on several metabolic and other pathways, e.g. in the process of blood coagulation and apolipoprotein regulation, is evident. The results from both murine and human studies presented here provide novel insights into changes in the plasma proteome in the context of thyroid diseases which might contribute to a better understanding of TH action on metabolism and other pathways.
Thermally treated kaolinite is used to develop a range of alumino‐silicate‐based precursor materials but its behavior during plasma spraying has not been well‐researched. In this study, two types of kaolinite samples were investigated in the form of low defect (KGa‐1b) and high defect (KGa‐2) varieties. The extreme temperatures of the plasma stream (up to 20 000 K) induced flash melting to produce a highly porous alumino‐silicate glass without any crystallization of new Al−Si oxide minerals. The glass is comprised largely of intact or deformed spheres (average diameters 1.14–1.44 μm), which indicates rapid quenching and solidification before impact. The subspherical structures contain up to 40 % closed pore space caused by the rapid escape of water during melting. The low‐density, porous alumino‐silicate glass coatings with predicted specific surface areas (>0.95 m2/g) and hardnesses >1.8 GPa represent a potentially reactive but physically stable substrate ideal for further chemical functionalization.
Im Rahmen der vorliegenden Arbeit wurden erstmalig systematische Untersuchungen zum Thema „Plasma-Flüssigkeits-Wechselwirkungen“ dargestellt. Es gelang mit Hilfe einer geeigneten kalten Atmosphärendruckplasmaquelle in Form einer dielektrisch behinderten Oberflächenentladung (DBE) ein Plasma in verschiedenen Arbeitsgasen zu zünden und Flüssigkeiten ohne direkten Plasmakontakt zu behandeln. Um einen Einblick in die komplexen Mechanismen zu bekommen, wurde im analytischen Teil dieser Arbeit das Plasma mittels OES untersucht, die angrenzende Gas-Phase mittels FT-IR-Spektroskopie und MS, und anschließend die Flüssigkeit unter Nutzung photometrischer Methoden und pH-Wert-Messungen. Auf der Basis dieser Untersuchungen folgten theoretische Ausführungen zu möglichen Wechselwirkungen der detektierten Komponenten mit der Flüssigkeit. Theoretisch entstehen bei der Luftplasmabehandlung von Wasser, über Zwischenprodukte (ROS und RNS) wie z. B. HO•, HOO•, NO•, NO2•, schlussendlich H+, NO3-, NO2- und H2O2. Bei der Argon- und Argon-Sauerstoff-Plasmabehandlung von Wasser dürften aufgrund des Stickstoffmangels nur ROS entstehen, die in der Entstehung von H+ und H2O2 enden. Diese Hypothesen zur Bildung der Spezies NO3-, NO2- und H2O2, sowie die Ansäuerung der Flüssigkeit wurden mittels photometrischer Methoden und pH-Wert-Messungen überprüft und bestätigt. Im anschließenden biologischen Teil der vorliegenden Arbeit wurde der Einfluss der Plasmabehandlungen in den verschiedenen Arbeitsgasen auf in physiologischer NaCl-Lösung und PBS suspendierte Mikroorganismen (E. coli, S. aureus und B. atrophaeus Sporen) untersucht. In ungepuffertem Medium wurden die vegetativen Mikroorganismen innerhalb weniger Minuten Plasmabehandlung inaktiviert. In PBS hingegen wurden längere Behandlungszeiten benötigt. Das Plasma hatte auf die suspendierten Sporen wie erwartet kaum eine inaktivierende Wirkung. Die zwei vermutlichen Hauptwege laufen einerseits über reaktive Stickstoffspezies (RNS) und andererseits über reaktive Sauerstoffspezies (ROS). RNS können in Wechselwirkung mit Wasser über diverse zelltoxische Zwischenprodukte wie z. B. NO•, NO2•, N2O3, ONOOH, ONOO- zu NO3- und NO2- umgesetzt werden. ROS in Interaktion mit Wasser resultieren in Bildung von H+ und H2O2. Auch hier wird angenommen, dass eine Vielzahl an antimikrobiellen Komponenten entsteht, z. B. HO•, HOO•, O2•-. Es gibt folglich sehr viele Reaktionen und Interaktionen zwischen plasmagenerierten reaktiven Spezies und Wasser, welche in zelltoxischen Komponenten enden und die inaktivierende Wirkung des Plasmas auf suspendierte Mikroorganismen erklären. Um die Interaktionen zwischen den Phasen Plasma-Gas-Flüssigkeit besser zu verstehen und Hypothesen zu prüfen, wurden einerseits die Mikroorganismen-Suspensionen und destilliertes Wasser nur mit plasmabehandeltem Gas behandelt und andererseits die Mikroorganismen plasmabehandelter Flüssigkeit ausgesetzt. Die Untersuchungen zeigten deutlich, dass die plasmainitiierte Chemie und damit die biologischen Effekte des Plasmas auf das Gas bzw. in Flüssigkeit übertragen werden. Folglich werden biologische Effekte des Plasmas über die Gas- und Flüssigkeits-Phase vermittelt und Plasma-Flüssigkeits-Wechselwirkungen müssen immer im zusammenhängenden chemischen System Plasma-Gas-Flüssigkeit(-Zelle) betrachtet werden. Weiterhin wurden chemische und mikrobiologische Effekte durch den Einfluss von gasförmigem NO• und O3 auf Wasser bzw. suspendierte Mikroorganismen untersucht und mit der Luftplasmabehandlung verglichen. Hierbei zeigten beide Begasung deutlich geringere inaktivierende Effekte als die Luftplasmabehandlung. Die Begasung von Mikroorganismen mit NO• und O3 und die Analytik von begastem Wasser geben einen detaillierten Einblick in die Chemie und Mechanismen der RNS und ROS. Die Behandlung mit der dielektrisch behinderten Oberflächenentladung in Luft vereint diese zwei Hauptwege über RNS und ROS und resultierte somit in effektiveren antimikrobiellen Wirkungen. Durch die Experimente der vorliegenden Arbeit wurde ein geeignetes biologisches Modell gefunden und validiert, um den Einfluss von Plasma auf lebende Zellen zu untersuchen und Mechanismen von Plasmabehandlungen aufzudecken. Anhand des Modells „Mikroorganismen-Suspension“ konnte gezeigt werden, dass die Gas-Phase (Behandlungen mit DBE-Abgas) und die extrazelluläre Flüssigkeit (Behandlungen mit plasmabehandelter Flüssigkeit) eine bedeutende Rolle bei der Vermittlung der Plasmaeffekte spielen. Die Spezies aus dem Plasma gelangten über die Gas-Phase durch Diffusion/Penetration/ Interaktion in die Flüssigkeits-Phase, reagierten teilweise zu anderen reaktiven Spezies weiter und erreichten so die Zellen. Die verschiedenen analytischen Methoden und anschließende theoretische Betrachtungen der Phasen Plasma-Gas-Flüssigkeit gaben einen detaillierten Einblick in die chemischen Mechanismen von Plasma-Flüssigkeits-Wechselwirkungen und zeigten biologisch wirksame Komponenten auf.
Background: Plasma-generated compounds (PGCs) such as plasma-processed air (PPA) or plasma-treated water (PTW) offer an increasingly important alternative for the control of microorganisms in hard-to-reach areas found in several industrial applications including the food industry. To this end, we studied the antimicrobial capacity of PTW on the vitality and biofilm formation of Listeria monocytogenes, a common foodborne pathogen.
Results: Using a microwave plasma (MidiPLexc), 10 ml of deionized water was treated for 100, 300, and 900 s (pre-treatment time), after which the bacterial biofilm was exposed to the PTW for 1, 3, and 5 min (post-treatment time) for each pre-treatment time, separately. Colony-forming units (CFU) were significantly reduced by 4.7 log10 ± 0.29 log10, as well as the metabolic activity decreased by 47.9 ± 9.47% and the cell vitality by 69.5 ± 2.1%, compared to the control biofilms. LIVE/DEAD staining and fluorescence microscopy showed a positive correlation between treatment and incubation times, as well as reduction in vitality. Atomic force microscopy (AFM) indicated changes in the structure quality of the bacterial biofilm.
Conclusion: These results indicate a promising antimicrobial impact of plasma-treated water on Listeria monocytogenes, which may lead to more targeted applications of plasma decontamination in the food industry in the future.
Diese Dissertation beschäftigt sich mit der Erzeugung von edelmetallfreien Katalysatoren für die Sauerstoffreduktion in Brennstoffzellen. Dabei wird ein neuartiger, dualer Plasmaprozess entwickelt, aufgebaut und die so-erzeugten Schichten mit verschiedenen elektrochemischen (CV, RDE und RRDE) und strukturanalytischen Methoden (SEM, EDX, IR, XPS, Leitfähigkeit, XRD, NEXAFS, EXAFS und TEM) untersucht. Auf diese Weise ist es erstmalig gelungen edelmetallfreie Katalysatoren mit einem Plasmaprozess herzustellen, ohne dass eine zusätzliche Pyrolyse benötigt wird. Die katalytische Aktivität der Schichten ist außerdem deutlich höher als die von rein chemisch hergestellten Metall–Polypyrrol-Schichten.
Plasmapolymerisation mit einem Atmosphärendruck-Mikroplasma-Jet zur Bildung funktioneller Schichten

In Rahmen dieser Arbeit wurde die Plasmapolymerisation von aminogruppenhaltigen und perfluorierten Kohlenwasserstoffen mit einem Atmosphärendruck Mikroplasma Jet untersucht, mit dem Ziel einer erstmaligen erfolgreichen Abscheidung von Teflon-artigen und aminogruppenhaltigen Schichten. Hierzu wurde ein Versuchsaufbau zur Schichtabscheidung mit einem Mikroplasma-Jet bei Atmosphärendruckbedingungen konzipiert und aufgebaut. Dieser besteht im Wesentlichen aus dem Plasma-Jet und der ihn umgebenden Glaskuppel, welche die Erzeugung definierter Umgebungsatmosphären bei Normaldruck gestattet sowie vor eventuell entstehenden toxischen Reaktionsprodukten schützt. Als erste Aufgabe wurde die Deposition mit den aminogruppenhaltigen Präkursoren Cyclopropylamin (CPA) und Ethylendiamin (EDA) bearbeitet. Es zeigte sich, dass die Abscheidung im selbstorganisierten Jet-Modus möglich war. Die abgeschiedenen Schichten besitzen trotz eines kuppelförmigen Abscheidungsprofils eine homogene chemische Struktur mit einem Stickstoffgehalt von bis zu 20%, wie durch Profilometrie beziehungsweise XPS ermittelt wurde. Es wurden Werte von [NH2]/[C] zwischen 5,5 % und 3 % (EDA) sowie 4 % und 1 % (CPA) erreicht, abhängig von der Behandlungszeit der Substrate und der verwendeten Umgebungsatmosphäre. Die Schutzgasatmosphäre, bestehend aus einem Gemisch aus Stickstoff und Wasserstoff, welche dazu gedacht war die Bildung primärer Aminogruppen zu unterstützen, hatte einen negativen Effekt auf die Abscheidung. Im Vergleich zu einem Prozess an Luft wurde die Depositionsrate halbiert. Weiterhin konnte ein positiver Effekt auf den Anteil der Aminogruppen nur bei CPA festgestellt werden. Bezüglich der chemischen Zusammensetzung der Schichten wird ein erstes Modell der Plasmapolymerisationsreaktionen vorgestellt, welches auf dem wiederholten Vorgang der Abspaltung einer Aminogruppe und der nachfolgenden Reaktion der so entstandenen Radikale basiert. Bei der Bearbeitung der zweiten Aufgabe, der Deposition von fluorierten Plasmapolymer-Schichten, wurde ein spezielles Entladungsregime des Jets entdeckt. Die hierbei identifizierten Konditionen ermöglichten erstmalig die Abscheidung von C:F-Schichten mit einem Atmosphärendruck Jet. Hierbei wurden mit Octafluorcyclobutan (c-C4F8) als Präkursor, mit hohen Wachstumsraten (bis zu 43 nm/s mit N2-Atmosphäre) Schichten erzeugt. In diesen wurde mitttels XPS eine homogene chemische Struktur mit einem [F]/[C]-Verhältnis von 1,4 und einem sehr geringen Gehalt an Stickstoff und Sauerstoff nachgewiesen. Fits des hoch aufgelöst gemessenen C 1s Peaks zeigen einen Vernetzungsgrad von 44 % und ein [CF2]/[CF3]-Verhältnis von rund 1,8. Der statische Wasserkontaktwinkel bei diesen Schichten lag im Bereich von 100° – 135°. Die geforderte Hydrophobie der Schichten wurde damit erreicht. Luft als Umgebungsatmosphäre während des Beschichtungsprozesses führt nicht zu einem überwiegend ätzenden Plasmaprozess, reduziert jedoch die Depositionsrate um Faktor vier. Änderungen der chemischen Zusammensetzung der Schicht im Vergleich zur Schutzgasatmosphäre wurden nicht festgestellt. Die Verwendung von Octafluorpropan (C3F8) als Präkursor ergab nur ein minimales Schichtwachstum unter Schutzgas- und kein Wachstum unter Luft-Atmosphäre. Basierend auf den Beobachtungen anderer Autoren, wurde dies durch für die Plasmapolymerisation ungünstigere Fragmentierung des Präkursors erklärt. Das spezielle Entladungsregime, die eingeschnürte und lokalisierte bogenähnliche Entladung, wird als die Ausprägung einer --Modus Atmosphärendruck Entladung erklärt, bei der das Substrat als zweite geerdete Elektrode fungiert. Hierzu ist eine ausreichende Leitfähigkeit des Substrats notwendig. Anhand eines vereinfachten Ersatzschaltbildes werden die beobachteten Abhängigkeiten von Substratmaterial und Entladungsregime modelliert
Platelet factor 4 (PF4, synonym: CXCL4) is an evolutionary old chemokine with proposed roles in hemostasis and antimicrobial defense. In addition, PF4 has attracted considerable attention as a crucial mediator of one of the most prothrombotic adverse drug effects affecting blood cells, heparin-induced thrombocytopenia (HIT). Interest in PF4 substantially increased in 2021 when it was identified as the target antigen in the life-threatening adverse effect, vaccine-induced immune thrombotic thrombocytopenia (VITT). We address the concept that a major biological function of PF4—a strongly cationic chemokine—is to bind to negatively-charged prokaryotic microorganisms, resulting in structural changes in PF4 that trigger a danger signal recognized by the adaptive immune system. Application of biophysical tools has provided substantial insights into the molecular mechanisms by which PF4 becomes immunogenic, providing insights into a new mechanism of autoimmunity. Binding of autoantibodies with high affinity induces conformational change(s) in the endogenous protein, which are then recognized as foreign antigen, as exemplified by the prothrombotic disorders, autoimmune HIT and VITT. The final part of our review summarizes current assays for HIT and VITT, explaining how structural aspects of anti-PF4 pathobiology relate to assay design and performance characteristics. Currently, functional (platelet activation) assays using washed platelets detect HIT antibodies when heparin is added, and VITT antibodies when PF4 is added. Solid-phase PF4-dependent immunoassays using microtiter plates are sensitive for both HIT and VITT antibodies, while rapid immunoassays, in which the PF4/heparin antigen is coated on beads, are sensitive and specific for HIT, but not for VITT antibodies.
Sphingosine-1-phosphate (S1P) is a versatile signaling lipid involved in the regulation of numerous cellular processes. S1P regulates cellular proliferation, migration, and apoptosis as well as the function of immune cells. S1P is generated from sphingosine (Sph), which derives from the ceramide metabolism. In particular, high concentrations of S1P are present in the blood. This originates mainly from erythrocytes, endothelial cells (ECs), and platelets. While erythrocytes function as a storage pool for circulating S1P, platelets can rapidly generate S1P de novo, store it in large quantities, and release it when the platelet is activated. Platelets can thus provide S1P in a short time when needed or in the case of an injury with subsequent platelet activation and thereby regulate local cellular responses. In addition, platelet-dependently generated and released S1P may also influence long-term immune cell functions in various disease processes, such as inflammation-driven vascular diseases. In this review, the metabolism and release of platelet S1P are presented, and the autocrine versus paracrine functions of platelet-derived S1P and its relevance in various disease processes are discussed. New pharmacological approaches that target the auto- or paracrine effects of S1P may be therapeutically helpful in the future for pathological processes involving S1P.
In der vorliegenden Arbeit wurde der Einfluss von Rosuvastatin auf die eNOS-Expression (endotheliale NO-Synthase), LOX-1, dem Rezeptor für oxidiertes LDL, und verschiedene Adhäsionsmoleküle (ICAM-1, VCAM-1) in humanen Endothelzellen analysiert. Rosuvastatin zeigte keine physiologisch relevante Abnahme der Zellviabilität. In Hinblick auf die eNOS-mRNA- und Proteinexpression führte Rosuvastatin zu einer signifikanten Verbesserung sowohl unter normalen physiologischen als auch unter inflammatorsichen Bedingungen, ausgelöst durch eine Koinkubation mit TNF-alpha. Auch war es in der Lage, die Aktiviät der endothelialen NO-Synthase signifikant zu erhöhen. Eine Inkubation humaner Endothelzellen mit TNF-alpha resultierte in einer signifikanten Expressionserhöhung von LOX-1 sowohl auf mRNA- als auch auf Proteinebene, welche durch eine gleichzeitige Gabe von Rosuvastatin sogar noch ausgeprägter ausfiel. Trotz dieser vermehrten Expression führte die Inkubation mit Rosuvastatin aber zu einer signifikant verminderten Aufnahme von oxLDL in die Zellen. Diese statinvermittelten Effekte ließen sich durch einen Inhibitor der Rho-assoziierten Proteinkinase (Y27632) imitieren. Des Weiteren konnte diese Arbeit zeigen, dass Rosuvastatin die Oberflächenexpression von ICAM-1 und VCAM-1 signifikant vermindert, wohingegen auf mRNA-Ebene eine Expressionssteigerung zu beobachten war. Untersuchungen im Rahmen dieser Arbeit konnten klären, dass dieser Widerspruch auf ein vermehrtes Shedding der löslichen Proteine zurückzuführen ist. Rosuvastatin vermittelt diese Effekte auf die Adhäsionsmoleküle möglicherweise durch das Eingreifen in den NF-kappaB-Signalweg, da es sowohl Effekte auf die Degradation von IkappaB-alpha als auch die Translokation der NF-kappaB-Untereinheit p50 ausübt.
Background: Protease-activated receptor 1 (PAR1) and toll-like receptors (TLRs) are inflammatory mediators contributing to atherogenesis and atherothrombosis. Vorapaxar, which selectively antagonizes PAR1-signaling, is an approved, add-on antiplatelet therapy for secondary prevention. The non-hemostatic, platelet-independent, pleiotropic effects of vorapaxar have not yet been studied. Methods and Results: Cellular targets of PAR1 signaling in the vasculature were identified in three patient cohorts with atherosclerotic disease. Evaluation of plasma biomarkers (n = 190) and gene expression in endomyocardial biopsies (EMBs) (n = 12) revealed that PAR1 expression correlated with endothelial activation and vascular inflammation. PAR1 colocalized with TLR2/4 in human carotid plaques and was associated with TLR2/4 gene transcription in EMBs. In addition, vorapaxar reduced atherosclerotic lesion size in apolipoprotein E–knock out (ApoEko) mice. This reduction was associated with reduced expression of vascular adhesion molecules and TLR2/4 presence, both in isolated murine endothelial cells and the aorta. Thrombin-induced uptake of oxLDL was augmented by additional TLR2/4 stimulation and abrogated by vorapaxar. Plaque-infiltrating pro-inflammatory cells were reduced in vorapaxar-treated ApoEko mice. A shift toward M2 macrophages paralleled a decreased transcription of pro-inflammatory cytokines and chemokines. Conclusions: PAR1 inhibition with vorapaxar may be effective in reducing residual thrombo-inflammatory event risk in patients with atherosclerosis independent of its effect on platelets.
The pathobiont Streptococcus pneumoniae causes life-threatening diseases, including pneumonia, sepsis, meningitis, or non-invasive infections such as otitis media. Serine proteases are enzymes that have been emerged during evolution as one of the most abundant and functionally diverse group of proteins in eukaryotic and prokaryotic organisms. S. pneumoniae expresses up to four extracellular serine proteases belonging to the category of trypsin-like or subtilisin-like family proteins: HtrA, SFP, PrtA, and CbpG. These serine proteases have recently received increasing attention because of their immunogenicity and pivotal role in the interaction with host proteins. This review is summarizing and focusing on the molecular and functional analysis of pneumococcal serine proteases, thereby discussing their contribution to pathogenesis.
This article gives an initial overview of the explanatory power of established approaches in comparative political science of various lockdown strategies in connection with the COVID-19 pandemic in 35 democracies. In a macro-comparative statistical analysis of the first wave of the pandemic, I test partisan and veto player theories. I distinguish two phases of the first wave of the COVID-19 pandemic, which show distinct patterns of political impacts. In the first phase of implementing lockdown strategies, central governments were relatively uncontested and partisan theory has strong explanatory power. In the second phase of lifting lockdowns, party differences lose relevance, but veto players have a strong influence during this time. The analysis shows that political science theories are useful for analysing political processes not only under normal conditions but also in extreme social crises. Moreover, it provides deeper insights into the democratic decision-making process of advanced democracies in exceptional situations.
Pollen productivity estimates (PPEs) are a key parameter for quantitative land-cover reconstructions from pollen data. PPEs are commonly estimated using modern pollen-vegetation data sets and the extended R-value (ERV) model. Prominent discrepancies in the existing studies question the reliability of the approach. We here propose an implementation of the ERV model in the R environment for statistical computing, which allows for simplified application and testing. Using simulated pollen-vegetation data sets, we explore sensitivity of ERV application to (1) number of sites, (2) vegetation structure, (3) basin size, (4) noise in the data, and (5) dispersal model selection. The simulations show that noise in the (pollen) data and dispersal model selection are critical factors in ERV application. Pollen count errors imply prominent PPE errors mainly for taxa with low counts, usually low pollen producers. Applied with an unsuited dispersal model, ERV tends to produce wrong PPEs for additional taxa. In a comparison of the still widely applied Prentice model and a Lagrangian stochastic model (LSM), errors are highest for taxa with high and low fall speed of pollen. The errors reflect the too high influence of fall speed in the Prentice model. ERV studies often use local scale pollen data from for example, moss polsters. Describing pollen dispersal on his local scale is particularly complex due to a range of disturbing factors, including differential release height. Considering the importance of the dispersal model in the approach, and the very large uncertainties in dispersal on short distance, we advise to carry out ERV studies with pollen data from open areas or basins that lack local pollen deposition of the taxa of interest.
Abstract
Antimicrobial coating of implant material with poly(hexamethylene biguanide) hydrochloride (PHMB) may be an eligible method for preventing implant‐associated infections. In the present study, an antibacterial effective amount of PHMB is adsorbed on the surface of titanium alloy after simple chemical pretreatment. Either oxidation with 5% H2O2 for 24 hr or processing for 2 hr in 5 M NaOH provides the base for the subsequent formation of a relatively stable self‐assembled PHMB layer. Compared with an untreated control group, adsorbed PHMB produces no adverse effects on SaOs‐2 cells within 48 hr cell culture, but promotes the initial attachment and spreading of the osteoblasts within 15 min. Specimens were inoculated with slime‐producing bacteria to simulate a perioperative infection. Adsorbed PHMB reacts bactericidally against Staphylococcus aureus, Staphylococcus epidermidis, and Pseudomonas aeruginosa after surface contact. Adhered SaOs‐2 cells differentiate and produce alkaline phosphatase and deposit calcium within 4 days in a mineralization medium on PHMB‐coated Ti6Al4V surfaces, which have been precontaminated with S. epidermidis. The presented procedures provide a simple method for generating biocompatibly and antimicrobially effective implant surfaces that may be clinically important.

After size and charge-state selection, the cluster properties are further investigated by various methods including photo-dissociation. Depending on the particular cluster species various decay modes are observed.
In der vorliegenden Arbeit wurde die Assoziation der Polymorphismen Exon 6 und 5UTR des IGF-II-Gens mit dem Phänotyp Small-for-gestational-age (SGA) überprüft. IGF-II als ein bedeutender pränataler Wachstumsfaktor stellt ein mögliches Kandidatengen für die Entstehung von SGA dar. SGA und ein damit verbundenes zu niedriges Geburtsgewicht bei Neugeborenen sind wichtige Risikofaktoren sowohl für postnatale als auch für später auftretende Erkrankungen. IUGR ist eine multifaktoriell verursachte Störung, der Umwelt- und genetische Faktoren auf verschiedenen Ebenen zu Grunde liegen können. Entsprechend wurden bekannte Einflussfaktoren für das Geburtsgewicht und die Körperlänge Neugeborener wie Geschlecht des Kindes, Alter der Mutter, mütterliches Gewicht vor sowie Gewichtszunahme während der Schwangerschaft, Körpergröße der Mutter, Nikotinkonsum sowie mütterliche Erkrankungen berücksichtigt. Es wurde die Häufigkeit des Vorkommens von Exon 6 g.4225A/G und 5UTR g.6814_6819delAGGGC im Patientenkollektiv im Vergleich zu nicht-wachstumsverzögerten Neugeborenen. Für die Analysen standen DNA und Daten aus dem SNiP, einer populationsbasierten Querschnittsstudie von 147 SGA-Neugeborenen und 258 nicht-wachstumsverzögerten Neugeborenen aus Ostvorpommern zur Verfügung. Die Fälle entstammen weitgehend der gleichen Population wie alle Kontrollen. Alle Neugeborenen waren gesunde, nicht miteinander verwandte Einlinge ohne Fehlbildungen. Die Verteilung der Genotypen wurde für Exon 6 g.4225A/G mittels PCR und anschließender SSCP sowie Silberfärbung und für 5UTR g.6814_6819delAGGGC mittels PCR und Heteroduplexanalyse untersucht. Bei der Kontrolle der PCR-Produkte von 5UTR g.6814_6819delAGGGC wurde die Fähigkeit des Polymorphismus, Heteroduplices zu bilden, entdeckt und nachfolgend die Methode der Heteroduplexanalyse für diesen Polymorphismus etabliert. Für die Berechnungen wurden die SGA-Neugeborenen in Fallgruppen eingeteilt: Neugeborene mit einem Geburtsgewicht unterhalb der 10. Perzentile mit normaler Geburtslänge (A), solche mit einer Geburtslänge unterhalb der 10. Perzentile mit normalem Geburtsgewicht (B), die Neugeborenen mit einem Geburtsgewicht und einer Geburtslänge jeweils unterhalb der 10. Perzentile (C)sowie die Neugeborenen mit einem BMI unterhalb der 10. Perzentile (D) bildeten je eine Gruppe. Die Gesamtheit der Gruppen A, B und C ergibt die SGA-Gruppe. Als Kontrollgruppe dienten Neugeborene mit einem Geburtsgewicht und einer Geburtslänge oberhalb der 10. Perzentile (Gruppen A-C und SGA-Gruppe) bzw. mit einem BMI oberhalb der 10. Perzentile (Gruppe D). Die statistische Auswertung erfolgte mittels U-Test nach Mann-Whitney für den Vergleich der Verteilung der Variablen Gestationsalter, Alter bzw. Größe der Mutter, Gewicht vor der Schwangerschaft und Gewichtszunahme jeweils zwischen den Fallgruppen und Kontrollen. Für die Berechnung der Unterschiede hinsichtlich der Allelfrequenz von Exon 6 g.4225A/G und 5UTR g.6814_6819delAGGGC sowie des Rauchverhaltens wurden der Chi-Quadrat-Test nach Pearson bzw. der Exakte Test nach Fisher verwendet. Zusätzlich wurden in einer logistischen Regressionsanalyse die Genotypen gleichzeitig mit den genannten Risikofaktoren für SGA überprüft, um ihren Einfluss auf das SGA-Risiko zu quantifizieren. Bezüglich Gestationsalter, Alter der Mutter sowie mütterlichem Gewicht vor der Schwangerschaft wurden keine signifikanten Unterschiede zwischen SGA-Kindern und Kontrollen festgestellt. Bei der Untersuchung von mütterlicher Gewichtszunahme, Größe der Mutter sowie Nikotinkonsum zeigten sich leichte, jedoch statistisch signifikante Unterschiede. Unter den SGA-Neugeborenen war die Gewichtszunahme und Größe der Mutter geringer und der angegebene Nikotinkonsum häufiger. Bei Analysen im multivariaten Modell wurden nur hinsichtlich des Rauchverhaltens statistisch signifikante Unterschiede zwischen SGA- und Kontrollgruppe in Bezug auf die übrigen Risikofaktoren für SGA gefunden. Demnach konnte ein wichtiger Teil der als Einflussfaktoren für SGA bekannten Variablen in der Patienten- und Normalgruppe kontrolliert werden. Die Analyse der Verteilung der Polymorphismen zeigte weder für Exon 6 g.4225A/G noch für 5UTR g.6814_6819delAGGGC eine signifikante Assoziation mit SGA. Ein nicht statistisch signifikanter Trend unterstrich den vermuteten Zusammenhang. Demnach war im Patientenkollektiv gegenüber den normalen Kontrollen ein erhöhtes Vorkommen des G-Allels in Exon 6 g.4225A/G bzw. der Deletion in 5UTR g.6814_6819delAGGGC zu verzeichnen. Beide Unterschiede erreichten jedoch nicht die statistische Signifikanz. Somit ist ein Beitrag der genannten Polymorphismen zur Entstehung von SGA wahrscheinlich, aber durch unsere Ergebnisse nicht statistisch signifikant belegt. Um dieses Ergebnis zu bestätigen sollten weitere populationsbasierte Untersuchungen mit größeren Fallzahlen mit einer noch umfassenderen Kontrolle der SGA-Risikofaktoren durchgeführt werden.
Bei ersten Betrachtungen zum Zelltransport nahm man an, dass Stoffe passiv über die Zellmembran diffundieren. Nach weiteren Forschungen musste dies revidiert werden. Heute weiß man, dass dies fast nur über spezielle Proteine möglich ist. Diese Proteine sind Kanäle und Transporter welche aufgrund ihrer Funktion eine Schlüsselposition in der zellulären Homöostaste darstellen. Sie sorgen für die Aufnahme und den Ausstrom fast aller wichtigen Substanzen.
Im Jahr 2007 wurde die Entdeckung eines neue Zelltransporters, TETRAN (Tetracycline transporter-like protein), veröffentlicht. In ersten Untersuchungen zeigte sich ein bevorzugter Transport von NSAIDs über die Zellmembran. NSAIDs haben bei unterschiedlichen Personen abweichende Wirkungen bzw. Nebenwirkungen deren Gund bis heute noch nicht endgültig geklärt ist.
In dieser Arbeit konnten polymorphe TETRAN überexprimierende Zellen erzeugt werden und der Nachweis unterschiedlich starker Expression des Proteins in verschiedenen Geweben nachgewiesen werden. Unter anderem zeigte sich eine gute Expression in Plazentagewebe. Welches auch wegen guter Verfügbarkeit zur Reihensequenzierung von 25 Proben genutzt wurde. Es zeigten sich 28 Synonyme und 85 Veränderungen im Gencode welche Einfluss auf die Primärsequenz des Proteins haben. Diese wurden alle in einer berechneten Grafik des Proteins zusammengefasst und dargestellt. In drei Situationen bildete sich anstatt des Wechsels einer Aminosäure ein Stopcodon. Dies trat jedoch nie homozygot auf. So das immer ein vollständiges Protein vorhanden war. In Anlehnung an vergleichbare Studien könnte dies mit ein Grund der differenten Wirkung von NSAIDs sein. Umfangreichere Beweise können in weiteren Studien noch folgen.
Polypharmacy in patients with multiple sclerosis and the impact on levels of care and therapy units

Background: The aim of this study was to examine the societal costs of polypharmacy in patients with multiple sclerosis (MS). We therefore focused on the association between the number of medications on the level of care (LOC), the German classification of the need for care, and the number of therapy sessions (TTU).
Methods: In addition to demographic information and medication, 101 MS patients performed the Multiple Sclerosis Health Resource Utilization Survey (MS-HRS). Medications were subdivided into a total number of medications (TD), MS-related medication [MSD, i.e., disease-modifying drugs (DMDs) and symptomatic treatment (SD)], and medication for comorbidities (CDs). Multivariate linear regression models were performed to estimate if the amount of each medication type affects LOC or TTU.
Results: Polypharmacy appeared in 54 patients at the time of the survey. The relative risk (RR) of LOC 1 increased significantly by 2.46 (p = 0.001) per TD and by 2.55 (p = 0.004) per MSD, but not per CD (RR 1.44; p = 0.092). The effect of RR on MSD was driven by SD (RR 2.2; p = 0.013) but not DMD (RR 2.6; p = 0.4). RR of MSD remained significant for LOC 2 (1.77; p = 0.009) and LOC 3/4 (1.91; p = 0.015), with a strong trend in RR of SD, but not DMD. TTU increased significantly per MSD (p = 0.012), but not per TD (p = 0.081) and CD (p = 0.724).
Conclusion: The number of MSDs is related to the likelihood of a higher level of care and the number of therapy sessions and is therefore a good indication of the extent of the societal costs.
Im Alltag des HNO-Arztes ist die Polyposis nasi eine häufige Erkrankung. Der Patient stellt sich in der Regel mit Symptomen wie behinderter Nasenatmung, Kopfschmerzen oder Nasenlaufen vor. Häufig findet sich eine Assoziation mit anderen Atemwegserkrankungen, wie Asthma oder Aspirinintoleranz. Therapeutisch stehen eine Behandlung mit lokalen oder systemischen Steroiden sowie operative Eingriffe zur Verfügung. Eine hohe Rezidivrate sowohl beim operativen als auch beim medikamentösen Vorgehen macht die Behandlung oft unbefriedigend. In den letzten Jahren wurden mehrere Studien zur Untersuchung der Häufigkeit der Polyposis nasi durchgeführt. Ziel war es nicht nur genauere Zahlen zur Prävalenz und Inzidenz dieser Erkrankung zu erhalten, sondern auch die Erforschung von Krankheitssymptomen, Begleitumständen wie die Belastung durch Umweltfaktoren, Medikamenten oder anderen Erkrankungen und demographischen Besonderheiten. Zur Abschätzung möglicher genetischer Zusammenhänge interessierte auch die familiäre Belastung. Einige Studien untersuchten nur symptomatische Patienten oder verwendeten einen Fragebogen. Nur eine schwedische Studie von Johansson et al. führte ebenfalls eine Reihenuntersuchung durch. Die Ergebnisse wiesen auf eine Zunahme der Erkrankung im Alter hin. Allerdings konzentrierten sich die vorliegenden Untersuchungen generell auf Erwachsene bzw. Kinder und Jugendliche. Diese Studie beschäftigt sich nun ausschließlich mit Personen ab dem 60. Lebensjahr. In einer Reihenuntersuchung von 500 Probanden wurde neben einer endoskopischen Untersuchung der Nase ein ausführlicher Fragebogen mit den Teilnehmern erarbeitet. Die gewonnenen Daten sollten helfen, die Häufigkeit der Polyposis nasi in einem bestimmten geographischen Areal zu schätzen, Begleitfaktoren zu erfassen und eine kritische Würdigung etablierter Behandlungsmethoden ermöglichen. Augrund der oft langen Krankheitsdauer und Rezidivfreudigkeit würde eine Bewertung der vorhandenen Therapien eine jahrzehntelange Nachbeobachtung erfordern. Die Konzentration auf Menschen ab dem 60. Lebensjahr sollte die Schwierigkeiten solcher Langzeitstudien umgehen. Bei der endoskopischen Rhinoskopie von 500 Probanden konnten Nasenpolypen bei 30 Teilnehmern (6 %) diagnostiziert werden. Diese Studie bestätigt somit ein erhöhtes Vorkommen der Polyposis nasi im Alter. Nasenpolypen kamen bei Männern, Kinderlosen und Rauchern in der vorliegenden Studie häufiger vor. Fast die Hälfte der Betroffenen kannte die Diagnose nicht, trotz einer gegenüber dem Studiendurchschnitt häufiger vorkommenden typischen Symptomatik. Die Vermutung, dass diese Patienten unter anderen belastenderen Erkrankungen litten, ließ sich nicht belegen. Die untersuchten chronischen Begleiterkrankungen kamen in dieser Gruppe seltener vor. Die Bewertung der heute zur Verfügung stehenden therapeutischen Möglichkeiten zeigt eine Überlegenheit der operativen Nasennebenhöhlensanierungen gegenüber weniger invasiven oder konservativen Methoden. Mit einer spontanen Remission der Erkrankung darf nicht gerechnet werden. Zusammen mit der steigenden Prävalenz der Polyposis nasi im Alter spricht dies für eine konsequente Therapie und eine lange Nachbetreuung der Betroffenen. Die hohe Zahl undiagnostizierter Patienten fordert eine gewisse Sensibilität gegenüber den krankheitstypischen Symptomen.
In den Weltmeeren findet rund die Hälfte der jährlichen globalen Kohlenstofffixierung statt, davon ein großer Anteil in küstennahen Regionen. Hier kommt es zu wiederkehrenden saisonalen Algenblüten, die durch eine zeitlich begrenzte explosionsartige Vermehrung von Mikroalgen (hauptsächlich Diatomeen und Coccolithophoren) charakterisiert sind. Vor allem Frühjahrsblüten (März-Mai) haben aufgrund ihrer zeitlichen und räumlichen Vorhersagbarkeit einen hohen Stellenwert als Modellsysteme, anhand deren sich der Kohlenstoffkreislauf der Meere untersuchen lässt.
Mikroalgen produzieren eine große Vielfalt an Makromolekülen, die für die mit ihnen vergesellschafteten Bakterien als Nahrungsgrundlage dienen. Besonders im Fokus stehen hier die für den Kohlenstoffkreislauf relevanten Polysaccharide. Im Gegensatz zu anderen natürlichen Makromolekülen wie DNA oder Proteinen können Polysaccharide aus vielen verschiedenen Monomeren mit unterschiedlichsten Bindungen bestehen. Zusätzlich finden sich an diesen Zuckermonomeren viele Modifikationen wie Acetylierungen, Methylierungen oder Sulfatierungen, die die Komplexität weiter erhöhen. Diese Variabilität bedingt eine hohe strukturelle und funktionale Diversität. So können Polysaccharide Speicherstoffe, Zellwandbestandteile oder Teile der extrazellulären Matrix darstellen.
Komplementär hierzu besitzen Polysaccharid-verwertende Bakterien entsprechend komplexe, enzymatische Abbaumechanismen. Besonders hervorzuheben sind hier die Bakterien des Phylums Bacteroidota, die sich in verschiedensten Nischen auf den Abbau von Polysacchariden spezialisiert haben. Sie finden sich in Bodenproben, als Teil der menschlichen Darmflora, oder eben auch als bedeutende Begleiter von Algenblüten.
Bacteroidota (und in marinen Systemen hauptsächlich die zu ihnen gehörenden Flavobakterien) besitzen zum Abbau diverser Polysaccharide sogenannte Polysaccharide utilization loci (PULs), genomische Inseln, die alle notwendigen Proteine zur Aufnahme und Abbau eines bestimmten Polysaccharids codieren. Hierzu gehören hochspezifische Enzyme (Carbohydrate-active enzymes, CAZymes), transkriptionelle Regulatoren sowie Transportersysteme, die initial gespaltene Oligosaccharide über die Membran in das Bakterium transportieren, wo sie von weiteren Enzymen vollständig abgebaut werden. Diese Co-Lokalisation der benötigten Gene und deren gemeinsame Regulation stellt einen enormen Selektionsvorteil der Bacteroidota dar und ist der Grund, warum sie, ähnlich wie Algen, einer jährlich wiederkehrenden Sukzession folgen, die sich gut untersuchen lässt.Die Forschungsartikel, die Teil dieser Doktorarbeit sind, untersuchen das Zusammenspiel von Polysaccharid-produzierenden Algen mit den Bakterien, die sie abbauen, aber auch darauf basierende Beziehungen der Bakterien untereinander. Die erste Publikation beschäftigt sich mit dem weit verbreiteten Speicherpolysaccharid α-Glucan, für das der Großteil der blütenbegleitenden Bakterien einen spezifischen aktiven PUL besitzt. Eine Untersuchung der in der Blüte vorhandenen Algenarten bestätigte, dass die Blüte von β-Glucan-produzierenden Algen dominiert wird. Da Bakterien aber selbst α-Glucane als Speicherpolysaccharide verwenden, konnte gezeigt werden, dass nicht die Algen selbst, sondern die Bakterien Hauptproduzent dieser Polysaccharide während einer Phytoplanktonblüte sind. Bakterielle Proteine, die dem Abbau von Algen-β-Glucan und dem daraus folgenden Aufbau von bakteriellem α-Glucan dienen, waren in Umweltproben und in Laborkulturen unter ähnlichen Bedingungen abundant. Die Untersuchung von extrahiertem bakteriellem Polysaccharid bewies, dass dieses nicht nur α-Glucan enthält, sondern dass dieses Polysaccharid auch in der Lage war, α-Glucan PULs mariner Bakterien zu induzieren. Hier zeigte sich ein innerhalb des marinen Kohlenstoffkreislaufs bisher wenig berücksichtigter Kreislauf, indem Bakterien Polysaccharide anderer Bakterien nutzen, die z.B. durch Viren lysiert wurden.
Die anderen zwei Artikel dieser Arbeit befassen sich mit dem Abbau von Zellwandpolysacchariden durch blütenassoziierte Modellbakterien. In einer der Studien wird detailliert der Abbau eines β-Mannans (ein Polysaccharid das hauptsächlich aus dem Monosaccharid Mannose besteht) durch ein Bakterium des Genus Muricauda beschrieben. Die PUL-Struktur dieses Bakteriums kam in mehreren anderen Phytoplanktonblüten-assoziierten Bakterien vor. Diese Beobachtung wies darauf hin, dass es sich hier um ein Mannan mit zusätzlichen Galactose- und Glucose-Substitutionen handelte. Proteom-Untersuchungen bestätigten, dass das Bakterium derartige Substrate unter Induktion des β-Mannan-PULs nutzen können. β-Mannan konnte durch Antikörpermarkierung in Blütenproben sowie spezifischen Mikroalgenarten (Chaetoceros, Coscinodiscus) nachgewiesen werden. Die in dieser Publikation charakterisieren β-Mannan-PUL-codierten Enzyme waren in der Lage, dieses Signal zu löschen, was bewies, dass Muricauda sp. Mannan-basierte Zellwandpolysaccharide bestimmter Arten von Mikroalgen abbauen kann.
Die dritte Studie geht näher auf den Abbau von Xylanen (bestehend aus Xylose) durch ein blütenassoziiertes Bakterium des Genus Flavimarina ein. In diesem Bakterium wurden anhand der enthaltenen Xylanasen zwei putative Xylan-PULs annotiert. Wachstumsexperimente und Proteom-Untersuchungen zeigten, dass einer dieser PULs hauptsächlich bei Wachstum auf Glucoronoxylan induziert wird, während der andere PUL aufArabinoxylane stärker reagierte. Untersuchung der PUL-CAZymes bestätigte diese Ergebnisse durch Charakterisierung mehrerer Xylanasen sowie Glucoronidasen und Arabinofuranosidasen. Zusätzlich codierten beide PULs für Esterasen, die eine Modifikation der natürlichen Substrate durch Acetylierungen oder Methylierungen nahelegen. Da all diese Merkmale von terrestrischen Xylanen geteilt werden und in Blütenproben aus Küstennahen Regionen Xylane nachgewiesen wurden, ist es möglich, dass Bakterien aus solchen Regionen sowohl Xylane terrestrischen Ursprungs (z.B. durch Flusseinspeisung) sowie marinen Ursprungs abbauen können.
Marine bacteria represent the most diverse organisms in the marine environment. The majority of these microbes is unknown and unculturable. Algae represent the main nutrient source for bacteria. Macro- and microalgae can consist to 70% of polysaccharides. The metabolic degradation of marine polysaccharides is underexplored and thus these mechanisms have to be investigated. These mechanisms are of high importance to generate defined oligosaccharides for the medical and pharmaceutical applications. The specific structure of marine poly- and oligosaccharides show antiviral activities, e.g. carrageenans from red algae are used for the inhibition of human papillomavirus. Another alginate derived marine polysaccharide show inhibition of the replication of the human immunodeficiency virus (HIV). The degradation mechanisms of marine CAZymes and the structure of marine polysaccharides should be further investigated for their high potential of antiviral activities and the creation of new marine drugs.
Many marine bacteria produce membrane extension like membrane vesicles or appendages but the function of these is poorly understood. In order to investigate their function, especially concerning polysaccharide utilization, proteomic analyses of subcellular compartments were performed. Microscopy analyses revealed that, beside MV, P. distincta forms different appendages, vesicle chains (VC) and thin filaments which were dedicated to extracellular polymeric substance. The formation of MV and VC was independent of growth phase or carbon source. The proteomic data showed that transporters end enzymes for the initial degradation of pectin and alginate were highly abundant in these membrane extensions and that there could be a kind of sorting for proteins in the membrane extensions. Additionally, two PUL encoded alkaline phosphatases and other phosphate acquiring enzymes were abundant in the MV and VC fractions. This indicates, that P. distincta constitutively produces enzymes for phosphate uptake, which would be necessary in the phosphate-limiting environment of the Southern Ocean. On the one hand marine bacteria produce membrane extensions in order to create a larger surface in the nutrient limiting marine environment for an increased chance to get in contact to nutrients and on the other hand the results indicate an accumulation of enzymes responsible for uptake and degradation of carbohydrates and phosphates in the MV and VC. Therefore, the membrane extensions act as nutrient traps and this might be beneficial for the bacteria in the diffuse aquatic environment.
The microbial community structure and the metabolism of bacteria in the Southern Ocean are very poorly investigated. The SO is a harsh environment for all organism but nevertheless, the SO is of high importance for the climate in the world due to the high carbon dioxide uptake. In this study water samples from two different sampling sites (S1 and S2) in the SO were investigated. With a metagenomic and metaproteomic approach the key players and the metabolic activity were analyzed. Additionally, the surface water was inoculated with pectin and incubated for several days in order to analyze polysaccharide utilization loci for pectin degradation and to isolate new pectin degraders. 16S-rDNA analyses revealed the bacterial community from the genomic data. Bacteria were separated in particle-associated and free-living bacteria. The overall particle associated bacterial community at both sampling sites was comparable, with Bacteroidetes and Gammaproteobacteria as the abundant phylum. Within the Gammaproteobacteria the Alteromonadaceae and Colwelliaceae were more abundant at S2 than at S1. The free-living bacteria at S1 were dominated by the Alphaproteobacteria, especially the SAR11 clade I. Metagenomic analyses showed that both sampling sites had comparable PUL composition, but taxonomical classification of PULs was differently. The metaproteome data revealed that PUL encoded enzymes were not highly abundant. Only few CAZymes were found, mostly TonB-dependent transporters belonged to the detected PUL proteins. Taxonomical classification of proteins showed differences between the sampling sites. At S2 the genus Colwellia and Arcobacter were highly increased compared to S1. At this location Candidatus Pelagibacter, Planktomarina and Polaribacter were the abundant taxa. The functional classification at both sampling sites was comparable. The only difference was the high abundance of Epsilonproteobacteria at S2 referable to the Arcobacter species. Nevertheless, the notably taxonomical differences could not be explained by the proteomic data and the functional classification, because no specific metabolic function could be highly addressed to these bacteria. These results assumed that different abundance of the key players could be explained by different environmental conditions. The pectin enriched cultured at both sampling sites were investigated for the functional potential of pectin degrading enzymes. No metaproteomic approach could be performed due to less sampling material. Only one PUL for the degradation of rhamnogalacturonan, a component of pectin, was found at S1. In contrast, bacteria grown on pectin could be isolated from these samples. Genome sequencing of five isolates showed that functional potential of pectin degradation is available. Due to the limitations of sequence alignments, it was not possible to detect a PUL responsible for pectin utilization in the metagenomic data. The results show that the polysaccharide degradation mechanism in the Southern Ocean has to be more investigated to get knowledge about the bacterial activity in the ocean’s surface and the carbon turnover in this underexplored environment.
Purpose
Outcomes of multiple myeloma (MM) patients who are refractory to daratumumab are dismal and no standard of treatment exists for this patients’ population. Here, we investigate the role of pomalidomide combinations in daratumumab-refractory MM patients.
Methods
We performed a retrospective analysis of myeloma patients treated at four referral centers (three in Germany and one in Italy). Review chart identified 30 patients with relapsed and refractory myeloma, who progressed during treatment with daratumumab and were treated with pomalidomide-based combinations in the subsequent lines of therapy.
Results
Responses improved from 37% with daratumumab to 53% with pomalidomide. Of seven patients with extramedullary MM (EMM), four achieved a clinical stabilization with pomalidomide, including one patient with a long-lasting complete response. Median progression-free survival and overall survival were 6 and 12 months, respectively. Pomalidomide combinations were well tolerated, no patient discontinued treatment due to adverse events.
Conclusion
These data show that pomalidomide-based combinations can be an effective and safe salvage regimen for daratumumab-refractory patients, including those with EMM.
Representative epidemiologic data on the average volume of the parotid gland in a large population-based MRI survey is non-existent. Within the Study of Health in Pomerania (SHIP), we examined the parotid gland in 1725 non-contrast MRI-scans in T1 weighted sequence of axial layers. Thus, a reliable standard operating procedure (Intraclass Correlation Coefficient > 0.8) could be established. In this study, we found an average, single sided parotid gland volume of 27.82 cm3 (95% confidence interval (CI) 27.15 to 28.50) in male and 21.60 cm3 (95% CI 21.16 to 22.05) in female subjects. We observed positive associations for age, body mass index (BMI), as well as male sex with parotid gland size in a multivariate model. The prevalence of incidental tumors within the parotid gland regardless of dignity was 3.94% in the Northeast German population, slightly higher than assumed. Further epidemiologic investigations regarding primary salivary gland diseases are necessary.
Die vorliegende Arbeit befasst sich mit der Verbreitung und Populationsgenetik der invasiven asiatischen Buschmücke Ae. j. japonicus, die sich seit 2008 durch Menschen-vermittelten Transport in Deutschland ausbreitet. Aedes j. japonicus ist unter Laborbedingungen Vektor für verschiedene Viren, unter anderem für das Dengue-Virus und das Chikungunya-Virus, und wurde im Feld mit dem Japanische Enzephalitis-Virus, dem West Nil-Virus und dem La Crosse-Virus infiziert gefunden. 2012 wurde aufgrund mehrerer unabhängiger Mücken-Einsendungen im Rahmen des Citizen-Science-Projekts “Mückenatlas” eine Population der Asiatischen Buschmücke in Westdeutschland entdeckt. Das Verbreitungsgebiet dieser Population befand sich weit nördlich der bisher angenommenen nördlichen Verbreitungsgrenze der Art in Süddeutschland. Das Ausmaß der Population wurde nach einem zeitlich begrenzten Monitoring auf eine Fläche von ca. 2000 km2 bestimmt. Aus dieser Population wurden Individuen von fünf Orten populationsgenetischen Analysen unterzogen, um verwandtschaftliche Beziehungen innerhalb der Population und im Vergleich zu anderen europäischen Populationen aufzudecken. Hierzu wurden sieben Mikrosatelliten-Loci untersucht. Zusätzlich wurde ein Teil der mitochondrialen nad4-Genregion der Individuen auf Nukleotid-Polymorphismen untersucht. Die Ergebnisse wurden mit bereits zuvor erhobenen Daten von Populationen aus der Schweiz, aus Österreich/Slowenien und Belgien verglichen. Die Mikrosatellitensignatur der westdeutschen Population unterschied sich deutlich von der der anderen europäischen Populationen. Weiterhin wurden verschiedene nad4-Haplotypen gefunden, die zuvor nirgendwo sonst in Europa aufgetreten waren. Demnach ist zu vermuten, dass diese Population auf eine unabhängige Einschleppung von Individuen aus Übersee zurückgeht. Der genaue Ursprung – USA oder Ostasien – konnte nicht bestimmt werden. 2013 wurden zwei weitere Ae. j. japonicus-Populationen in Europa entdeckt: eine in Norddeutschland und eine weitere in den Niederlanden. Die genetischen Signaturen von Individuen dieser Populationen wurden wie beschrieben analysiert. Zusätzlich wurde das genetische Material einer größeren Menge von Individuen aus Slowenien sowie von Individuen aus Kroatien und Süddeutschland untersucht. Die Ergebnisse wurden mit denen aus der vorigen Studie verglichen und zeigten aufgrund einer ähnlichen Mikrosatellitensignatur und gleicher nad4-Haplotypen klar, dass die norddeutsche Population eine Subpopulation der westdeutschen ist. Die geringe Populationsdichte und die vergleichsweise kleine Ausdehnung der norddeutschen Population deuten außerdem darauf hin, dass die Abspaltung nicht lange zurückliegt. Die niederländische Population scheint hingegen auf einer weiteren Einschleppung von Individuen aus Übersee zu basieren. Im Spätsommer 2015 wurde die bisher letzte deutsche Ae. j. japonicus-Population in Oberbayern und dem angrenzenden Österreich entdeckt. Populationsgenetischen Analysen zufolge ist diese Population eng mit der früher beschriebenen österreichisch-slowenischen Population verwandt und unterscheidet sich von allen anderen deutschen Populationen, was darauf schließen lässt, dass es sich bei ihr um eine Abspaltung von der österreichisch-slowenischen Population handelt. Die Ver- und Ausbreitung von Ae. j. japonicus in West- und Norddeutschland wurde vom Zeitpunkt der Entdeckung in 2012/2013 bis 2015 beobachtet. In dieser Periode erweiterte die westdeutsche Population ihr Verbreitungsgebiet beträchtlich, während die norddeutsche überhaupt nicht zu expandieren schien. Dies ist möglicherweise darauf zurückzuführen, dass die norddeutsche Population jünger als die westdeutsche ist, das Verschleppungsereignis noch nicht so weit zurückliegt und die Population sich noch in der Gründerphase befindet. Die passive weltweite Verschleppung von Stechmücken wird in der Zukunft vermutlich zunehmen, und die Etablierung und Ausbreitung invasiver Spezies, inklusive der Asiatischen Buschmücke und anderer potenzieller Überträger von Krankheitserregern, werden Europa und Deutschland weiterhin vor herausfordernde Probleme stellen. Das Monitoring der Ausbreitung von Populationen und die Durchführung populationsgenetischer Analysen zur Ermittlung von geographischen Ursprüngen sowie von Wanderungs- und Transportrouten werden helfen, weitere Einschleppungs- und Ausbreitungsereignisse nachzuvollziehen und zu unterbinden und sind daher essenzielle Instrumente des Managements von Mückenvektoren.
To uncover the genetic structure of Populus euphratica forests along the Tarim River in Xinjiang, China, a PCR set of eight microsatellite markers was established. 18 primer pairs originally developed for P. tremuloides and P. trichocarpa were screened for amplification in P. euphratica. The eight most variable loci were selected for further genotyping experiments. Subsequently, two multiplex PCR assays, each containing four loci, were set up and optimized. Three populations containing altogether 436 trees were used to characterize the selected loci. The set was found to be moderately polymorphic (mean expected heterozygosity = 0.57). The resolution was sufficient to discriminate even siblings with high confidence (PID = 1.81x10-5). Cumulative exclusion probabilities were 0.89 (single parent), 0.98 (paternity), and 1.00 (parent pair) and proved the set’s suitability for parentage analysis. Practical and theoretical analysis of consequences of genotyping errors in this semi-clonal plant showed that the vast majority of errors (62.1%) lead to division of identical genotypes. Merging of different genotypes was found to be a very rare case (0.4%). This always leads to an overestimation of genotypes. A similarity threshold of one allele difference between two genotypes to be regarded as being identical lead to an underestimation of clonal richness and genotype number of one per cent compared to an overestimation of more than 20 per cent without such a threshold. Allowing a certain amount of variation is therefore expected to reflect the clonal structure better than an analysis that considers exact matches only. Using a combination of morphological and molecular analyses, a first study demonstrated that root suckers are clearly distinguished from seedlings in their root architecture. Root suckering starts when trees are 10–15 years old and bridges distances of up to 40 m at a time. Root suckers depend on their parent tree for at least five years and are expected to have a higher mortality than generatively grown trees. Molecular analysis of old growth stands revealed a highly variable proportion of clonal growth between different stands. In the study area, the proportion of clonality decreases with distance to the main river bed (R = 0.31 at the site closest to the main river, R = 0.97 at the site farthest away from the river). An analysis of the history of river movements at different sites indicates a dependency of clonal growth on the frequency of ground water replenishment by the yearly floods. Genetic differentiation among the stands in the study area is low (FST = 0.055), and isolation by distance was not detectable (P = 0.058). Also, the river does not function as a vector for directed gene flow in downstream direction (P > 0.11). The forests are therefore considered to be one large panmictic metapopulation with unrestricted gene flow. Clonal growth does not lead to higher final stand densities (P = 0.99) and is obviously not of crucial importance for stand survival. Furthermore, analysis of vitality measures and size differences indicate that root suckers are in disadvantage both in vitality and in survival rate compared to seedlings. In this light, a possible function of clonal growth as a luxury strategy to enhance a genetic individual’s reproduction success under good site conditions can be discussed. The genetic structure of the (meta)population bears direct implications for management and conversation of the Tugai forest in Xinjiang. Due to the low degree of differentiation and the unhindered gene flow even small, fragmented, or isolated populations have conservational value, thereby clearly answering the SLOSS question (a single large or several small protected areas) in the latter sense. More than that, non-clonal stands with the highest amount of genotypic diversity can be easily identified on satellite and aerial images. Selection of such stands for conservation is therefore possible without expensive and time-consuming molecular analyses.
In rural areas, healthcare providers, patients and relatives have to cover long distances. For specialised ambulatory palliative care (SAPV), a supply radius of max. 30 km is recommended. The aim of this study was to analyse whether there are regional disparities in the supply of SAPV and whether it is associated with the distance between the SAPV team’s site and the patient’s location. Therefore, anonymised data of the Association of Statutory Health Insurance Physicians of the Federal State of Mecklenburg-Western Pomerania (M-V) were retrospectively analysed for the period of 2014–2017. Identification as a palliative patient was based on palliative-specific items from the ambulatory reimbursement catalogue. In total, 6940 SAPV patients were identified; thereof, 48.9% female. The mean age was 73.3 years. For 28.3% of the identified SAPV patients (n = 1961), the SAPV teams had a travel distance of >30 km. With increasing distance, the average number of treatment days per patient increased. It was found that there are regional disparities in the provision of SAPV services in M-V and that local structures have an important impact on regional supply patterns. The distance between the SAPV team’s site and the patient’s location is not the only determining factor; other causes must be considered.
Die vorliegende kumulative Promotionsarbeit basiert auf drei Originalmanuskripten, die am Ende der Arbeit eingebunden sind. Arterielle Hypertonie ist eine der häufigsten Erkrankungen bei Erwachsenen in Deutschland und ein wichtiger Prädiktor für kardiovaskuläre Morbidität und Mortalität. Das Renin-Angiotensin-Aldosteron System (RAAS) nimmt bei der Blutdruckregulation sowie bei der Aufrechterhaltung des Wasser- und Elektrolythaushalts eine Schlüsselrolle ein. Das RAAS kann durch verschiedene Ursachen gestört werden und dadurch zum Auslöser einer arteriellen Hypertonie werden. So weisen Patienten mit primärem Hyperaldosteronismus (PAL) eine vom RAAS autonome, erhöhte Aldosteronproduktion auf. PAL wird im Großteil der Fälle durch Aldosteron-produzierende Adenome in der Nebenniere bzw. uni- oder bilaterale Hyperplasie der Nebennieren verursacht. Man vermutet, dass der PAL die häufigste Form der sekundären Hypertonie ist. Die exakte Prävalenz des PAL ist allerdings unbekannt. Als Screeningtest wird der Aldosteron-Renin-Quotient (ARR) empfohlen, der einen relativen Aldosteronüberschuss im Vergleich zum Renin anzeigen kann. Die in diversen Studien ermittelten ARR Grenzwerte für das PAL Screening unterscheiden sich deutlich. Diese Grenzwerte wurden überwiegend anhand von Vergleichen des ARR von Patienten mit und ohne PAL ermittelt. Eine andere Möglichkeit um zwischen einem physiologischen und pathophysiologischen ARR zu unterscheiden liefern Referenzbereiche. Ziel der ersten Analyse war es Referenzwerte für den ARR aber auch die Plasma Aldosteronkonzentration (PAC) sowie die Plasma Reninkonzentration (PRC) zu ermitteln. Aldosteron nimmt auch bei der Pathogenese weiterer kardiovaskulärer Erkrankungen eine bedeutende Rolle ein, da es prooxidative, proinflammatorische und profibrotische Effekte ausübt. Die exakten Mechanismen die diesen Effekten unterliegen, sind bisher nur teilweise geklärt. Ziel der vorgestellten Studie war es herauszufinden, ob die PAC oder der ARR in einer großen Studie mit Probanden aus der Hintergrundsbevölkerung mit einer eingeschränkten flussvermittelten Vasodilatation (FMD) einhergehen. Neben der Vielzahl an Studien die Effekte des RAAS auf das kardiovaskuläre System beschreiben, mehren sich in den letzten Jahren Untersuchungen, die den Einfluss des RAAS auf den Metabolismus analysieren. Das RAAS wird dabei im Zusammenhang mit pathologischen Veränderungen des Glukosemetabolismus, des Fettstoffwechsels und der Insulinresistenz gesehen. Das Metabolische Syndrom (MetS) bezeichnet das gemeinsame Auftreten von multiplen metabolischen Veränderungen. Zur Definition des MetS werden die Faktoren viszerale Adipositas, erhöhter Blutzucker, erhöhte Triglyceride, erniedrigtes HDL-Cholesterol sowie erhöhter Blutdruck herangezogen. Bei Vorliegen von mindestens drei dieser Komponenten kann ein MetS festgestellt werden. Ziel der vorgestellten Analyse war es die Assoziation zwischen der PAC und dem MetS sowie seinen Einzelkomponenten in zwei großen deutschen Studien zu prüfen. Die der Arbeit zugrundeliegenden Daten stammen aus der ersten Follow-up Untersuchung der Study of Health in Pomerania (SHIP-1). Die Assoziation zwischen der PAC und dem MetS wurde in SHIP-1 und dem F4 Survey der Kooperativen Gesundheitsforschung in der Region Augsburg (KORA F4) analysiert. Die Referenzwerte für die PAC, PRC und den ARR wurden in einer Population bestehend aus 1347 Probanden ermittelt. Es wurden geschlechts- und altersgruppenspezifische (25-54 Jahre und 55-74 Jahre) Referenzwerte berechnet. Der Zusammenhang zwischen der PAC bzw. dem ARR und der FMD in der SHIP-1 Studie wurde anhand der Daten von 972 Probanden im Alter zwischen 25 und 88 Jahren geprüft. Es wurde ein inverser Zusammenhang zwischen dem ARR und der FMD bei Probanden beobachtet. Der Zusammenhang zwischen der PAC und dem MetS wurde in SHIP-1 anhand der Daten von 2830 Probanden und in KORA F4 anhand der Daten von 2901 Probanden geprüft. Sowohl in SHIP-1 als auch in KORA F4 wurden Zusammenhänge zwischen der PAC und dem MetS sowie Fettstoffwechselstörungen detektiert. Eine hohe PAC war mit erhöhten Odds Ratios für ein MetS, niedriges HDL-Cholesterol und erhöhte Triglyceride assoziiert. Zusammenfassend bekräftigen die durchgeführten Analysen die Hypothese, dass Störungen des RAAS mit pathophysiologischen kardiovaskulären und metabolischen Veränderungen in der Allgemeinbevölkerung einhergehen.
Populationsbasierte Studie zu Prädispositionsfaktoren und Häufigkeit der Hüftgelenksdysplasie

6.1 Zusammenfassung Ziel der prospektiven Dissertation war die Analyse von Korrelationen zwischen sonographischem Hüftbefund zum Zeitpunkt der U2 (4.-6. Lebenstag), familiärer Vorbelastung, Geschlecht und intrauterinem Lagetyp. Im Rahmen der Surveillance of Neonates in Pommerania (SNiP), einer populationsbasierten Studie zu Prävalenz und Inzidenz neonataler Morbiditätsfaktoren, wurden zwischen Mai 2002 und März 2004 insgesamt 2256 Neugeborene Kinder registriert (2030 termingeborene Kinder und 226 Frühgeborene Kinder). Davon erhielten 1053 Reifgeborene einen Hüftgelenksultraschall zum Zeitpunkt der U2 (4.-6. Lebenstag), sowie seit Oktober 2003 auch Frühgeborene (n=45) im korrigierten Alter von 4-5 Tagen. Die sonographische Einteilung der Befunde erfolgte nach Graf. Bei 4,9% der untersuchten Kinder wurde ein therapiepflichtiger Befund (ab Typ IIc und höher nach Graf) diagnostiziert. Bei 3,2% der Kinder fand sich ein einseitiger, bei 1,7 % ein beidseitiger pathologischer Befund. Die Analyse der Geschlechtsverteilung zeigte ein signifikant gehäuftes Auftreten bei Mädchen (6,5% w / 3,4% m, p <0,023). Es ergaben sich keine signifikanten Unterschiede bezüglich betroffener Seite und intrauterinen Lagetyp. Analysen zur familiären Belastung wiesen eine nicht-signifikante mütterliche Prädisposition (p<0.083) aus. Signifikanter Risikofaktor für eine therapiepflichtige Hüftgelenksdysplasie ist das weibliche Geschlecht. Daneben findet sich intrafamiliär eine durch die Mutter vermittelte Prädisposition. Der frühzeitige Nachweis einer therapiepflichtigen Hüftdysplasie (zum Zeitpunkt U2) in Kenntnis relevanter Risikofaktoren erbringt möglicherweise einen prognostischen Vorteil für das Behandlungsergebnis. Prospektive Untersuchungen auf Basis größerer Fallzahlen sind erforderlich.
Ziel: Das Ziel der Studie ist es, den Zusammenhang von maternalem Diabetes und fetalem und maternalem Schwangerschaftsoutcome zu beschreiben. Methode: Insgesamt wurden n=4593 Mütter und ihre Kinder in der populationsbasierten SNiP Studie, Survey of Neonates in Pomerania eingeschlossen. Die Datenerhebung erfolgte im Zeitraum vom 05/2002 – 11/2008 in Universitäts- und Kreiskrankenhäusern im Nordosten von Mecklenburg-Vorpommern durch face-to-face Interviews, Fragebögen und Einsicht in die Krankenakten und Labordaten. Der Vergleich erfolgte zwischen Diabetes-erkrankten Müttern mit präexistenter oder sich erstmanifestierender Blutglukoseintoleranz (n=243), die klinisch an Hand des oralen Glukosetoleranztest diagnostiziert sind und Non-Diabetes-erkrankten Müttern (n=4350). Ergebnisse: Die Prävalenz für eine maternale Blutglukoseintoleranz beträgt 5,3%. Als Komorbiditäten mit der maternalen Blutglukoseintoleranz sind ein höheres Alter, eine höhere Anzahl von vorausgegangenen Schwangerschaften (Parität der Mutter) und Übergewichtigkeit (Relative Risiko 1,97) assoziiert. Als fetale Risikoparameter kann eine gehäufte Makrosomie (OR=2,28) aufgezeigt werden. Als maternaler Risikoparameter zeigt sich ein gehäuft positiver Nachweis vaginaler Erreger (OR=1,97). Als negativer Outcome zeigt sich eine insgesamt gesteigerte Sectiorate und Einsatz geburtsmechanischer unterstützender Maßnahmen (OR=1,96) und eine Häufung pathologischer Geburtslagen (15,3%Diabetes/9,3%kein Diabetes) beim Kind, sowie ein nahezu viermal so hohes Risiko einer postnatalen stationären Aufnahme des Neugeborenen (OR=3,70) und Notwendigkeit einer präpartalen ärztlichen Behandlung (OR=4,78) bei der Mutter mit diabetischer Stoffwechsellage. Bei einem Drittel (32,2%) der Neugeborenen diabetischer Mütter findet sich eine Hypoglykämie gegenüber 2,3% der übrigen Neugeborenen. Trotz erhöhter Sectio-Rate findet sich keine erhöhte Inzidenz von Atemstörungen bei den Neugeborenen diabetischer Mütter. Keine Unterschiede finden sich beim Apgar, arteriellen Nabelschnur-pH und der Inzidenz einer Hyperbilirubinämie. Fazit: In der SNiP-Kohorte liegt die Prävalenz des Diabetes in der Schwangerschaft klar im oberen Bereich der in der Literatur berichteten Häufigkeiten. Der Gestationsdiabetes ist mit erhöhten Raten pathologischer Geburtslagen, Kaiserschnittentbindungen und neonataler Makrosomie assoziiert. Das Risiko für eine stationäre Behandlung ist für das Neugeborene einer diabetischen Mutter um den Faktor 4 erhöht. Die Hauptursache liegt in dem Auftreten neonataler Hypoglykämien bei einem Drittel dieser Kinder. Diese Ergebnisse zeigen, dass der Diabetes in der Schwangerschaft weiterhin, auch bei verbesserter Therapie und Überwachung, eine relevante Risikoerhöhung für Mutter und Kind mit sich bringt.
Ziel der vorliegenden Untersuchung war die Entwicklung und Validierung eines pla-nimetrischen MRT-Messprotokolls zur Erhebung von Normwerten für das menschli-che Bulbusvolumen und die Sulcustiefe in der deutschen Bevölkerung. Neben der Häufigkeitsangabe zum Vorkommen von Bulbusaplasien wurden ferner Seitendis-krepanzen sowie eine mögliche Beeinflussung durch das Alter und Geschlecht un-tersucht. Aus vorhergehenden Untersuchungen ist bekannt, dass das Bulbusvolu-men und die Sulcustiefe in direktem Zusammenhang mit dem Riechvermögen ste-hen. Aufgrund der starken Prävalenz von Riechstörungen in der Bevölkerung wurden die Strukturen weiterhin auf einen möglichen morphologischen Zusammenhang mit dem Vorkommen von Stirn- und Kieferhöhlenverschattungen analysiert.
Für die Auswertung mittels der Bildbetrachtungssoftware OsiriX standen 250 MRT-Datensätze der SHIP-Studie zur Verfügung.
Mit Volumina von 10,8 mm³ bis 251,2 mm³ und einer Standardabweichung von 44,9 mm³ erwies sich der BO als äußerst variabel. In zwei Fällen konnte eine linksseitige Aplasie beobachtet werden. Die Sulcustiefe unterlag mit Messwerten von 0,14 mm bis 13,87 mm und einer SD von 1,71 mm ebenso großen Schwankungen. Die ermit-telten Normwerte des BO (104,6 mm³, SD = 44,9 mm³) und des SO (8,39 mm, SD = 1,71 mm) sind vergleichbar mit den Ergebnissen aus Untersuchungen von Bur-meister et. al und Rombaux et al. (Burmeister et al. 2011d; Rombaux et al. 2010). Die statistische Analyse zeigte, dass das Bulbusvolumen unabhängig von Seite, Alter und Geschlecht ist. Auf die Sulcustiefe dagegen wirken mehrere Einflussfaktoren. Der SO auf der linken Seite war um durchschnittlich 0,47 mm signifikant flacher als auf der rechten Seite. Zudem konnte mit zunehmendem Alter eine Abflachung der Sulcustiefen nachgewiesen werden. Männer verfügten über durchschnittlich 0,71 mm signifikant flachere Sulci als Frauen. Ferner wurde untersucht, ob Stirn- und Kiefer-höhlenverschattungen als Einflussfaktoren auf den SO und BO wirken. Männer wie-sen signifikant häufiger Verschattungen der Stirnhöhlen auf als Frauen. Auf das Bul-busvolumen bewirkten Verschattungen jedoch keine Veränderung.
Mit der vorliegenden Arbeit wurde eine standardisierte Methode vorgestellt, die es ermöglicht, den BO und SO quantitativ zu analysieren. Die Tiefenmessung des SO erwies sich als reliabel und ist geeignet um im Klinikalltag angewendet zu werden. Ebenso wie die Tiefenmessungen des SO in einer definierten Ebene erfolgten, wur-den für die planimetrischen Bulbusmessungen die Lage und Begrenzung des BO präzise beschrieben. Das hier entwickelte Messprotokoll kann als Anleitung für wei-tergehende Untersuchungen dienen. Perspektivisch kann zur Erleichterung und Ver-kürzung der aufwendigen und zeitintensiven Vermessungen des BO das vorgestellte Messprotokoll als Grundlage für eine semiautomatische Methodik genutzt werden. Außerdem kann geprüft werden, ob die Messungen in 3T-MRTs und in T1- und T2-Wichtung leichter fallen.
Die Vergleiche mit anderen Arbeiten lassen aufgrund der Probandenauswahl, deren geringeren Probandenanzahl und der abweichenden Vorgehensweisen bei den Mes-sungen nur bedingt Schlussfolgerungen zu. Daraus resultierend existieren bisher keine einheitlichen Normwerte für den BO und SO. Aus den Ergebnissen der vorlie-genden Arbeit ergeben sich neue Anreize für künftige Studien, die sich mit phyloge-netischen Aspekten befassen könnten. Weiterhin sollte der Frage nachgegangen werden, ob bei neurodegenerativen, psychischen und internistischen Krankheiten abweichende Werte für den BO oder SO beobachtet werden können und inwiefern das Maß der Abweichung als prognostisches Instrument genutzt werden kann. An-hand der vorliegenden Untersuchung kann zudem keine Aussage getroffen werden, inwiefern sich symptomatische Verschattungen der Stirn- und Kieferhöhlen von asymptomatischen Verschattungen hinsichtlich des Bulbusvolumens und der Sulcus-tiefe unterscheiden. Weiterführende Studien sollten deshalb neben der klinischen Relevanz auch den Zustand der gesamten Nasennebenhöhlen, insbesondere der Siebbeinzellen, betrachten, um zu überprüfen, ob dortige Veränderungen den BO oder SO beeinflussen.
Weltumspannende Krisen, die alle signifikanten Wirtschaftsnationen vollständig erfassen, sind eher selten. Vielmehr blieben in der Vergangenheit schwerwiegende Krisen überwiegend auf einen Staat oder Region begrenzt. Mit Ausbruch der Finanz- und Wirtschaftskrise in den Vereinigten Staaten kam es zur sukzessiven Ansteckung entwickelter und aufstrebender Volkswirtschaften. Als Reaktion darauf senkte die US Zentralbank die Federal Fund Rate von 4,75% im September 2007 auf 0-0,25% im Dezember 2008. Zur Unterstützung konventioneller geldpolitischer Maßnahmen wurde in den USA verstärkt auf Forward Guidance und zusätzlich das Large Scale Asset Purchase Program als unkonventionelle Maßnahme gesetzt. In ähnlicher Weise reagierten die Zentralbanken des europäischen Währungsraums, Großbritanniens und Japans. Während die expansive Geldpolitik in den entwickelten Volkswirtschaften eine sukzessive Zinssenkung bis zum jetzigen Niedrigzinsniveau auslöste, blieb in den Schwellenländern der G20 das Zinsniveau zunächst unverändert und resultierte in einem Anstieg der Zinsdifferenz zwischen den Ländergruppen. Infolgedessen verstärkten Investoren aus den entwickelten Volkswirtschaften bei der Suche nach zinstragenden Aktiva ihr Engagement in den Schwellenländern der G 20. Gleichzeitig veränderte sich als Reaktion auf die Finanzkrise die Risikowahrnehmung der Investoren aus entwickelten Volkswirtschaften auf unerwartete Informationen und negative Preisentwicklungen. Eine Umkehr der Zinsentwicklung in Verbindung mit einer Stagnation bzw. Rezession in den entwickelten Volkswirtschaften kann Kapitalabzug aus den Schwellenländern der G 20 auslösen. Der Einsatz von weitreichenden Kapitalverkehrskontrollen zur Abschwächung oder Verhinderung des Kapitalabzuges durch die betroffenen Staaten kann aufgrund krisenbedingter Preisverfälle zu schweren Verlusten der Investoren und erhebliche Auswirkungen auf die Finanzmärkte der entwickelten Volkswirtschaften besitzen.
Chronic alcohol abuse is one of the most common addictions and one of the most substantial public health problems as it affects millions of people physically as well as mentally around the world. Globally more than 3 million deaths are assignable to alcohol intake each year. Chronic alcoholism is a multi-component disease and its development is associated with both environmental as well as genetic factors. However, the key mechanisms underlying an addiction, especially on a cellular and physiological basis, are still unknown. Bio-medically an influence of chronic alcohol consumption on synaptic plasticity in the brain of humans as well as rodents has been proven.
On the dendritic shaft of nervous brain cells, small membrane protrusions called dendritic spines can be found. These spines possess the capacity to change their morphology and quantity and are thought to play an important role in learning and memory forming, and seem to be impaired in multiple neurological disorders. These dynamics are called synaptic plasticity. Most of these studies however, were carried out on the cortex. These previous observations raise the question whether such alterations in synaptic plasticity can also be observed in regions of the brain that contribute to the limbic system and therefore to the processing of emotional responses, learning and decision making. The amygdala is of special interest when trying to understand the neurobiology and pathophysiology that lead to the emergence and up keeping of an alcohol addiction. In this thesis a closer look has been taken at possible alterations in synaptic plasticity within different amygdaloid nuclei by the help of a rat model. These rats were put into the so called postdependent state, one of the most common animal models to investigate excessive ethanol intake in rodents. The postdependent state is a model in which the key driving force to obtain alcohol as part of a preserved addiction cycle is based on negative affect. Studies showed differences in the behavioural outcome of those animals that were exposed to chronic intermittent alcohol consumption compared to a control group, so it was of special interest to see whether those behavioural changes also show on a cellular basis.
In the study, a morphological comparison of the spine length as well as the spine density of alcohol dependent rats with a comparable control group has been made. The medial, the central, the lateral and the basolateral amygdaloid nucleus were of special interest in this research project.
The results showed no significant difference of the spine densities in any of the four amygdaloid regions. When comparing the spine morphology within the ethanol and the control group, differences showed in the lateral amygdaloid nucleus. In this region the spines of the ethanol group were significantly smaller. This leads to the conclusion that chronic alcohol intake can have an influence on the spine morphology and hence alter anatomical brain structures.
Objective
Congenital hyperinsulinism (CHI) is a rare disease characterized by persistent hypoglycemia as a result of inappropriate insulin secretion, which can lead to irreversible neurological defects in infants. Poor efficacy and strong adverse effects of the current medications impede successful treatment. The aim of the study was to investigate new approaches to silence β-cells and thus attenuate insulin secretion.
Research Design and Methods
In the scope of our research, we tested substances more selective and more potent than the gold standard diazoxide that also interact with neuroendocrine ATP-sensitive K+ (KATP) channels. Additionally, KATP channel-independent targets as Ca2+-activated K+ channels of intermediate conductance (KCa3.1) and L-type Ca2+ channels were investigated. Experiments were performed using human islet cell clusters isolated from tissue of CHI patients (histologically classified as pathological) and islet cell clusters obtained from C57BL/6N (WT) or SUR1 knockout (SUR1-/-) mice. The cytosolic Ca2+ concentration ([Ca2+]c) was used as a parameter for the pathway regulated by electrical activity and was determined by fura-2 fluorescence. The mitochondrial membrane potential (ΔΨ) was determined by rhodamine 123 fluorescence and single channel currents were measured by the patch-clamp technique.
Results
The selective KATP channel opener NN414 (5 µM) diminished [Ca2+]c in isolated human CHI islet cell clusters and WT mouse islet cell clusters stimulated with 10 mM glucose. In islet cell clusters lacking functional KATP channels (SUR1-/-) the drug was without effect. VU0071063 (30 µM), another KATP channel opener considered to be selective, lowered [Ca2+]c in human CHI islet cell clusters. The compound was also effective in islet cell clusters from SUR1-/- mice, showing that [Ca2+]c is influenced by additional effects besides KATP channels. Contrasting to NN414, the drug depolarized ΔΨ in murine islet cell clusters pointing to severe interference with mitochondrial metabolism. An opener of KCa3.1 channels, DCEBIO (100 µM), significantly decreased [Ca2+]c in SUR1-/- and human CHI islet cell clusters. To target L-type Ca2+ channels we tested two already approved drugs, dextromethorphan (DXM) and simvastatin. DXM (100 µM) efficiently diminished [Ca2+]c in stimulated human CHI islet cell clusters as well as in stimulated SUR1-/- islet cell clusters. Similar effects on [Ca2+]c were observed in experiments with simvastatin (7.2 µM).
Conclusions
NN414 seems to provide a good alternative to the currently used KATP channel opener diazoxide. Targeting KCa3.1 channels by channel openers or L-type Ca2+ channels by DXM or simvastatin might be valuable approaches for treatment of CHI caused by mutations of KATP channels not sensitive to KATP channel openers.
Ac(et)ylation is a post-translational modification present in all domains of life. First identified in mammals in histones to regulate RNA synthesis, today it is known that is regulates fundamental cellular processes also in bacteria: transcription, translation, metabolism, cell motility. Ac(et)ylation can occur at the ε-amino group of lysine side chains or at the α-amino group of a protein. Furthermore small molecules such as polyamines and antibiotics can be acetylated and deacetylated enzymatically at amino groups. While much research focused on N-(ε)-ac(et)ylation of lysine side chains, much less is known about the occurrence, the regulation and the physiological roles on N-(α)-ac(et)ylation of protein amino termini in bacteria. Lysine ac(et)ylation was shown to affect protein function by various mechanisms ranging from quenching of the positive charge, increasing the lysine side chains’ size affecting the protein surface complementarity, increasing the hydrophobicity and by interfering with other post-translational modifications. While N-(ε)-lysine ac(et)ylation was shown to be reversible, dynamically regulated by lysine acetyltransferases and lysine deacetylases, for N-(α)-ac(et)ylation only N-terminal acetyltransferases were identified and so far no deacetylases were discovered neither in bacteria nor in mammals. To this end, N-terminal ac(et)ylation is regarded as being irreversible. Besides enzymatic ac(et)ylation, recent data showed that ac(et)ylation of lysine side chains and of the proteins N-termini can also occur non-enzymatically by the high-energy molecules acetyl-coenzyme A and acetyl-phosphate. Acetyl-phosphate is supposed to be the key molecule that drives non-enzymatic ac(et)ylation in bacteria. Non-enzymatic ac(et)ylation can occur site-specifically with both, the protein primary sequence and the three dimensional structure affecting its efficiency. Ac(et)ylation is tightly controlled by the cellular metabolic state as acetyltransferases use ac(et)yl-CoA as donor molecule for the ac(et)ylation and sirtuin deacetylases use NAD+ as co-substrate for the deac(et)ylation. Moreover, the accumulation of ac(et)yl-CoA and acetyl-phosphate is dependent on the cellular metabolic state. This constitutes a feedback control mechanism as activities of many metabolic enzymes were shown to be regulated by lysine ac(et)ylation. Our knowledge on lysine ac(et)ylation significantly increased in the last decade predominantly due to the huge methodological advances that were made in fields such as mass-spectrometry, structural biology and synthetic biology. This also includes the identification of additional acylations occurring on lysine side chains with supposedly different regulatory potential. This review highlights recent advances in the research field. Our knowledge on enzymatic regulation of lysine ac(et)ylation will be summarized with a special focus on structural and mechanistic characterization of the enzymes, the mechanisms underlying non-enzymatic/chemical ac(et)ylation are explained, recent technological progress in the field are presented and selected examples highlighting the important physiological roles of lysine ac(et)ylation are summarized.
Summary
Streptococcus pneumoniae (the pneumococcus), a bacterium belonging to the normal flora in the human respiratory tract, continues to be an important pathogen due to its contribution to morbidity and mortality among children, the elderly, and immunocompromised persons. Global estimates of pneumococcal deaths among children declined by 51% between 2000 and 2015. This achievement was mainly due to the introduction of pneumococcal conjugate vaccines (PCVs) in countries with the highest pneumococcal burden. Since May 2012, children in Ghana have been receiving PCV vaccination as part of routine immunization. The continuous monitoring of the pneumococcus after PCV introduction is essential to understand the changing epidemiology of the pathogen in the population.
This study therefore, aims to determine the (1) prevalence, serotypes, and sequence types of pneumococcal isolates, (2) antibiotic susceptibility patterns and the genetic basis for the antibiotic resistance among these pneumococcal isolates, and (3) prevalence of selected virulence genes that have been identified as potential vaccine candidates. Nasopharyngeal swabs were obtained from vaccinated children under five years of age in Cape Coast, Ghana. Six years after PCV implementation, we provide data on the epidemiology of pneumococcal strains circulating among children in Cape Coast Ghana. Standard microbiological and molecular techniques were used to identify and characterize the pneumococcal strains.
Overall, pneumococcal carriage prevalence was 29.4% (151/513). All participating children were fully vaccinated. Of the 26 different serotypes identified, the top five PCV13 serotypes (VT) were 6B, 23F, 19F, 3, 6A and non-PCV13 vaccine serotypes (NVT) were 23B, 13, 11A, 15B, and 34. PCV13 coverage was 38.4%, however, more than half of the isolates were NVT with a coverage rate of 61.6%. The isolates were highly susceptible to levofloxacin, ceftriaxone, vancomycin, and erythromycin. However, marked resistance to cotrimoxazole and tetracycline was observed. The reduction in penicillin resistance (35.8%) as compared to pre-vaccination data (45% - 63%) suggests an attributable effect from PCV13 vaccination. However, penicillin resistance was also detected in some NVT serotypes. Overall, 28.5% of the isolates resistant to three or more different classes of antibiotics were classified as multidrug-resistant (MDR). To analyze the genetic basis for resistance to penicillin, erythromycin and tetracycline, pbp2b, ermB, mefA, and tetM genes were amplified.
Thirty-eight (70%) out of the 54 penicillin-resistant isolates contained the pbp2b resistance gene. Out of the 11 erythromycin-resistant isolates, 7 (63.6) and 4 (36.4%) were positive for the ermB and mefA genes, respectively. The tetM gene was detected in 85 (98.8%) of the 86 tetracycline resistance isolates.
To determine the extent to which potential protein-based vaccines could be protective in Ghanaian children, we sought to determine the prevalence of selected virulence genes among the isolates. The lytA, pavB, and cpsA genes were present in all the carrier isolates. However, psrP, pcpA, pilus islet (PI) PI-1, and PI-2 were present in 62.7%, 87.5%, 11.8%, and 6.5% of the strains, respectively. The psrP and pcpA virulence genes were evenly distributed among all the serotypes. However, the pilus islets were detected in only seven serotypes namely 19F, 6B, 9V, 6A, 13, 11A, and 23B. Five serotype 19F isolates possessed both PI-1 and PI-2. Furthermore, the pilus islets were associated with multidrug resistance.
The predominant NVT serotype 23B and isolates resistant to ≥ 4 antibiotics were analysed by multilocus sequence typing (MLST). Nine known sequence types (STs) and 10 novel STs were identified. Seven out of the 10 new STs belonged to serotype 23B, while the remaining 3 were VTs 6B and 19F. A capsular switch was identified among isolates of ST802, which comprised of both serotype 23F and 19F. The majority of serotype 23B strains belonged to ST172. The ST172 is associated with serotype 23F and a single locus variant (SLV) of internationally disseminated clone ST338 (Colombia23F-26). Consequently, ST172 was characterised with marked antibiotic resistance and with traits of capsular switching. One serotype 6B strain was identified as a SLV of ST273 (Greece6B-22) while two serotype 9V strains belonged to the internationally disseminated clone ST156 (Spain9V-3).
In conclusion, this study showed a marginal decline in overall pneumococcal carriage prevalence, persistence of VTs despite the increase in NVTs, and the occurrence of serotype replacement and capsular switching. In addition, sequence types related to internationally disseminated clones are circulating in Ghana. With the high pcpA and psrP coverage detected,including these genes in protein-based vaccines could provide adequate protection for Ghanaian Children.
Background: Although 20-30% of all strokes occur in the posterior circulation, few studies have explored the characteristics of patients with strokes in the posterior compared to the anterior circulation so far. Especially data on young patients is missing. Methods: In this secondary analysis of data of the prospective multi-centre European sifap1 study that investigated stroke and transient ischemic attack (TIA) patients aged 18-55 years, we compared vascular risk factors, stroke aetiology, presence of white matter hyperintensities (WMH) and cerebral microbleeds (CMB) between patients with ischaemic posterior circulation stroke (PCS) and those having suffered from anterior circulation stroke (ACS) based on cerebral MRI. Results: We diagnosed PCS in 612 patients (29.1%, 407 men, 205 women) and ACS in 1,489 patients (70.9%). Their age (median 46 vs. 47 years, p = 0.205) and stroke severity (modified Rankin Scale: both 2, p = 0.375, Barthel Index 90 vs. 85, p = 0.412) were similar. PCS was found to be more frequent among the male gender (66.5 vs. 60.1% with ACS, p = 0.003). Vertebral artery (VA) dissection was more often the cause of PCS (16.8%) than was carotid artery dissection of ACS (7.9%, p < 0.001). Likewise, small vessel disease (Trial of Org 10172 in Acute Stroke Treatment [TOAST] = 3, PCS: 14.7%, ACS: 11.8%) and stroke of other determined aetiology (TOAST = 4, PCS: 24.5%, ACS: 16.0%) were more frequent in those with PCS. Furthermore, patent foramen ovale (PFO; PCS: 31.1%, ACS: 25.4%, p = 0.029) was more often detected in patients with PCS. In contrast, large-artery atherosclerosis (TOAST = 1, PCS: 15.4%, ACS: 22.2%) and cardio-embolic stroke (TOAST = 2, PCS: 15.6%, ACS: 18.0%) were less frequent in those with PCS (p < 0.001) as were preceding cerebrovascular events (10.1 vs. 14.1%, p = 0.014), TIA (4.8 vs. 7.7%, p = 0.016) and smoking (53.2 vs. 61.0%, p = 0.001). The presence, extent, and location of WMH and CMB did not differ between the 2 groups. Conclusions: Our data suggested a different pattern of aetiology and risk factors in young patients with PCS compared to those with ACS. These findings especially call for a higher awareness of VA dissection and potentially for more weight of a PFO as a risk factor in young patients with PCS. The structure and content of the training phase following completion of medical school, referred to in most countries as postgraduate medical training, varies between countries. The purpose of this article is to give national and international readers an overview of the organisation and structure of postgraduate medical training in Germany.
The content and duration of postgraduate training in Germany are stipulated by state medical boards, officially termed associations (Landesärztekammer). In a periodically updated decree, the federal German medical association (Bundesärztekammer) provides a template for postgraduate medical training structure (Musterweiterbildungsordnung), which is adapted by the state medical associations. Admission to postgraduate medical training in Germany takes place by way of open, free-market selection. Based on the traditional assumption that junior doctors acquire all necessary clinical skills “on the job”, formal education in the form of seminars, lectures, or preorganised, detailed rotation plans through various specialties or wards is largely absent. Requirements for postgraduate medical training focus on the fulfilment of broad categories of rotations rather than specific content or gaining competencies. With few exceptions, no structured educational programs with curricular learning objectives exist. Limited funding impedes program development and expansion. Junior doctors bear the primary organisational responsibility in their training, which often results in extended training times and dissatisfaction. Structured training programs which prioritise skill-building and formal education are needed to support junior doctors and ensure their competence in primary and specialty care.
Postoperative Immune Suppression in Visceral Surgery: Characterisation of an Intestinal Mouse Model

Background: Postoperatively acquired immune dysfunction is associated with a higher mortality rate in case of septic complications. As details of this severe clinical problem are still unknown, animal models are essential to characterise the mechanisms involved. Methods: Mice were laparotomised and the small intestine was pressed smoothly in antegrade direction. For extension of trauma, the intestine was manipulated three times consecutively. Following this, the ex vivo cytokine release of splenocytes was determined. The degree of surgical trauma was analysed by detection of HMGB1 and IL-6 in serum and by neutrophil staining in the muscularis mucosae. Results: We adapted the previously described animal model of intestinal manipulation to provide a model of surgically induced immune dysfunction. Following intestinal manipulation, the mice showed elevated serum levels of HMGB1 and IL-6 and increased infiltration of granulocytes into the muscularis mucosae. Ex vivo cytokine release by splenocytes was suppressed in the postoperative period. The degree of suppression correlated with the extent of surgical trauma. Conclusions: In this study, we describe a surgically induced immune dysfunction animal model, in which a significant surgical trauma is followed by an immune dysfunction. This model may be ideal for the characterisation of the postoperative immune dysfunction syndrome.
Postoperative Therapien, rezidivfreies Intervall und Gesamtüberleben von uterinen Adenosarkomen

Uterine Adenosarkome (AS) sind eine sehr seltene Variante der uterinen Sarkome. Sie bestehen aus einer benignen, epithelialen und einer malignen, mesenchymalen Komponente. Ein wichtiges histopathologisches Merkmal ist das sarcomatous overgrowth (SO), welches bei den für gewöhnlich low-grade Tumoren mit einer aggressiveren Verlaufsform assoziiert ist. Aktuell existieren für Deutschland praktisch keine Daten zu wirksamen Therapieregimen und zum Langzeitüberleben.
In diese Studie konnten 53 AS, eine für diese Entität hohe Fallzahl, aus der Bundesrepublik Deutschland eingebunden werden. Die Datenerhebung erfolgte gemischt retro- und prospektiv über neun Jahre. Mittels der Kaplan-Meier-Methode und des multivariaten Cox-Regression-Modells wurden mögliche Prognosefaktoren für das rezidivfreie Intervall (RFI) und das Gesamtüberleben (OS) identifiziert. Zum Vergleich der einzelnen Überlebenskurven wurde der log-Rank-Test verwendet. Das Signifikanzniveau aller Tests lag bei p=0,005.
Von den 53 beobachteten AS konnten 33 (62,3%) als AS ohne SO und 20 (37,7%) als AS mit SO (AS-SO) diagnostiziert werden. Unser medianes Follow-up beträgt 19 Monate (Spanne 1-144 Monate). Insgesamt verstarben elf Frauen (20,8%), wobei in neun Fällen das AS todesursächlich war. Es wurde ein medianes OS von 68 Monaten bzw. ein tumorspezifisches Überleben von 34 Monaten ermittelt. Das SO ist sowohl hinsichtlich des OS (p=0,195) als auch des tumorspezifischen Gesamtüberlebens (p=0,083) mit einer schlechteren Prognose assoziiert. Während das 1- und 2- Jahresüberleben beider Gruppen sehr ähnlich liegt, ist das Langzeitüberleben nach fünf Jahren für AS-SO drastisch reduziert, sodass insbesondere hier eine lange Nachbeobachtungszeit erforderlich ist.
Insgesamt entwickelten 15 Frauen (28,3%) nach median 79 Monaten (95%-KI) ein Rezidiv, welches einer Rezidivrate von 27,3% (n=9) für AS ohne SO und 30% (n=6) für AS-SO entspricht. Zudem kann näherungsweise ein negativer Einfluss für das Vorliegen eines SO auf das RFI registriert werden (p=0,098).
Eine R1/2-Resektion und eine G3-Klassifizierung sind mit einem signifikant schlechteren Outcome (RFI und OS) verbunden. Zudem zeigen AS mit einem Ki67-Proliferationsindex ≥ 30 einen Trend früher Rezidive zu entwickeln (p=0,164). Prognostisch vorteilhaft, mit längerem RFI und OS, ist das Fehlen eines Lymphknotenbefalls zu werten. Jedoch führt eine systematische Lymphonodektomie nicht zu einem Überlebensvorteil. Im Gegenteil: Die Lymphonodektomie führt sogar zu einem 5,6-fach höheren Sterberisiko als ein Unterlassen der selbigen. Eine BSO erbringt ebenfalls keinen Überlebensvorteil.
Zusammenfassend ist die Therapie der Wahl die totale Hysterektomie. Eine R0-Resektion ist bei ausgedehnten AS anzustreben. Eine Lymphonodektomie bzw. bilaterale Salpingoophorektomie hingegen erbringen keinen prognostischen Vorteil. Grundsätzlich führt die generelle Anwendung einer postoperativen Therapie zu keiner Verbesserung des RFI und OS. Bei ausgeprägtem postoperativen Therapiewunsch oder fortgeschrittenen Stadien kann am ehesten eine Radiotherapie zur Reduktion von Lokalrezidiven angewendet werden.
Sieben (n=7/15; 46,6%) Frauen mit einem Rezidiv verstarben (medianes Überleben 64 Monate), wobei auch hier das prognostisch schlechtere AS-SO mit einem kürzeren Überleben einhergeht. Prozentual wurden mehr Fernmetastasen (60,9%) gegenüber Lokalrezidiven (39,1%) registriert, wobei am häufigsten eine Fernmetastasierung in die Lunge stattfand. Die Rezidivlokalisation zeigt keinen Einfluss auf das Outcome der Frauen. Analog zur Primärtherapie ist das maximale Tumordebulking mit einer Resektion aller Tumorherde die beste Rezidivtherapie. Multimodale Therapiekonzepte, die ein maximales Tumordebulking beinhalten, erzielen ähnlich gute Ergebnisse. Chemotherapeutika waren nur in Einzelfällen wirksam und haben insgesamt den gleichen Effekt wie eine reine Beobachtung. Häufig musste eine Chemotherapie aufgrund der Toxizität und starken Nebenwirkungen, bei meist unter Therapie progredienter Krankheit, abgebrochen werden. Die längste partielle Remission konnte mit einer Kombination aus Carboplatin und Paclitaxel bei einer Patientin mit AS ohne SO für 13 Monate erreicht werden.
Zukünftig werden noch größere Studien benötigt um konsequente Therapiestrategien, insbesondere postoperative Therapien nach inadäquat erfolgter Operation, zur Verbesserung des OS und des RFI abzuleiten oder zu entwickeln. Um auch verlässlichere Aussagen bezüglich des Rezidivverhaltens und –therapie machen zu können, muss ein langes Follow-up möglichst vieler Frauen gewährleistet werden. Insbesondere sollte auch ein stadienabhängige Betrachtung der einzelnen Therapieverfahren erfolgen, um einen besseren Vergleich zu ermöglichen.
Mit dem Ziel, eine evidenzbasierte Grundlage für die Rehabilitation nach VKB-Ersatz zu erarbeiten, wurde eine Fragebogenumfrage unter erfahrenen Kreuzbandchirurgen durchgeführt. Neben der Auswertung der Antworten wurden die einzelnen Nachbehandlungsansätze mit der Literatur in Vergleich gestellt. Für die gängigen und bekanntesten Nachbehandlungsmodule herrscht eine weitgehende Übereinstimmung zwischen Praxis und Empfehlung in der Literatur. Bei den eher unüblicheren Modulen konnte kein Trend abgeleitet werden bzw. war die Antwortquote zu niedrig oder die Verfahren den Teilnehmern zu unbekannt. Allerdings wird auch die Schwierigkeit deutlich, auf evidenzbasierter Grundlage Ergebnisse zu erhalten. Eine weitere Schwierigkeit besteht vor allem darin, viele einzelne Variablen unter Studienbedingungen mit ausreichender statistischer Aussagekraft auf ihre Effizienz hin zu untersuchen, da individuelle Aspekte der Patienten und patientenspezifische operationstechnische Verfahren zu berücksichtigen sind. Unbestritten bleibt, dass für die meisten operativen Therapieverfahren die Nachbehandlung für den Gesamterfolg sehr bedeutsam ist. Daher ist es erstrebenswert und notwendig, das Ziel einer evidenzbasierten Grundlage für Rehabilitationskonzepte nach VKB-Ersatzplastik und anderen operativen Therapien zu verfolgen. Im Hinblick auf die erheblichen Kosten im Gesundheitssystem sollten die Effekte der verschiedenen Rehabilitationsmaßnahmen nachhaltig überprüft werden.
Die Bedeutung des (postoperativen) Schmerzes sowie dessen adäquate Behandlung nimmt im klinischen Alltag eine zentrale Stellung ein.
Zur Verbesserung der Schmerztherapie wurde im Juli 2009 das Akutschmerzzertifizierungsprogramm „Qualitätsmanagement Akutschmerz“ in der Universitätsmedizin Greifswald eingeführt. Die Intention dieser Maßnahme war, mit Hilfe der Implementierung der S3-Leitlinie zur „Behandlung akuter perioperativer und post- traumatischer Schmerzen“ das Schmerzmanagement zu optimieren. Dies sollte durch folgende Grundpfeiler erreicht werden: Durch die Zunahme der interdisziplinären Zusammenarbeit seitens aller am Schmerzmanagement beteiligten Fachrichtungen, der Einführung von standardisierten verfahrensspezifischen Behandlungsalgorithmen mit definierten Interventionsgrenzen, einer umfangreichen Schmerzanamnese, -erfassung und -dokumentation sowie der Weiterbildung der am Schmerzmanagement beteiligten Berufsgruppen.
Ziel der vorliegenden Arbeit war es, die postoperative Schmerzsituation am ersten Tag nach einer Wirbelsäulenoperation vor und nach Einführung des „Qualitätsmanagements Akutschmerz“ in der Klinik und Poliklinik für Orthopädie und orthopädische Chirurgie der Universitätsmedizin Greifswald zu untersuchen und zu prüfen, ob eine Verbesserung des postoperativen Schmerzmanagements durch die Einführung des Zertifizierungsprogramms „Qualitätsmanagement Akutschmerz“ gelungen ist. Des Weiteren wurde analysiert, ob Faktoren wie Operationsart, Geschlecht, Rauchen, psychische Komorbiditäten und chronische Schmerzen das postoperative Schmerzempfinden nach Eingriffen an der Wirbelsäule beeinflussen.
Hierzu wurden die Daten retrospektiv von 1322 Patienten ausgewertet, die sich im Zeitraum vom 01.01.2008 bis zum 31.12.2013 in der Klinik und Poliklinik für Orthopädie und orthopädische Chirurgie der Universitätsmedizin Greifswald einer Wirbelsäulenoperation unterzogen hatten. Zur Evaluierung des postoperativen Schmerzempfindens wurde die Schmerzstärke (n=893) mittels Numerischer Ratingskala (NRS) und die Zufriedenheit mit der aktuellen Schmerzsituation (Schmerzzufriedenheit) (n=438) anhand eines 5-stufigen Skalierungssystems erhoben.
In dem untersuchten Zeitraum konnte eine kontinuierliche Verbesserung der postoperativen Schmerzstärke festgestellt werden. Die Ergebnisse bezüglich der postoperativen Schmerzzufriedenheit zeigten, abgesehen von einer einmaligen Verbesserung zwischen 2009 und 2010 zum Zeitpunkt der Einführung des „Qualitätsmanagements Akutschmerz“, eine stetige Verschlechterung der Schmerzzufriedenheit. Daraus lassen sich zwei Schlussfolgerungen ableiten:
1. Durch die Einführung des „Qualitätsmanagements Akutschmerz“ wurde eine Verbesserung des postoperativen Schmerzmanagements erreicht.
2. Die Schmerzstärke und Schmerzzufriedenheit sind nicht miteinander assoziiert.
Die Analyse der Faktoren, die möglicherweise Schmerzstärke und Schmerzzufriedenheit beeinflussen, ergab bei chronischen Schmerzpatienten eine signifikant höhere Schmerzstärke im Vergleich zu Patienten ohne chronische Schmerzen. Die Resultate der Schmerzzufriedenheit bezüglich chronischer Schmerzen zeigten keinen signifikanten Unterschied. Bei der Untersuchung der Operationsart zeigten sich signifikante Unterschiede mit der höchsten Schmerzstärke sowie geringsten Schmerzzufriedenheit bei offenen Operationen, gefolgt von perkutanen Eingriffen und kombinierten perkutanen Operationen mit Mini-open-Synovektomien. Bei den Ergebnissen hinsichtlich des Geschlechts, Rauchens sowie psychischer Komorbiditäten kristallisierten sich bei der Schmerzstärke und Schmerzzufriedenheit jeweils unterschiedliche Tendenzen heraus; es ließen sich jedoch keine signifikanten Unterschiede feststellen.
Die Ergebnisse bezüglich der Schmerzstärke im gesamten untersuchten Zeitraum und der Schmerzzufriedenheit zwischen 2009 und 2010 bestätigen die Empfehlungen der S3-Leitlinie zur „Behandlung akuter perioperativer und posttraumatischer Schmerzen“, dass eine optimale postoperative Schmerztherapie durch ihre Integration in ein multimodales perioperatives Gesamtkonzept erreicht wird. Dabei haben neben den oben genannten Grundpfeilern der Leitlinie die Integration des Patienten in das perioperative Schmerzmanagement und die Berücksichtigung dessen individueller Bedürfnisse einen hohen Stellenwert.
Die Einführung von standardisierten Behandlungsalgorithmen haben sich im klinischen Alltag bewährt. Allerdings zeigen die Ergebnisse an der Universitätsmedizin Greifswald, dass für chronische Schmerzpatienten zusätzlich eine individuell angepasste Schmerztherapie notwendig ist. Neben den positiven Ergebnissen hinsichtlich der Schmerzstärke im gesamten untersuchten Zeitraum, bedarf es zukünftig weiterer Untersuchungen, um der festgestellten abnehmenden Schmerzzufriedenheit ab 2010 entgegenzuwirken.
Mussel farming, compared to marine finfish aquaculture, represents an environmentally friendly alternative for a high quality protein source and can at the same time be a measure to remove excess nutrients in eutrophic areas. As such, it is considered as a promising “blue growth” potential and promoted within the European Union. To expand mussel aquaculture, new regions have to be considered because there are multiple marine usages, and spatial limitations occur in coastal areas. The brackish Baltic Sea might be considered for expansion of mussel aquaculture. This study focusses on estimated production potential, economic profitability and nutrient remediation potential of mussel farming at different salinities. Four experimental mussel farms were set up along the German Baltic coast at salinities ranging from 7 to 17 psu. Collected growth data was used to calibrate and validate a Dynamic Energy Budget model and to predict the potential mussel production at 12 sites along the German coast. The estimated production and nutrient removal was used to assess economic profitability, assuming two usages of the harvest: human consumption and mussel meal production. Measured mussel specific growth rates increased with salinity from 0.05 mm d–1 in Greifswald Bay to 0.11 mm d–1 in Kiel Fjord. Within 6 months, a 1-ha farm could produce from 1 t (Darss-Zingst-Bodden-Chain) to 51 t (Flensburg) fresh mussels and remove 1.1 to 27.7 kg P and 24.7 to 612.7 kg N, respectively. Mussel farms at sites west of Rostock at salinities >10 psu could produce 5 cm mussels within 18 months, but only farms at Flensburg, Eckernförde and Kiel Fjord became profitable at a farm size of 4 ha (160,000 m3) at current market prices of 2.2 € kg–1. Regardless of the farm size, none of the farm sites could operate profitable if fresh mussels were sold for animal feeding at sales price of 0.06 € kg–1. Yearly nutrient removal costs at a small-scale farm (1 ha) ranged between 162 € (Flensburg) and 4,018 € (Darss-Zingst-Bodden-Chain) kg–1 nitrogen, and 3,580 € and 88,750 € kg–1 phosphorus, respectively.
In times of recent climate change, mechanisms to deal with different environments (e.g. via dispersal to other habitats, or via in-situ responses such as genetic adaptation or phenotypic plasticity) are essential. In regions showing seasonality, organisms are already adapted to regular and, thus, often predictable environmental changes. One well-known strategy to survive periods of food shortage, especially during the winter, is hibernation. Although hibernation is already an adaptation to overcome unfavourable conditions, the optimal timing of hibernation to match for example food abundance peaks is likely to be influenced by changing climatic conditions, as expected during human-induced global change. Thus, the ability to respond to changes in optimal timing of hibernation can be crucial for organisms. All hibernators are positioned at the slow end of the slow-fast life history continuum. Longevity combined with a low annual reproductive output can result in slow recovery from population crashes and is expected to be associated with slow genetic adaptation. Therefore, it is assumed that phenotypic plasticity, a rather rapid and sometimes reversible process, is a crucial mechanism in long-lived organisms to adapt to changing environments. However, how differences in individual hibernation behaviour influence mortality and whether individuals are plastic with respect to their hibernation behaviour are largely unknown.
Recent studies suggest that climatic change can influence hibernation behaviour in various species differently, in a positive or negative way. Female Columbian ground squirrels (Urocitellus columbianus) delayed their emergence from hibernation with later snow melt and lower spring temperatures. Next to the environmental impact, emergence date showed a moderate heritability in female Columbian ground squirrels. Yellow-bellied marmots (Marmota flaviventris) emerged earlier from hibernation with warmer spring temperatures which resulted in a longer growing period for their offspring and, therefore, higher survival rates. In contrast, in alpine marmots (Marmota marmota) lower snow cover due to higher temperatures and, thus, less isolation led to lower juvenile survival. Negative effects, such as reduced juvenile survival, would be of high concern, especially for long-lived species with a low reproductive output.
Bats are exceptionally long-lived compared to other mammals of the same size and often show a low reproductive output with one offspring per year. This is especially true in the temperate zone where bats, furthermore, are characterized by seasonality and depend on hibernation during winter period to survive food and water shortage. Because bats are of high conservation concern it is of prime importance to understand their ability to respond to different climatic conditions and associated mortality costs.
The basis of this study was a five-year data set of 1047 RFID-tagged individuals from two bat species, Natterer’s bats (Myotis nattereri) and Daubenton’s bats (Myotis daubentonii), that were automatically tracked when entering or leaving the joint hibernaculum, “Brunnen Meyer”, located in north-western Germany. The two species are similar sized, share demographical traits and often occupy the same areas. Nevertheless, they differ in their foraging strategy and activity pattern during hibernation period. Natterer’s bats are able to glean insects from surfaces, even at low temperatures. Daubenton’s bats depend on flying arthropods and, thus, warmer temperatures. And indeed there is evidence that Natterer’s bats are able to hunt during hibernation period, while in Daubenton’s bats a lack of feeding during the hibernation period is suggested. Furthermore, Natterer’s bats are characterized by a higher activity at the hibernaculum throughout the hibernation period, while Daubenton’s bats on average arrive earlier, stay inactive through the winter and leave later in spring.
In both species, the aim was to investigate the impact of their individual hibernation behaviour, precisely the timing of departure in late winter and early spring, on mortality, their adjustment of departure timing to the North Atlantic Oscillation Index (NAO), as well as differences within and between the two species from 2011 until 2015.
To later on estimate the potential mortality costs of departure timing, gaining knowledge about the seasonal survival pattern (winter vs. summer) in the two species was a first necessity. In birds, particularly small species were described as winter-regulated populations with a higher mortality during winter. In contrast, in hibernating mammal species, such as bats, a relatively lower or similar winter survival compared to summer survival was shown. In this study, the analysed data demonstrated that the winter 2010/2011 was exceptionally catastrophic in Natterer’s bats and did not impact Daubenton’s bats. When excluding this catastrophic winter in Natterer’ bats, our results revealed a stable winter-summer-survival difference (higher winter and lower summer survival) in adult Natterer’s and Daubenton’s bats, with inter-annual variation in the level of survival which indicates a potential environmental impact on survival. This winter-summer survival pattern is in line with the survival pattern shown for other hibernators. Juveniles always had a lower survival rate than adult bats in both species. Nevertheless, the extent to which the species differ between seasons and age classes was stronger in Daubenton’s bats. They always showed a slightly higher winter survival and a lower summer survival than Natterer’s bats. Together with the catastrophic winter 2010/2011 in Natterer’s bats, this indicates a species-specific sensitivity to the timing of specific weather events which is in line with their foraging strategies and activity pattern during hibernation period.
With respect to emergence behaviour from the hibernaculum, the results of this study suggest considerable differences among individuals within as well as between bat species. In comparison to Daubenton’s bats, Natterer’s bats tuned their emergence more closely to weather conditions, specifically the NAO, a large scale weather index related to winter severity, and showed individual variation in behavioural plasticity. In Daubenton’s bats only the females responded to changing conditions and left earlier in individually-experienced warmer and milder winters, comparable to Natterer’s bats females. A potential reason might be reproductive advantages for the females resulting in a longer growing period for their offspring. The shown higher winter survival in adult bats of both species indicated already higher energy expenditure outside the hibernaculum. Thus, leaving early, being active and staying outside longer by itself bore a risk (exposure risk effect). Under consideration of longer exposure times, early departing individuals had on top of that an increased risk to die. This was not given in each year, but a species- and year-specific pattern was revealed. Natterer’s bats were only significantly affected by early departure in 2011, while the remaining years show no significant additional risk of leaving early. In Daubenton’s bats, the years 2014 and 2015 were associated with a significantly higher mortality of leaving early. This is in line with the hypothesis that Daubenton’s bats might not be able to hunt for insects leaving too early and do so as a best out of a bad job. Nevertheless, the year-specific pattern suggests that early bats might profit from advantageous weather conditions during early spring.
An additional hint for an environmental impact on early bat survival in at least Daubenton’s bats is that the median proportion of night hours above 3 °C within five days after departure was included in the model with the lowest AIC. However, the effect was not strong enough to be selected as the best model and, therefore, further analyses are needed to investigate this first hint.
In conclusion, the reduced winter survival of juveniles compared to adults highlights the importance of considering age class effects in studies that investigate seasonal survival patterns. The stable species-specific winter-summer-survival difference with a higher winter survival compared to summer survival, as well as the one catastrophic winter in Natterer’s bats underline the importance of including seasonal survival patterns in assessing potential fitness costs of changed behaviour. Furthermore, our results suggest that long-lived hibernating bat species have the potential to plastically adjust to changing climatic conditions, but this potential differs between species. Among-individual differences in emergence together with species-specific mortality costs of early emergence suggest the potential for natural selection to shape hibernation phenology. In summary, our findings suggest species-, population- and group-specific differences in the ability to respond to changing environments and, therefore, underline the necessity to further investigate local responses in various organisms to estimate consequences of recent climate change on a wider range.
To this day, the patient’s outcome after any form of cerebral ischemia is often mediocre
at best. The added damage that occurs at reperfusion after ischemia seems to be as
important as the ischemic injury itself. New therapeutic strategies targeted at this
critical issue are therefore crucial. P188, an amphiphilic triblock copolymer, has risen
to be one of the most promising pharmacological therapeutics, as its capability to insert
into injured cell membranes seems to perfectly fit the needed criteria to protect against
I/R injury. Lately, it has become apparent that mitochondrial function particularly profits
from P188 treatment after I/R injury. Therefore, the question arose, if P188 may
interact directly with mitochondria.
In the present study, rat isolated brain mitochondria were injured and then treated with
P188. The injury took place either in vivo by asphyxial cardiac arrest before isolation
of mitochondria or in vitro after isolation by addition of the ROS H2O2. After treatment
with P188, mitochondrial function was evaluated through the assessment of ATP
synthesis, O2 consumption and CRC.
10 or 15 min of asphyxia in vivo as well as 200 μM H2O2 for 10 min in vitro significantly
impaired mitochondrial function. Furthermore, a damaging effect of RT on isolated
mitochondria became apparent. Contrary to the underlying hypothesis, P188 did not
preserve mitochondrial function independently of the injury mechanism chosen.
In conclusion, in the context of studying P188, two new methods of I/R injury
simulation, namely asphyxial cardiac arrest in vivo and the injury with H2O2 in isolated
mitochondria in vitro, have been established. However, it is not yet conclusive, if P188
does or does not directly improve mitochondrial function after I/R injury. Further
research looking at different injuring methods as well as modulating the treatment
method is needed to ultimately clarify this question.
Potential of nitrous oxide sedation in pedodontics to reduce the need of dental general anesthesia
(2022)
Aim: To investigate the possible reduction of the need for dental general anesthesia (DGA) through nitrous oxide sedation (NOS), when is used in combination with behavior management techniques among patients younger than 12 years of age referred to a specialized pedodontics practice due to the high dental treatment need and poor cooperation level.
Materials and methods: Retrospective analysis was performed of the digital medical records of all children treated under NOS in a specialized pedodontics clinic between 2012 and 2017. The potential reduction of the need for dental general anesthesia was assessed depending on the success rate of NOS on patient-level with relations to multiple related factors such as age, reason of referral and treatment need.
Results: out of 510 patients, 228 meet the inclusion criteria. Nitrous oxide was used in 406 dental treatment sessions on 228 pre-cooperative and/or anxious patients aged 3-12 years (mean 6.4±1.7; 43.4% female). 91.9% of the NOS sessions were successful in achieving the intended dental treatment. Complete oral rehabilitation was possible for 84% of the patients using NOS without the need of DGA. Regarding the age, DGA reduction among pre-school children was lower than in schoolchildren (77.8% and 87.9%, respectively).
Conclusion: A high fraction of anxious or semi-cooperative children with high dental treatment needs can be treated without the use of DGA when a comprehensive concept of caries management is combined with the use of NOS and behavior management techniques. NOS should, therefore, always be considered as an option for dental treatment of semi-cooperative children with high dental treatment needs before making a decision towards DGA.
Pharmaceutical residues are found in increasing concentrations in the environment and in potable water where they have verifiable effects on aquatic life. Conventional methods for water treatment are not able to sufficiently abate these generally stable compounds. It was found that physical plasma generated directly in water can degrade several of these recalcitrant organic pollutants. Studies on the basic plasma chemical processes for the model system of phenol showed that the degradation is primarily caused by hydroxyl radicals. This was confirmed by reaction chemistry and spin trap enhanced electron paramagnetic resonance spectroscopy (EPR). The degradation of diclofenac and its by-products were investigated in detail to perform a first risk-assessment of the new technology. Findings are not limited to the application of plasma but applicable to other advanced oxidation processes (AOP) that are based on the generation of hydroxyl radicals as well. Additionally, pulsed corona plasma and pulsed electric fields were assessed for their capacity to kill Legionella pneumophila in water. Whereas it was possible to kill L. Pneumophila with both methods, plasma treatment resulted in an enhanced bacterial killing. Therefore, advanced oxidation processes (AOP) and plasma treatment in particular are some of the few feasible approaches to decompose recalcitrant compounds in water.
Dendrochronology, the science of tree-rings is a tool which has been widely used for many years for understanding changes in the environment, as trees react to environmental changes over time. In the contemporary situation, where climate warming in the Arctic is unequivocal and its effects on the Alpine and tundra ecosystems are seen pronouncedly in the past decade, the role of dendro-studies and the use of trees and shrubs alike as proxies of change has become critical. Studies clearly indicate that warming in the Arctic and Alpine tundra has resulted in increased vegetation in recent years. Shrubs, in these sensitive ecosystems, have proven to be highly instrumental as they likely benefit from this warming and hence are good indicators and auditees of this change. Therefore, in this study, we investigate the potential of shrubs in the evolving field of dendro-ecology/climatology.
Studies from classical dendrochronology used annual rings from trees. Further, because of shrub sensitivity to contemporary change, shrub-based dendrochronological research has increased at a notable scale in the last decade and will likely continue. This is because shrubs grow even beyond the tree line and promise environmental records from areas where tree growth is very limited or absent. However, a common limitation noted by most shrub studies is the very hard cross-dating due to asynchronous growth patterns. This limitation poses a major hurdle in shrub-based dendrochronological studies, as it renders weak detection of common signals in growth patterns in population stands. This common signal is traced by using a ‘site-chronology’.
In this dissertation, I studied shrub growth through various resolutions, starting from understanding radial growth within individuals along the length of the stem, to comparison of radial growth responses among male and female shrubs, to comparing growth responses among trees and shrubs to investigation of biome-wide functional trait responses to current warming. Apart from Chapter 4 and Chapter 6, I largely used Juniperus communis sp. for investigations as it is the most widely distributed woody dioecious species often used in dendro-ecological investigations in the Northern Hemisphere.
Primarily, we investigated radial growth patterns within shrubs to better understand growth within individuals by comparing different stem-disks from different stem heights within individuals. We found significant differences in radial growth from different stem-disks with respect to stem heights from same individuals. Furthermore, we found that these differences depending on the choice of the stem-disk affect the resulting site-chronology and hence climate-sensitivity to a substantial extent and that the choice of a stem-disk is a crucial precursor which affects climate-growth relationships.
Secondly, we investigated if gender difference – often reported causing differential radial growth in dioecious trees – is an influential factor for heterogeneous growth. We found that at least in case of Juniperus communis. L and Juniperus communis ssp nana. WILLD there is no substantial gender biased difference in radial growth which might affect the site-chronology. We did find moderate differences between sexes in an overall analysis and attribute this to reproductive effort in females.
In our study to test the potential of shrubs for reconstruction, we used a test case of Alnus viridis ssp crispa. We found a strong correlation between ring-width indices and summer temperature. Initially, the model failed the stability tests when we tested the stability of this relation using a response function model. However, using wood-anatomical analysis we discovered that this was because of abnormal cell-wall formation resulting in very thin rings in the year 2004. Pointer year analysis revealed that the thin rings were caused because of a moth larval outbreak and when corrected for these rings the model passed all stability tests.
Furthermore, to see if trees and shrubs growing in same biomes react to environmental changes similarly, a network analysis with sites ranging from the Mediterranean biome to the Ural Mountains in Russia was carried out. We found that shrubs react better to the current climate warming and have a decoupled divergent temperature response as compared to coexisting trees. This outcome reiterated the importance of shrub studies in relation to contemporary climate change. Even though trees and shrubs are woody forms producing annual rings, they have very different growth patterns and need different methods for analysis and data treatment.
Finally, in a domain-wide network analysis from plant-community vegetation survey, we investigated functional relationships between plant traits (leaf area, plant height, leaf nitrogen content, specific leaf area (SLA), and leaf dry matter content (LDMC)) and abiotic factors viz. temperature and soil moisture. We found a strong relation between summer temperature and community height, SLA and LDMC on a spatial scale. Contrarily, the temporal-analysis revealed SLA and LDMC lagged and did not respond to temperature over the last decade. We realized that there are complex interactions between intra-specific and inter-specific plant traits which differ spatially and temporally impacting Arctic ecosystems in terms of carbon turn over, surface albedo, water balance and heat-energy fluxes. We found that ecosystem functions in the Arctic are closely linked with plant height and will be indicative of warming in the short term future becoming key factors in modelling ecosystem projections.
Background
Telephone-Cardiopulmonary Resuscitation (T-CPR) significantly increases rate of bystander resuscitation and improves patient outcomes after out-of-hospital cardiac arrest (OHCA). Nevertheless, securing correct execution of instructions remains a difficulty. ERC Guidelines 2021 recommend standardised instructions with continuous evaluation. Yet, there are no explicit recommendations on a standardised wording of T-CPR in the German language. We investigated, whether a modified wording regarding check for breathing in a German T-CPR protocol improved performance of T-CPR.
Methods
A simulation study with 48 OHCA scenarios was conducted. In a non-randomised trial study lay rescuers were instructed using the real-life-CPR protocol of the regional dispatch centre and as the intervention a modified T-CPR protocol, including specific check for breathing (head tilt-chin lift instructions). Resuscitation parameters were assessed with a manikin and video recordings.
Results
Check for breathing was performed by 64.3% (n = 14) of the lay rescuers with original wording and by 92.6% (n = 27) in the group with modified wording (p = 0.035). In the original wording group the head tilt-chin manoeuvre was executed by 0.0% of the lay rescuers compared to 70.3% in the group with modified wording (p < 0.001). The average duration of check for breathing was 1 ± 1 s in the original wording group and 4 ± 2 s in the group with modified wording (p < 0.001). Other instructions (e.g. check for consciousness and removal of clothing) were well performed and did not differ significantly between groups. Quality of chest compression did not differ significantly between groups, with the exception of mean chest compression depth, which was slightly deeper in the modified wording group.
Conclusion
Correct check for breathing seems to be a problem for lay rescuers, which can be decreased by describing the assessment in more detail. Hence, T-CPR protocols should provide standardised explicit instructions on how to perform airway assessment. Each protocol should be evaluated for practicability.
In general, concepts such as chemical equilibria and classical analytical chemistry, as well as instrumental methods such as potentiometry and flow injection analysis (FIA), are taught separately in the curricula of chemistry-related degree programs. However, FIA provides an opportunity to connect all of these important topics. This article shows how simple experiments can be used to link the fundamentals of FIA and potentiometry to the fundamentals of chemical equilibria and teach them in a sustainable way. The experiments were designed for undergraduate environmental science students with basic knowledge of chemical equilibria and classical analytical chemistry and minimal knowledge of electrochemistry. However, they can also be integrated into various courses on instrumental analysis in chemistry-related degree programs.
Für den erfolgreichen Erhalt von Natur und Landschaft sowohl in Schutzgebieten als außerhalb davon ist die Akzeptanz der jeweiligen Gebietsbewohner notwendig und ihr aktives Engagement hilfreich. Häufig gibt es jedoch Widerstand gegenüber Naturschutzmaßnahmen und wenige aktiv Engagierte. Ein Grund wird in der überwiegend naturwissenschaftlichen Argumention gesehen, die offenbar die Bedeutung von Natur und Landschaft für die Bevölkerung nicht ausreichend widerspiegelt. In der vorliegenden Arbeit wird daher der Frage nachgegangen, ob und wie ästhetische und emotionale Wertschätzungen das Portfolio von Argumenten für den Natur- und Landschaftserhalt bereichern können. Das empirische Material wurde im Rahmen eines Forschungsprojektes (gefördert von der Deutschen Bundesstiftung Umwelt, DBU, 2009-2012) gewonnen, dessen Ziel die Entwicklung und Erprobung von Erhebungsmethoden für ein sozioökonomisches Monitoring in den vier UNESCO-Biosphärenreservaten Mittelelbe, Schaalsee, Schorfheide-Chorin und Südost-Rügen war. Der zentrale Fokus der vorliegenden Arbeit mit soziologischer Forschungsperspektive liegt darauf, wie die Bewohner der vier untersuchten Biosphärenreservate ihre Region emotional und ästhetisch wertschätzen. Neben geographischen wurden vor allem die soziologischen Theorieansätze Social Identity Theory (SIT) und die Identity Process Theory (IPT) erörtert, um mögliche emotionale und ästhetische Argumente fundiert einordnen zu können. Es wurde eine quantitative CATI-Befragung mit mindestens 300 Interviews pro Biosphärenreservat auf der Grundlage einer Zufallsstichprobe durchgeführt. Die Daten wurden neben deskriptiven Untersuchungen mittels binär logistischer Regressionsanalysen und Varianzpartitionierungen als schließende statistische Methoden untersucht. Ein grundlegendes Ergebnis der Analysen ist, dass die Bewohner in allen vier Gebieten eine hohe regionale Verbundenheit aufweisen. Weiterhin scheinen Natur und Landschaft maßgeblich zur emotionalen Verbundenheit zur Region beizutragen. Außerdem stellte sich heraus, dass regionale Verbundenheit sowohl allgemein als auch mit explizitem Landschaftsbezug wichtige Themen für viele Menschen sind und kein Nischenthema einer speziellen Personengruppe darstellen. Denn zwar scheinen einige soziodemographische und andere Faktoren die emotionale Verbundenheit zu verstärken, diverse Parameter, wie etwa Geschlecht und ehrenamtliches Engagement, spielen dagegen aber kaum eine Rolle. Aus den Ergebnissen wurden folgende Handlungsempfehlungen für die Argumentation für Natur- und Landschaftserhalt abgeleitet: (1) Natur und Landschaft haben eine hohe emotionale und ästhetische Bedeutung für viele Bewohner, daher sollten alle raumwirksamen Aktivitäten umfangreich und so zeitig wie möglich erläutert und diskutiert werden. (2) Landschaftliche Vielfalt wird geschätzt und Natur und Landschaft werden nicht als rein stereotype Ansammlung von Landschaftselementen wahrgenommen, sondern mit regionsspezifischen Charakteristika. Wenn Maßnahmen in Natur und Landschaft zu diesen ästhetischen Werten beitragen haben sie ein großes Potenzial, die Unterstützung für Naturerhalt zu erhöhen. Daher wird empfohlen, diese Einflüsse der Maßnahmen besonders zu betonen. (3) Der Bildungsstand scheint grundsätzlich ein besonders wichtiger Faktor für die untersuchten Phänomene zu sein. Daher wird geraten, wenn möglich, die zielgruppenspezifische Kommunikation mit ästhetischen und emotionalen Argumenten sowie Angebote für ein Engagement so auszugestalten, dass Personen aus verschiedenen Bildungsmilieus erreicht werden. (4) Um Menschen für ein naturerhaltendes Engagement zu motivieren wird empfohlen, allgemein an der Region interessierte Personen zu suchen und anzusprechen. (5) Es scheint ratsam, bei der Formulierung von emotionalen Argumenten für den Erhalt von Natur und Landschaft auf den Begriff Stolz zu verzichten. Der Terminus findet unter Akademikern weniger Zustimmung, wenngleich sie eine hohe regionale Verbundenheit haben können. Der Heimatbegriff sollte nur benutzt werden, solange er sich auf kollektiv geteilte, eudaimonistische Werte bezieht und zukunftsgerichtet verwendet wird. (6) Neben der regionalen Verbundenheit können besonders die eudaimonistischen Werte Ruhe, Erholung und andere nicht mit Naturschutzzwecken konfligierende Freizeitnutzungen zielführend in der Argumentation für den Naturerhalt sein. Insgesamt wird für den Natur- und Landschaftserhalt tätigen Akteuren empfohlen, das offensichtlich bestehende Potenzial ästhetischer und emotionaler Argumente stärker zu nutzen und naturwissenschaftliche Argumente damit sinnvoll zu ergänzen. Die in dieser Arbeit diskutierten Ergebnisse liefern dafür eine fundierte Grundlage, da sich aus der nun vorliegenden vergleichenden Analyse von vier unterschiedlichen UNESCO-Biosphärenreservaten in Deutschland belastbare Empfehlungen ableiten lassen, die deutlich über die Untersuchungsgebiete hinaus anwendbar sind.
In this work, studies with respect to the exhaust problem were performed
in the stellarator experiment Wendelstein 7-X with different target concepts and different magnetic field geometries. Different infrared cameras were used to study the heat flux from the plasma onto the PFC. In the first publication, the limiter set-up was used with a simpler magnetic topology in the plasma edge. The radial fall-off of the parallel heat flux for inboard limiters in W7-X shows, similar to inboard limiters in tokamaks, two different radial fall-off lengths, a short (narrow) one, characterizing the near-SOL, and a long (broad) characterizing the far-SOL. For the far-SOL, the heating power and connection length have been identified as the main scaling parameters, while for the near-SOL, the electron temperature close to the LCFS has been identified as the main scaling parameter. The two fall-off lengths differ by a factor 10, and the found scalings for both regimes differ from known models and experimental scalings in tokamaks. A turbulent-driven feature was discussed in the publication as a possible explanation for the behavior of the fall-off length in W7-X.
The gained information and data have been further used to support many
other publications, covering the symmetry of the heat loads, the
energy balance of the machine, and seeding experiments.
The heat exhaust in W7-X with an island divertor was studied in the second
and third publication. Definitions of parameters such as peaking factor and
wetted area were applied for the heterogeneous heat flux pattern on the
W7-X divertor. It was shown that the island divertor concept is capable
of spreading out the heat efficiently, resulting in large wetted areas of up to 1.5 m2. The reached values for the wetted area are comparable to the ones of the larger tokamak JET but with a much smaller ratio of wetted
area to the area of the last closed flux surface. Furthermore, a positive
scaling of the wetted area with the power in the SOL was observed. This
scaling is beneficial for future reactors but needs further investigation of the involved transport processes. The peaking factor (discussed in the second publication) describes how concentrated the heat load is within the region of the strike line. It was shown that this factor is decreasing for increasing densities without affecting the wetted area. The present work paves the way for further analysis of the transport processes of the heat flux towards the island divertor of Wendelstein 7-X.
Aim
To determine the long-term effects of the use of powered tooth brush (PTB) in comparison to manual tooth brush (MTB) on periodontitis severity, coronal caries experience, and the number of missing teeth using in a population-based cohort study.
Materials and Methods
Using 7-year follow-up data of 2214 participants of the Study of Health in Pomerania (SHIP-TREND), comprehensively adjusted linear models using generalized least squares and ordinal regression models estimated the effects of PTB usage on dental outcomes in complete case and imputed data.
Results
At follow-up, PTB users had lower medians for mean probing depth (PD; 2.21 mm) and mean clinical attachment levels (1.73 mm) than MTB users (2.30 and 1.96 mm, respectively). Adjusted models revealed the beneficial effects of PTB usage on follow-up levels of plaque, bleeding on probing, mean PD, percentage of sites with PDs ≥4 mm, mean clinical attachment levels (all, interdental, and non-interdental sites, respectively), and the number of missing teeth. For the number of missing teeth, the effects were more pronounced in participants aged ≥50 years. No significant effects of PTB usage on the number of decayed or filled surfaces (all and interdental sites) were found.
Conclusions
A recommendation of PTB usage in dental practice could contribute to the long-term promotion of oral health.
Riociguat is one of several approved therapies available for patients with pulmonary arterial hypertension (PAH). Treatment should be initiated and monitored at an expert center by a physician experienced in treating PAH, and the dose adjusted in the absence of signs and symptoms of hypotension. In certain populations, including patients with hepatic or renal impairment, the elderly, and smokers, riociguat exposure may differ, and dose adjustments should therefore be made with caution according to the established scheme. Common adverse events are often easily managed, particularly if they are discussed before starting therapy. Combination therapy with riociguat and other PAH-targeted agents is feasible and generally well tolerated, although the coadministration of phosphodiesterase type 5 inhibitors (PDE5i) and riociguat is contraindicated. An open-label, randomized study is currently ongoing to assess whether patients who do not achieve treatment goals while receiving PDE5i may benefit from switching to riociguat. In this review, we provide a clinical view on the practical management of patients with PAH receiving riociguat, with a focus on the opinions and personal experience of the authors.

In den letzten Jahrzehnten hat sich durch randomisiert-kontrollierte Studien (RCTs) eine breite Evidenzbasis von Psychotherapie mit mittleren bis großen Effekten für verschiedene psychische Störungen gebildet. Neben der Bestimmung dieser Wirksamkeit („Efficacy“) ebneten Studien zur Wirksamkeit unter alltäglichen Routinebedingungen („Effectiveness“) historisch den Weg zur Entwicklung eines praxisorientierten Forschungsparadigmas. Im Beitrag wird argumentiert, dass im Rahmen dieses Paradigmas praxisbasierte Studien eine wertvolle Ergänzung zu RCTs darstellen, da sie existierende Probleme in der Psychotherapieforschung adressieren können. In der gegenwärtigen praxisorientierten Forschung liefern dabei neue Ansätze aus der personalisierten Medizin und Methoden aus der ‚Computational Psychiatry‘ wichtige Anhaltspunkte zur Optimierung von Effekten in der Psychotherapie. Im Kontext der Personalisierung werden bspw. klinische multivariable Prädiktionsmodelle entwickelt, welche durch Rückmeldeschleifen an Praktiker_innen kurzfristig ein evidenzbasiertes Outcome-Monitoring ermöglicht und langfristig das Praxis-Forschungsnetzwerk in Deutschland stärkt. Am Ende des Beitrags werden zukünftige Richtungen für die praxisorientierte Forschung im Sinne des ‘Precision Mental Health Care’ -Paradigmas abgeleitet und diskutiert.
Background
The ‘wallflower’ hypothesis proposes females mate indiscriminately to avoid reproductive delays. Post-copulatory mechanisms may then allow ‘trading up’, favouring paternity of future mates. We tested links between pre- and post-copulatory choice in Latrodectus geometricus female spiders paired sequentially with two males. These females copulate as adults or as subadults and store sperm in paired spermathecae. Choosy adults have a higher risk of delays to reproduction than subadults.
Results
We predicted low pre-copulatory, but high post-copulatory choice at first matings for adults and the opposite for subadults. At second matings, we expected all females would prefer males superior to their first. We found all females mated indiscriminately at their first pairing, but in contrast to subadults, adults usually allowed only a single insertion (leaving one of their paired spermatheca empty); a mechanism of post-copulatory choosiness. Adult-mated females were more likely to remate than subadult-mated females when they became adults, showing a preference for larger males, while subadult-mated females tended to prefer males of greater size-corrected mass.
Conclusions
Our results show that the ‘wallflower’ effect and ‘trading up’ tactics can be utilized at different life stages, allowing females to employ choice even if rejecting males is costly.
Abstract
Background
The CRISPR/Cas9 system has opened new perspectives to study the molecular basis of cerebral cavernous malformations (CCMs) in personalized disease models. However, precise genome editing in endothelial and other hard‐to‐transfect cells remains challenging.
Methods
In a proof‐of‐principle study, we first isolated blood outgrowth endothelial cells (BOECs) from a CCM1 mutation carrier with multiple CCMs. In a CRISPR/Cas9 gene correction approach, a high‐fidelity Cas9 variant was then transfected into patient‐derived BOECs using a ribonucleoprotein complex and a single‐strand DNA oligonucleotide. In addition, patient‐specific CCM1 knockout clones were expanded after CRISPR/Cas9 gene inactivation.
Results
Deep sequencing demonstrated correction of the mutant allele in nearly 33% of all cells whereas no CRISPR/Cas9‐induced mutations in predicted off‐target loci were identified. Corrected BOECs could be cultured in cell mixtures but demonstrated impaired clonal survival. In contrast, CCM1‐deficient BOECs displayed increased resistance to stress‐induced apoptotic cell death and could be clonally expanded to high passages. When cultured together, CCM1‐deficient BOECs largely replaced corrected as well as heterozygous BOECs.
Conclusion
We here demonstrate that a non‐viral CRISPR/Cas9 approach can not only be used for gene knockout but also for precise gene correction in hard‐to‐transfect endothelial cells (ECs). Comparing patient‐derived isogenic CCM1+/+, CCM1+/−, and CCM1−/− ECs, we show that the inactivation of the second allele results in clonal evolution of ECs lacking CCM1 which likely reflects the initiation phase of CCM genesis.
Abstract
Alkali ion beams are among the most intense produced by the ISOLDE facility. These were the first to be studied by the ISOLTRAP mass spectrometer and ever since, new measurements have been regularly reported. Recently the masses of very neutron-rich and short-lived cesium isotopes were determined at ISOLTRAP. The isotope 148Cs was measured directly for the first time by Penning-trap mass spectrometry. Using the new results, the trend of two-neutron separation energies in the cesium isotopic chain is revealed to be smooth and gradually decreasing, similar to the ones of the barium and xenon isotopic chains. Predictions of selected microscopic models are employed for a discussion of the experimental data in the region.
The rapid neutron-capture or the r-process is responsible for the origin of about half of the neutron-rich atomic nuclei in the universe heavier than iron. For the calculation of the abundances of those nuclei, atomic masses are required as one of the input parameters with very high precision. In the present work, the masses of the neutron-rich Zn isotopes (A=71 to 81) lying in the r-process path have been measured in the ISOLTRAP experiment at ISOLDE/CERN. The mass of 81Zn has been measured directly for the first time. The half-lives of the nuclides ranged from 46.5 h (72Zn) down to 290 ms (81Zn). In case of all the nuclides, the relative mass uncertainty (∆m/m) achieved was in the order of 1E-8 corresponding to a 100-fold improvement in precision over previous measurements.
The aim of this study was to analyse the predictive power of several clinical baseline parameters and the de-/remineralisation properties of in vivo etched sites measured with quantitative light-induced fluorescence (QLF) for subsequent 2-year caries increment. At baseline, in 44 children (8.23 ± 1.5 years) two areas (diameter 2 mm) of the buccal surface of a primary posterior tooth were etched with 36% phosphoric acid gel for 1 and 4 min, respectively. The etched sites were analysed immediately after etching (ΔQ1) and 24 h (ΔQ2) later by QLF. Additionally, caries status (deft/DMFT and initial caries), approximal plaque, bleeding on probing, and the patient’s current use of fluorides were recorded. In the 2-year follow-up, 29 children were re-assessed. After clinical examination, the caries increment was calculated (ΔDMFT) and correlated with the baseline clinical variables and the QLF readings. Results showed a significant positive correlation between ΔQ<sub>1 min</sub> and the ΔDMFT (r = 0.44, p = 0.02). The ΔDMFT was significantly correlated with the baseline deft (r = 0.56, p = 0.002), cavitated active caries lesions (r = 0.52, p = 0.003), and filled teeth (r = 0.53, p = 0.003). In a regression analysis the use of fluoridated salt (SC = –0.10) and fluoride gel (SC = –0.14) were negatively associated with ΔDMFT. In conclusion, these findings suggest that the demineralisation properties of the etched sites and the outcome of the 24-hour measurements with QLF are significantly associated with caries increment. Previous caries experience strongly correlated with caries increment in this group of children.
Predicting complications in pediatric Crohn's disease patients followed in CEDATA-GPGE registry

Background
Complications of Crohn's disease (CD) often impair patients' quality of life. It is necessary to predict and prevent these complications (surgery, stricturing [B2]/penetrating [B3] disease behavior, perianal disease, growth retardation and hospitalization). Our study investigated previously suggested and additional predictors by analyzing data of the CEDATA-GPGE registry.
Methods
Pediatric patients (< 18 years) diagnosed with CD with follow up data in the registry were included in the study. Potential risk factors for the selected complications were evaluated by performing Kaplan-Meier survival curves and cox regression models.
Results
For the complication surgery, the potential risk factors older age, B3 disease, severe perianal disease and initial therapy with corticosteroids at the time of diagnosis were identified. Older age, initial therapy with corticosteroids, low weight-for-age, anemia and emesis predict B2 disease. Low weight-for-age and severe perianal disease were risk factors for B3 disease. Low weight-for-age, growth retardation, older age, nutritional therapy, and extraintestinal manifestations (EIM) of the skin were identified as risk factors for growth retardation during the disease course. High disease activity and treatment with biologicals were predictors for hospitalization. As risk factors for perianal disease, the factors male sex, corticosteroids, B3 disease, a positive family history and EIM of liver and skin were identified.
Conclusion
We confirmed previously suggested predictors of CD course and identified new ones in one of the largest registries of pediatric CD patients. This may help to better stratify patients’ according to their individual risk profile and choose appropriate treatment strategies.
(1) Background: Predicting chronic low back pain (LBP) is of clinical and economic interest as LBP leads to disabilities and health service utilization. This study aims to build a competitive and interpretable prediction model; (2) Methods: We used clinical and claims data of 3837 participants of a population-based cohort study to predict future LBP consultations (ICD-10: M40.XX-M54.XX). Best subset selection (BSS) was applied in repeated random samples of training data (75% of data); scoring rules were used to identify the best subset of predictors. The rediction accuracy of BSS was compared to randomforest and support vector machines (SVM) in the validation data (25% of data); (3) Results: The best subset comprised 16 out of 32 predictors. Previous occurrence of LBP increased the odds for future LBP consultations (odds ratio (OR) 6.91 [5.05; 9.45]), while concomitant diseases reduced the odds (1 vs. 0, OR: 0.74 [0.57; 0.98], >1 vs. 0: 0.37 [0.21; 0.67]). The area-under-curve (AUC) of BSS was acceptable (0.78 [0.74; 0.82]) and comparable with SVM (0.78 [0.74; 0.82]) and randomforest (0.79 [0.75; 0.83]); (4) Conclusions: Regarding prediction accuracy, BSS has been considered competitive with established machine-learning approaches. Nonetheless, considerable misclassification is inherent and further refinements are required to improve predictions.
The effects of climate change lead to increasing social injustice and hence justice is intrinsically linked to a socio-ecological transformation. In this study, we investigate whether justice sensitivity motivates pro-environmental intention (PEI) and behavior (PEB) and, if so, to what extent emotions and moral disengagement determine this process. For this purpose, we conducted two quota-sampling surveys (Study 1: N = 174, Study 2: N = 880). Multiple regression analyses in both studies suggest that a higher perception of injustice from a perpetrator’s, beneficiary’s, and observer’s perspective is associated with an increased PEI. However, moral disengagement best predicted PEB and PEI. Guilt and authentic pride were found to be emotional predictors of PEI. Additionally, mediation analyses demonstrated that guilt mediates the connection between both perpetrator and beneficiary sensitivity and PEI. These results suggest that when the predominant originators of climate change (i.e., individuals from industrialized countries) perceive global climate injustice from the perspective of a beneficiary or a perpetrator, they experience guilt and have a higher PEI. Based on this mechanism, it seems promising to render global injustice more salient to those responsible for activities that lead to climate change to motivate them to adapt their behavior. The role of moral disengagement and victim sensitivity as barriers to pro-environmental behavior is discussed in this context.
